In order to presume the relationships between two species of P. ternata and P. tripartia, and their populations of the Korean Pinellia, RAPD analysis was performed. The length of the amplified DNA fragments ranged from 300 to 2,500bp. Seventy scorable RAPD makers were found from the PCR reactions with 7 random oligoprimers and were analyzed by Nei-Li's genetic coefficient. Also, some regional groups instead of same taxa were clustered from the phenogram of UPGMA analysis and NJ tree. Populations within each species were clustered at low genetic distance, there had the closed relationship. According to the regional individuals, Pinellia ternata was showed the variation pattern of morphological (leaf shape and flower color) and cytological characters(somatic chromosome numbers). So we suggested to difference of characteristic variety based on variety of habitat. According to this study, new species (Pinellia sp.) was affiliated with Pinellia and had the closest relationship with Hallasan and Japan population. The RAPD data was very useful to define the genetic variation and to discuss the relationships among the intraspecific taxa and their populations of the Korean Pinellia.
The flora of vascular plants in Sumeunmulbaengdui wetland located in Hallasan National Park was surveyed 9 times from September, 2010 to May, 2014. This survey resulted in 236 taxa in total composed of 202 species, 2 subspecies, 27 varieties and 5 forms of 148 genera under 69 families. They were categorized by their life forms into 8 groups of 37 megaphanerophytes, 26 nanophanerophytes, 2 epiphytes, 3 chamaephytes, 84 hemicryptophytes, 33 geophytes, 21 hydrophytes, 30 therophytes. Among them, 10 taxa of Korean endemic plants and 9 taxa of rare and endangered plants were included. In addition, the floristic regional indicator plants were 43 taxa comprised of 3 taxa of grade V, 7 taxa of grade IV, 12 taxa of grade III, 8 taxa of grade II and 13 taxa of grade I. 2 species of alien plants, Erechtites hieracifolia Raf. and Taraxacum officinale Weber were found. Sustainable interest and protective measures to Sumeunmulbaengdui wetland are required, since this area is comparatively larger in size and higher in plant diversity than other Ramsar wetlands in Jeju-do Province.
BACKGROUND: Yeast isolates associated with the leaves, stems, and flowers of the tiger lily needed to be identified using isolation methods that have previously been used effectively in yeast biotechnology. A culture-based approach was necessary for the isolation of many yeast strains associated with tiger lily. METHODS AND RESULTS: In this study, the homogenized leaves, stems, and flowers of tiger lily were spreaded onto GPY medium containing chloramphenicol, streptomycin, Triton X-100, and L-sorbose. A total of 82 yeast strains from the leaves, 94 and 97 yeast strains from the stems and flowers were isolated, respectively. Yeast isolates were identified by phylogenetic analysis based on internal transcribed spacer region sequencing. The yeast species isolated from the leaves comprised of 31 isolates of the genus Pseudozyma, 28 of Aureobasidium pullulans, and 11 of the genus Cryptococcus. Those isolated from the stems comprised of 40 of A. pullulans and 11 of Cryptococcus, and 95 of A. pullulans While, 1 isolate each of the genera Rhodotorula and Metschnikowia were isolated from the flowers. CONCLUSION: We identified site-specific yeast communities associated with tiger lily. These yeast isolates may have high potential for application in the field of biotechnology.
The effects of ground vibration on lava tubes during construction were studied to aid design of management and preservation measures for lava tubes. Ground conditions were assessed by RMR (Rock mass rating) and Q-system classifications for the Geomunoreum lava tubes, and vibration velocity was measured during in situ blasting tests in the Manjanggul and Yongcheondonggul lava tubes. Results indicate that the higher the rock quality, the greater the effect of vibration, although there is no clear linear relationship due to ground heterogeneity. A relationship derived between vibration velocity (PPV) and intensity (dB(V)) on the basis of blasting tests indicates that a vibration level of < 0.285 cm/sec meets the regulatory limit of 0.371 cm/sec and 65 dB(V) during daytime, and 0.285 cm/sec and 60 dB(V) during night. For blasting vibrations, square- and cube-root scaled distances are linearly correlated, with R2 ≥ 0.76. On the basis of this correlation, explosive-charge weights meeting the 0.2 cm/sec vibration criterion for cultural heritage were estimated to be 2.88 kg at 50 m distance, and 11.52 kg at 100 m.

본 연구는 한라산국립공원 동사면에 위치한 구상나무숲 장기생태 모니터링 5ha 조사지역 ($20{\times}20m$, 125개 방형구)에서 구상나무의 유묘 및 치수 발생 현황을 파악하기 위하여 수행되었다. 조사는 2016년 6~8월 사이에 $5{\times}5m$ 방형구 125곳에 대해 계통추출법을 적용하여 실시하였다. 조사한 전체 125개 방형구 중에서 54개에서만 유묘가 관찰되었으며, 그 내용은 유묘 615개체 및 치수 1개체로 총 616개체가 발견되었다. 그러나 과반수의 71개 방형구에서 치수뿐만 아니라 유묘도 출현하지 않았다. 심지어 전체 125개 방형구에서 유묘가 가장 많이 관찰된 방형구(2-6지역, 95개체)를 제외하면, 나머지 124개 방형구의 평균 유묘 개체수는 4.2개로 매우 적었고 높이도 35cm 이하에 불과했다. 이와 함께 장기생태 모니터링 조사 지역 내에서 $1{\times}1m$ 방형구 183곳에 대해 단순 무작위 추출법으로 발생지 조사를 수행하였다. 발생지 조사 결과 유묘 2,518개체, 치수 2개체 및 유목 2개체 등 총 2,522개체가 발견되어 계통추출법을 적용한 조사와 유사한 결과가 나타났다. 본 연구를 통해 한라산국립공원 동사면 구상나무숲 내에서 구상나무 유묘의 발생은 어느 정도 이루어지고 있으나, 교란으로 인하여 치수로의 발달이 제한되는 것으로 판단된다. 여러 교란 요인 중에서 조사된 총 3,138개체 중 12.1%, 381개체에서 유제류인 노루에 의한 섭식이 확인되어 노루의 섭식이 유묘 발생 및 치수 발달에 교란을 야기하는 가장 중요한 요인으로 추정해볼 수 있다. 한반도 특산식물 구상나무의 보전 대책 수립을 위한 유묘 및 치수 교란 원인에 대한 지속적인 모니터링이 진행되어야 한다고 사료된다.
We conducted taxonomical investigations based on morphological characteristics, fruit morphology, and literature research on the tribe Potentilleae (Rosaceae) in Korea covering seven genera and 24 species. The style position on the ovary and the shape of style were useful characteristics for the classification of subtribal and generic levels in the tribe Potentilleae. The subtribe Fragariinae is characterized by subbasal or lateral style on the ovary and anthers with one theca. The subtribe Potentillinae has a subterminal style on the ovary, except for Argentina, which presents a subterminal and lateral style and anthers with two thecae. These results support the recent taxonomic recognition that i) the tribe Potentilleae consists of two subtribes, and ii) genera such as Dasiphora, Comarum, and Sibbaldianthe sometimes included in Potentilla s.l. are treated as independent genus. In the subtribe Potentillinae, Argentina, which has been treated as Potentilla, is supported as a distinct genus according to the characteristics of the subterminal and lateral style position and the ventral stipular auricles. In Fragaria, F. nipponica subsp. chejuensis, which has generally small leaves and a limited distribution only on Hallasan Mt., is supported by treatment as an endemic species. Duchesnea chrysantha is distinguished from D. indica by the characteristics of light green or yellowish green leaves, thin and somewhat membranous leaflets, and broad ovate or obovate leaflets. Each complex of P. dickinsii and P. chinensis remains unresolved with regard to controversy over the taxonomic circumscription due to their external morphological variations. Additional taxonomical research and molecular population studies are required for a more in-depth understanding of the tribe Potentilleae in Korea.
To conduct ethnobotanical study that grow wild in biosphere reserves or are traded in nearby traditional markets, we collected and analyzed traditional knowledge information on vascular plants that live in Mt. Seorak and Mt. Halla. As a result, the ethnobotanical plants investigated in Mt. Seorak consisted of 102 families and 489 taxa. The ethnobotanical plants investigated in Mt. Halla consisted of 118 families and 374 taxa. Both ethnobotanical plants from Mt. Seorak and Mt. Halla were most widely used for oriental medicine and treatment, and leaves were the most used organ. Differences in diversity by family of ethnobotanical plants inhabiting Mt. Seorak and Mt. Halla showed significant differences only in other use. The ethnobotanical plants traded at the traditional market near Mt. Seorak consists of 59 species in 34 families and 56 genera. The ethnobotanical plants traded at the traditional market near Mt. Halla consists of 102 species in 49 families and 94 genera. All ethnobotanical plants traded in traditional markets were most often used for oriental medicine and treatment, and stems were used most often in the traditional market near Mt. Seorak, and leaves were used most often in the traditional market near Mt. Halla.
This study was conducted to investigate the difference in feeding habits of Red-Tongued Viper Snakes, according to available foods sources and areas. The effects of differences in food sources were found on Red-Tongued Viper Snake inhabited in the Jeju Island and its islet Gapado, from May 2006 to Nov. 2010. The food sources for the Red-Tongued viper snake population in the Jeju Island were found to be as follows: Chinese red-headed centipedes (Scolopendra subspinipes mutilans), Jeju Salamanders (Hynobius quelpaertensis), Japanese tree Frogs (Hyla japonica), Narrow-mouthed Toad (Kaloula borealis), Dybowski's Brown Frogs (Rana dybowskii), Black-spotted Pond Frogs (Rana nigromaculata), Smooth Skinks (Scincella vandenburghi), Asian Keelback Snakes (Amphiesma vibakari), Lesser White-toothed Shrews (Crosidura shantungensis), Hallasan Shrews (Sorex caecutiens hallamontanus), and Jeju Striped Field Mice (Apodemus chejuensis). This implies that Red-Tongued Viper Snakes mainly feed on amphibians, reptiles, and small mammals. Among these, amphibians occupied the highest portion at 55.2% followed by mammals at 20.7%, centipedes at 13.8%, and reptiles at 10.3%. On the contrary, Red-tongued viper snake population in Gapado only feed on Chinese red-headed centipedes and Smooth Skinks (S. vandenburghi). Since only a small amount of nutrient can be obtained from Chinese red-headed centipeds or Smooth Skinks, this feeding habit for Red-tongued viper snake would adversely effect on the growth or regeneration. The reason why Red-Tongued viper snake population in the Gapado mainly feed on Lizard and Centipedes in spite of relatively various available food sources, might be due to the low density of other food sources in the Gapado. Red-Tongued viper snake could be feeding on foods that are low in quality but are easily accessible, to minimize energy consumption on searching for other more nutritious foods. A snake tends to select the size of its food depending on the size of its own head. The positive correlation was found between the size of the heads of Red-Tongued viper snakes from the Jeju island and the diameter of their foods. The head size was larger in the males than females in viper snake population from the Jeju Island, which might effect on their selection of foods. However, no significant difference was found between the sizes of the head and the food in the Red-Tongued viper snake population from the Gapado. The findings of this study would provide meaningful data, which directly shows that even within the same viper species they choose different available food sources according to their inhabitance. This leads to their growth and adaptation to their environment which is beneficial for sustaining of its population.
For management and preservation measures of lava tube, it is studied how the blasting vibration by constructions near Geomunoreum lava tubes in Jeju affect lava tube. 11 boreholes were drilled in study area, and in-situ blasting tests were conducted by changing from 0.5 kg to 10 kg charge per delay in those boreholes. The vibration velocity, which meets the regulatory vibration criterion during daytime, was estimated as below 0.276 cm/sec by analyzing the relationship between vibration velocity and vibration level. In addition, SRE and CRE were calculated from the results of in-situ blasting tests, and k-values were shown as 130.04 in SRE, 199.71 in CRE, respectively. Also, n-values were shown as -1.717 in SRE, -1.711 in CRE, respectively. Charge per delay were assessed based on these equations, and charges per delay had ranges of 0.57~7.42 kg/delay in estimation equation of vibration velocity, 0.21~5.29 kg/delay in SRE, and 0.04~5.51 kg/delay in CRE, considering the 0.2 kine vibration criterion for cultural heritage and the 20~100 m distance from vibration source. Additionally, the relationships which meet the criteria of 0.2 kine, were calculated by combining CRE in this study with the result of previous study. Allowable charges per delay, which meet the criteria of 0.2 kine, were evaluated as 1.07 kg/delay in 50 m, 5.13 kg/delay in 100 m and 22.26 kg/delay in 200 m distances. These relationships for each vibration velocity are useful to deduce charge per delay for the ground near Geomunoreum lava tube.
Among protected areas, National Parks are rich in biodiversity, and the benefits of ecosystem services provided to human are higher than the others. Ecosystem service evaluation is being used to manage the value of national parks based on objective and scientific data. Ecosystem services are classified into four services: supporting, provisioning, regulating and cultural. The purpose of this study is to evaluate habitat quality among supporting services. Habitat Quality Model of InVEST was used to analyze. The coefficients of sensitivity and habitat initial value were reset by reflecting prior studies and the actual conditions of protected areas. Habitat quality of 21 national parks except Hallasan National Park was analyzed and mapped. The value of habitat quality was evaluated to be between 0 and 1, and the closer it is to 1, the more natural it is. As a result of habitat quality analysis, Seoraksan and Taebaeksan National Parks (0.90), Jirisan and Odaesan National Parks (0.89), and Sobaeksan National Park (0.88) were found to be the highest in the order. As a result of comparing the area and habitat quality of 18 national parks except for coastal-marine national parks, the larger the area, the higher the overall habitat quality. Comparing the value of habitat quality of each zone, the value of habitat quality was high in the order of the park nature preservation zone, the park nature environmental zone, the park cultural heritage zone, and the park village zone. Considering both the analysis of habitat quality and the legal regulations for each zone of use, it is judged that the more artificial acts are restricted, the higher the habitat quality. This study is meaningful in analyzing habitat quality of 21 National Parks by readjusting the parameters according to the situation of protected areas in Korea. It is expected to be easy to intuitively understand through accurate data and mapping, and will be useful in making policy decisions regarding the development and preservation of protected areas in the future.
