Effect of Dietary Pigment Sources on the Growth and Shell Color of Abalone (Haliotis discus hannai) (배합사료에 색소원료 첨가가 참전복 치패의 성장 및 패각 색깔에 미치는 영향)

  • This study investigated the effect of dietary pigment sources on growth and shell color of juvenile abalone(Haliotis discus hannai). Three replicate groups of the abalone (average weight 173 mg) were fed diets containing various pigment sources such as Porphyra powder, Spirulina, yeast astaxanthin, and paprika extract for 16 weeks. Survival and weight gain were not affected by dietary pigment sources (P>0.05). Shell color of abalone fed diets containing Porphyra powder and Spirulina approached the yellow-red and orange, colors similar to wild abalone. However, shell color of abalone fed the diets containing yeast astaxanthin and paprika extract were similar to the bright green control group. These results should be useful for changing the shell color of abalone in aquaculture.

Molecular Characterization and Expression Analysis of a Glutathione S-Transferase cDNA from Abalone (Haliotis discus hannai) (북방전복 (Haliotis discus hannai)에서 분리한 Glutathione S-transferase 유전자의 분자생물학적 고찰 및 발현분석)

  • Glutathione S-transferases (GSTs) are a superfamily of detoxification enzymes that primarily catalyze the nucleophilic addition of reduced glutathione to both endogenous and exogenous electrophiles. In this study, we isolated and characterized a full-length of alpha class GST cDNA from the abalone (Haliotis discus hannai). The abalone GST cDNA encodes a 223-amino acid polypeptide with a calculated molecular mass of 25.8 kDa and isoelectric point of 5.69. Multiple alignments and phylogenetic analysis with the deduced abalone GST protein revealed that it belongs to the alpha class GSTs and showed strong homology with disk abalone (Haliotis discus discus) putative alpha class GST. Abalone GST mRNA was ubiquitously detected in all tested tissues. GST mRNA expression was comparatively high in the mantle, gill, liver, and digestive duct, however, lowest in the hemocytes. Expression level of abalone GST mRNA in the mantle, gill, liver, and digestive duct was 182.7-fold, 114.8-fold, 4675.8-fold, 406.1-fold higher than in the hemocytes, respectively. Expression level of abalone GST mRNA in the liver was peaked at 6 h post-infection with Vibrio parahemolyticus and decreased at 12 h post-infection. While the expression level of abalone GST mRNA in the hemocytes was drastically increased at 3 h post-infection with Vibrio parahemolyticus. These results suggest that abalone GST is conserved through evolution and may play roles similar to its mammalian counterparts.

Seasonal Changes in Physiology of the abalone Haliotis discus hannai reared from Nohwa Island on the South Coast of Korea (북방전복 Haliotis discus hannai의 계절별 생리적 변화)

  • Growth, oxygen consumption, ammonia excretion, feeding and assimilation rate were examined from May 2010 to February 2011 in order to assess the physiological changes of Haliotis discus hannai in accordance with changes in season. The water temperature was in the range of $8-23.2^{\circ}C$ and the salinity in the range of 31.9-34.1psu during the examination period. The length of shell of Haliotis discus hannai grew from 36.3 mm to 66.1 mm in the 1 year old entities and from 60.6 mm to 66.1 mm for the 2 year old entities, while the weight of the meat increased from 3.16 g to 12.04 g and from 15.8 g to 21.5 g, respectively. The oxygen consumption and ammonia excretion rate displayed trend of increase in accordance with the increase in water temperature, while the feeding rate was high during the period from July to October. The assimilation rate was in the range of 68%-71% without significant difference between the age of the entities and seasons. SFG displayed +value throughout the year for the 1 year old entities of Haliotis discus hannai as well as for the 2 year old entities with the exception of the period of July, thereby showing that they are growing throughout the year. Therefore, there was no environmental effect including water temperature and salinity during the examination period.

  • 생물에게 물리적인 손상(physical damage)이나 stress를 주지 않기 위한 마취제로서의 역할은 매우 중요하다. 특히 전복은 어류와는 달리 부착기질과 은신처를 이용하여 부착생활을 하기 때문에 무게측정(weighting), 표지(tagging), 선별(selection), 밀도 조절 등을 위해서 박리작업은 반드시 필요하다. 따라서 본 연구는 요소비료(CO(NH$_2$)$_2$)를 마취제로 이용하여 요소비료의 농도와 수온 등 물리적 요인이 시볼트 전복 Haliotis gigantea의 박리율과 회복율에 미치는 영향을 조사함으로서 경제적이며 효과적인 박리 기술을 개발하고자 하였다.   • This study was conducted to investigate the effects of nickel chloride ($NiCl_2$) exposure on oxidative stress of the abalone Haliotis discus hannai. Experimental groups were composed of one control condition and five nickel chloride exposure conditions (5.9, 8.8, 13.3, 20.0, 30.0 mg/L). Superoxide dismutase (SOD) activity was increased in the foot and hepatopancreas, but decreased in the gill. Catalase (CAT) activity was increased in all exposure groups except 20.0 mg/L and 30.0 mg/L. In the gill, CAT activity was similar to the control group for all exposure groups. In the hepatopancreas, CAT activity was increased compared with the control group. However SOD and CAT activity showed not significant differences (P > 0.05).

  • Genetic parameters for growth-related traits were estimated in 9-month old of two Korean abalone subspecies, Haliotis discus hannai and H. discus discus, using multiple traits of animal model. The data were collected from the records of 3,504 individuals produced from 16 sires and 17 dams in H. discus hannai and 821 individuals produced from 3 sires and 4 dams in H. discus discus, which was evaluated at the Bukjeju branch, NFRDI, from May 20, 2004 to February 14, 2005. The heritability estimates obtained from restricted maximum likelihood (REML) method range from 0.29 to 0.31 for three growth traits (shell length, shell width and body weight) in H. discus hannai and from 0.22 to 0.28 in H. discus discus, respectively. The heritabilities for shell shape and condition factor were lower than others of growth traits such as ranging from 0.03 to 0.24 in H. discus hannai and from 0.06 to 0.11 in H. discus discus, respectively. Genetic and phenotypic correlations were >0.91 between shell parameters and weight in two abalone subspecies, respectively, indicating that breeding for weight gains could be successfully achieved by selecting for shell length.
