Insulin-like growth factor-binding protein-5 (IGFBP-5) is one of the six members of IGFBP family, important for cell growth, apoptosis and other IGF-stimulated signaling pathways. In order to explore the significance of IGFBP-5 in cells of the Inner Mongolian Cashmere goat (Capra hircus), IGFBP-5 gene complementary DNA (cDNA) was amplified by reverse transcription polymerase chain reaction (RT-PCR) from the animal's fetal fibroblasts and tissue-specific expression analysis was performed by semi-quantitative RT-PCR. The gene is 816 base pairs (bp) in length and includes the complete open reading frame, encoding 271 amino acids (GenBank accession number JF720883). The full cDNA nucleotide sequence has a 99% identity with sheep, 98% with cattle and 95% with human. The amino acids sequence shares identity with 99%, 99% and 99%, respectively. The bioinformatics analysis showed that IGFBP-5 has an insulin growth factor-binding protein homologues (IB) domain and a thyroglobulin type-1 (TY) domain, four protein kinase C phosphorylation sites, five casein kinase II phosphorylation sites, three prenyl group binding sites (CaaX box). The IGFBP-5 gene was expressed in all the tested tissues including testis, brain, liver, lung, mammary gland, spleen, and kidney, suggesting that IGFBP-5 plays an important role in goat cells.
Objective: The aim of this study was to investigate the effect of single nucleotide polymorphisms (SNPs) of the melanogenesis associated transcription factor (MITF) and dopachrome tautomerase (DCT) genes on plumage coloration in Asian native duck breeds. MITF encodes a protein for microphthalmia-associated transcription factor, which regulates the development and function of melanocytes for pigmentation of skin, hair, and eyes. Among the tyrosinase-related family genes, DCT is a pigment cell-specific gene that plays important roles in the melanin synthesis pathway and the expression of skin, feather, and retina color. Methods: Five Asian duck varieties (black Korean native, white Korean native, commercial Peking, Nageswari, and Bangladeshi Deshi white ducks) were investigated to examine the polymorphisms associated with plumage colors. Among previously identified SNPs, three synonymous SNPs and one indel of MITF and nine SNPs in exon regions of DCT were genotyped. The allele frequencies for SNPs of the black and white plumage color populations were estimated and Fisher's exact test was conducted to assess the association between the allele frequencies of these two populations. Results: Two synonymous SNPs (c.114T>G and c.147T>C) and a 14-bp indel (GCTGCAAAC AGATG) in intron 7 of MITF were significantly associated with the black- and white-colored breeds (p<0.001). One non-synonymous SNP [c.938A>G (p.His313Arg)] in DCT, was highly significantly associated (p<0.001) and a synonymous SNP (c.753A>G) was significantly associated (p<0.05) with black and white color plumage in the studied duck populations. Conclusion: The results of this study provide a basis for further investigations of the associations between polymorphisms and plumage color phenotypes in Asian duck breeds.
The backs with a hair cut of 6-week-old healthy ICR male mice were once exposed to a dose of $400mJ/cm^2$ UVB. An acute dermal inflammation was observed, and the inflamed skins were almost completely cured after 6 days of the exposure. At 24 hours after exposure, the epidermal keratinocytes showed a cell-membrane damage with the destruction of intercellular junctions, agglutination of tonofilaments within the cytoplasm and nucleus damage. The activity of XO showed a significant increase (p<0.05) in up to 144 hours. The activities of CAT and SOD showed a significant decrease (p<0.05) in up to 96 hours, but they were not significantly different from the normal value at 144 hours. The GST activity was significantly decreased (p<0.01) in up to 96 hours, not so at 24 hours. However, that was not significantly different from the normal value at 144 hours. There was a significant decrease (p<0.01) in the contents of TBARS at 48 and 96 hours, without any significant difference at 144 hours. While the content of GSH was significantly lower (p<0.05) at 24 hours, that was not significantly different thereafter up to 144 hours from the normal value. Therefore, it is assumed that skin damage with a dose of $400mJ/cm^2$ UVB irradiation might be caused by the oxidative stress which was resulted from the unbalance of oxygen fret radical generating and scavenging enzymes.

Background and Objectives : Despite of high prevalence, Tinnitus remains obscure because the cause and mechanism is poorly understood. In the absence of a suitable animal model, past investigations tool place in humans. Recently, the animal model with sodium salicylate ototoxicity is broadly used because of its reversibility. Balyangtongum-Tang had been used clinically to treat tinnitus and other hearing problems. We investigated the effects of Balyangtongum-Tang on cochlear morphologic change induced by sodium salicylate ototoxicity and were to find out its therapeutic effects on ototoxicity in rat model. Materials and Methods : Healthy ten Sprague-Dawley rats were divided into normal(2), control(4) and sample(4) groups. The sample group was given extract of Balyangtongum - Tang(1cc/100g) once a day for 4 days. After 3 hours when last medication were given, the sample and control groups were injected intraperitoneally with sodium salicylate(500mg/kg). We observed the cochlear morphologic changes of rats every 1, 2, 3 and 5 hours after injection. Results : The electron microscopic finding of outer hair cell shows some changes in the curticular plate and cytoplasm. Some vacuoles were found in the control and sample groups. Vacuolization in the curticular plate and cytoplasm of the sample group after 3, 5 hours were similar to the control group. But the curticular plate of the sample group after 1, 2 hours did not from vacuole. On the other hand the control group after 1, 2 hours formed vacuoles in the curticular plate. Light microscopic findings of cochlear duct in control and sample groups didn't find any difference. Conclusion : The results suggest that extract of Balyangtongum-Tang reduces the morphologic changes induced by sodium salicylate ototoxicity and the effects are remarkable in frist 2 hours. However the effective times are different with previous studies, it seems to due to the difference of tolerance and sensitivity of laboratory animals.

Klinefelter syndrome is the most common chromosomal abnormality, with a 47, XXY karyotype and typical clinical findings of infertility, hypogonadism, reduced body hair, gynecomastia, tall stature, and incresed gonadotropins and decreased testosterone levels. In addition to this classic description, several other diseases have been discribed in Klinefelter syndrome such as unilateral renal aplasia, autoimmune disease, diabetes mellitus, sexual precoxity, renal cell carcinoma, intravesical ureterocele, and osteoporosis. The incidence is 1 in 400-1,000 of the population and urological abnormalities are not common. However a case of Klinefelter syndrome associated with multicystic dysplastic kidney has not been not reported up to date. Therefore, we describe a 1-day-year old baby boy who presented with Klinefelter syndrome with unilateral multicystic kidney dysplastic disease, plus with a brief review of the literature.
The purpose of this study was to clarify the effect of light emitting diode (LED) irradiation on healing of impaired wound and alteration of mast cells in experimental diabetic rats. Twenty-four male Sprague-Dawley rats were divided into four groups: excision (Ex), excision-LED irradiation (Ex-LED), diabetes + excision (DM) and diabetes + excision + LED irradiation (DM-LED). Diabetes was induced in rats by streptozotocin (STZ) injection (70 mg/kg, single dose) and 6 mm punch excision wounds were created on the back after shaving hair. The LED-irradiated rats were treated to a daily dose of $5\;J/cm^2$ LED (630 nm) light for 11 days after surgery, and were killed at day 1, 3, 7 and 11. The lesion and adjacent skin tissues were excised, fixed with 10% buffered formalin and embedded with paraffin. For evaluation of wound healing, hematoxylin-eosin (HE) and Masson trichrome staining were performed. Mast cells (MCs) were stained with toluidine blue (pH 0.5) and quantified using a computerized image analysis system. The proliferation activity of keratinocyte in skin tissues was analyzed on sections immunostained with proliferative cell nuclear antigen (PCNA). The results showed that wound healing rate, collagen density and neo-epidermis length, number of PCNA-positive cells, fibroblasts and mast cells were significantly higher in the LED-irradiated rats than in the DM and Ex rats throughout the periods of experiment. Exceptionally, the number of MCs was significantly lower at day 11 compared with day 7 after surgery in the all groups. These findings suggest that the LED irradiation may promote the tissue repair process by accelerating keratinocyte and fibroblast proliferation and collagen production in normal rats as well as in diabetic rats, and MCs may play an important role at an early stage of skin wound healing in normal and diabetic rats.

There are many synthetic chemicals, such as di(2-ethylhexyl) phthalate (DEHP) and dibutyl phthalate (DBP), used in chemical reaction processes in industry. The establishment of toxicity and detection of synthetic chemicals that may pose a genetic hazard in our enviornment is subjects of great concern at present DEHP, a ubiquitous phthalate plasticizer, induces a wide range of developmental and reproductive toxicities in mammals. DEHP belongs to the large diverse class of peroxisome proliferator compounds, which include herbicides, hypolipidemic drugs. DBP is a plasticizer used to products containing nitrocellulose, polyvinyl acetate, and polyvinyl chloride such as food wraps and blood bags. DBP is also used in cosmetics as a solvent and fixative for perfumes, a suspension agent for solids, an antifoamer, a skin emollient, and hair spray The present study was performed to examine patterns of gene expression in MCF-7 cells following DEHP and DBP exposure. Changes in gene expression were determined by microarray analysis using KISTCHIP-400 including 401 endocrine related genes based on public database and research papers. Of the genes analysis, we determined that genes detected by array showed a 2-fold or greater change in their expression level(increase or decrease). The results of this study demonstrate that a number of genes were differentially expressed in MCF-7 cells but these changes were not significant. Therefore, we keep going this study using microarray analysis and future studies will examine changes of gene expression on time-course and does treatment in variable cell lines.
Background Scalp defects may be caused by various etiological factors, and they represent a significant surgical and aesthetic concern. Various surgical techniques can be applied for reconstructive work such as primary closure, skin grafting, pedicled or free flaps. In this article, the authors share their clinical experience with scalp operations using the technique of local flaps and discuss the application of this method from the perspective of not only the size of the defect, but also in relation to the anatomical area, quality of surrounding tissue, and patient's condition. Methods During the period from December 2007 to December 2012, 13 patients with various scalp defects, aged 11 to 86 years, underwent reconstruction with local pedicle flaps. The indications were based on the patients' condition (age, sex, quality of surrounding tissue, and comorbidities) and wound parameters. Depending on the size of the defects, they were classified into three groups as follows: large, 20 to 50 $cm^2$; very large, 50 to 100 $cm^2$; extremely large, 100 $cm^2$. The location was defined as peripheral (frontal, temporal, occipital), central, or combined (more than one area). We performed reconstruction with 11 single transposition flaps and 1 bipedicle with a skin graft on the donor area, and 2 advancement flaps in 1 patient. Results In all of the patients, complete tissue coverage was achieved. The recovery was relatively quick, without hematoma, seroma, or infections. The flaps survived entirely. Conclusions Local flaps are widely used in scalp reconstruction since they provide healthy, stable, hair-bearing tissue and require a short healing time for the patients.
A 5-year-old Pekinese dog was presented for evaluation of a back mass of 1 year duration. Fine needle aspiration cytology of the mass revealed numerous keratinocytes, and a lesser number of multinucleated giant cells along with a low number of spindle cells, which were suggestive of epidermal cyst or hair follicle tumor with secondary granulomatous inflammation. The mass was surgically removed and examined histologically. Microscopically there was partially encapsulated well-demarcated nodule in deep dermis and subcutis. The nodule consisted of central cyst and secondary cysts that were lined by a single or multiple layers of basaloid cells and squamous epitheliums with central laminated keratin. Multifocal aggregation of numerous macrophages and multinucleated giant cells were present. The final diagnosis was keratoacanthoma with granulomatous inflammation. The mass did not recur 5 months after surgery.

Tall fescue(Festuca amndinacea Schreb.) is a productive pasture grass that is widely adapted to the soil and climate of Korea. It is regarded, however, as relatively low in palatability and nutritive value. On the other hand, Italian ryegrass(lo1ium rnultz$om Lam.) is highly nutritious and palatable, but is essentially weak in cold tolerance. Numerous investigators have succeeded in producing hybrids between Lolium spp. and Festuca spp. but no improved varieties have resulted. Therefore, intergeneric hybridization of Italian ryegrass with tall fescue is employed in 1984 to improve the adaptive and cold resistance quality of Italian ryegrass. In the mean time, F1-hybrid plants obtained by crossing Italian ryegrass x tall fescue were observed in open field. The results are summarized as follows: 1. Leaf-size and silica cell hair were intermediate between the types of parents but the shape of auricle tended to show Italian ryegrass and auricle-cilia to show tall fescue. 2. Roots of $F_1$-hybrid plants were mainly distributed at top soil and did not produced florescence material. 3. Head type of hybrids was intermediate between the types of parents but ray length was somewhat less than the length of tall fescue ray. Typical Italian ryegrass or tall fescue-like phenotypes were not observed. 4. Anthers of $F_1$-hybrid plants were formed but pollen were not formed. Even though pollen were formed, they were all empty. 5. Hybrid plants were more winterhardy and summer persistency than Italian ryegrass. They showed a equivalent growth to Italian ryegrass in spring and to tall fescue in summer. 6. Chromosome of $F_1$-hybrids was allopolyploid(2n=4~=28) and all $F_1$-hybrid plants were sterile.
