TNF-α 자극에 활성화된 HaCaT 세포주에서 Yakuchinone-A에 의한 건선 피부염 개선 효과 (Improving effect of psoriasis dermatitis by yakuchinone A in the TNF-α stimulated HaCaT cells)

  • 건선(psoriasis)은 인체 피부조직 중 표피의 과증식 및 다양한 크기의 홍반, 인설 등이 동반되는 난치성 자가면역 피부질환이다. 건선 피부염 발병 기작은 명확히 규명되지 않았으나 각질 형성세포의 과분화 과정에 관여하는 염증성 cytokine과 Th17 세포를 포함한 면역세포를 염증부위로 유인하는 chemokine (C-C motif) ligand 20 (CCL20)이 발병과정에 관여하는 것으로 알려진다. 따라서 건선치료에 효과적인 천연 소재를 발굴하기 위해 예로부터 항염증 활성이 알려진 익지인(Alpiniaoxyphylla Miquel)의 유효성분인 yakuchinone-A의 건선 피부염 개선효과를 연구하였다. 먼저 CCK-8 assay 통해 human keratinocyte (HaCaT) 세포에 tumor necrosis factor-alpha (TNF-α)와 yakuchinone-A를 동시 처리하여 세포독성을 관찰한 결과, yakuchinone-A는 10 ㎍/mL까지 세포독성이 관찰되지 않았다. TNF-α를 HaCaT 세포에 처리하여 염증을 유발한 후 yakuchinone-A를 농도별로 처리한 결과 IL-6, IL-8, TNF-α 등 건선 피부염 유발 cytokine의 mRNA 발현이 각각 61.4±7.5, 23.6±1.5, 46.0±4.8% 수준으로 감소하였고, Th17 세포를 유인하는 chemokine인 CCL20 또한 yakuchinone-A에 의해 유의적으로 억제되었다. 또한 CCL20 발현에 관여하는 NF-κB/IκB pathway에서 IκB 인산화 및 STAT3 인산화가 yakuchinone-A에 의해 79.1±5.0, 80.8±2.3% 수준만큼 농도 의존적으로 억제되었다. 마지막으로 Th17 세포에 의해 분비되는 IL-17A에 의해 활성화된 HaCaT 세포에 yakuchinone-A를 처리한 결과, CCL20 mRNA발현이 농도의존적으로 감소하였다. 이러한 결과들을 토대로 yakuchinone-A는 건선 피부염 개선 활성을 가지며, 향후 새로운 건선 피부염 개선 소재로 개발될 수 있을 것으로 기대된다.

Curcumin ameliorates TNF-α-induced ICAM-1 expression and subsequent THP-1 adhesiveness via the induction of heme oxygenase-1 in the HaCaT cells

  • Adhesion molecules such as ICAM-1 are important in the infiltration of leukocytes into the site of inflammation. In this study, we investigated the inhibitory effects of curcumin on ICAM-1 expression and monocyte adhesiveness as well as its underlying action mechanism in the TNF-${\alpha}$-stimulated keratinocytes. Curcumin induced expression of heme oxygenase-1 (HO-1) in the human keratinocyte cell line HaCaT. In addition, curcumin induced Nrf2 activation in dose- and time-dependent manners in the HaCaT cells. Curcumin suppressed TNF-${\alpha}$-induced ICAM-1 expression and subsequent monocyte adhesion, which were reversed by the addition of tin protoporphyrin IX (SnPP), a specific inhibitor of HO-1, or HO-1 knockdown using siRNA. Furthermore, Nrf2 knockdown using siRNA reversed the inhibitory effect of curcumin on the TNF-${\alpha}$-induced ICAM-1 expression and adhesion of monocytes to keratinocytes. These results suggest that curcumin may exert its anti-inflammatory activity by suppressing the TNF-${\alpha}$-induced ICAM-1 expression and subsequent monocyte adhesion via expression of HO-1 in the keratinocytes.

Cadmium으로부터 손상을 유도한 HaCaT 세포에서 머위(Petasites japonicus) 추출물의 세포보호효과 (Cytoprotective Effect of Petasites japonicus Extract on Cadmium-induced Cytotoxicity in HaCaT cell)

  • 본 연구는 머위 추출물의 화장품소재로서의 가능성을 확인하기 위하여 자체의 독성과 카드뮴으로부터 유도된 세포손상에 미치는 영향에 대하여 평가하였다. 카드뮴으로부터 손상을 유도한 각질형성세포에 머위 추출물을 처리하여 세포사멸 인자인 Bcl-2와 procaspase-3의 단백질 발현을 측정하였다. 그 결과 머위 추출물 $200{\mu}g/mL$를 제외한 모든 농도에서 98% 이상의 높은 생존율을 나타내었으며 세포사멸인자인 Bcl-2와 procaspase-3 단백질 발현이 증가한 것으로 보아 머위 추출물이 카드뮴 독성 시 일어나는 세포자멸사에 대한 보호기전을 나타낸 것으로 평가되었다. 또한 카드뮴으로 12 h 동안 PARP cleavage를 유도한 각질형성세포에 머위 추출물을 전처리한 결과, 카드뮴을 매개로 하는 PARP cleavage를 억제하는 것으로 나타났다. 이러한 결과를 통해 머위 추출물이 피부세포 보호 효능을 나타내는 천연소재로서의 활용가능성을 제안한다.

  • Up-regulation of adhesion molecules plays an important role in the infiltration of leukocytes into the skin during the development of various inflammatory skin diseases, such as atopic dermatitis. In this study, we investigated the modulatory effects of 2,3-dimethoxy-2′-hydroxychalcone (DMHC) on tumor necrosis factor (TNF)-α-induced intercellular adhesion molecule-1 (ICAM-1) expression and monocyte adhesiveness, as well as the molecular mechanisms underlying its action in the HaCaT human keratinocyte cell line. Pre-treating HaCaT cells with DMHC significantly suppressed TNF-α-induced ICAM-1 expression and subsequent monocyte adhesiveness. DMHC inhibited TNF-α-induced activation of NF-ᴋB. In addition, DMHC induced HO-1 expression as well as NRF2 activation. Furthermore, HO-1 knockdown using siRNA reversed the inhibitory effect of DMHC on TNF-α-induced ICAM-1 expression and adhesion of monocytes to keratinocytes. These results suggest that DMHC may inhibit TNF-α-induced ICAM-1 expression and adhesion of monocytes to keratinocytes by suppressing the signaling cascades leading to NF-ᴋB activation and inducing HO-1 expression in keratinocytes. 각질형성세포에서 MMP-1 활성 및 자외선 유도 무모쥐 피부손상에 대한 카테킨 고함유 녹차추출물의 영향 (Effects of Catechin-rich Green Tea Extract on the MMP-1 Activity of HaCaT Keratinocyte Cells and on UVB-induced Skin Damage in Hairless Mice)

  • Background: Skin is an organ that protects the human body from various environmental stimuli that can induce immune system activation. Skin aging can be largely divided into two categories: physiological aging, which is caused by the a decreased physiological function of the skin and structural changes with aging, and photoaging, which is caused by the chemical stress induced by external stimuli such as ultraviolet (UV) radiation. Methods and Results: The objective of this study was to investigate the anti-wrinkle and UV protective effect of catechin-rich green tea extract (CGTE) in activated keratinocyte (HaCaT cells) and UV-induced skin damage in hairless mice. The results showed that CGTE inhibits the tumor necrosis factor-alpha interferon-gamma ($TNF-{\alpha}+IFN-{\gamma}$)-induced expression of matrix metalloproteinase (MMP)-1 in HaCaT cells. In addition, the CGTE treatment significantly reduced wrinkle formation, epidermal thickness, collagen deposition, and transepidermal water loss in dorsal skin irradiated with UVB. However, the ${\beta}$-glucosidase activity was significantly increased. The CGTE treatment inhibits mRNA expression and enzyme activity of MMP-2 and MMP-9 in the dorsal skin irradiated with UVB. Conclusions: It is expected that CGTE can be effectively used as a functional food and cosmetic ingredient to improve skin moisture retention and reduce wrinkle formation.

진세노사이드 Rb1과 Rg1에 의한 HaCaT 피부각질세포의 전사체 분석 (Transcriptome Analysis of Human HaCaT Keratinicytes by Ginsenosides Rb1 and Rg1)

  • 인삼(Panax ginseng C. A. Meyer)의 주요 생리활성물질인 진세노사이드(ginsenoside) Rb1과 Rg1의 효능검증 및 작용점을 규명하고자 HaCaT 피부각질세포에서 유전체 분석(gene expression profiles)을 실시하였다. 진세노사이드 Rb1과 Rg1 각각의 처리 농도 및 시간에 따른 HaCaT 세포에 대한 세포독성은 나타나지 않았으며, $10{\mu}g/mL$의 진세노사이드 Rb1과 Rg1 각각을 6 및 24 시간 처리하여 유전체 분석 결과, 진세노사이드 Rb1과 Rg1의 24 시간 처리군에서 항노화 및 피부탄력 관련 유전자인 fibroblast growth factor (FGF2)의 활성이 증가된 것으로 나타났다. 또한 진세노사이드 Rb1의 24 시간 처리군에서는 항산화 작용점에 있는 일련의 유전자군, FANCD2, FGF2, LEPR, FAS 등의 활성을 확인하였다. 향후 확인된 항노화 및 피부탄력 관련 주요인자들의 작용 및 상관관계를 구체적으로 확인하고, 특히 진세노사이드 Rb1의 신호전달을 완성하고자 한다.

종대황과 선복화 에탄올 추출물의 인간 피부 세포주인 HaCaT 세포에서 NRF2/ARE에 의존적인 유전자 발현의 유도를 통한 항산화 효과 (Ethanol Extracts of Rheum undulatum and Inula japonica Protect Against Oxidative Damages on Human Keratinocyte HaCaT cells through the Induction of ARE/NRF2-dependent Phase II Cytoprotective Enzymes)

  • 본 연구진은 HaCaT-ARE-luciferase 세포를 이용하여 400 여개의 약용식물 에탄올 추출물 중 NRF2/ARE 유도효과가 있는 신규 추출물을 검색하였고 이를 통하여 종대황(Rheum undulatum)과 선복화(Inula japonica)의 주정 추출물이 HaCaT-ARE-luciferase 세포에서 ARE 활성을 강하게 유도하는 것을 관찰하였다. 종대황과 선복화 에탄올 추출물은 HaCaT 세포에서 생존(viability)을 증가시켰고 NRF2/ARE에 의존적인 phase II cytoprotective 효소인 heme oxygenase-1 (HO-1)와 NADPH:quinone oxidoreductase-1 (NQO1)의 전사 및 단백질 발현을 강하게 유도하였다. 또한 종대황과 선복화 추출물은 HaCaT 세포에서 TPA로 유도한 세포 내 활성 산소 및 이를 통하여 생성되는 스트레스 마커인 8-hydroxydeoxyguanosine (8-OH-dG)과 4-hydroxynonenal (4HNE)의 발생을 강하게 억제하였다. 본 연구는 종대황과 선복화의 에탄올 추출물이 인간 피부 세포주인 HaCaT 세포에서 NRF2/ARE에 의존적인 유전자 발현의 유도를 통하여 강력한 항산화 효과를 발휘한다는 것을 증명한다.

삼잎국화 추출물의 피부세포 생리활성 효과 (Effect of Rudbeckia laciniata Extract on Physiological Activity of HaCaT Cells)

  • The objective of the present investigation was to obtain vitamin, mineral, flavonoid, and polyphenol profiles of Rudbeckia laciniata (RL), and to examine the effects of extract of RL (RLE) on various physiological activities of HaCaT keratinocyte for the utilization of RL as natural raw materials to develop functional food. To accomplish this purpose, we checked the contents of the general nutrients of RL. The contents of vitamin A, vitamin $B_1$ and vitamin $B_2$ were $7.49{\mu}g/g$, $51.96{\mu}g/g$, and $132{\mu}g/g$ respectively, while vitamin C and vitamin $D_3$ were not detected. The contents of mineral such as Ca, K and Fe were 2.01 mg/g, 6.06 mg/g and 0.03 mg/g respectively. Total flavonoid contents of RLE were 0.25 mg/g, and total polyphenol were estimated as 1.43 mg/g. Because RL contains high levels of vitamin A which is associated with skin aging, we investigated the effect of RLE on physiological function of keratinocytes with respect to skin aging. We found that RLE significantly increased the growth rate of HaCaT cells and reduced ultraviolet radiation B (UVB)-induced cellular toxicity. Also, the extract of Rudbeckia laciniata attenuated the UVB-induced reactive oxygen species (ROS) generation in a dose-dependent manner in HaCaT cells. In addition, treatment with the extract dose-dependently increased migration activity of HaCaT cells. Thus, these findings indicated that RLE could regulate the physiological activity of keratinocytes, and may be used to develop functional foods.

HaCaT 세포주에서 캐모마일 (Matricaria chamomilla) 추출물의 항병원 성 및 항염 효과에 관한 연구 (Study of Anti-microbial Activities and Anti-inflammatory Effects of Chamomile (Matricaria chamomilla) Extracts in HaCaT cells)

  • Chamomile (Matricaria chamomilla), a member of the Asteraceae family, is a well-known for medicinal plant and can be found in India and Europe. Chamomile is an effective sedative and various medical effects. But, the effects of acne treatment by chamomile were not investigated. Therefore, we assessed the anti-oxidant effects, anti-microbial activity and anti-inflammatory effects by chamomile extracts in HaCaT keratinocyte cells. Anti-oxidant effects of chamomile extracts were investigated by DPPH assay. Also, results of MTT assay was demonstrated that chamomile extracts did not have a cytotoxic effect in HaCaT cells. To assess the antimicrobial activity, we determined formation of inhibition zone of Propionibacterium acnes by extracts from chamomile. Tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) induces production of inflammatory cytokines such as interleukin-$1{\beta}$ (IL-$1{\beta}$), IL-6 and IL-8 and expression of COX-2. Chamomile extracts could inhibit TNF-${\alpha}$-induced mRNA expression levels of IL-$1{\beta}$, IL-6, IL-8 and COX-2 gene. These results demonstrated the possibility of chamomile for prevention and treatment of skin inflammatory diseases such as acne.

Acidic pH-activated $Cl^-$ Current and Intracellular $Ca^{2+}$ Response in Human Keratinocytes

  • The layers of keratinocytes form an acid mantle on the surface of the skin. Herein, we investigated the effects of acidic pH on the membrane current and $[Ca^{2+}]_c$ of human primary keratinocytes from foreskins and human keratinocyte cell line (HaCaT). Acidic extracellular pH ($pH_e{\leq}5.5$) activated outwardly rectifying $Cl^-$ current ($I_{Cl,pH}$) with slow kinetics of voltage-dependent activation. $I_{Cl,pH}$ was potently inhibited by an anion channel blocker 4,4'-diisothiocyanostilbene-2,2'-disulphonic acid (DIDS, 73.5% inhibition at 1${\mu}$M). $I_{Cl,pH}$ became more sensitive to $pH_e$ by raising temperature from $24^{circ}C$ to $37^{circ}C$. HaCaT cells also expressed $Ca^{2+}$-activated $Cl^-$ current ($I_{Cl,Ca}$), and the amplitude of $I_{Cl,Ca}$ was increased by relatively weak acidic $pH_e$ (7.0 and 6.8). Interestingly, the acidic $pH_e$ (5.0) also induced a sharp increase in the intracellular [$Ca^{2+}$] (${\triangle}[Ca^{2+}]_{acid}$) of HaCaT cells. The ${\triangle}[Ca^{2+}]_{acid}$ was independent of extracellular $Ca^{2+}$, and was abolished by the pretreatment with PLC inhibitor, U73122. In primary human keratinocytes, 5 out of 28 tested cells showed ${\triangle}[Ca^{2+}]_{acid}$. In summary, we found $I_{Cl,pH}$ and ${\triangle}[Ca^{2+}]_{acid}$ in human keratinocytes, and these ionic signals might have implication in pathophysiological responses and differentiation of epidermal keratinocytes.
