p13 gene was first described in Leucania separata multinuclear polyhedrosis virus (Ls-p13) several years ago, but the function of P13 protein has not been experimentally investigated to date. In this article, we indicated that the expression of p13 from Heliothis armigera single nucleocapsid nucleopolyhedrovirus (Ha-p13) was regulated by both early and late promoter. Luciferase assay demonstrated that the activity of Ha-p13 promoter with hr4 enhancer was more than 100 times in heterologous Sf9 cells than that in nature host Hz-AM1 cells. Both Ls-P13 and Ha-P13 are transmembrane proteins. Confocal microscopic analysis showed that both mainly located in the cytoplasm membrane at 48 h. Results of RNA interference indicated that Ha-p13 was a killing-associated gene for host insects H. armigera. The AcMNPV acquired the mentioned killing activity and markedly accelerate the killing rate when expressing Ls-p13. In conclusion, p13 is a killing associated gene in both homologous and heterologous nucleopolyhedrovirus.
$\alpha$-Amylase gene of B. amyloliquefaciens was cloned to E. coli-yeast shuttle vector YEp-13 and expressed in E. coli. Chromosomal DNA of B. amyloliquefaciens was partially digested with Sau3Al and YEp13 plasmid was cleaved with BamH1. The hybrid plasmid, pHA28, was constructed by shotgun method and transformed to E. coli C600 and HB101. The amount of $\alpha$-amylase produced by transformants of E. coli was about 20% to 30% of that produced by B. amyloli-quefaciens. About 65% of $\alpha$-amylase produced by transformant was secreted into periplasm and the others were located in cytoplasm. $\alpha$-Amylase production was maximal when transformants were cultivated for 15hr to 20hr. As the result of agarose gel electrophoresis, pHA28 plasmid was found to be various in its size. This result suggested that pHA28 plasmid was segregated.
This study aimed at improving the $TiO_2$ photocatalytic degradation of HA. A set of tests was first conducted in the dark to study the adsorption of HA at different coexisting material concentration. Adsorption rate increased with adding cation ion but decreased with adding bicarbonate ion. The photodegradation of HA in the presence of UV irradiation was investigated as a function of different experimental condition: initial concentration of HA, $TiO_2$ weight, pH, air flow rate and coexisting material. It was increased either at low pH or by adding cation ion. The increase of cation strength in aqueous solution could provide a favorable condition for adsorption of HA on the $TiO_2$ surface and therefore enhance the photodegradation rate. It was found that bicarbonate ions slowed down the degradation rate by scavening the hydroxyl radicals.

In this study, the nutrient removal capacity of paddy field was investigated. Paddy field was considered as a constructed wetland. The present study was conducted during a cropping period in a rice cultivation area located in Hampyung-gun, Jeollanamdo, Korea. Water balance and mass balance of nutrients were estimated after intensive field monitoring. The surface runoff losses of T-N and T-P were 20.82 kg/ha and 1.55 kg/ha, respectively. The losses were 13.2% and 6.4% of T-N and T-P input into paddy fields, respectively. The results showed that the paddy fields have nutrient removal function.
Nitrogen and P in surface runoff and eroded sediment from cropland areas can contaminate streams and lakes. Runoff losses of N and P were determined in a small field plot $(14.3{\times}24.8\;m)$ of peach orchard from March to November in 1999. Nitrogen and P were applied in the rate of 172 and 46 kg/ha using chemical fertilizer and mixed oil cake fertilizer. During the season, in 26 rainfall events, $421.5\;m^3/ha$ of runoff including 1,989 kg/ha of soil loss was collected. Concentrations of total-N, $NO_3-N$, $NH_4-N$, total-P and $PO_4-P$ in runoff samples were in the range of $4.7{\sim}171.0,\;0.1{\sim}188.0,\;0.13{\sim}3.36$, $0.58{\sim}4.99$ and $0.05{\sim}3.71\;mg/l$, respectively. Total loss of N was 16.39 kg/ha and 75% of the loss was $NO_3-N$. Total loss of P was 1.04 kg/ha, and $PO_4-P$ and sediment bound P accounted for 47 and 27% of the total loss, respectively. The losses of N and P were about 9.5 and 2.3% of the applied N and P in the plot, respectively. Although the loss of N or P would be relatively small in agricultural aspect, considering the high concentrations of N and P in runoff, loss of N and P from croplands should be controlled to reduce the eutrophication problem of stream waters.

This experiment was carried out to study the effects of different environmental conditions and cultivation techniques on productivity of grasslands in central and southern area of Korea on 2017 and 2018. Average dry matter yield of grasslands at 10 actual production sites was 7,496 kg/ha. that was ranged from 4,652 to 13,292 kg/ha with least significant difference(LSD) of 1,577kg/ha between grasslands (p<0.05) on 2017. Average dry matter yield of grasslands at 10 actual production sites was 7,914 kg/ha. that was ranged from 3,927 to 12,372 kg/ha with LSD of 1,577kg/ha between grasslands(p<0.05) on 2018. Dry matter(DM) yield of grasslands have positive correlation with soil fertility (p<0.01) but not correlated with rainfall and air temperature among cultivation environments. And also DM yield of grasslands have positive correlation with grassland management techniques(p<0.01). These results suggest that practices of grassland management techniques and improvement of soil fertility are more important than cultivation environments by climate change for increasing the DM yield of grassland in central and southern area of Korea.
Peat humin(p-Humin), humic acid(p-HA) and fulvic acid(p-FA) were isolated from Canadian Sphagnum peat moss by dissolution in 0.1M NaOH followed by acid precipitation. After purification cycles, they are characterized for their elemental compositions and, acid/base properties. Functionalities and carbon structures of the humic fractions were also characterized using FT-IR and solid state $^{13}C$-NMR spectroscopy. Those results are compared with one another and with soil humic substances from literatures. Main purpose of this study was to present a chemical and spectroscopic characterization data of humic substance from peat moss needed to evaluate its environmental applicability. The relative proportions of the p-Humin, p-HA and p-FA in the peat moss was $76\%,\;18\%,\;and\;3\%$, respectively, based on the total organic matter content ($957{\pm}32\;g/kg$). Elemental composition of p-Humin were found to be $C_{1.00}H_{1.52}O_{0.79}N_{0.01}$ and had higher H/C and (N+O)/C ratio compared to those of p-HA($C_{1.00}H_{1.09}O_{0.51}N_{0.02}$) and p-FA($C_{1.00}H_{1.08}O_{0.65}N_{0.01}$). Based on the analysis of pH titration data, there are two different types of acidic functional groups in the peat moss and its humic fractions and their proton exchange capacities(PEC, meq/g) were in the order p-FA(4.91) >p-HA(4.09) >p-Humin(2.38). IR spectroscopic results showed that the functionalities of the peat moss humic molecules are similar to those of soil humic substances, and carboxylic acid(-COOH) is main function group providing metal binding sites for Cd(II) sorption. Spectral features obtained from $^{13}C$-NMR indicated that peat moss humic molecules have rather lower degree of humification, and that important structural differences exist between p-Humin and soluble humic fractions(p-HA and p-FA).

To find out the effects of phosphatic fertilizer application level on the dry matter yield and nutrient contents of grasses, this experiment was arranged as a randomized complete block design with six treatments(0, 50, 100, 150, 200 and 250kg $P_2O_5$/ ha) and conducted at hill land in Gwangju, Gyeonggi Province from September, 1989 to October, 1992. The results obtained are summarized as follows : 1. As the phosphatic fertilization level increase, the early growth and cover degree of grasses wintered were more favorable and plant length was higher. 2. With 150kg $P_2O_5$/ ha application for three years, the average dry matter yield of grasses, 10,112kgl ha was sirnilared to that of 200kg $P_2O_5$/ ha and the yield was composed of 77.2% grasses(7,808kg/ ha), 19.2% legumes(l,937kg 1 ha) and 3.6% weeds(367kg / ha). 3. The crude protein and energy productivity of grasses increase as phosphatic fertilization level was increased, but those were not different between 150 and 250kg $P_2O_5$ 1 ha. Therefore, phosphatic fertilization level must be applied with 150kg / ha for a reasonable management in hilly grassland.

To investigate the effects of inorganic phosphate ($P_2O_5$ 200kg/ha), fermented cow manures($P_2O_5$ 200kg/ha, 400kg/ha) and poultry manures($P_2O_5$ 200kg/ha, 400kg/na) levels mixed with sawdust on the soil changes and productivity of alfalfa monoculture, a field experiment arranged by randonmized complete block design was conducted at National Livestock Research Institute in Suwon, September, 1993 to December 1995. The results are as follows. S1. Soil pH was changed from 5.40(before experiment) to 6.17(fermented cow manure, $P_2O_5$ 400kg/ha), 6.76 (fermented poultry manure, $P_2O_5$400kg/ha) and 6.3l(inorganic phosphate, $P_2O_5$ 200kg/ha) at 0~5cm depth of subsoil, respectively. Organic contents after experiment were lower as a range of 2.41%~3.15% than those before experiment. 2. Total nitrogen contents of each treatment showed as 0.13%~0.16% at 0~5cm depth of subsoil comparing with before experiment(0.15%). Available phosphate content of inorganic phosphate($P_2O_5$200kgha) was highest as 405ppm. lowest as 125ppm(fermented poultry manure, $P_2O_5$ 200kg/ha) than before trial(205ppm). 3. K and Ca contents of after experiment were appeared to 0.22%~0.78% and 6.45%~9.45% comparing with before experiment(0.10% and 3.56%), respectively. 4. During two years, average DM yield of fermented cow rnanure($P_2O_5$ 400kg/ha) was highest as 11, 731kg/ha and lowest as 9, 017kg/ha at fermented poultry manure plot($P_2O_5$ 200kg/ha) with as sawdust, but no significant difference was found among treatment(P>0.05). 5. CP production, NEL and TDN yield of fermented cow manure($P_2O_5$ 400kg/ha) were highest as 2, 282kg/ha 69, 900kg/ha and 6, 392kg/ha, respectively, but no significant difference among treatment(P>0.05). 6. In inorganic contents of alfalfas, there was not show same tendency in accordance with treatments. As a results mentioned above, it was possible to use 200kg $P_2O_5/ha$ of fermented cow or poultry manure with sawdust instead of inorganic phosphae fertilizer.
