질소시비 수준이 수수 $\times$ 수단그라스 교잡종과 대두와의 간작재배에 미치는 영향 (Effects of Nitrogen Level on Intercropping Cultivation of Sorghum $\times$ Sudangrass Hybrid and Soybean)

  • A field experiment was conducted in Chungiu and Jungwon to evaluate growth characteristics, root development, dry matter yield, crude protein yield and palatability according to nitrogen fertilizer level at intercropping cultivation of sorghum $\times$ sudangrass hybrid(hereinafier referred as SSH) and soybean. Nitrogen level was six treatment of Okg, 30kg 60kg, 90kg 120kg, and 150kg per hectare, and cutting date (sorghum $\times$ sudangrass and soybean) was july 28. 1. In the SSH, plant length was increased with increasing nitrogen fertilizer from 0kg/ha to 120kg/ha, but 150kgha treatment was decreased cornpared to 120kg/ha treatment. In the soybean, plant length did not show difference among 0kg, 30kg 60kg, and 90kg per hectare, but 120kg and 150kgfha treatment was on the contrary short. In the SSH, leaf length, leaf width and leaf number were the highest in 120kg/ha treatment, but soybean showed multifarious. 2. Stem diameter of SSH was increased with increasing nitrogen fertilizer. But soybean was thick from 0kg/ha treatment to 60kg/ha treatment, while above 90kg/ha treatment was on the contrary thin. Stem of SSH was hard with increasing nitrogen fertilizer, but soybean was soft with increasing nitrogen fertilizer. Root weight (Dry matter) of SSH was increased with increasing nitrogen fertilizer. On the contrary, soybean was decreased with increasing nitrogen fertilizer. 3. In the dairy cattle and Korean native cattle, palatability according to nitrogen fertilizer was the highest at 90kgha treatment, while 120kgha treatment and 150kgha treatment was the lowest. But in the deer, 30kg/ha treatment of low nitrogen fertilizer was the highest in palatability. 4. Nitrogen content of root of SSH was increased with increasing nitrogen fertilizer(P<0.01). But in soybean, 30kg and 60kg/ha treatment were higher than 0kg, 90kg, 120kg and 150kg/ha treatment(P<0.01). Total niwogen content of soil were high at 90kg 120kg 150kgha trea~ment(P<0.01). 5. Dry matter yield was high at the 90kg 120kg and 150kgha treatment as range from 15, 262kg/ha to 15, 614kg/ha without significant difference among those, but Okgha treatment was the lowest as 11, 183kg/ha (PC 0.05). Protein content of SSH was the highest at 90kg/ha treatment as 8.3 percentage. Soybean was highest at 60kgha treatment as 22.9 percentage. Protein yield was the highest at 90kg/ha treatment as 1, 547.6kg/ha, but Okgha treatment was the lowest as 1, 022.8kg/ha (P<0.01).

콜라겐 코팅된 결정화 하이드록시아파타이트 담체를 이용한 골아세포의 성장 (The Effect of Collagen Coating in Crystallized Hydroxyapatite Scaffold)

  • Purpose: Hydroxyapatite(HA) has been widely used due to its chemical similarity to bone and good biocompatibility. HA is composed of macropores and micropores. Too much irregularities of the micropores are ineffective against the adhesion and proliferation of osteoblast. Many efforts have been tried to overcome these drawbacks. HA crystal coating on the irregular surface of HA scaffold, crystallized HA, is one of the method to improve cell adhesion. Meanwhile, the collagen has been incorporated with HA to create composite scaffold that chemically resembles the natural extracellular matrix components of bone. The authors proposed to examine the effect of collagen - coated crystallized HA on the adhesion and proliferation of osteoblast. Method: HA powder containing $10{\mu}m$ pore size was manufactured as 1 cm pellet size. For the making crystallized HA, 0.1 M EDTA solution was used to dissolve HA powder and heated $100^{\circ}C$ for 48 hours. Next, the crystallized HA pellets were coated with collagen (0.1, 0.5, and 1%). The osteoblasts were seeded into HA pellets and incubated for the various times (1, 5, and 9 days). After the indicating days, methylthiazol tetrazolium (MTT) assay was performed for cell proliferation and alkaline phosphatase (ALP) activty was measured for bone formation. Result: In SEM study, the surface of crystallized HA pellet was more regular than HA pellet. MTT assay showed that the proliferation of osteoblasts increased in a collagen dose - dependent and time - dependent manner and had a maximum effect at 1% collagen concentration. ALP activity also increased in a collagen dose - dependent manner and had a highest effect at 1% collagen concentration. Conclusion: These data showed that crystallization and collagen coating of HA was effective for osteoblast proliferation and ALP activity. Therefore, our results suggest that crystallized - HA scaffold with collagen coating is may be a good strategy for tissue engineering application for bone formation.

저분자량 히알루론산의 제조 및 화장품에의 응용 (Preparation of Oligo Hyaluronic Acid by Hydrolysis and Its Application as a Cosmetic Ingredient)

  • 이온교환수지를 이용하여 말단기에 이중결합 생성이나 고리열림반응 없이 저분자 히알루론산(oligo HA)을 제조하였다. 제조된 oligo HA의 화장품소재로서의 활용 가능성 및 그 효능을 평가하기 위하여 fibroblast, keratinocyte 및 SIRC cell을 이용하여 독성을 평가하였고, Caco-2 cell과 인공피부를 이용하여 피부 투과도를 평가하였다. Oligo HA는 fibroblast와 keratinocyte cell에서 각각 300 ${\mu}g/mL$ 및 1,000 ${\mu}g/mL$까지의 농도에서 독성이 없었으며 in vitro ocular test에서도 2,000 ${\mu}g/mL$의 높은 농도에서까지 자극에 의한 세포독성이 관찰되지 않았다. Caco-2 cell을 이용한 세포 투과실험에서는 HA는 거의 투과되지 않는 것에 비해 oligo HA은 16.0 %까지 투과되었고, 인공피부를 이용한 세포투과 실험에서도 약 90 %의 상당히 높은 투과도를 보였다. 사람 피부에서 보습효과를 확인하기 위하여 oligo HA를 함유한 제형을 피부에 도포한 후 피부 수분량과 경피수분 손실량을 측정한 결과 HA와 비슷하게 우수한 보습 효과를 확인할 수 있었다. 인체 피부 누적 첩포 실험 결과, 특별한 피부 자극이 확인되지 않았다. Oligo HA는 HA의 우수한 보습력을 유지하면서 높은 피부 투과도를 갖는 보습소재로써 화장품에 유용하게 활용될 수 있음을 확인하였다.

The analysis of the cultivation status of the upland crops in the paddy field using unmanned aerial vehicle

  • Recently, the South Korean government encourages the cultivation of upland crops in the paddy field to maintain an adequate level of rice production and then to balance the demand and supply of rice. This is mainly because the rice consumption per capita per year has continued to decline from 135 kg in 1979 to 61.9 kg in 2016, although the rice production was relatively stable. As a result, the rice overproduction became a big social problem. As a part of that, various upland crops such as soybean, maize, minor cereals and forage crops are planted in the paddy field 10 years ago. The cultivation of these crops may settle the problem of short supply and mass import of the crops to some extent. However, a systematic remote observation of upland crops in the paddy field is very scarce. This study investigated the cultivation status of upland crops and any changes of crop harvesting in the paddy field by using an unmanned aerial vehicle (UAV). Also, we analyzed the kind of upland crops and cultivation area in the paddy field by utilizing time series observation images. A fixed wing UAV is used for the investigation. This is because it is easy to use the flight operation and to control flight management software, and it can automatically cope with various emergency states such as a strong wind and battery discharge. The material of UAV is expanded polypropylene, which has an advantage of less equipment damage and risk during takeoff and landing. We acquired observed images in Buljeong-myeon, Goesan-gun, Chungcheongbuk-do, South Korea by using fixed wing UAV in 2015 and 2016. The total investigated area reaches 6,045 ha, and among them the agricultural area was 1,377 ha. For the next step, we created an orthoimage from all images taken using Pix 4D mapper program. According to the results of image analyses in 2015, the paddy field covered total 577 ha (75.9%) with crop plant. The cultivation area of beans, ginseng, maize, tobacco and peach was 256 ha (36.6%), 63 ha (9.2%), 37 ha (5.4%), 31 ha (4.5%) and 27 ha (3.8), respectively. And in 2016, the total covered area was 586 ha (77.1%), and it was comprised of 253 ha (35.5%), 88 ha (12.3%), 29 ha (4.1%), 22 ha (3.1%) and 32 ha (4.5%) in the same order. In this study, we focused on identifying the paddy field which was converted to the cultivation of upland crops by using UAV. And, it has been indicated that the cultivation area of rice decreased from 141 ha in 2015 to 127 ha in 2016, although that of ginseng increased by 25 ha. As a result, it is expected that a lot of paddy field could be replaced by high-income crops such as ginseng and fruit tree (peach) instead of relative low-income rice. More specific and widespread research on the remote sensing in the paddy field needs to be done.

평창 및 제천 지역 신갈나무림의 바이오매스와 연간 순생산량 (Biomass and Annual Net Production of Quercus Mongolica Stands in Pyungchang and Jecheon Areas)

  • 강원도 평창군 중왕산 지역 60~70년생 신갈나무림과 충북 제천 월악산 지역 35년생 신갈나무림에서 사면과 지역에 따른 바이오매스와 연간 순생산량을 비교하였다. 강원 중왕산 해발 1000m 지역에 있는 60~70년생 신갈나무림의 바이오매스는 북사면에서 252.9 ton/ha (지상부 212.2 ton/ha), 남사면에서 212.3 ton/ha (지상부 177.4 ton/ha)이며, 충북 월악산 해발 300m 북사면 지역의 35년생 신갈나무림은 198.7 ton/ha (지상부 157.3 ton/ha)로 추정되었다. 또한 연간 순생산량은 중왕산 북사면 지역 17.3 ton/ha/yr (지상부 14.5 ton/ha/yr), 남사면 지역 14.2 ton/ha/yr (지상부 11.9 ton/ha/yr), 윌악산 지역 21.2 ton/ha/yr (지상부 16.8 ton/ha/yr)로 추정되어, 중왕산 지역 북사면에서 남사면보다 바이오매스와 연간 순생산량이 더 크게 나타났다. 엽면적 지수의 경우 중왕산 지역 북사면 (5.77)이 남사면(3.97)보다 더 많으며, 월악산 35년생 신갈나무림은 중왕산 지역보다 잎의 중량이 더 많고 엽면적 지수도 11.17로 높게 나타났다. 반면 잎의 순동화율은 중왕산 해발 1000m 북사면 지역 4.26kg/kg/yr, 남사면 지역 6.06 kg/kg/yr, 월악산 지역 2.60 kg/kg/yr으로 나타나, 중왕산 남사면 지역이 가장 높고 월악산 지역이 가장 낮아 중왕산 지역에서 잎의 생산효율이 더 좋은 것으로 나타났다.

무기태인산과 톱밥발효가축분이 Alfalfa 단파초지의 토양변화와 생산성에 미치는 영향 (Effects of Productivity of Alfalfa Monoculture by Inorganic Phosphaate and Fermented Cow and poultry manure Levels Mixed with Sawdust)

  • To investigate the effects of inorganic phosphate ($P_2O_5$ 200kg/ha), fermented cow manures($P_2O_5$ 200kg/ha, 400kg/ha) and poultry manures($P_2O_5$ 200kg/ha, 400kg/na) levels mixed with sawdust on the soil changes and productivity of alfalfa monoculture, a field experiment arranged by randonmized complete block design was conducted at National Livestock Research Institute in Suwon, September, 1993 to December 1995. The results are as follows. S1. Soil pH was changed from 5.40(before experiment) to 6.17(fermented cow manure, $P_2O_5$ 400kg/ha), 6.76 (fermented poultry manure, $P_2O_5$400kg/ha) and 6.3l(inorganic phosphate, $P_2O_5$ 200kg/ha) at 0~5cm depth of subsoil, respectively. Organic contents after experiment were lower as a range of 2.41%~3.15% than those before experiment. 2. Total nitrogen contents of each treatment showed as 0.13%~0.16% at 0~5cm depth of subsoil comparing with before experiment(0.15%). Available phosphate content of inorganic phosphate($P_2O_5$200kgha) was highest as 405ppm. lowest as 125ppm(fermented poultry manure, $P_2O_5$ 200kg/ha) than before trial(205ppm). 3. K and Ca contents of after experiment were appeared to 0.22%~0.78% and 6.45%~9.45% comparing with before experiment(0.10% and 3.56%), respectively. 4. During two years, average DM yield of fermented cow rnanure($P_2O_5$ 400kg/ha) was highest as 11, 731kg/ha and lowest as 9, 017kg/ha at fermented poultry manure plot($P_2O_5$ 200kg/ha) with as sawdust, but no significant difference was found among treatment(P>0.05). 5. CP production, NEL and TDN yield of fermented cow manure($P_2O_5$ 400kg/ha) were highest as 2, 282kg/ha 69, 900kg/ha and 6, 392kg/ha, respectively, but no significant difference among treatment(P>0.05). 6. In inorganic contents of alfalfas, there was not show same tendency in accordance with treatments. As a results mentioned above, it was possible to use 200kg $P_2O_5/ha$ of fermented cow or poultry manure with sawdust instead of inorganic phosphae fertilizer.

붉가시나무 간벌강도에 따른 지상부 탄소고정량에 관한 연구 (Aboveground Carbon Storage of Quercus acuta Stands by Thinning Intensity)

  • 본 연구는 전라남도 완도수목원의 붉가시나무 간벌처리구 임분의 4년 뒤 지상부 탄소고정량을 추정하기 수행되었다 간벌 처리구별 4년 뒤의 임분 생장은 임분예측표를 이용하였으며, 지상부의 건중량 분포는 $W=aD+bD^2$의 모형식을 이용하여 추정하였다. 간벌 처리구별 지상부 생체량은 대조구(148.4 ton/ha), 약도처리구(105.6 ton/ha), 중도처리구(68.7 ton/ha) 그리고 강도처리구(39.1 ton/ha) 분포를 보였다. 붉가시나무의 4년 뒤 간벌처리구별 탄소고정량은 건중량에 0.5를 곱하여 구하였으며, 대조구(74.2tonC/ha), 약도처리구(52.8tonC/ha), 중도처리구(34.3 tonC/ha) 그리고 강도처리구의 탄소고정량은 38.2 tonC/ha으로 조사되었다.

당귀(當歸), 황기(黃芪), 약침(藥鍼)이 고지방(高指肪) 식이(食餌)로 유발(誘發)된 비만(肥滿) 백서(白鼠)의 식이, 지질대사, ALT에 미치는 영향(影響) (Effect of Herbal Acupuncture (Angelica gigas, Astragalus membranaceus) on Diet, Serum Lipid Metabolism and ALT in Obese Rats Induced by High Fat Diet)

  • Objectives : This study was performed to investigate the effect of herbal acupuncture(Angelica gigas, Astragalus membranaceus) at Chun-wan(CV-12) and Chok-Samni(ST36) on weight gain, food intake, food efficiency, serum of lipid concentrations, liver function and HDL-cholesterol to total cholesterol ratio of rats fed high fat diet for diet for 5 weeks. Methods : Experimental groups were divided into normal group, high fat diet group(Control), high fat diet and Angelica gigas herbal acupuncture at CV12 and ST36 treated group(HA-1), high fat diet and Astragalus membranaceus herbal acupuncture at CV12 and ST36 treated Group(HA-2), high fat diet and Astragalus membranaceus herbal acupuncture at CV12 and ST36 treated Group(HA-3). Herbal acupuncture was bilaterally treated at the level of 16.7 ul/kg body weight per day. Results : Body weight were significantly decreased on 15th and 20th day in HA-1, HA-2, and decresed on 25th and 30th day in all the groups. Food intake were increased on 20th day and after were decreased on 30th day in HA-1 and HA-2. And food intake of HA-3 were decreased on 10th day and after were increased on 20th and 25th day. Total cholesterol, LDL-cholesterol, triglyceride and ALT were significantly decreased in HA-1, HA-2 and HA-3 groups compared with control group. HDL-cholesterol to total cholesterol ratio were significantly increased in HA-1, HA-2 and HA-3 groups compared with control group. And food efficiency were significantly decreased in HA-1 and HA-3 groups compared with control group. Especially HDL-cholesterol were increased in HA-1 Group. Conclusions : Herbal acupuncture using Angelica gigas and Astragalus membranaceus at CV12 and ST36 can manage obesity by controlling body weight, food intake and food efficiency ratio.

유기물원이 다른 퇴비연용시 퇴비시용 한계량 연구 (Amount of Maximum Compost Application on the Long-term Application with Different Organic Material Sources in Upland Soil)

  • 본 연구는 밭토양에서 종류가 상이한 퇴비시용 시 퇴비시용 한계량을 구명하기 위하여 수행하였다. 공시토양은 양토와 사양토를 사용하였고, 시험에 사용한 퇴비의 종류는 계분퇴비, 우분퇴비, 분뇨잔사 및 식품오니퇴비 이었다. 퇴비 시용량은 0, 40, $80Mg\;ha^{-1}$ 수준이었고, 공시작물은 콩과 옥수수로서 1994년부터 1997년도까지 4년간 연작을 하였으며 주요 연구결과를 요약하면 다음과 같다. 옥수수의 건물생산량은 퇴비 시용량이 증가될수록 증가되었으나 콩은 퇴비 $60Mg\;ha^{-1}$ 시용까지는 증가되다가 그 이상은 감소되었다. 질소와 인산 이용율은 옥수수가 콩보다 많았으며, 질소 이용율은 옥수수가 21~31%, 콩은 8~19%였고, 인산 이용율은 옥수수와 콩이 각각 5~7%. 1~2%로 낮은 편이었다. Biomass기준 계분퇴비 시용 한계량은 옥수수재배시는 $39{\sim}47Mg\;ha^{-1}$ 콩 재배시는 $8{\sim}13Mg\;ha^{-1}$범위 이었다. 작물의 질소 요구량을 기준으로 하였을 경우는 옥수수 $123Mg\;ha^{-1}$, 콩은 $87Mg\;ha^{-1}$였고, 인산기준으로는 옥수수 $29Mg\;ha^{-1}$ 콩은 $14Mg\;ha^{-1}$범위였다. 토양의 염기포화도 80%를 한계수준으로 평가한 토성별 퇴비시용 한계량은 옥수수재배시 양토는 $69{\sim}93Mg\;ha^{-1}$, 사양토는 $49{\sim}69Mg\;ha^{-1}$범위이었다.

HaCaT cell에서 황련 추출물(Coptis chinensis)의 피부보습과 항염증 효과 (Skin Moisturizing Properties and Anti-Inflammatory effects of extracts from Coptis chinensis in HaCaT cells)

  • 황련은 한의학에서 진정, 소염, 항균 및 해열 등의 여러 질병의 치료에 이용되어져 왔다. 본 연구에서는 UVB와 TNF-α/IFN-γ로 유도된 HaCaT 세포에서 피부장벽과 염증관련 인자에 대한 황련 열수 추출물(CCW)의 효과를 조사하고, 보습 및 항염증 소재로서의 가능성을 알아보고자 하였다. 각 질형성세포인 HaCaT 세포에서 CCW의 보습 및 항염 효과를 확인하기 위해 MTT assay를 통한 시료의 독성 평가를 실시하였다. 이를 바탕으로, HA(Hyaluronic acid) 생성량과 filaggrin의 protein 및 mRNA 발현을 측정하였다. TNF-α/IFN-γ로 자극된 HaCaT 세포에서 CCW는 농도 의존적으로 HA의 생성량을 증가 시켰고, filaggrin의 protein 및 mRNA 발현 측정 결과 CCW의 농도 증가에 따라 발현율도 증가하였다. UVB로 유도된 HaCaT 세포에서 CCW는 ROS의 생성을 감소 시켰고, 양성대조군(positive control)인 EGCG( (-)-epigallocatechin-3-gallate)와 유의한 결과를 보여 주었다. 또한, CCW는 염증성 사이토카인 TNF-α, IL-1β, IL-6, IL-8의 발현을 억제 시켰고, TNF-α/IFN-γ로 자극된 HaCaT 세포에서 피부 염증에 따른 주요 인자인 COX-2의 protein 및 mRNA 발현이 농도 의존적으로 감소되는 것을 확인할 수 있었다. 이러한 결과는, 황련 열수 추출물의 보습 및 항염증 효과를 가지는 화장품 소재로서의 가치를 제안할 수 있다.
