Modulation of the inflammatory process and interaction of THP-1 monocytes with intestinal epithelial cells by glasswort (Salicornia herbacea L.) extracts (인간 단핵구 THP-1의 염증반응 및 장관상피세포와의 상호작용에 미치는 퉁퉁마디 추출물 분획의 영향)

  • The glasswort is an edible halophyte demonstrating various physiological effects including anti-inflammatory activity. In the present study, the effects of glasswort extracts on inflammatory events and interactions of THP-1 monocytes with intestinal epithelial cells were investigated. Five solvent fractions, including the ethylether fraction (Fr.E), were prepared from a 70% methanol extract of glasswort. THP-1 monocytes underwent differentiation by phorbol 12-myristate 13-acetate treatment and were then activated by lipopolysaccharide (LPS), which induced cyclooxygenase (COX)-2 expression. None of the glasswort fractions tested alone induced COX-2 in differentiated THP-1 cells. Fr.E, however, enhanced LPS-induced COX-2 expression in differentiated THP-1 cells. Culture media of THP-1 cells treated with each fraction stimulated the growth of normal intestinal INT-407 cells more prominently than that of HT-29 colon cancer cells. COX-2 expression in HT-29 cells was inhibited when the cells were exposed to the THP-1 culture medium treated with Fr.E. Thus, glasswort could modulate the interaction between immune cells and intestinal cells.

Cytotoxic Effect of Extracts from Styela clava against Human Cancer Cell Lines (암세포주에 대한 미더덕 추출물의 세포독성 효과)

  • The present study describes the preliminary evaluation of the cytotoxic effect from Styela clava (Korean name : miduduk) extracts. S. clava was treated with methanol, ethanol, acetone, and water, then cytotoxic effect of the extracts were evaluated by the MTT reduction assay. The ethanol extracts from S. clava showed the cytotoxic activity on the HT-29 human colon cancer cells. The ethanol extracts was further fractionated with n-hexane, diethyl ether, ethyl acetate, and water according to the degree of polarity. The diethyl ether fraction showed high cytotoxic activity of HT-29 cells. however, the other fractions showed low cytotoxicity. The diethyl ether layer also showed the cytotoxic activity against SW620, HeLa, and MCF-7 cells. These studies support that extracts from S. clava may be a potential candidate as a possible chemotherapeutic agent against human cancer cells.

The Cytotoxicity of Kahweol in HT-29 Human Colorectal Cancer Cells Is Mediated by Apoptosis and Suppression of Heat Shock Protein 70 Expression

  • Although coffee is known to have antioxidant, anti-inflammatory, and antitumor properties, there have been few reports about the effect and mechanism of coffee compounds in colorectal cancer. Heat shock proteins (HSPs) are molecular chaperones that prevent cell death. Their expression is significantly elevated in many tumors and is accompanied by increased cell proliferation, metastasis and poor response to chemotherapy. In this study, we investigated the cytotoxicity of four bioactive compounds in coffee, namely, caffeine, caffeic acid, chlorogenic acid, and kahweol, in HT-29 human colon adenocarcinoma cells. Only kahweol showed significant cytotoxicity. Specifically, kahweol increased the expression of caspase-3, a pro-apoptotic factor, and decreased the expression of anti-apoptotic factors, such as Bcl-2 and phosphorylated Akt. In addition, kahweol significantly attenuated the expression of HSP70. Inhibition of HSP70 activity with triptolide increased kahweol-induced cytotoxicity. In contrast, overexpression of HSP70 significantly reduced kahweol-induced cell death. Taken together, these results demonstrate that kahweol inhibits colorectal tumor cell growth by promoting apoptosis and suppressing HSP70 expression.

Comparisons of Properties, In Vitro Anti-Mutagenicity, and Anti-Cancer Effects of Short-Term Fermented Soybean Foods (Chungkukjang, Shuidouchi, and Natto) (콩 발효 식품(청국장, 물두시, 낫또)의 특성, In Vitro 항돌연변이 및 항암 효과)

  • The general characteristics, anti-mutagenicity, and in vitro anti-cancer effects of Chungkukjang, Shuidouchi, and Natto were studied. Representative brands of the three types of soybean-based short-term-fermented foods were chosen and evaluated. Chungkukjang showed the highest amino and ammonia type nitrogen contents, although Natto exhibited lower levels, and Shuidouchi showed the lowest pH and high acidity. Anti-mutagenicity was assessed by the Ames test and in vitro anti-cancer effects were assessed in HT-29 human colon cancer cells. Chungkukjang exhibited the highest anti-mutagenicity and anti-cancer effects. Based on these results, Chungkukjang showed the best functional effects, and Shuidouchi showed better functional effects than Natto. Even if such efficacies depend on raw materials and processing methods, our comparison of three types of foods from representative brands shows that Chungkukjang was the best soybean-derived product in terms of overall quality, followed by Shuidouchi and Natto. These results might be due to differences in duration of fermentation, processing method, and quality of raw materials, but further research on the basis of ethnic culture and administration method of each nation should follow.

Suppressive effects of pinosylvin on prostaglandin E$_2$and nitric oxide production in lipopolysaccharide-stimulated RAW 264.7 cells

  • The inhibitors of prostaglandin biosynthesis and nitric oxide production by corresponding inducible isozyme have been considered as potential anti-inflammatory and cancer chemopreventive agents. In our continuous search for cancer chemopreventive agents from natural products, we have evaluated the inhibitory potential of PGE$_2$and NO production in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7 cells. As a result, pinosylvin (3,5-dihydroxy-trans-stilbene), a stilbenoid, mainly found from the heartwood and leaves of the Pinus sylvestris, showed potential inhibitory activity of LPS-induced PGE$_2$and NO production in a dose-dependent manner. Pinosylvin also suppressed the LPS-induced iNOS protein expression. Further study revealed that pinosylvin exhibited antioxidant activity by the DPPH free radical scavenging potential and inhibitory effect of xanthine oxidase activity. In addition, pinosylvin inhibited COX -2 overexpressed human colon cancer cell (HT-29) growth in a time- and dose-dependent manner. These findings suggest that pinosylvin might be a promising candidate for developing cancer chemopreventive agent.

Heavy Metal Contents and Antioxidant Activity and Cytotoxic Effect of Red Sea Bream (Pagrus major): Comparative Studies in Domestic and Imported Red Sea Bream (Pagrus major) (국내산 및 수입산 참돔의 중금속 함량 및 항산화 활성과 세포독성 효과 비교)

  • This study compared the heavy metal contents and the effects of extracts from domestic and imported red sea bream on the antioxidant activity and cytotoxicity of human cancer cell lines. The antioxidant activity was measured using the fluorescently sensitive dye, 2’-7’ dichlorofluorescein-diacetate (DCFH-DA), and antiproliferative activity against AGS human gastric adenocarcinoma and HT-29 human colon cancer cell lines, which was determined by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Domestic red sea bream had a higher mercury content when compared to imported red sea bream, but there was no significant difference in the lead content. Treatments with acetone/methylene chloride (A+M) and methanol (MeOH) extracts from domestic and imported red sea bream dose-dependently decreased the H2O2 induced ROS production, compared to the control. The cell viability showed that treatments with the A+M and MeOH extracts had cytotoxicity in the growth of AGS and HT-29 cancer cells. In the case of AGS, the extracts from the domestic red sea bream were higher in inhibiting cancer cell growth, compared to imported red sea bream. Our results demonstrate that the heavy metal contents of domestic and imported red sea bream were below the limit of the Food Code of Korea. The results of the biological activities indicate that the antioxidant activity of extracts from imported red sea bream was more effective, while the extracts from the domestic red sea bream were stronger in cytotoxic activity.

NADPH oxidase inhibitor diphenyleneiodonium induces p53 expression and cell cycle arrest in several cancer cell lines (NADPH oxidase 저해제인 diphenyleneiodonium의 p53 발현 및 암세포의 성장억제에 대한 연구)

  • The Diphenyleneiodonium (DPI) is widely used as an inhibitor of flavoenzymes, particularly NADPH oxidase. In this study, we investigated the effect of DPI on the cell growth progression of human colon cancer cells HCT-116 (wild-type p53), HT-29 (p53 mutant) and human breast cancer cells MCF-7 (wild-type p53). DPI treatment in cancer cells evoked a dose- and time-dependent growth inhibition, and also induced the cell cycle arrest in C2/M phase. The peak of cell population arrested in C2/M phase was observed at12 hr after treatment of DPI. In addition, DPI significantly induced the expression of p53, which induces proapoptotic genes in response to DNA damage or irreparable cell cycle arrest, at 6 hr in DPI-stimulated cells. However, a catechol apocynin, which inhibits the assembly of NADPH oxidase, did not induce p53 expression. This suggest that p53 expression induced by DPI is not associated with the inhibition of NADPH oxidase. In conclusion, we suggest that DPI induces the expression of wild-type p53 by ROS-in-dependent mechanism in several cancer cells, and upregulated p53 may be involved in regulatory mechanisms for growth inhibition and cell cycle arrest at C2/M phase in DPI-stimulated cells.

Inhibition effects of extracts and its solvent fractions isolated from Limonium tetragonum on growth of human cancer cells (염생식물 갯질경 [Limonium tetragonum]의 추출물이 암세포성장에 미치는 효과)

  • In this study, crude extracts of the halophyte Limonium tetragonum and their solvent fractions were evaluated on anticancer activity in AGS and HT-29 human cancer cells using MTT assay. Each of the crude extracts (MeOH and $CH_2Cl_2$) of Limonium tetragonum showed a significant inhibitory effect on the growth of human cancer cells. The combined crude extracts of MeOH and $CH_2Cl_2$ were partitioned between $CH_2Cl_2$ and water. The organic layer was further partitioned between 85% aq. MeOH and n-hexane, and then the aqueous layer was fractionated with n-BuOH and $H_2O$, successively. Growth inhibition effects of crude extracts and their solvent fractions from Limonium tetragonum increased in a dose-dependent manner. Among them, 85% aq. MeOH, n-hexane and n-BuOH fractions revealed very good inhibition effects on the growth of human cancer cells. These results suggest that we can isolate active compounds from Limonium tetragonum to show much more strong anticancer activity.

Antioxidative Effects of Purple Sweet Potato Extracts (자색고구마 추출물의 항산화 효과)

  • The colored sweet potato, particularly purple sweet potato, has been well known to contain anthocyanins abundantly. This study was conducted to examine the antioxidant properties of purple sweet potato. The chopped purple sweet potato was extracted 2 times with water or acetone for 18 hours at $28^{\circ}C$. The antioxidative potential of each solvent extract was assessed by DPPH free radical scavenging activity assay, FRAP assay, and total phenolic contents. The results showed that both extracts had not only high DPPH free radical scavenging activity but had high level of total phenolic compounds. Furthermore, both solvent extracts were found to have antioxidative effects in human colon cancer cells (HCT 116, HT 29) in DCFDA assay. The notable antioxidant activity of purple sweet potato suggests its significant health benefit and deserves further study to develop into functional food ingredient.

Growth-inhibitory Effect of the Solar Salt-Doenjang on Cancer Cells, AGS and HT-29 (천일염으로 제조한 된장의 암세포 성장 억제효과)

  • Doenjang was prepared by using Bacillus subtilis DJI starter and purified salt or solar salt with 12% (w/w) concentration. The prepared Doenjangs were fermented and ripened for 2 month and 16 month, respectively. MTT assay was used to measure the growth-inhibitory effect of Doenjang extracts (water, methanol) on BJ (human foreskin normal cell), HT-29 (human colon cancer cell) and AGS (human gastric adenocarcinoma cell). The anticancer effect increased in both purified salt-Doenjang and solar salt-Doenjang as fermentation period increased. Moreover in the case of water extracts, solar salt-Doenjang (growth inhibitory rate; AGS: 50%, HT-29: 44%) showed definitely higher anticancer effect than purified salt-Doenjang (AGS: 32%, HT-29: 32%). In addition, apoptosis were observed when the water extracts of the Doenjangs were treated into AGS. In particular, it was observed that more apoptosis occured in solar salt-Doenjang treats than purified salt-Doenjang treats. These results suggested that prolonging the fermentation with addition of solar-salt when making Doenjang increased its anticancer effect via cancer cell growth inhibition induced by apoptosis process.
