본 연구에서는 headspace-solid phase microextraction(HS-SPME)-GC/MS법을 이용하여 PVC 벽지의 원자재에서 방출되는 TVOC를 분석하여 기여도를 평가하였다. 시료로 사용한 원자재는 6종으로 희석제, 안정제, 가소제, 충전제, 발포제와 PVC 레진이다. 각 원자재 시료를 22 mL 바이알에 9 mL씩 취한뒤 $100^{\circ}C$에서 1시간동안 평형을 이룬 다음 $75{\mu}m$ Carboxen-PDMS 화이버로 $25^{\circ}C$에서 1시간 흡착하여 GC/MS로 분석하였다. 정성결과 toluene, ethylbenzene 그리고 xylene과 같은 방향족 화합물과 acetone, methoxyacetone, 2-butanone 등의 케톤류와 nonane, decane, undecane 등의 알켄류가 검출되었다. 그 밖에도 ethanol, butanol 등의 알콜류와 알데히드류 등이 검출되었다. TVOC 방출량은 희석제 $54.20{\mu}g/g$, 안정제 $32.88{\mu}g/g$, 가소제 $0.50{\mu}g/g$, PVC 레진 $0.88{\mu}g/g$, 발포제 $0.22{\mu}g/g$, 충전제가 $0.11{\mu}g/g$로 나타났다. 각 원자재의 TVOC 방출량과 배합비를 고려한 TVOC 기여도는 희석제 0.708, 안정제 0.129, PVC 레진 0.115 순으로 높게 나타났다. 따라서 희석제와 안정제, PVC레진의 질적인 개선을 통해 TVOC 방출량을 줄이는 노력이 필요하다. 또한 본 연구에서 개발한 HS-SPME-GC/MS 방법은 원자재의 TVOC 분석에 유용하게 사용될 것으로 생각된다.
In this study, the volatile compounds in 9 commercial fermented soybean pastes were extracted and analyzed by headspace-solid phase microextraction (HS-SPME) and gas chromatography-mass spectrometry (GC-MS), respectively. A total of 63 volatile components, including 21 esters, 7 alcohols, 7 acids, 8 pyrazines, 5 volatile phenols, 3 ketones, 6 aldehydes, and 6 miscellaneous compounds, were identified. Esters, acids, and pyrazines were the largest groups among the quantified volatiles. About 50% of the total quantified volatile material was contributed by 5 compounds in 9 soybean paste samples; ethyl hexadecanoate, acetic acid, butanoic acid, 2/3-methyl butanoic acid, and tetramethyl-pyrazine. Three samples (CJW, SIN, and HAE) made by Aspergillus oryzae inoculation showed similar volatile patterns as shown in principal component analyses to GC-MS data sets, which showed higher levels in ethyl esters and 2-methoxy-4-vinylphenol. Traditional fermented soybean pastes showed overall higher levels in pyrazines and acids contents.
간 및 간에 함유된 불포화지방산인 arachidonic, linolenic, linoleic 및 oleic acid의 철 이온 존재하에서의 headspace 산화 반응생성물을 SPME법을 이용하여 포집, 분석하는 것에 의해 간에 함유된 불포화지방산의 산화생성물과 간의 비린내 불쾌취 형성과의 관련성을 검토하였다. SPME fiber중 불포화지방산의 headspace 산화생성물인 휘발성 알데히드류, 알코올류 및 지방산 등 대다수의 화합물을 검출에는 PDMS/DVB fiber가 적합하였다. 간의 headspace 산화생성물은 arachdonic acid을 비롯한 간에 함유된 각 불포화지방산의 head-space 산화생성물과 일치하였으며, 간의 불쾌취 형성은 간에 함유된 불포화지방산의 산화로 생성되는 산화 반응생성물에 의한 것이 시사되었다. 간의 불쾌취를 형성하는 주요성분으로는, arachidonic acid에서 생성되어 간의 금속취를 형성하는 화합물로 생각되어지는 1-octen-3-one 및 hexanol과 linolenic acid에서 형성되어 간의 비린내를 형성하는 (E,E)-2,4-heptadienal이 그 원인물질인 것으로 사료된다. 산화에 의해 생성된 각 불포화지방산의 산화 반응생성물은 간의 불쾌취인 비린내, 금속취 등을 생성하며, 주로 arachidonic acid 및 linolenic acid의 산화에 의해 생성된 산화 반응생성물에 의한 것으로 밝혀졌다.
우리나라의 자생식물인 털진달래(Rhododendon mucronulatum Turcz. var. ciliatum Nakai)지상부위로부터 정유를 얻고, 휘발성분을 headspace(HS)와 solid-phase microextraction(SPME)법으로 흡착시킨 다음 GC-MS로 분석하였다. 털진달래 정유에는 탄화수소 15종, 알코올 3종, 아세테이트 1종 등 총 19종의 화학성분이 함유되어 있었으며, 다함량 성분은 2-${\beta}$-pinene(16.1%), camphene(11.9%), ${\delta}$-3-carene(11.4%), d,l-limonene(9.5%), and ${\gamma}$-terpinene(9.5%)이었다. SPME법으로 흡착된 시료에는 탄화수소 13종, 알코올 1종, 질소화합물 1종 등 총 17종의 휘발성 화학물질이 함유되어 있었고, 이들 중 다함량 성분은 camphene(19.6%), 2-${\beta}$-pinene(18.0%), ${\delta}$-3-carene(17.4%), timethyl hydrazine(9.7%), ${\gamma}$-terpinene(8.5%), and d,l-limonene(5.5%)이었다. 그리고 HS법으로 흡착된 시료에는 탄화수소 11종, 알코올 1종, 질소화합물 1종 등 총 13종의 휘발성 화학물질이 함유되어 있었으며, 이들 중 다함량 성분은 camphene(25.8%), ${\delta}$-3-carene(24.8%), 2-${\beta}$-pinene(20.2%), d,l-limonene(5.4%), tricyclene(5.1%), trimethyl hydrazine(4.6%)이었다. 털진달래 정유의 향취는 coniferous, balsamic, woody하였으며, HaCaT 각질형성세포에 대한 $IC_{50}$ 값은 0.030 ${\mu}g/mg$이었다.
Fresh japanese anise (Illicium anisatum L.) tree leaves were collected and ground after drying. The essential oils of the leaves were analyzed by gas chromatography-mass spectrometry (GC-MS) using headspace (HS) and solid phase-microextra (SPME) methods. Volatile components of the leaves were identified 21 and 65 components in HS and SPME, respectively. The main components of the essential oils obtained by HS method were eucalyptol (36.7%), (+)-sabinene (15.61%), ${\delta}$-3-carene (6.87%), ${\alpha}$-pinene (6.07%), ${\gamma}$-terpinen (5.72%), ${\alpha}$-limonene (5.26%), ${\beta}$-myrcene (4.13%), ${\alpha}$-terpinene (4.04%) and ${\beta}$-pinene (3.73%). The other components were less than 3.5%. SPME method also showed that eucalyptol (17.88%) was main. The other were 5-allyl-1-methoxy-2 (13.29%), caryophyllene (6.09%), (+)-sabinene (5.60%), ${\alpha}$-ocimene (4.89%) and ${\beta}$-myrcene (3.73%), and the rest were less amounts than 3.5%. This work indicated that many more volatile components were isolated, comparing to the previous literature data and that SPME method was much more effective than HS method in the analysis of the volatile components.
In many drinking water treatment plants, chlorination process is one of the main techniques used for the disinfection of water. This disinfecting treatment leads to the formation of haloacetic acid (HAAs). In this study, headspace solid-phase microextraction (HS-SPME) was studied as a possible alternative to liquid-liquid extraction for the analysis of HAAs in drinking water. The method involves direct derivatization of the acids to their methyl esters without methyl tert-butyl ether (MTBE) extraction, followed by HS-SPME with a $2cm-50/30{\mu}m$ divinylbenzene/carboxen/polydimethylsiloxane fiber. The effects of experimental parameters such as selection of SPME fiber, the volume of sulphuric acid and methanol, derivatization temperature and time, the addition of salts, extraction temperature and time, and desorption time on the analysis were investigated. Analytical parameters such as linearity, repeatability and limit of detection were also evaluated. The $2cm-50/30{\mu}m$-divinylbenzene/carboxen/polydimethylsiloxane fiber, sulphuric acid of 1ml, methanol of 3ml, derivatization temperature of $50^{\circ}C$ derivatization time of 2hrs, sodium chloride salt of 10g, extraction time of 30 minutes, extraction temperature of $20^{\circ}C$ and desorption time of 1 minute at $260^{\circ}C$ were selected as the optimal experimental conditions for the analysis of HAAs. The linearities ($r^2$), relative standard deviations (%RSD) and limits of detection (LOD) for HAAs were 0.9978~0.9991, 1.1~9.8% and $0.05{\sim}0.2{\mu}g/l$, respectively.
죽초액은 대나무 숯 제조시 발생하는 강한 산성을 가진 부산물로 80-90%의 수분과 200 종 이상의 유기화합물로 구성되어 있으며, 일반적으로 토양 질 개선, 식물 생육 조절, 아토피 피부염 개선 및 항균효과 등 피부관련 질환에 널리 활용되고 있다. 본 연구에서는 담양군에서 제조한 죽초액 휘발성성분을 direct analysis (DA)법, head space solid phase microextraction (HS-SPME)법, stir bar sorptive extraction (SBSE)법을 이용하여 GC/MS로 분리하고 동정하였다. 위 세 가지 방법을 이용하여 103 종의 화합물을 검출하였으며, HS-SPME와 SBSE방법에서 페놀류 화합물의 선택적 분석이 가능하였다. 죽초액의 주요한 화합물은 유효한 생리활성 물질로 알려진 cresol, guaiacol, p-ethyl guaiacol, syringol이 분석되었다.
혈액 중에서 톨루엔을 HS-SPME 법을 이용하여 GC/MSD로 분석하였으며, 부탄가스류의 분석은 HS 법을 이용하여 CC/FID로 분석하였다. 우리 나라의 경우 지난 3년 동안 청소년들이 본드류나 부탄가스류 등을 흡입한 상태에서 사망한 경우가 총 75건이 발생하였다. 본드류를 흡입하다가 사망한 사람은 27명이었으며, 그 중에서 9명은 톨루엔 중독상태에서 자제력의 상실 등으로 인하여 고층에서 떨어지는 추락사를 하였으며, 이때의 혈중 농도 분포는 $1.3{\sim}21.6{\mu}g/mL$으로 상당히 높은 농도였다. 또한 부탄가스를 흡입하다가 질식 사망 또는 흡입 사망한 사람은 54명이었으며, 이 중에는 본드류와 부탄가스를 동시에 흡입하다가 사망한 6명도 포함되었다.
해수 내 유류계 방향족탄화수소인 BTEX(benzene, toluene, ethylbenzene, xylene)와 다환방향족탄화수소(polycyclic aromatic hydrocarbons: PAHs)를 동시에 분석할 수 있는 기술 개발을 위해 GC/MS에서 고체상미량추출법(solid phase microextraction: SPME)을 이용하여 최적의 분석기법을 정립하였다. SPME 기법은 전통의 분석 방법과 비교할 때 조작이 간단하고, 파이버를 재사용할 수 있고, 휴대하기 쉽고, 시료의 운반이나 저장하는 동안 오염을 최소화 할 수 있는 장점이 있다. 최적의 SPME 조건을 정립하기 위해 여러 변수 즉, SPME 수착제, 흡착 시간, 흡착 온도, 교반 속도, GC 탈착 시간들을 확인하였다. 다양한 SPME 수착제($100{\mu}m$ PDMS, $75{\mu}m$ CAR/PDMS, $65{\mu}m$ PDMS/DVB)를 이용하여 BTEX와 PAHs(분자량 78부터 202까지)를 동시에 분석한 결과 $65{\mu}m$ PDMS/DVB를 최적의 수착제로 선정하였다. 최적의 수착제로 $65{\mu}m$ PDMS/DVB 선정한 다음 순차적으로 다른 변수들을 확인하였다. 그 결과 BTEX와 PAHs 동시 분석하기 위한 최적의 SPMD 조건은 흡착시간 60분, 흡착온도 $50^{\circ}C$, 교반속도 750 rpm, GC 탈착시간 3분으로 결정되었다. 최적화한 HS-SPME-GC/MS 분석법을 이용하여 인공오염해수 내 유류계 방향족탄화수소 분석 결과 이전 연구 방법과 유사하였다. HP-SPME-GC/MS 분석법은 기존에 유기용매를 사용한 방법이 가졌던 단점과 제한점을 보완할 수 있으며, 해수 내 유류에 의한 BTEX 및 PAHs 분석에 효율적으로 적용할 수 있다.
In many drinking water treatment plants, chlorination process is one of the main techniques used for the disinfection of water. Analytical parameters such as linearity, repeatability and detection limits were also evaluated. The $50/30{\mu}m$-divinylbenzene/carboxen/polydimethylsiloxane fiber, extraction time of 30 minutes, extraction temperature of $20^{\circ}C$ and desorption time of 1 minute at $260^{\circ}C$ were the optimal experimental conditions for the analysis of HANs. The correlation coefficients ($r^2$) for HANs was 0.9979~0.9991, respectively. The relative standard deviations (%RSD) for HANs was 2.3~7.6%, respectively. Detection limits (LDs) for HANs was $0.01{\sim}0.5{\mu}g/L$, respectively.
