Chronopharmacokinetics of Vancomycin in Normal Volunteers (반코마이신의 시간 약물동태학)

  • Carcadian rhythm dependence of vancomycin pharmacokinetics was evaluated in 5 normal volunteers receiving a single intravenous 1.0 g dose of vancomycin at 8 o'clock in the morning and another occasion at 8 o'clock in the evening in a crossover manner. The serum data were subjected to simultaneous computer nonlinear least squares regression analysis using a two-compartment pbarmacokinetic model. The mean half-life of vancomycin was $4.78\pm0.81$ hr in the morning and $4.25\pm0.51$ hr in the evening. The mean total body clearance of vancomycin was $1.29\pm0.58$ hr in the morning and $5.58\pm0.48$ hr in the evening. No circadian rhythm was found to be apparent in normal volunteers. The mean in intrasubject difference in the half-life between 8 A.M. and 8 P.M. was $15.4\%$ with fluctuations ranging from $10.4\sim33.8\%$, It is reasonable to consider individual circadian rhythm for effective dosage regimen of vancomycin in clinical chronotherapeutics.

  • In this study, flow-sediment discharge characteristics and relationships are estimated for the Cheonggyecheon basin, newly restored in 2005. Flow and sediment discharge measurement is performed on Ogansugyo, Majanggyo 2 and Yongdugyo during the rainy seasons in 2005-2006. The parameters of Clark UH are estimated using measured data, and sediment rating curves are derived by using grading and concentration analysis. Concentration time and storage coefficient of Clark UH are estimated 0.5 hr and 0.63 hr for Majanggyo 2, and 0.4 hr and 0.45 hr for Yongdugyo, respectively. Grain size of sediments are composed of fine silt to fine sand (0.008-0.25mm) based on the sediment grain size classification (Lane, 1947). Flow-sediment relationships derived by using concentration analysis shows that sediment discharge has no obvious relationship with flow discharge.

Optimization of Tri-enzyme Extraction Procedures for the Microbiological Assay of Folate in Red Kidney Bean and Roasted Peanut Using Response Surface Methodology

  • Total folate content was determined by microbiological assay using Lactobacillus casei spp. rhamnosis (ATCC 7469) with a 96-well microplate technique. Using roasted peanut and red kidney beans as representative legume samples, response surface methodology (RSM) was supplied to optimize the trienzyme procedures for the determination of folate in legumes. After response surface regression (RSREG), the second-order polynomial equation was fitted to the experimental data. Ridge analysis showed that the optimal digestion times were <2 hr for $Pronase^{(R)}$ and $\alpha$-amylase, and <5 hr for conjugase to obtain maximal folate values for legume samples. This study confirms that established digestion times for cereal products (AOAC Method 2004.05) of 3 for protease and 2 hr for $\alpha$-amylase are applicable to legumes. Conjugase treatment can be reduced to 5 from 16 hr and the conjugase level to 5 from 20 mg per sample, providing significant cost saving.

Morphological and Genetic Characterization of Caffeine-Rich and -Poor Tea Tree (Camellia sinensis L.) Lines

  • In this study, 160 tea tree (Camellia sinensis L.) lines were classified by caffeine content using colorimetric methods. Among them, caffeine-rich lines (HR-78, HR-137, HR-82 and HR-123) and poor lines (HP-85, HP-88, HP-19, and HP-131) were selected. To know the difference in morphological and genetic characters between caffeine-rich and poor lines, we used leaf/shoot growth and RAPD methods. Cluster pattern of morphological characters (leaf width, leaf length, leaf area and shoot length) showed that shoot length was longer in caffein-rich lines than in -poor lines. In genetic analysis, amplified DNA bands having various sizes were detected in RAPD analysis where 30 random primers were used. However, the discriminated primer set that distinguish caffein-rich tree line from -poor lines was not found. These results can be used as the basic data to determine the morphological and genetic differences among caffein-rich and -poor lines.

Effects of Pellet Moisture Content on the Physical Properties of Vacuum-puffed Yukwa

  • The effects of pellet moisture content on physical properties (expansion ratio, density and breaking strength) of vacuum-puffed Yukwa (non-oil puffed Yukwa) were investigated in this study. The Yukwa was made from the waxy rice steeped at 25 and $30^{\circ}C$ for 3, 5 and 10 days with pellet drying times (6, 8 and 10.5 hr). As the drying time increased from 6 to 10.5 hr at $50^{\circ}C$, the highest value of pellet moisture content (29.4%) was found in the samples made from the steeped waxy rice at $25^{\circ}C$ for 5 days after 6 hr drying, while the lowest value (16.3%) was found at $25^{\circ}C$ for 3 days after 10.5 hr drying. Both redness and yellowness values of vacuum-puffed Yukwa increased as the drying time increased. The expansion ratio of Yukwa was greatly affected by drying time, ranging from 2.07 (26.8% pellet moisture content) to 7.01 (24.0% pellet moisture content). From the data, it can be concluded that the pellet moisture content had a significant influence on the physical characteristics of vacuum-puffed Yukwa. With vacuum puffing condition of 3 min heating and 2 min puffing, the pellets with about 25% moisture content showed higher expansion ratio, and lower density and breaking strength.

Determination of flood-inducing rainfall and runoff for highly urbanized area based on high-resolution radar-gauge composite rainfall data and flooded area GIS Data

  • This study derived the Flood-Inducing-Rainfall (FIR) and the Flood-Inducing-Runoff (FIRO) from the radar-gage composite data to be used as the basis of the flood warning initiation for the urban area of Seoul. For this, we derived the rainfall depth-duration relationship for the 261 flood events at 239 watersheds during the years 2010 and 2011 based on the 10-minute 1km-1km radar-gauge composite rainfall field. The relationship was further refined by the discrete ranges of the proportion of the flooded area in the watershed (FP) and the coefficient variation of the rainfall time series (CV). Then, the slope of the straight line that contains all data points in the depth-duration relationship plot was determined as the FIR for the specified range of the FP and the CV. Similar methodology was applied to derive the FIRO, which used the runoff depths that were estimated using the NRCS Curve Number method. We found that FIR and FIRO vary at the range of 37mm/hr-63mm/hr and the range of 10mm/hr-42mm/hr, respectively. The large variability was well explained by the FP and the CV: As the FP increases, FIR and FIRO increased too, suggesting that the greater rainfall causes larger flooded area; as the rainfall CV increases, FIR and FIRO decreased, which suggests that the temporally concentrated rainfall requires less total of rainfall to cause the flood in the area. We verified our result against the 21 flood events that occurred for the period of 2012 through 2015 for the same study area. When the 5 percent of the flooded area was tolerated, the ratio of hit-and-miss of the warning system based on the rainfall was 44.2 percent and 9.5 percent, respectively. The ratio of hit-and-miss of the warning system based on the runoff was 67 percent and 4.7 percent, respectively. Lastly, we showed the importance of considering the radar-gauge composite rainfall data as well as rainfall and runoff temporal variability in flood warning system by comparing our results to the ones based on the gauge-only or radar-only rainfall data and to the one that does not account for the temporal variability.

The Weekly and Daily Energy Expenditure and Nutrition Survey on the Republic of Bores Army Cadets (육군 사관생도의 에너지소비량 및 영양섭취량에 관한 연구)

  • Determination of weekly and daily energy expenditure was made on 62 Republic of Korea Army cadets who were selected at random in order to estimate the weekly and daily ealorie expenditure. Basal metabolic rate (B.M.R.), and energy cost of various military and daily activities were measured by indirect calorimetry using open circuit method. Time-motion studies were also carried on using a stop-watch. The total weekly energy expenditure was calculated by summation of data using energy cost per minute, and the time spent on each activity. Determination of daily energy expenditure was deduced from each data of weekly energy expenditure. Food survey was also carried on for a week, and daily calorie intake was determined by a weekly average discounting loss in cooking. All measurements were determined from the Standard Table of Food Composition published by the Ministry of National Defense (1961). Following data were observed. 1. Physical status of cadets are as follows. Please note that the height and weight averages are 1-2cm and 4-5kg respectively over that of the Seoul National University students. First year Height 167.92 cm $(S.D.{\pm}4.09)$ Weight 61.72 kg $(S.D.{\pm}4.53)$ Second year Height 167.89 cm $(S.D.{\pm}3.46)$ Weight 63.01 kg $(S.D.{\pm}4.61)$ Third year Height 168.15 cm $(S.D.{\pm}4.24)$ Weight 43.48 kg $(S.D.{\pm}5.03)$ Fourth year Height 168.10 cm $(S.D.{\pm}3.70)$ Weight 64.02kg $(S.D.{\pm}5.10)$ 2. The B.M.R. of cadets averaged $36.57\;Cal./m^2/hr.(S.D.{\pm}3.63\;Cal./m^2/hr.)$ is almost equal with data on the same ages of civilians and the Japanese, but a lower average of $5.1\;Cal./m^2/hr.$ than that of a common soldier. 3. The energy expenditure during various military activities is close agreement with Consolazio. Passmore and Durnin, and Japanese reports.

Performance Evaluation of Rainfall Disaggregation according to Temporal Scale of Rainfall Data (강우자료의 시간해상도에 따른 강우 분해 성능 평가)

  • In this study, rainfall data with various temporal scales (3-, 6-, 12-, 24-hr) are disaggregated into 1-hourly rainfall data to evaluate the performance of rainfall disaggregation technique. The rainfall disaggregation technique is based on a database generated by the stochastic point rainfall model, the Neyman-Scott Rectangular Pulse Model (NSRPM). Performance evaluation is carried out using July rainfall data of Ulsan, Changwon, Busan and Milyang weather stations in Korea. As a result, the rainfall disaggregation technique showed excellent performance that can consider not only the major statistics of rainfall but also the spatial correlation. It also indirectly shows the uncertainty of future climate change scenarios with daily temporal scale. The rainfall disaggregation technique is expected to disaggregate the future climate change scenarios, and to be effective in the future watershed management.

A Relationship of Constitution Type, Lifestyle Status and Metabolic Syndrome Incidence in Korean Adults (우리나라 성인의 사상체질과 생활습관 상태에 따른 대사증후군 발생 위험 상관성)

  • Objectives We aimed to identify the incidence of metabolic syndrome (MetS) and its clustering components according to constitution type and lifestyle risk factors in Korean adults. Methods This study included 1,978 adults aged 30-55 years from the Korean Medicine Daejeon Citizen Cohort (KDCC) study. We defined lifestyle factors including smoking, alcohol consumption, physical activity, sleep, dietary quality, and weight status. Total lifestyle scores were created based on the six lifestyle factors (ranging from 0 to 5 factors) and classified into two groups: unhealthy (0-2 factors), or healthy (3-5 factors). Cox proportional hazard regression was used to estimate the hazard ratio (HR) and 95% confidence intervals (CIs) of primary endpoints: MetS events and their clustering components. Results During a median follow-up of 2.2 years, we documented 125 new onsets of MetS. Compared with participants with healthy, the HR of unhealthy participants was 2.401 (95% CI: 1.497-3.851) for MetS incidence. After adjusting for covariates, TE type with unhealthy was higher HR values of abdominal obesity (HRs: 1.499, 95%CI: 1.061-2.117) and hypertension (HRs: 1.840, 95%CI: 1.032-3.277), respectively. Conclusion Unfavorable lifestyle factors were highly associated with the prevalence of MetS and its clustering such as abdominal obesity and hypertension in Korean adults with TE. Tailored health management is needed to consider individual traits and healthy lifestyles to prevent cardiometabolic diseases.

Analysis of Extreme Rainfall Distribution Scenarios over the Landslide High Risk Zones in Urban Areas (도심지 토사재해 고위험지역 극치강우 시간분포 시나리오 분석)

  • In this study, we analyzed the extreme rainfall distribution scenarios based on probable rainfall calculation and applying various time distribution models over the landslide high risk zones in urban areas. We used observed rainfall data form total 71 ASOS (Automated Synoptic Observing System) station and AWS (Automatic Weather Station) in KMA (Korea Meteorological Administration), and we analyzed the linear trends for 1-hr and 24-hr annual maximum rainfall series using simple linear regression method, which are identified their increasing trends with slopes of 0.035 and 0.660 during 1961-2014, respectively. The Gumbel distribution was applied to obtain the return period and probability precipitation for each duration. The IDF (Intensity-Duration-Frequency) curves for landslide high risk zones were derived by applying integrated probability precipitation intensity equation. Results from IDF analysis indicate that the probability precipitation varies from 31.4~38.3 % for 1 hr duration, and 33.0~47.9 % for 24 hr duration. It also showed different results for each area. The $Huff-4^{th}$ Quartile method as well as Mononobe distribution were selected as the rainfall distribution scenarios of landslide high risk zones. The results of this study can be used to provide boundary conditions for slope collapse analysis, to analyze sediment disaster risk, and to use as input data for risk prediction of debris flow.
