ASE 및 SPE 복합정제법을 이용한 친환경농업토양의 다성분잔류농약 분석 (Pesticide Multiresidues Analysis of Environmental-friendly Agricultural Soils by the Complex Cleanup Method of Accelerated Solvent Extraction (ASE) and Solid Phase Extraction (SPE))

  • 국내의 친환경농산물과 토양에서 자주 검출되는 농약 50성분을 선정하고, 경남지역의 친환경농산물 재배농지의 토양시료 40점을 대상으로, 가속용매추출법(accelerated solvent extraction)과 고상추출법(solid-phase extraction)의 복합처리법으로 정제하고 GC/ECD/NPD, HPLC/UV/FL, GC/MSD 및 HPLC/MSD를 이용한 분석법을 적용하여 잔류농약의 잔류실태를 조사하였다. 적용한 분석법에 의한 농약 회수율은 72~118%의 범위로 평균 95.5%이었고, CV(%)값 평균은 3.0%로 나타났다. 토양 40점 중 잔류농약 검출은 21점에서 20성분이 검출되었으며 검출토양의 농약별평균 검출량은 endosulfan 0.035, ethoprophos 0.043, chlorpyrifos 0.020, chlorfenapyr 0.023, flufenoxuron 0.047, fenvalerate 0.070, cypermethrin 0.266, lufenuron 0.016, bifenthrin 0.022, fenobucarb/BPMC 0.025, difenoconazole 0.043, fenarimol 0.059, kresoxim-methyl 0.020, tetraconazole 0.026, isoprothiolane 0.039, iprobenfos 0.017, nuarimol 0.014, fluquinconazole 0.156, tebuconazole 0.047 및 oxadiazon이 0.045 mg/kg으로 나타나서 친환경재배농지의 토양환경기준에 잔류농약의 설정이 시급하다고 판단되었다.

서울시 유통 건조농산물 중의 농약잔류 실태 연구 (A Study of Current Status on Pesticide Residues in Commercial Dried Agricultural Products)

  • 서울지역에 유통되고 있는 국내산 및 수입산 건조농산물 58건을 대상으로 다성분 동시분석방법에 따라 GC-NPD, $GC-{\mu}ECD$, HPLC-UV와 HPLC-FLD로 분석가능한 총 253종의 농약잔류량을 분석한 후 GC-MSD로 확인한 결과 총 14건의 건조농산물에서 잔류농약이 검출되어 24.1%의 검출률을 나타내었고, 이 중 건조 채소류의 검출률은 25.5%였다. 품목별 잔류농약의 검출률은 건조과채류, 건조엽채류, 건조엽경채류 순으로 높았으며, 특히 건조 과채류 중 건조고추의 잔류농약 분포가 다양하고 검출빈도가 상당히 높았다. 건조 농산물의 기준에 없는 12종의 잔류농약이 검출되었으며, 합성피레스로이드계, 유기염소계, 유기인계 순으로 검출빈도가 높았고, 특히 합성피레스로이드계 중 cypermethrin의 검출 횟수가 가장 많았다. 또한 대상목적에 따라 분류하면, 살충제, 살균제, 제초제, 살선충제 순서로 검출건수가 많았다. 산지별로 보면, 국내산, 중국산 ,북한산, 미국산, 베트남산의 순서로 잔류농약의 검출건수가 많았으며, 국내산은 과채류에서 수입산은 엽경채류에서 잔류농약의 검출빈도가 높았다.

Charateristics of Voltage Dependent Calcium Uptake and Norepinephrine Release in Hypothalamus of DOCA-salt Hypertensive Rats

  • Purpose of the present study was to clarify the role of noradrenergic neural activities in hypothalamus for either triggering or maintaining hypertension in deoxycorticosterone (DOCA)-salt hypertensive rats. Two groups of animals were prepared: 1) normotensive Wistar rats and 2) DOCA-salt induced hypertensive rats. Voltage dependent $^{45}Ca^{++}$ uptake, endogenous norepinephrine release, and the catecholamine content in the hypothalamus of DOCA-salt hypertensive and normotensive Wistar rats were compared. Animals at 4, 6 and 16 week-old of two groups were sacrificed by decapitation and hypothalamus was dissected out. Voltage dependent calcium uptake and norepinephrine release were determined from hypothalamic synaptosomes either in low potassium or high potassium stimulatory condition by using $^{45}Ca^{++}$ isotope and HPLC-ECD technique. Degrees of voltage dependent $^{45}Ca^{++}$ uptake and norepinephrine release in hypothalamic synaptosomes of 16-week-old DOCA-salt hypertensive rats were significantly greater than those of age matched normotensive control rats. The norepinephrine and dopamine contents of hypothalamus were about the same in two groups of animals. These results suggest that the alteration of evoked norepinephrine release related to calcium uptake in hypothalamus may play a role in the maintenance of hypertension in DOCA-salt hypertensive rats.

Relationship between the Changes of Catecholamines and Blood Pressure Induced by Exposure to Low- and High-levels of Lead in Rats

  • In this study, it was tested whether the changes of catecholamines and its metabolites are related with the changes of blood pressure(BP) induced by different levels of lead exposure. Adult male SD rats were exposed to lead by giving drinking water containing 50(low doses), 200 and 1,000 ppm(high doses) of lead(as lead acetate) or sodium acetate(for control groups, supplying an identical amount of acetate) for 7 or 16 weeks. The systolic BP was measured in the unanesthetized state by the tail-cuff technique. Levels of catecholamines and its metabolites in urine were measured by HPLC-ECD. Rats receiving 200 and 1,000 ppm developed an elevation of systolic BP at 3 and 7 weeks compared with week 0, but blood pressure levels at 16 weeks returned to normal. For the 50 ppm lead treated group, systolic BP increased significantly at 7 weeks and 16 weeks. The concentrations of norepinephrine and VMA in the urine of lead exposed rats changed similarly to the changes of blood pressure, but blood viscosity levels in all lead treated rats increased continuously during all lead treatment periods. This result suggests that the changes of catecholamines and its metabolites in urine by lead intoxication may influence the changes of blood pressure.

식품 중의 유기인제, 유기염소계 및 카바메이트계 농약의 다성분 분석법 비교 (A Study on the Multiresidue Analytical Methods for Organophosphorus, Organochlorine, and n-Methyl carbamate Pesticides in Food)

  • A multiresidue method(MRM) for pesticides must be rapid and must test a wide variety of pesticides at relevant toxicological concentrations. In this study, three kinds of major analytical methods such as Korean Food Code Method, CDFA MRM, and Holstege's method are tested to compare the average recoveries, solvent consumption, and required time for the analysis of 18 organochlorines, 18 organophosphates, and 6 carbamates in spinach samples. Samples for pesticides analysis were extracted and cleaned up according to the respective methods and detected by gas chromatography with selective detectors, ECD and NPD, HPLC with postcolumn reaction system(PCRS). Average recovery of 42 pesticides by Korean Food Code method, CDFA method, and Holstege's method were 91.3%, 88.1%, 89.0%, respectively. Amount of solvent consumption and required time for the analysis of Korean Food Code method were from two and a half times to three times as much as those of another two methods. For the development and legal application of more rapid and effective MRMs, prolonged study is necessary.

The Effects of Lead Exposure on Glutamatergic Nervous System in Rat Cerebellar Cells

  • Changes in glutamate release and uptake on cerebellar cells after the chronic exposure to lead were investigated. Rats were received 0.25% lead acetate in drinking water from the beginning of the pregnancy. The control group was given 0.125% sodium acetate in drinking water. The cerebellar cells from 7 or 8 day-old pups were cultured. Amino acid release from cerebellar granule cells and the glutamate uptake into cerebellar glial cells were measured using HPLC-ECD. Basal glutamate release and NMDA-induced glutamate release didn't show significant difference. However, the other amino acids in the granule cells obtained from lead exposed pups were less released than the control after the stimulation by NMDA (50$\mu$M). SNAP-induced (50$\mu$M) glutamate release was significantly reduced in granule cells prepared from lead exposed pups. The basal glutamate uptake in glial cells didn't show any difference. However, the uptake in glial cells prepared from lead exposed pups was significantly less blocked by PDC (24$\mu$M) compared to the control group. These results indicate that lead exposure to the mother might affect the Excitatory amino acid system during the development of the offspring.

  • Lithium, a simple monovalent cation, is the mainstay in the treatment of manic-depressive illness, but despite extensive research, its mechanism of action remains to be elucidated. Because lithium requires chronic administration for therapeutic efficacy and because its beneficial effects last well beyond its discontinuation, it has been postulated that lithium may exert major effects at the genomic level. We have previously shown that Yungmijihwangwon (YM) increase TH protein expression through antioxidant activity and unknown effects in vivo. In the pressent study, we have sought to determine if lithium in herbal medicine also increases the expression of endogenous gene known to be regulated by AP-1 and have therefore investigated the effects of herbal medicine on tyrosine hydroxylase (TH) levels. Male mice were treated with LiCl and herbal medicine for 30 days, and TH levels and dopamine level were measured in striatum using immunoblotting and ECD-HPLC. Herbal medicine treatment resulted in statistically nonsignificant increase in TH levels in mouse striatum. The precise therapeutic relevance of these effects is presently unknown.

대전 지역 유통 식용 한약재의 잔류농약 실태 연구 (A Study on the Pesticide Residues Monitoring of Medicinal Herbs which has marketed in the Daejeon)

  • This study was conducted to investigate the residue amount of pesticide on the 41 medicinal herbs in Daejeon area. This study was carried out to monitor the current status of pesticide residues in commercial medicinal herbs for sale of food use in 2012. It was performed using GC/ECD, GC/NPD, HPLC to analyze pesticides residues. Residues of 283 pesticides were analyzed by a simultaneous multiresidue method in 41 medicinal herbs being on sale in Daejeon. The medicinal herbs detected pesticides in 10 of 41 cases, showed a detection rate of 24.39%. The medicinal herbs which exceed the maximum residue limit were five cases as Cnidii Rhizoma, Osterici Radix, Artemisiae Capillaris Herba, Zizyphi Fructus and Alismatis Rhizoma. And pesticide residue of Cnidii Rhizoma and Alismatis Rhizoma exceeds the limit standard presented in only medicine use of KFDA. The residual pesticides which had the high detection rate were Chlopyrifos, Tebuconazole and Endosulfan in the detection of medicinal herbs. For further research, standards of Pesticide Residues in medicinal herbs should be added and more research of pesticide residues in medicinal herbs required. And standards of pesticide residues in medicinal herbs should be applied equally as medicines and food.

Characteristics of Voltage-Dependent Clacium Uptake and Norepinephrine Release in Hypothalamus of SHR

  • The characteristics of voltage-dependent ^{45}Calcium$ uptake and norepinephrine release as factors controlling neural activities in the hypothalamus which is an important regulatory site for cardiovascular function wre studied. Two groups of animals : male spontaneously hyperterisive rat (SHR) and age-matched nomotensive wistar rat (NW) were used in this study. Animals at 4, 6 and 16 weeks of age were sacrificed by decapitiation and the hypothalamus was dissected out. Voltage-dependent calcium uptake and norepinephrine release were determined from hypothalamic synaptosomes either in low potassium (5 mM) or high potassium (41 mM) stimulatory conditions by using ^{45}Ca$ isotope and HPLC-ECD techniques. Degrees of voltage-dependent ^{45}Calcium$ uptake and norepinephrine release evoked by calcium uptake in the hypothalamus of prehypertensive phase (4 weeks old) of SHR were significantly smaller than those in NW of the same age. However, in the developmental phase (6 weeks old) and the established phase (16 weeks old) of hyperrtension in SHR, degrees of voltage-dependent ^{45}Calcium$ uptake and norepinephrine release were similar to those of age-matched normotensive wistae eats. These data imply that the deficit in hypothalamic norepinephrine release might be an important underlying factor for the development of hypertension in SHR.

Extremely Low Frequency Magnetic Fields Modulate Bicuculline-Induced-Convulsion in Rats

  • The effect of extremely low frequency (ELF,60Hz) magnetic fields (MFs) on convulsions was investigated in rats. We determined the onset arid duration of convulsions induced by bicuculline alone or by co-exposure to MFs and bicuculline. In addition, we measured the GABA concentrations in the rat brains using HPLC-ECD. MFs strengthened the convulsion induced by bicuculline (0.3, 1, and 3${\mu}g$, I.c.v.), with a shortening of the onset time, but lengthening of the duration time. Co-exposure to MFs and bicuculline decreased the GABA levels in the cortex, hippocampus and hypothalamus, whereas MFs alone reduced the level of GABA only in the hippocampus. These results suggest that the exposure to MFs may modulate bicuculline-induced convulsions due to GABA neurotransmissions in rat brains.
