Simultaneous Determination of Avicularin and Quercitrin in Astilbe chinensis by HPLC

  • This study developed a HPLC analysis method for the determination of avicularin (AL) and quercitrin (QT) in Astilbe chinensis by HPLC. HPLC was performed on a Capcell Pak C18 MGII column ($4.6{\times}250mm$, $5{\mu}m$) with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and acetonitrile at a flow rate of 1.0 mL/min at $30^{\circ}C$ and a UV detection wavelength of 254 nm. The analytical method was validated for its specificity, precision, accuracy, and linearity. The limits of detection and quantitation were AL 0.09 and 0.28 mg/mL, QT 0.03 and 0.09 mg/mL respectively. Calibration curves showed good linearity (AL r2 > 0.99990 and QT r2 > 0.99994), and the precision of analysis was satisfied (less than AL 0.59%, QT 0.63%). Recoveries of quantified compounds ranged from AL 100.57 to 102.08%, QT 99.94 to 100.64%. These results indicate that the developed HPLC method was applied successfully to the detection of AL and QT in A. chinensis.

Quantitative Analyses for the Quality Evaluation of Salviae Miltiorrhizae Radix by HPLC

  • In this study, quantitative analysis for the quality evaluation of Salviae Miltiorrhizae Radix using HPLC/UV was developed. For quantitative analysis, six major bioactive compounds were determined. The separation conditions employed for HPLC/UV were optimized using ODS $C_{18}$ column ($250{\times}4.6\;mm$, $5\;{\mu}m$) with gradient condition of A (1% formic acid in $H_2O$) and B (acetonitrile : methanol : formic acid = 100 : 75 : 1) as the mobile phase at a flow rate of 1.0 mL/min and a detection wavelength of 280 nm. These methods were fully validated with respect to the linearity, accuracy, precision and recovery. The HPLC/UV method was applied successfully to the quantification of six major compounds in the Salviae Miltiorrhizae Radix. The results indicate that the established HPLC/UV method is suitable for the quantitative analysis.

누에에 함유된 1-Deoxynojirimycin의 분석을 위한 HPLC-ELSD 분석법 밸리데이션 (Development and Validation of a Unique HPLC-ELSD Method for Analysis of 1-Deoxynojirimycin Derived from Silkworms)

  • A simple and accurate assay was developed for the quantitative analysis of 1-deoxynojirimycin (1-DNJ) derived from the silkworm (Bombyx mori). Normal-phase high-performance liquid chromatography coupled with an evaporative light scattering detector (HPLC-ELSD) and a hydrophilic interaction liquid chromatography column was used. Various parameters were applied to optimize the analysis method. The limits of detection and quantification of 1-DNJ were 2.97 × 10-3 and 9.00 × 10-3 mg/mL, respectively. The calibration curve showed good linearity results. The concentration range and the r2 value were 0.0625-1.0 mg/mL and 0.9997, respectively. The accuracy test demonstrated a significantly high recovery rate (89.95-103.22%). The relative standard deviation was ≤ 1.00%. Thus, a method for the accurate identification and quantitative analysis of 1-DNJ in silkworms was developed. Moreover, in this procedure, the process of derivatization of 1-DNJ, which was required in previous experiments, could be eliminated. This technique may be actively utilized for the development of pharmaceuticals and health functional foods using 1-DNJ.

A New HPLC Condition for the Analysis of Aclacinomycins A and Y in the Mixtures of Aclacinomycins

  • As the previous HPLC method (Ogasawara et al., 1983) was described for the analysis of aclacinomycin A and some of its analogue compounds but not for aclacinomycin Y, we developed novel HPLC condition by optimizing solvent system. The newly developed solvent system allowed a complete separation of aclacinomycins Y and A, as opposed to the incomplete resolution of these two compounds in conventional method. The amounts of aclacinomycins Y and A could be accurately determined when the fermentation broth of Streptomyces lavendofoliae DKRS was analyzed by the newly developed method.

  • Sulfonamides, a therapeutically important group of antimicrobial drugs, are widely used to treat and prevent the acute systemic and skin infections in dairy cattle. They also pose an economic hazard through inhibition of growth of dairy starter cultures. This study was carried out to compare four screening methods for detection of sulfamethazine in milk and determine the positive milk sample by HPLC method. Sulfamethazine was used to spike at five levels of sulfamethazine. The Lac-Tek test and CharmII test were also consistent better than TTCII test and Delvo SP test in sulfamethazine detection. Analysis probabilities of obtaining a positive response with TTCII test and Delvo SP test assay at 50 ppb sulfamethazine level in milk samples were only 14%, 42% each. Whereas using the Lac-Tek test and CharmII test would have resulted in 100% identification of the five levels. Determination of sufamethazine using the HPLC method in the spiked milk were 10.64, 19.30, 30.76,38.83 and 50.23 ppb, respectively.

  • Carbohydrate analysis is important in studying structure and activity of complex polysaccharides. New analytical method was applied to get an information on the composition of polysaccharides showing antitumor activity. Monosaccharides were labeled with 7-amino-1, 3-naph-thalenedisulfonic acid (7-AGA) by reductive amination and separated by HPLC. Five kinds of polysaccharides from Basidiomycetes were hydrolyzed and analyzed in combination with electrophresis and HPLC. At the same time, alditol acetate derivatives were prepared and analyzed by gas chromatography. Two different techniques using different derivatization methods showed very similar results. The monosaccharides from Coriolus versicolor and Cordyceps militaris were glucose and galactose. Phellinus linteus composed of glucose, glactose, mannose, arabinose and fucose. The HPLC method with fluorescence detector was very sensitive compared to other methods.

Chiral Drugs의 광학분할을 위한 HPLC Column의 응용 (The Application of Chiral HPLC Columns for Enantiomer Separation of Chiral Drugs)

  • In terms of chiral issue, two enantiomers of chiral drugs often differ significantly in their pharmacological, toxicological and pharmacokinetic profile. Chiral switches of racemic drugs have been redeveloped as single enantiomers. Several chiral resolution techniques in chirotechnology are introduced and the most used chiral HPLC chromatographic method among several chiral analysis techniques is described with its several advantages. Several types of chiral HPLC columns derived from their chiral selectors are discussed with their property and applications for enantiomer separation.

추출 용매와 분석 기법에 따른 된장의 유리아미노산 비교 (Comparison of Free Amino Acids in Soybean Paste (Doenjang) by Different Extraction Solvents and Analytical Methods)

  • This work was conducted to obtain a rapid, accurate, and precise procedure for free amino acids analysis in Doenjang with HPLC-OPA (high performance liquid chromatography using-phthalaldehyde) and AAA (automatic amino acid analyzer) methods. Different sample extraction procedures among water, 0.1 M perchloric acid, and 0.1% meta-phosphoric acid were also compared. The optimal extraction solvent was 0.1% meta-phosphoric acid for both the HPLC-OPA and AAA methods. Good recoveries for glycine and methionine were observed using the 0.1% meta-phosphoric acid extraction with HPLC-OPA method. Method precisions (% relative standard deviation) for the free amino acids ranged for 1.62% to 8.27%, in which the HPLC-OPA method with water extraction showed the lowest value at 1.62%. Inhibition rates of the free amino acids in Doenjang were greatest with an addition of NaCI at a 1% concentration.

Quantitative HPLC Analysis and Extraction of 2,6-dimethoxy-1,4-benzoquinone from Ficus foveolata Stems

  • An antibacterial benzoquinone, 2,6-dimethoxy-1,4-benzoquinone, isolated from Ficus foveolata stems was used as a standard marker for establishment of quantitative HPLC analysis for the stem extracts of F. foveolata. The method utilized a TSK-gel ODS-80Ts column (5 μm, 4.6 × 250 mm) with the mixture of methanol and 5% acetic acid in water (24:76, v/v) as the mobile phase at a flow rate of 1 mL/min, and quantitative detection at 289 nm. The parameters i.e. linearity, intraday and interday precision, accuracy, specificity and sensitivity of the method were evaluated for method validation. The recoveries of the method were 99.5 - 103.6% and good linearity (R2 ≥ 0.9999) was obtained. A high degree of specificity, sensitivity as well as repeatability and reproducibility (RSD less than 2 and 5%, respectively) were also achieved. Chloroform was served as the most suitable solvent for extraction of 2,6-dimethoxy-1,4-benzoquinone. The optimised sample preparation and HPLC method can be practically used in the routine quality control process of F. foveolata stem extracts.

Recent Developments in High-performance Liquid Chromatography of Lipids

  • The possibilities for HPLC analysis of lipids have been revolutionised by the availability of evaporative light-scattering detectors, with which the response is independent of the nature of the mobile phase and does not depend On the presence of specific chromophores in the lipids. It was thus possible to develop an HPLC procedure, involving ternary gradient elution, for separating all the lipid classes in animal tissues in a single step. Although reversed-phase HPLC has been widely used for the analysis of molecular species of lipids, sliver ion chromatography can be a valuable alternative. For example, a stable silver ion column for HPLC was developed which permitted resolution of molecular species of triacylglycerols, even from such complex samples as fish oils, again With light-scattering detection and gradient elution. The capacity for HPLC resolution of diastereomeric diacyl-sn-glycerol derivatives, prepared from triacylglycerols. has lead to a new simple method for stereospecific analysis of the latter.
