High performance liquid chromatographic separation is described for the analysis of bile acids after hydrolysis in seven commercial crude bile drugs and ox and pig galls. They are simultaneously separated with HPLC mobile phase of acetonitrile/0.5% ammonium carbonate (pH 6.7) (25.5 : 74.5) at a flow rate $(1.0{\rightarrow}1.5ml/min.)$ and differential refractometer. The linearity of calibration curve and recovery test are good by using the method. The analysis of major bile acids in seven commercial crude bile drugs using the described method is presented. Sample no. 1 of them is similar to separation pattern of ox gall. Sample no. 6 of them is supposed to be genuine bear gall on the basis of identification of ursodeoxycholic acid. Sample $no.\;2{\sim}5$ and 7 of them are supposed to be pig gall on the basis of identification of hyodeoxycholic acid which is a characteristic component of pig gall.
Six total mixed rations (TMR) containing 0, 4, 6, 8, 10, 12% tannin (TMR I-VI), using Accacia nilotica pods as a source of tannin, were used to study the effect of Acacia tannin on in vitro nutrient digestibility and gas production in goats. This study also investigated the degraded products of Acacia nilotica tannin in goat rumen liquor. Degraded products of tannins were identified using high performance liquid chromatography (HPLC) at different hours of incubation. In vitro digestibility of dry matter (IVDMD) and organic matter (IVOMD) were similar in TMR II, and I, but declined (p<0.05) thereafter to a stable pattern until the concentration of tannin was raised to 10%. In vitro crude protein digestibility (IVCPD) decreased (p<0.05) with increased levels of tannins in the total mixed rations. Crude protein digestibility was much more affected than digestibility of dry matter and organic matter. In vitro gas production (IVGP) was also reduced (p<0.05) with increased levels of tannins in the TMR during the first 24 h of incubation and tended to increase (p>0.05) during 24-48 h of incubation. Gallic acid, phloroglucinol, resorcinol and catechin were identified at different hours of incubation. Phloroglucinol and catechin were the major end products of tannin degradation while gallate and resorcinol were produced in traces. It is inferred that in vitro nutrient digestibility was reduced by metabolites of Acacia nilotica tannins and ruminal microbes of goat were capable of withstanding up to 4% tannin of Acacia nilotica pods in the TMR without affecting in vitro nutrient digestibility.
Triglyceride compositions of cottonseed, soybean, sesame, perilla corn and rapeseed oils have been determined by a high performance liquid chromatographic analysis. An optimum condition was obtained by using a ALC/GPC 244 type, from Waters Association, Japan with $\mu$ Bondapak $C_{18}(1/4^{'}\times1^{'})$ column. A similar distribution pattern of triglycerides was found in cottonseed, soybean, sesame, rapeseed and corn oils. It was noted that $C_{40}$, $C_{42}$ and $C_{44}$ were the major components in these seed oils, except perilla oil. The results showed that contents $O_{40}-C_{48}$ triglyceride types in cottonseed, sesame and corn oil were within $2.23{\sim}41.24%$ and $C_{38}-C_{48}$ triglyceride types in soybean oil were within $3.01{\sim}10.02%$ and $C_{34}-C_{46}$ triglyceride types in rapeseed oil were within $2.38{\sim}28.68%$.
Comparative effects of electron beam and gamma-ray irradiation (25 ∼15 kGy) were investigated on saponin stability and some physiological and chemical properties of white and red ginseng powders. Saponin components were found stable upon irradiation of both energies when determined by TLC and HPLC, after 4 months of storage at room temperature as well as immediately after treatment The contents of total phenolics and acidic polysaccharides of the samples were higher in red ginseng than in white. Polysaccharide contents increased with irradiation doses. Amylase activity of white ginseng was stimulated by irradiation but decreased with the lapse of storage time. There were no apparent differences in electrophoresis patterns of extracted proteins depending on irradiation doses and energy sources.
Objectives: To determine the standards for discrimination of Magnoliae Cortex, the experiment of specific external-internal characters and the physicochemical pattern analysis were performed. Methods: External characteristics was observed using a stereoscope. Paraffin-mediated sectioned materials were stained by Ju's method. Physicochemical patterns of materials were analyzed using HPLC. Results: 1. Botanical characteristics: Magnolia officinalis had one seed and a white flower, while M. obovata had two seeds and a white flower. Machilus thunbergii had berry and spherical fruits and yellowish green panicles. 2. External characteristics: M. officinalis and M obovata were dark and thick. M. officinalis was gray brown and greasy while M. obovata was light-gray, less oily and smoothly sectioned. Machilus thunbergii was thin and relatively light or yellow-brown, coarsely sectioned and faintly specific scents. 3. Internal characteristics: The bast parts of M. officinalis and M. obovata were commonly wider than Machilus thunbergii The cork cortex of M. officinalis was $10{\sim}mg/L$ cell layers with many oil cells, while that of M. obovata was $4{\sim}7$ cell layers with less oil cells. Machilus thunbergii's xylem which consisted of ring-shaped cambium at 1st and 2nd part was occupied in large portion. 4. Physicochemical pattern: Both M. officinalis and M. obovata involved honokiol and magnolol. All kinds of M. officinalis involved Magnatriol B but one kind of M. ovobata and all of Machilus thunbergii didn't. Machilus thunbergii showed different pattern of chromatogram from that of 2 species above. Conclusions: These results could be used as standards for discrimination of Magnoliae Cortex and as the method of objectification in medicinal herbs giving the basic resource for bioactivity research.

Objectives : To determine the discriminative criteria for Atractylodes rhizomes, the experiment of externalinternal characteristics and physicochemical pattern analysis were performed. Methods : External characteristics was observed using stereoscope. The sectioned materials which were covered with parffin were stained by Ju's method. Physicochemical patterns were analyzed using HPLC/DAD. Results : 1. External shape of original plant : Atractylodes maaocephala and A. japonica had relatively long petioles and 3-5 parted leaves. A. macrocephala had big purple flowers whereas A. japonica had relatively small white flowers and pinnate bracts. A. lancea had sessile leaves and white flowers, and the end parts of degenerated stamens were bent. 2. External shape of herbal medicine: A. macrocephala which was fist-shaped rhizome had pa-pillate processes and the cross section was light gray and sulcate. A. japonica and A. lancea were connected-beady or tubercular rhizomes, and the cross sections were both yellow-colored white. However, the cross section of A. japonica was fibrous, the width of cortex was narro-wer than that of stele, and radial shape in cortex was rare, whereas the width of cortex in A. lancea was similar to that of stele in size, and radial shape in cortex was obvious. 3. Internal shape of herbal medicine: A. macrocephala and A. lancea did not have lignified fascicles in cortex. However, the vascular bundles and vessels of A. macrocephala were wedge shaped and radial arrangement, and vascular bundles were densely populated in stele whereas those of A. lancea were repeatedly arranged and thinly extended to cortex. A. japonica had lignified fascicles in cortex and the width of vascular bundles was conspicuously thick with narrow intervals. 4. Physicochemical pattern analysis : A. macrocephala and A. lancea contained atractylenolide I and atractylenolide Ill whereas A. japonica contained atractylenolide I, atractylenolide Ill. diacetyl-atractylodiol, compound-4, compound-5. The three species of Atractylodes rhizomes showed different chromatogram patterns. Conclusions : The results could be used as discriminative criteria for Atractylodes rhizomes and as fundamental materials to researches of further pattern analysis and biological reaction.
Tandem HPLC and atmospheric pressure chemical ionization(APcI) LC/MS method was used for the determination and confirmation of carbendazim residues in soybean sprout. Fluorescence(FL) detector was connected in tandem with the ultraviolet(UV) detector for dual detection of the carbendazim residue at the excitation and emission wavelength of 280 nm and 310 nm, respectively. The limit of detection for carbendazim was $0.1{\mu}g/kg$ sample. Recoveries of carbendazim from fortified soybean sprout at 0.5, 1.0 and 2.0 ppm were averaged 89.1%. Mass spectrometry using a APcI source confirmed the carbendazim residue in the soybean sprout sample. Fragmentation pattern on the APcI LC/MS spectrum of carbendazim was simpler than that from electron impact(EI) mass spectrum. Carbendazim produced 3 major ions including m/z 133, m/z 159 and m/z 191($M^{+}$). This method was sensitive enough to provide reliable and reproducible results for practical applications.
Cortex Phellodendri (CP) is derived from the dried bark of Phellodendron amurense. It has been widely used as a drug in traditional Korea medicine for treating diarrhea, jaundice, swelling pains in the knees and feet, urinary tract infections, and infections of the body surface. Many analytical methods have been used to study oriental herbal medicines, such as thin-layer chromatography, column liquid chromatography, and high performance liquid chromatography (HPLC). In this study, preparative centrifugal partition chromatography (CPC) was successfully carried out in order to separate pure compounds from a CP methanol extract. The optimum two-phase CPC solvent system was composed of n-butanol: acetic acid: water (4:1:5 v/v/v). The flow rate of the mobile phase was 3 mL/min in ascending mode with rotation at 1,000 rpm. The CPC-separated fraction and purification procedures were carried out by preparatory HPLC. The $^1H$ NMR spectrum revealed that the resonances at ${\delta}$ 4.10 and 4.20 ppm corresponded to three protons ($-OCH_3$), whereas those at ${\delta}$ 6.10 ppm corresponded to two protons ($-OCH_2O-$). Further, two aromatic protons (H-11 and H-12) conveys a doublet-doublet pattern. The H-11 doublet and H-12 doublet appear at ${\delta}$ 7.98 and 8.11, respectively. The $^{13}C$ NMR. spectrum showed a tetrasubstituted with a methylenedioxy group at C2 and C3, and two methoxy groups at C9 and C10. The chemical structure of the berberine was identified by $^1H$, $^{13}C$-nuclear magnetic resonance and electrospray ionization-mass spectroscopy spectral data analysis.
The new anti-impotency analogue was identified in food source. Detection of this analogue was accomplished through screening of food samples by liquid chromatography/photodiode array detector. The spectrum pattern of analogue compound was similar to that observed for hongdenafil which was analogue of sildenafil. This new compound was isolated and purified using the liquid-liquid extraction, thin layer chromatography, column chromatography and preparative HPLC. And then those structure were identified using analytical instruments such as HPLC/PDA, LC/MS/MS and NMR. The compound was given a name to oxohongdenafil which was replaced with acetyl oxoethylpiperazinyl residue instead of sulfonyl piperazine group of sildenafil. The regulation for the abovementioned analogue, oxohongdenafil, was established by Standard of Korean food code.

The changes of feeding pattern and the amount of insecticide penetrated into the leaf were monitored for 420 min after treatment of three concentrations of dinotefuran. At about 30 min after treatment of insecticide, the overall feeding pattern in EPG (Electrical Penetration Graph) monitoring was started to change, and there was significant difference in proportions of NP (non-penetration), PA (pathway activity), and phloem-feeding patterns among the different concentrations of dinotefuran treatment. Especially, as the amount of insecticide penetrated into a leaf increased, the reactive behaviors against this insecticide such as withdrawal of proboscis and movements of stylet of aphid were more quickly exhibited. And also, total time at which the proboscis of the aphids did not penetrate the plant was getting longer. The amount of dinotefuran penetrated into a leaf was monitored with HPLC. When the recommended concentration (100 mg/L) of the insecticide was treated, 5.24${\sim}$7.24 mg/L of the insecticide was detected from the leaf, and the proboscis of the aphids was apparently withdrawn from the leaf at approximately 60 minutes after treatment of this insecticide.
