Isolation of a starfish myorelaxant peptide (SMP) isotype from the pyloric caeca of Patiria pectinifera

  • Peptides are naturally occurring biological molecules that are found in all living organisms. These biologically active peptides play a key role in various biological processes. The aim of this study is the extraction and the purification of bioactive materials that induce relaxation of an apical muscle from the pyloric caeca of Patiria pectinifera. The acidified pyloric caeca extract was partially separated by the solid phase extraction using a stepwise gradient on Sep-Pak C18 cartridge. Among the fractions, materials eluted with 60% methanol/0.1% trifluoroacetic acid was put a thorough of a series of high performance liquid chromatography (HPLC) steps to isolate a neuropeptide with relaxation activity. The purified compound was eluted at 28% acetonitrile in 0.1% trifluoroacetic acid with retention time of 25.8 min on the CAPCELL-PAK C18 reversed-phase column. To determine the molecular weight and the amino acid sequence of the purified peptide, LC-MS and Edman degradation method were used, respectively. The primary structure of the peptide was determined to be FGMGGAYDPLSAGFTD which corresponded to the amino acid sequence of a starfish myorelaxant peptide (SMP) isotype (SMPb) found in the cDNA sequence encoding SMPa and its isotypes. In this study, a muscle relaxant neuropeptide (SMPb) has been isolated from pyloric caeca of starfish P. pectinifera. This is the first report of SMPb isolation on the protein level from P. pectinifera.

유채 전초 추출물 유래 항산화 및 항염 활성 성분 (Anti-oxidative and Anti-inflammatory Constituents from the Extracts of Brassica napus L. Whole Plant)

  • In this study, we investigated anti-oxidative and anti-inflammatory efficacy, and identified their constituents from Brassica napus L. (Korean name: Yuchae) whole plant. Upon the anti-oxidative activities screening, the ethanol extract exhibited potent DPPH and ABTS+ radical scavenging activities. On the anti-inflammation studies using LPS-induced RAW264.7 cells, the extract inhibited the production of NO and pro-inflammatory cytokines (TNF-α, IL-1β, IL-6) effectively. To identify major constituents of B. napus extract, further purification was performed and led to isolation of two compounds; isorhamnetin 3,7-O-diglucoside(1) and isorhamnetin 3-O-glucoside(2). Quantitative analysis by high pressure liquid chromatography (HPLC) determined the flavonoid 1 as the major constituent. Isolated compounds showed DPPH radical scavenging effects and decreased NO levels without causing cell toxicities. These results indicate that the extract of Yuchae, a rich plant resource in Jeju Island, could be potentially applicable as an anti-oxidative and/or anti-inflammatory ingredients.

제비콩 잎의 isoflavone 함량 및 성분 분석 (Determination and Isolation of leaf Isoflavone in Hyacinth Bean)

  • Legume seed isoflavones may have a variety of desirable physiological effect on the human health including both the circulatory and skeletal systems. The present study was performed to determine the isoflavone content of leaf and seed as well as to purify and identify the types of isoflavone from leaf extract of hyacinth bean (Lablab purpureus (L.) Sweet). Reverse phase HPLC revealed six different types of isoflavone such as daidzin, genistin, daidzein, genistein, 6"-o-acetyl genistin and 6"-o-acetyl daidzin in aqueous methanol extract from seeds and leaves of the hyacinth bean. Relatively, leaf isoflavone content of hyacinth bean was greater than seed isoflavone content. Using DiAion HP-20 silica gel and sephadex LH-20 chromatography, pure daidzein was identified in the ether layer, whereas genistin was in the EtOAC fraction.

Biocontrol of Peach Gummosis by Bacillus velezensis KTA01 and Its Antifungal Mechanism

  • Peach tree gummosis is a botanical anomaly distinguished by the secretion of dark-brown gum from the shoots of peach trees, and Botryosphaeria dothidea has been identified as one of the fungal species responsible for its occurrence. In South Korea, approximately 80% of gummosis cases are linked to infections caused by B. dothidea. In this study, we isolated microbes from the soil surrounding peach trees exhibiting antifungal activity against B. dothidea. Subsequently, we identified several bacterial strains as potential candidates for a biocontrol agent. Among them, Bacillus velezensis KTA01 displayed the most robust antifungal activity and was therefore selected for further analysis. To investigate the antifungal mechanism of B. velezensis KTA01, we performed tests to assess cell wall degradation and siderophore production. Additionally, we conducted reverse transcription-quantitative polymerase chain reaction (RT-qPCR) analysis based on whole-genome sequencing to confirm the presence of genes responsible for the biosynthesis of lipopeptide compounds, a well-known characteristic of Bacillus spp., and to compare gene expression levels. Moreover, we extracted lipopeptide compounds using methanol and subjected them to both antifungal activity testing and high-performance liquid chromatography (HPLC) analysis. The experimental findings presented in this study unequivocally demonstrate the promising potential of B. velezensis KTA01 as a biocontrol agent against B. dothidea KACC45481, the pathogen responsible for causing peach tree gummosis.

국내 유통 약용식물 중 잔류농약 모니터링 및 위해성 평가 (Monitoring of Pesticide Residues and Risk Assessment for Medicinal Plants)

  • 약용식물 중 잔류농약의 안전성을 평가하고자 2012년 전국 9개 도시에서 유통되고 있는 인삼과 도라지에 대하여 전체 112점의 시료를 수거하여 잔류농약을 분석하였다. 122종의 농약에 대해 GC-ECD, GC-NPD 및 HPLC-UVD를 이용한 다종농약 다성분 분석법으로 잔류농약을 분석하였고, 분석 결과 12점의 시료에서 7종의 농약이 검출되어 10.7% 검출률을 보였다. 농약 성분별 검출 빈도는 procymidone, kresoxim-methyl, endosulfan, cypermethrin, tralomethrin, tetraconazole, chlorfluazuron 순이었다. 농약이 검출된 시료 중 잔류허용기준을 초과한 시료는 2점으로 1.8% 검출률을 보였으며, 도라지 1점에서 tetraconazole, 인삼 1점에서 cypermethrin이 검출되었다. 해당 작물에 대한 잔류허용기준이 설정되어 있지 않거나 품목고시 되어 있지 않은 시료는 10점에서 5종의 농약이 검출되어 8.9% 검출률을 보였다. 본 연구에서 검출된 농약이 해당 약용 식물의 섭취로 인체에 유입될 일일섭취허용량 대비 일일 섭취추정량은 최저 0.006%에서 최고 0.333%로 낮은 %ADI 값을 보여 인체 위해도는 낮은 것으로 판단되었다.

Liquid Chromatography Quadrupole Time-Of-Flight Tandem Mass Spectrometry for Selective Determination of Usnic Acid and Application in Pharmacokinetic Study

  • A rapid and sensitive method for determining usnic acid of Lethariella cladonioides in rat was established using high performance liquid chromatography (HPLC) quadrupole time-of-flight (QTOF) tandem mass (MS/MS). Rat plasma was pretreated by mixture of acetonitrile and chloroform to precipitate plasma proteins. Chromatographic separation was achieved on a column ($50{\times}2.1$ mm, $5{\mu}m$) with a mobile phase consisting of water (containing $5{\times}10^{-3}$ M ammonium formate, pH was adjusted to 3.0 with formic acid) and acetonitrile (20:80, v/v) at a flow rate of 0.3 mL/min. A tandem mass spectrometric detection with an electrospray ionization (ESI) interface was conducted via collision induced dissociation (CID) under negative ionization mode. The MS/MS transitions monitored were m/z 343.0448 ${\rightarrow}$ m/z 313.2017 for usnic acid and m/z 153.1024 ${\rightarrow}$ m/z 136.2136 for protocatechuic acid (internal standard). The linear range was calculated to be 2.0-160.0 ng/mL with a detection limit of 3.0 pg/mL. The inter- and intra-day accuracy and precision were within ${\pm}7.0%$. Pharmacokinetic study showed that the apartment of usnic acid in vivo confirmed to be a two compartment open model. The method was fully valid and will probably be an alternative for pharmacokinetic study of usnic acid.

  • Transforming growth factor $\beta$1(TGF-$\beta$1)은 여러 가지 생물학적 활성을 가지는 관계로 의학적 치료제로서 사용될 가능성이 크다. 본 연구에서는 혈소판추출, 젤여과, 양이온교환 크로마토그래피 및 역상 HPLC등 네 단계의 정제과정으로 이루어져 있는 정제공정을 이용한 TGF-$\beta$1을 값싸고 효울적으로 정제하였다. 이 과정을 거쳐 최종적으로 얻어진 TGF-$\beta$1은 비환원조건하에서 SDS-PAGE를 행한 결과 구매된 TGF-$\beta$1 표준품과 일치한 위치에서 한 개의 band가 관찰되어 순수하다는 것을 확인하였으며 또한 이것이 Westem blot를 통하여 TGF-$\beta$1 항체와 결합하는 것으로부터 TGF-$\beta$1임을 확인하였다 또한, mink lung epithelial cell line 을 이용한 성장저해 실험을 통해 정제된 TGF-$\beta$1이 구매되TGF-$\beta$1 표준품보다 조금 높은 활성을 가지는 것을 확인하였다 최종적으로 농축혈소판 10단위로부터 약 3.7$\mu$g의 정제된 TGF-$\beta$1이 얻어져 그 최종수율은 약 21%였다.

Development of Gradient Centrifugal Partition Chromatography Method and Its Application for the Isolation of 3,5-Dimethoxyphenanthrene-2,7-diol and Batatasin-I from Dioscorea opposita

  • Gradient centrifugal partition chromatography (GCPC) method was developed and applied to isolate 3,5-dimethoxyphenanthrene-2,7-diol (DMP) and batatasin-I (BA-I) from the dichloromethane soluble extract of Dioscorea opposita. In this method, the lower phase of n-hexane-methanol-water system (HMW, 10 : 9 : 1, v/v) was used as a mobile phase A (MpA) and water was used as a mobile phase B (MpB). This gradient CPC method is comparable to that of reversed-phase HPLC method in that the stationary upper-phase of HMW (10 : 9 : 1 v/v) works as if it were reversed-phase silica gel due to its hydrophobic property, while the lower phase (MpA) and water (MpB) functioned as hydrophilic mobile phases. The initial condition of the mobile phase was 20% MpA/80% MpB and maintained for 150 min to obtain DMP (1.2 mg), and then MpA was increased up to 50% to elute BA-I (1.7 mg). The purities of DMP and BA-I were 94.1% and 98.3% with the recovery yields of 83% and 86%, respectively. Similar results were obtained by linear-gradient CPC. The CPC peak fractions were identified by comparing their retention time to those of authentic samples of DMP and BA-I and their spectroscopic data ($^1$H NMR and $^{13}$C NMR) to those of literature values.

복분자(Rubus coreanus Miquel) 와인의 ethyl acetate 획분으로부터 저분자 휘발성 화합물들의 분리 및 구조해석 (Isolation and Identification of Low Molecular Volatile Compounds from Ethyl Acetate Layer of Korean Black Raspberry (Rubus coreanus Miq.) Wine)

  • 복분자 와인은 복분자 열매를 발효 숙성시켜 제조한 것으로, 소비자에게 널리 음용되고 있으나 그 원재료인 복분자 열매에 대한 연구에 비해 복분자 와인에 존재하는 성분들에 대한 체계적인 연구는 거의 전무한 실정이다. 최근 우리는 복분자주에 함유되어 있는 화합물들의 분자 수준에서의 연구를 통하여 4-hydroxybenzoic acid, 3,4-dihydroxybenzoic acid, 4-(2-hydroxyethyl)-phenol, pyrocatechol, ethyl gallate 등의 5종의 저분자 페놀성 화합물을 단리 구조해석하여 보고한 바 있다. 그 연속된 연구로써 본 논문에서는 복분자주에 존재하는 성분들에 대한 추가적인 분리 및 구조해석을 행하였다. 제조된 복분자 와인(11 L, 복분자 열매 15.7 kg)을 용매분획하여 얻어진 EtOAc층(56.2 g)의 일부(20 g)를 silica gel column chromatography와 ODS-HPLC로 정제하여 5종의 화합물을 단리하였다. 이 화합물들을 대상으로 MS 및 NMR 등의 기기분석을 행한 결과, ethyl succinate(1, 13.1 mg), vanillic acid(2, 2.6 mg), ethyl 3,4-dihydroxybenzoate(3, 13. 1 mg), furan-2-ol(4, 1.3 mg), 그리고 4-(4-hydroxyphenyl) butan-2(S)-ol(5, 1.1 mg)로 동정하였다. 이 화합물들 대부분은 발효식품에서 향기성분으로 동정된 바 있으며, 화합물 2는 복분자 열매에 존재함이 이미 보고되어 있으나, 화합물 1과 3-5는 복분자 열매 및 복분자와인으로부터 처음으로 동정되었다.

  • 녹황색 세균인 Chlorobium limicola f. thiosulfatophilum NCIB 8327을 glutamate 가 질소원으로 포함된 변형 Pfennig 배지에서 배양한 다음 균체를 얻고, glutamine synthetase 효소를 초원심분리, DEAE-Sepharose CL-6B 이온교환 크로마토그래피, Sephacryl S-300 젤여과 크로마토그래피, 분취용 액체크로마토그래피의 과정을 통해 2% 수율에 46.3배의 정제배수로효소를 분리정제하였다. UV-VIS 흡수스펙트럼과 polycrylamide 젤 정기영동을 통해분리된 효소의 순수도는 확인하였다. Sephacryl S-3000 젤을 이용하여 얻은 효소의 분자량은 280 kDa 정도였으며, SDS-polyacrylamide 젤 전기영동 결과, 30 kDa 정도의 분자랴ㄹ을 갖는 단위체의 심합체로 추정된다. 이효소의 최적 온도와 pH 는 각각 $30^{\circ}C$ 와 pH 7.0 이었으나, 두기질 L-glutamine 과 hydroxylamine-HCI 에 대한 km 값은 각각 27.9 mM 과 0.92 mM 이었다. 이 효소의 활성은 alamine, glycine, tryptophan 등의 아미노산에 의해 상당한 저해를 받는 것으로 나타났으며, asparagin, lysine, leucine, valine 등에 의해서는 거의 영향을 받지 않았다.
