Production of Glutamic Acid by Pseudomonas sp. L-10 (Pseudomonas sp. L-10에 의한 글루탐산의 생산)

  • A bacterium L-10 which produce mush of glutamic acid was Isolated from soil and identified as the genus Pserdomonas. The maximal glutamic acid production was obtained when the strain was cultured at 3$0^{\circ}C$ for 30 hrs in the optimal medium containing 5% glucose, 0.5% each of urea and yeast extract, 0.1% K2HP04, 0.02% MgSO4.7H20, 0.3% (NH, )rHP04, 0.5ug/l biotin and Initial pH 7.0, and then final glutamic acid production under the above conditions was 1.2mg/ml of cell cultures.

  • Directional effect of applied pressure during sintering on the microstructure and fracture toughness of the heat-treated silicon nitride ceramics has been investigated. The specimens with a composition of 92Si3N4-8Y2O3(in wt%) were sintered at 172$0^{\circ}C$ by a hot press (HP ) and a hot isostatic press (HIP) and heat-treated for grain growth at 1800~20$0^{\circ}C$. The fracture toughness of the HP samples increased with the grain size while the fracture toughness of the HIP treated samples remained the same even though the grain growth occurred. This discrepancy was explained by a bimodal grain size distribution and large aspect ratio of the HPed samples and a monomodal grain size distributjion and samll aspect ratio of the HIP treated samples.

Correlation between Property of Bituminous Concrete and Chromatogram of Asphalt Cement Used (역청콘크리트의 성질과 아스팔트 시멘트 크로마토그램과의 상관관계)

  • Chromatograms of asphalt cements were observed using high pressure - gel permeation chromatography (HP-GPC). The chromatogram characterzed profile of molecular size distribution of asphalt cements. Bituminous concretes using the asphalt cements were prepared in laboratory to measure mechanical properties, tensile strength (TS) and resilient modulus (MR), in dry and wet conditions. Three sources of AC-20 Asphalt cements and one aggregates were used for GPC test and strength test. Results of mechanical property tests and GPC tests were analyzed to evaluate correlation between those two results. Regression analysis showed strong correlation between a mechanical property and sliced percent areas of GPC profile. Mathematical models were developed for estimating mechanical properties of asphalt cement concrete based on HP-GPC data.

  • After modeling an aircraft turboprop engine using SIMULINK$\circledR$, performance simulation of PT6A-62 engine, which is main power plant of KT-1, was performed. For validation, performance parameters of the SIMULIINK model were compared with the simulated results by GASTURB program. It was confirm that the results by the SIMULINK model were well agreed with those by GASTURB within 1.07%, It was assumed that installation losses were bleed-air exteraction with a range from 0% to 5%, and power for accessories with a range from 0 to 20hp. In this investigation, it was found that the shafthorsepower was decreased by maxium 0.68%, but specific fuel consumption ratio was not effected nearly by these losses.

  • The production of pullulan by Aureobasidium pullulans HP-2001 was investigated under various ratios of glucose as carbon source and yeast extract as the nitrogen source, Highest conversion rate (productivity) of glucose to pullulan was 40.0% when concentrations of glucose and yeast extract were 5% and 0.15%, respectively. Maximal production of pullulan was 29.3g/1 when the concentration of glucose was 8%(w/v) and that of yeast extract was 40:1. On basis of the result that production of pullulan was found in a medium which concentration of glucose as carbon source was up to 20%(w/v), Aureobasidium pullulans HP-2001 seemed to overcome the catabolite repression. Conversion rate of pullulan from 20%(w/v) of glucose was 11.1%.

  • In the screening for lipid peroxidation inhibitors from edible mushroom, Agrocybe cylindracea, a bioactive compound AG 8 was isolated. The AG 8 was purified from methanol extract of its fruit body by Diaion HP-20 column chromatography, ethyl acetate extraction, and silica gel column chromatography, consecutively. Based on various NMR studies including $^1$H irradiation and HMBC experiments, the AG 8 was identified as MTA, 5'-deoxy-5'-methylthioadenosine. This compound inhibited lipid peroxidation with an $IC_{50}$/ value of 3.2 $\mu\textrm{g}$/$m\ell$. The MTA was isolated for the first time from basidiomycetes.

  • Moleclar size distribution of three AC-20 asphalt cements were observed using high pressure - gel permeation chromatograph (HP-GPC). Bituminous concrete specimens were made using the asphalt cements and one aggre¬gate in laboratory to measure mechanical properties, tensile strength CIS) and resilient modulusCM,J. in dry and wet conditons. Results of mechanical property tests and profile of molecular size distribution of asphalt cements were analyzed to evaluate the relatonship of mechanical property with chromatogram. Regression analy¬sis showed a strong correlation between a mechanical property and sliced percent areas of chromatogram. Mathematical prediction models were developed based on the regression analysis. It was shown that characteris¬tics of asphalt cement chromatogram could be successfully used for estimation of selected mechanical property of asphalt concrete .

  • Aspergillus sp. F184 was isolated from soil for the development of new xanthine oxidase inhibitor. This xanthine oxidase inhibitor was sequentially purified by filtration, HP-20 adsorption column chromatography, ethyl acetate extraction, silica gel column chromatography and crystallization, and was named as YUX 104. YUX 104 was identified to be 5,6-epoxy-2-hydroxy-3-methyl-2-cyclohexene-1,4-dione(terreic acid) by NMR and mass spectroscopic sudies.

  • In order to reduce systemic side effects following administration, flurbiprofen was formulated as O/W microemulsion consisting of the surfactant, oil phase and aqueous phase. Particle size distribution, apparent viscosity, solubility and skin permeation of flurbiprofen in various vehicles and microemulsion were evaluated. The domain of O/W microemulsion s phase diagram had difference between oil types and the area of O/W microemulsion was wide distributed by adding to PG and cosurfactant than that of water alone. As increasing 10, 15 and 20% of Brij 97 content and 1, 2.5, 5% of oil content, the solubility of flurbiprofen in O/W microemulsions and various vehicles was $400{\sim}1,000$ and $10{\sim}500$ times higher than that of control. Also, apparent viscosity of soybean oil microemulsions was higher than that of IPM microemulsions and that of vehicle were increased as increasing vehicle content. Since skin permeation of flurbiprofen decreased as increasing viscosity, in each vehicle, it was not affected 2% ${\beta}-CD$ and decreased as increasing PG content and to 2, 5 and 10% of $HP-{\beta}-CD$. In O/W microemulsion, 5% soybean oil. 20% Brij 97 and 75% water(A-1) with high viscosity showed low skin penetration.
