Effect of different levels of protein concentrates supplementation on the growth performance, plasma amino acids profile and mTOR cascade genes expression in early-weaned yak calves

  • Objective: This study evaluated the effects of different levels of protein concentrate supplementation on the growth performance of yak calves, and correlated the growth rate to changes occurring in the plasma- amino acids, -insulin profile, and signaling activity of mammalian target of rapamycin (mTOR) cascade to characterize the mechanism through which the protein synthesis can be improved in early weaned yaks. Methods: For this study, 48 early (3 months old) weaned yak calves were selected, and assigned into four dietary treatments according to randomized complete block design. The four blocks were balanced for body weight and sex. The yaks were either grazed on natural pasture (control diet) in a single herd or the grazing yaks was supplemented with one of the three protein rich supplements containing low (17%; LP), medium (19%; MP), or high (21%; HP) levels of crude proteins for a period of 30 days. Results: Results showed that the average daily gain of calves increased (0.14 vs 0.23-0.26 kg; p<0.05) with protein concentrates supplementation. The concentration of plasma methionine increased (p<0.05; 8.6 vs $10.1-12.4{\mu}mol/L$), while those of serine and tyrosine did not change (p>0.05) when the grazing calves were supplemented with protein concentrates. Compared to control diet, the insulin level of calves increased (p<0.05; 1.86 vs $2.16-2.54{\mu}IU/mL$) with supplementation of protein concentrates. Addition of protein concentrates up-regulated (p<0.05) expression of mTOR-raptor, mammalian vacuolar protein sorting 34 homolog, the translational regulators eukaryotic translation initiation factor 4E binding protein 1, and S6 kinase 1 genes in both Longissimus dorsi and semitendinosus. In contrast, the expression of sequestosome 1 was down-regulated in the concentrate supplemented calves. Conclusion: Our results show that protein supplementation improves the growth performance of early weaned yak calves, and that plasma methionine and insulin concentrations were the key mediator for gene expression and protein deposition in the muscles.

  • Weld repair of ASTM A-470 class 8 high pressure (HP) steam turbine rotor steel has been performed to extend the service life of older fossil units. Microhardness measurements were conducted across the weldment from unaffected base metal (BM) to weld metal (WM). The hardness of the BM was VHN 253, however it dropped up to VHN 227 at the heat affected zone (HAZ) close to unaffected BM for multipass SAW. This area of hardness drop is called "siftening zone" and has a width of 0.5-0.6mm. During creep rupture test, failure occurred around the softening zone and rupture time was 772.4hr at 19Ksi (132 Mpa) and 593.deg. C. Multipass MIG and TIG welding have been employed to reduce the softening zone width. The softening zone width for MIG was 0.3-0.4mm and for TIG was zero-0.4mm depending on heat inputs. However creep rupture time was decreased as softening zone width reduced. Creep rupture time also showed a close relationship with heat inputs in TIG process. The higher heat input, the longer rupture time. Most failure occurred at intercritical HAZ (ICHAZ), however rupture location was shifted to coarse grained HAZ (CGHAZ) as heat input decreased. The rupture surface showed tearing and dimple which indicated transgranular fracture. fracture.

  • The root of Panax ginseng C.A. Meyer were extracted with 70% aqueous EtOH and the concentrates were partitioned into MeOH and H2O fractions using Diaion HP-20. The repeated SiO2 or octadecyl SiO2 column, and MPLC for the MeOH fraction led to isolation of four malonyl ginsenosides. The chemical structures of these compounds were determined as malonyl ginsenoside Rd (1) malonyl ginsenoside Rc (2) malonyl ginsenoside Rb2 (3) malonyl ginsenoside Rb1 (4) based on spectroscopic analyses including Nuclear magnetic resonance and HR-TOF/MS. The contents of malonyl ginsenoside Rb1 was highist as 5.44 mg/g of five years of ginseng. And malonyl ginsenoside Rd was lowest as 0.11 mg/g of six years of ginseng. Additionally, the malonyl ginsenoside Rd exhibited hepatoprotective effect against ethanol-induced hepatotoxicity in HepG2 cell line.

작업관련성을 고려한 U라인 밸런싱+

  • 본 연구는 U자형 생산라인(U라인)에서의 라인밸런싱 문제를 해결하기 위한 유전알고리즘을 제시한다. U라인에서는 사이클 타임 내에 제품이 들어가는 방향과 나오는 방향의 작업을 한 작업자가 수행 할 수 있어서, 재공품 수량을 일정하게 유지한다거나 작업부하를 평활화 하는 등의 라인관리가 기존 직선라인에 비해 용이하다. U라인은 JIT(Just-ln-Time)생산 시스템에서 흔히 볼 수 있다. 본 연구는 U라인에서 사이클 타임이 고정되었을 때 작업장 또는 작업자의 수를 최소로 하면서 동시에 작업장에 할당된 작업들 간의 관련성을 최대화하는 라인밸런싱 문제를 다루었다. 라인밸런싱에 관한 기존 연구는 대부분 직선라인에 관한 것으로 U라인의 장점을 충분히 활용하지 못한다. 특히, 라인의 작업장의 수를 최소화하는 문제는 많은 대안해가 있음에도 불구하고, 작업관련성을 고려하여 해를 구하는 기법에 관한 연구는 아직 미미한 실정이다. 실제 조립라인에서는 가능한 한 관련된 작업들을 동일한 작업자에 할당하는 것이 바람직하며, 이러한 작업편성은 작업자의 작업능률을 향상시킬 수 있다. 유전알고리즘은 자연계의 적자생존과 생물학적 진화과정을 모방한 탐색기법으로 조합최적화 문제에 효과적인 기법으로 널리 알려져 있다. 본 연구는 유전알고리즘을 이용하여 U라인에서 작업관련성을 고려한 라인밸런싱 문제를 해결하기 위한 기법을 개발하였다. 문제의 목적에 적합한 개체의 평가함수가 제시되었으며, 개체의 형질을 효과적으로 자손에 유전할 수 있고 유전 연산이 용이한 개체의 표현방법과 개체의 해석방법이 제시되었다. 컴퓨터 실험을 통하여 개발한 알고리즘의 성능을 보였다.월 초순부터 중순에 각각 최고 성기를 나타내었다. H. papariensis의 암컷과 수컷의 발광양상을 분석하고자 정지발광과 구애 발광을 구분하여 조사하였고 각각의 발광지속시간과 발광주기를 구분하여 측정하였다. 수컷의 발광지속시간은 정지발광(0.12초)보다 구애발광(0.17초)에서 1.4배 증가하였으며 암컷의 발광지속시간은 정지발광(0.15초)보다 구애발광(0.19초)에서 1.5배 증가하였다. 발광주기는 수컷에서 정지발광(1.26초)보다 구애발광(1.12초)에서 0.88배 감소하였고, 암컷에서 정지발광(2.99초)보다 구애발광(1.06초)에서 0.35배 감소하였다. 발광양상에서 발광주파수는 수짓의 정지발광에서 0.8 Hz, 수컷 구애발광에서 0.9 Hz, 암컷의 정지발광에서 0.3 Hz, 암컷의 구애발광에서 0.9 Hz로 각각 나타났다. H. papariensis의 발광파장영역은 400 nm에서 700 nm에 이르는 모든 영역에서 확인되었으며 가장 높은 첨두치는 600 nm에 있고 500에서 600 nm 사이의 파장대가 가장 두드러지게 나타났다. 발광양상과 어우러진 교미행동은 Hp system과 같은 결과를 얻었다.하는 방법을 제안한다. 즉 채널 액세스 확률을 각 슬롯에서 예약상태에 있는 음성 단말의 수뿐만 아니라 각 슬롯에서 예약을 하려고 하는 단말의 수에 기초하여 산출하는 방법을 제안하고 이의 성능을 분석하였다. 시뮬레이션에 의해 새로 제안된 채널 허용 확률을 산출하는 방식의 성능을 비교한 결과 기존에 제안된 방법들보다 상당한 성능의 향상을 볼 수 있었다., 인삼이 성장될 때 부분적인 영양상태의 불충분이나 기후 등에 따른 영향을

Enzymatic Characterization of a Thermostable 4-α-Glucanotransferase from Thermotoga neapolitana (Thermotoga neapolitana 유래 내열성 4-알파-글루칸전이효소의 효소적 특성)

  • The gene encoding 4-$\alpha$-glucanotransferase (mgtA) from Thermotoga neapolitana was cloned and expressed in Escherichia coli in order to investigate whether this enzyme was capable of producing cycloamylose for industrial applications. MgtA was purified to homogeneity by HiTrap Q HP and Sephacryl S-200 HR column chromatographies. The size of the enzyme as determined by SDS-PAGE was about 52 kDa, which was in good agreement with its deduced molecular mass of 51.9 kDa. The optimal temperature and pH for the activity of the 4-$\alpha$-glucanotransferase was found to be $85^{\circ}C$ and 6.5, respectively. The enzyme hydrolyzed the 1,4-$\alpha$-glucosidic bonds in oligomeric 1,4-$\alpha$-glucans and transferred oligosaccharides (maltotriose being the shortest one) to acceptor maltodextrins. However, the enzymes had no activity against pullulan, glycogen, and other di- or trioligosaccharides with rare types of $\alpha$-bond. MgtA is distinguished from 4-$\alpha$-glucanotransferase from Thermotoga maritima in that it can convert maltotriose into maltooligosaccharides. The treatment of glucoamylase after the reaction of MgtA with maltotriose, maltotetraose, maltopentaose, or maltohexaose as sole substrate revealed that MgtA yielded linear maltooligosaccharides instead of cycloamylose.

The Effects of Levan on Blood Lipids and the Absorption of Calcium in Rats Fed a Low Calcium Diet (레반의 혈장지질 감소와 칼슘흡수율 증가 효과)

  • Levan, the fructose polymer is an indigestible carbohydrate regularly consumed by humans. Its physiological functions, in terms of hypocholesterolemic effect and calcium metabolism, have not been well documented. The objective of this study was to investigate the effects of levan on blood lipids and the calcium absorption ratio in rats fed a 0.1% low calcium diet. Thirty rats were divided into three groups and fed a 0.1% low calcium diet (control) or low calcium diets containing either 2.5% levan or 5% levan for eight weeks. The blood lipid and biomarkers relevant to Ca metabolism (urinary Ca and hydroxyproline), the femoral weight and the Ca contents were determined. The body weight gains were lower in the 5% levan group than the control group. Plasma concentrations of triacylglycerol and LDL-cholesterol decreased in the 5% levan group, compared to the control group, but the atherogenic indice were not affected. Blood alkaline phosphatase activity, Ca and urinary hydroxyproline excretion levels were not different in the three groups. The net calcium absorption in rats fed a 5% levan diet was greater than rats fed the control diet, while the femoral weights and Ca contents were not significantly different in the three groups. We concluded that a 5% levan diet could both enhance the calcium absorption and improve the lipid profiles in rats fed a low calcium diet.

Development of Simultaneous Analysis for Multiple Agricultural Pesticides in Raw Milk Products using GC-MS/MS (GC-MS/MS를 이용한 원유 원료 중 농약 동시분석법 확립)

  • GC-MS/MS using liquid-liquid extraction (LLE) and C18 cartridges was used to identify and quantify levels of chlorpyrifos, chlorpyrifos-methyl, cypermethrin, deltamethrin and permethrin in bulk raw milk. A calibration curve spanning 10 ng/mL to 200 ng/mL was obtained with a satisfactory correlation coefficient of 0.99. The limits of detection (LOD) and limits of quantitation (LOQ) for chlorpyrifos, chlorpyrifos-methyl, cypermethrin, deltamethrin, and permethrin in the matrix ranged from 0.06 to 1.81 ng/mL and 0.19 to 6.04 ng/mL, respectively. The recoveries of 5 pesticides from spiked samples at 37.5-125 ng/mL ranged from 86.1 to 102.1%. The measurement of uncertainty of the GC-MS/MS method for these five pesticides was developed based on the analytical process and quantification. An analysis method that is easier and faster than the method specified in the Korean food standards codes for analyzing these five pesticides in raw material milk was developed. Moreover, the analytical method for chlorpyrifos, chlorpyrifos-methyl, cypermethrin, deltamethrin, and permethrin in bulk raw milk by GC-MS/MS was established.

Biological Activities of Pharbitis nil and Partial Purification of Anticancer Agent from Its Extract (견우자의 생리활성 분석과 추출물로부터 항암 활성물질의 분리)

  • This study aimed to evaluate several biological activities of Pharbitis nil and to isolate an anticancer agent from its methanol extract. Pharbitis nil seeds were extracted with methanol (PNM). Then, PNM was fractionated into solvent layers such as ethyl acetate fraction (PNE), butanol fraction (PNB), and water fraction (PNW). The biological activities of the fractions were analyzed for tyrosinase inhibition, lipase inhibition, DPPH-free radical scavenging, and cell growth inhibition. PNM showed strong growth inhibition of prostate cancer PC-3 cells. PNM was subjected to Diaion HP-20 and eluted stepwise with 50%, 80%, and 100% methanol. Then, for activity-guided fraction, each fraction was analyzed for growth inhibition of prostate cancer PC-3 cells by using an MTT assay. Because the 100% fraction showed significantly strong inhibitory activity, the fraction was further separated in the reverse phase C18, which was eluted with 80% and 90% methanol. The 90% fraction was further subjected to Sephadex LH-20 using a mobile solvent of 100% methanol. Finally, the compound PN was partially purified for HPLC analysis. PN showed cell growth inhibitory activity and induced the apoptosis and cell cycle arrest of prostate cancer PC-3 cells, as measured by flow cytometry. The results together suggest that Pharbitis nil possesses various biological activities, especially the inhibitory activity for the proliferation of prostate cancer PC-3 cells, suggesting the possibility of its use as an anticancer agent.

The Effect of Veneering Techniques on the Color Parameters of Y-TZP Based All Ceramic Restoration of Varying Ceramic Core Thickness (지르코니아 코어 두께에 따른 전부도재관의 상부도재 축성방법이 보철물 색조에 미치는 영향)

  • The aim of study was to compare the color parameters and mean color difference of porcelain specimens by different veneering technique in order to examine the effect of veneering technique on esthetics of yttria-stabilized tetragonal zirconia polycrystalline(Y-TZP) all ceramic restoration. Three groups of square-shaped core ceramic specimens(14mm in diameter and 0.3, 0.5, 0.7 mm) and two groups of veneering ceramic specimen were prepared for analysis. Color parameter($L^*,a^*,b^*$) and color difference of zirconia core squares and core-veneer specimens were measured with ShadeEye $NCC^{(R)}$ spectrophotometer, respectively. Mean color difference(${\Delta}E^*$)were calculated using color difference formula. Two-way analysis of variance(ANOVA) combined with a Tukey multiple-range test were used to analysis the data(${\alpha}=0.05$). ${\Delta}E^*$ values were not significantly affected by core thickness and veneering porcelain(p=0.083). The color differences(${\Delta}E^*$) of core-veneer specimens with 0.5, 0.7 mm-A1,A2,A3.5 shade were mostly below 3.7 which was within the clinically acceptable range, while color differences between 0.3 mm-A1,A2 showed more than 3.7. All-ceramic system has color characteristics that clinicians have to consider when selecting materials. Also, manufacturers of different porcelain systems must make every effort to achieve color reproducibility.

Characterization of Ethanol Fermentation with Wild Type Yeast Strains (야생 효모 종류에 따른 알코올 발효 특성)

  • The objective of this study was to improve the quality of Korean rice wine with wild type yeast strains isolated from various traditional Korean fermented foods. Herein the fermentation and sensory characterization of wild yeast, for the purposes of brewing Korean rice wine, was investigated. 12 yeast strains were examined for their ethanol and glucose tolerance. In addition, the pH, soluble solids, acidity, amino acidity, ethanol content, organic acids, and volatile compounds were also studied for the alcoholic beverages made with the wild yeasts. Almost all Saccharomyces genera yeasts were showed to have a tolerance at 10% ethanol, but non-Saccharomyces genera yeasts displayed a low tolerance. The alcoholic beverages fermented by non-Saccharomyces yeasts demonstrated higher levels of soluble solids, titratable acidity, amino acids, and lower ethanol content, when compared with the alcoholic beverages fermented by Saccharomyces genera yeasts. The organic acid content, such as malic acid, acetic acid, and succinic acid, was seen to also be higher. The electronic nose was analyzed, and discriminant function analysis (DFA) was used for discriminating wild yeast strains. The DFA plots indicated a significant separation of Saccharomyces genera and non-Saccharomyces yeast strains. For volatile compounds, ethyl acetate from non-Saccharomyces yeasts, and ethanol from Saccharomyces genera yeast, a high area ratio was observed.
