Effect of Glycosaminoglycans on In vitro Fertilizing Ability and In vitro Developmental Potential of Bovine Embryos

  • The glycosaminoglycans (GAGs) present in the female reproductive tract promote sperm capacitation. When bovine sperm were exposed to 10 ${\mu}g/ml$ of one of four GAGs (Chondroitin sulfate, CS; Dermatan sulfate, DS; Hyaluronic acid, HA; Heparin, HP) for 5 h, the total motility (TM), straight-line velocity (VSL), and curvilinear velocity (VCL) were higher in the HP- or HA-treated sperm, relative to control and CS- or DS-treated sperm. HP and HA treatments increased the levels of capacitated and acrosome-reacted sperm over time, compared to other treatment groups (p<0.05). In addition, sperm exposed to HP or HA for 1 h before IVF exhibited significantly improved fertilizing ability, as assessed by 2 pronucleus (PN) formation and cleavage rates at d 2. Exposure to these GAGs also enhanced in vitro embryo development rates and embryo quality, and increased the ICM and total blastocyst cell numbers at d 8 after IVF (p<0.05). A real-time PCR analysis showed that the expression levels of pluripotency (Oct 4), cell growth (Glut 5), and anti-apoptosis (Bax inhibitor) genes were significantly higher in embryos derived from HA- or HP-treated sperm than in control or other treatment groups, while pro-apoptotic gene expression (caspase-3) was significantly lower in all GAG treatment groups (p<0.05). These results demonstrated that exposure of bovine sperm to HP or HA positively correlates with in vitro fertilizing ability, in vitro embryo developmental potential, and embryonic gene expression.

Sustained $K^+$ Outward Currents are Sensitive to Intracellular Heteropodatoxin2 in CA1 Neurons of Organotypic Cultured Hippocampi of Rats

  • Blocking or regulating $K^+$ channels is important for investigating neuronal functions in mammalian brains, because voltage-dependent $K^+$ channels (Kv channels) play roles to regulate membrane excitabilities for synaptic and somatic processings in neurons. Although a number of toxins and chemicals are useful to change gating properties of Kv channels, specific effects of each toxin on a particular Kv subunit have not been sufficiently demonstrated in neurons yet. In this study, we tested electro-physiologically if heteropodatoxin2 ($HpTX_2$), known as one of Kv4-specific toxins, might be effective on various $K^+$ outward currents in CA1 neurons of organotypic hippocampal slices of rats. Using a nucleated-patch technique and a pre-pulse protocol in voltage-clamp mode, total $K^+$ outward currents recorded in the soma of CA1 neurons were separated into two components, transient and sustained currents. The extracellular application of $HpTX_2$ weakly but significantly reduced transient currents. However, when $HpTX_2$ was added to internal solution, the significant reduction of amplitudes were observed in sustained currents but not in transient currents. This indicates the non-specificity of $HpTX_2$ effects on Kv4 family. Compared with the effect of cytosolic 4-AP to block transient currents, it is possible that cytosolic $HpTX_2$ is pharmacologically specific to sustained currents in CA1 neurons. These results suggest that distinctive actions of $HpTX_2$ inside and outside of neurons are very efficient to selectively reduce specific $K^+$ outward currents.

  • Objectives : The Major Essentials of Huangdi's Internal Classics Plain Questions (黃帝內經素問大要, MEHP) is one of the late-Chosun's literate physician Lee Gyoojoon (李圭晙, 1885-1923)'s main work, which is known to have logically proofread the Huangdi's Internal Classics Plain Questions (黃帝內經素問, HP). This study aims to examine two elements of the text: (1) the types of MEHP currently in circulation; (2) the types of publications of HP to be the MEHP's original script. Methods : In this study, basically bibliographical analyses of the form and contents was utilized about the types of MEHP and HP. However, to compare the sentences and phrases between prints, I've used 20 examples that Qian Chaochen (钱超尘) had proposed in his preceding studies. Also, regarding Lee Wonse (李元世)'s proofreading on the MEHP in 1999, I've used interviews of his students. Results : First, I've discovered that there are three versions MEHP in circulation: the woodblock printed version; Lee Wonse's handwritten version; Lee Wonse's proofreading version; and confirmed that Lee's proofreading version should be regarded as good version of MEHP. Also, I've discovered the possibility of other types of printed versions of the MEHP in existence, which is considered Lee's handwritten version's original draft. Second, I've confirmed that the original script of HP, which Lee Gyoojoon utilized for MEHP, is indeed not Gu Congde (顧從德) printed version HP, however, is the Chosun's bureau for military drill (訓練都監) printed version HP. Conclusion : Through this study, I've provided strong evidence that Lee Gyoojoon's MEHP is a unique and original research completed within the traditional realm of Korean medicine, which possesses the universality of Eastern Asian medicine represented by Huangdi's Internal Classics (黃帝內經).

Deodorization of Gastrodia elata Blum Extracts using Diaion HP-20 Resin (Diaion HP-20 레진을 이용한 천마 추출물의 불쾌취 제거)

  • Background: Although Gastrodia elata contains the valuable, bio-active compounds gastrodin and 4-hydroxybenzyl alcohol (4-HBA), it also contain ρ-cresol, which is a major source of an objectionable pig slurry-like odor. Accordingly, this study was performed to determine the deodorization effect of Diaion HP-20 resin on a G. elata extract. Methods and Results: Using Diaion HP-20 with an increasing concentration of ethanol, an extract of G. elata was fractionated into 15 fractions, which were anlayzed to determine their concentrations of active compounds and ρ-cresol. The gastrodin, 4-HBA, total phenolics, and ρ-cresol concentrations of the G. elata extract were 403.13, 408.15, 2,476.09, and 11.77 ㎍/㎖, respectively. Among the different fractions, gastrodin and 4-HBA were detected in fractions 3 to- 6, and 4 to- 10, respectively whereas total phenolics were detected in all samples, with fractions 5 to- 7 being characterized by higher concentrations than were the other fractions. ρ-Cresol was detected in fractions 12 to- 15, at concentrations ranging from 0.39 ㎍/㎖ to 6.05 ㎍/㎖ with the highest concentration detected in fraction 14. The fractions containing ρ-cresol could be selectively separated from the G. elata extract without the loss of major bio-active compounds such as gastrodin and 4-HBA. Conclusions: The results of this study indicate that a deodorizing method using Diaion HP-20 resin can be considered for improving the sensory qualities of G. elata extract without a loss of the major bio-active compounds.

The effect of tooth bleaching agent contained 35% hydrogen peroxide on the color, microhardness and surface roughness of tooth-colored restorative materials (35% 과산화수소를 함유한 치아미백제가 심미수복재의 색, 미세경도 및 표면 거칠기에 미치는 영향)

  • Objectives : The purpose of this study was to evaluate the effects of tooth bleaching agent contained 35% hydrogen peroxide on the color, microhardness and surface roughness of tooth-colored restorative materials. Methods : Four types of tooth-colored restorative materials, including a composite resin(Filtek Z350 ; Z350), a flowable composite resin(Filtek P60 : P60), a compomer(Dyract$^{(R)}$ AP ; DY), and a glass-ionomer cement(KetacTM Molar Easymix ; KM) were used in the study. The specimens($8mm{\times}5mm$) were made by using a customized acrylic mold. Each material was divided into two groups equally(n=40) : experimental group(35% HP) and control group(distilled water). 35% HP group was treated 30 mim/5 days for 15 days. Each 30 minute treatment session consisted of two 15 minute cycles of gel application with 20 second light exposure. The authors measured the color, microhardness, and roughness of the specimens before and after bleaching. The data were analyzed with ANOVA and T-test. Results : 35% HP group showed an apparent color change(${\Delta}E^*$) than control group. In particular, DY and KM showed a noticeable color change and statistically significant differences(p<0.05). 35% HP group showed a reduction in microhardness. Z350 and P60 does not have a statistically significant difference(p>0.05), DY and KM showed a statistically significant difference(p<0.05). Percentage microhardness loss(PML) of control group was 0.6 to 5.5% in the group, 35% HP group was 6.6 to 34.6%. Roughness was increased in 35% HP group after bleaching. Especially DY and KM were significantly increased(p<0.05). Conclusions : Bleaching agents may affect the surface of existing restorations; therefore, they should not be used indiscriminately when tooth-colored restorations are present.

Establishing HP/LP-EGR System and Founding Operating Strategy of Low Temperature Combustion Engine to Improve Fuel Consumption (연료소비율 개선을 위한 고압/저압 배기재순환 시스템 구축 및 저온연소 엔진의 운전전략 수립)

  • This study researched on the effect of HP/LP-EGR system to improve fuel consumption of Low Temperature Combustion Engine. Firstly, low temperature combustion engine with HP/LP-EGR system was established using 6.0L wastegate turbocharger HDDI engine. And suppliable EGR rate of the engine was proven to be enough to realize stable low temperature combustion. Then, optimum operating strategy was founded to develop fuel consumption of the engine. Control parameters were HP/LP-EGR valve and IPCV(Intake Pressure Control Valve) duty. Experiments method was that characteristics of the engine were measured and analyzed according to HP/LP-EGR strategies while EGR rate was fixed. Operating range for the strategy were divided into three parts, low load for low temperature combustion, high load for conventional diesel combustion, and transient condition. Finally, with the above strategy of this study, BSFC of the engine was improved about 2% compared to the base engine, and emission level, NOx and PM, met Tier4Final emission regulation.
