In vitro infection of Cryptosporidium parvum to four different cell lines

  • To determine a suitable condition for in vitro infection model of cryptosporidium parvum, four different cell lines, AGS, MDCK, HCT-8 and Caco-2, were used as host cell lines which were cultured at various concentrations of added supplements. These supplement include fetal bovine serum (FBS), sodium choleate, ascorbic acid, folic acid, calcium pantothenate, para-aminobenzoic acid and pyruvate and their effects on the cell lines which were infected with C. parvum were evaluated. The results of this study showed that the AGS cell line was most susceptible to C. parvum whereas the Caco-2 cells appeared to be least susceptible to C. parvum. In regards to the serum condition, 10% FBS was suitable for the growth of AGS and HCT-8 cells, and 1% FBS was good for the growth of the MDCK cells when they were inoculated with C. parvum. Vitamines had a positive effect on the AGS cells, and pyruvate also showed positive effects on all of the cell lines except for Caco-2. Modified medium for each cell line was prepared by adding appropriate amounts of each supplement which resulted in the highest parasite infection number. Modified media increased the number of parasites infected on AGS cells to 2.3-fold higher when compared to the control media. In this study, we found that the AGS cell line was a suitable host model for evaluating C. parvum in vitro study and the media contents for the optimal infection conditions were suggested.

  • The small type piroplasma which is known to be prevailing in Korean cattle was injected to un-susceptible hosts such as mice, guinea pigs, rats, rabbits, fowls and developing chicken eggs by i.v., i.p, and intra C.A.M. routes, and its morphological development in each animal species was observed. 1. The small type piroplasma of cattle did not produce any pathogenicity in the experimental animals such as mice, guinea pigs, rats, rabbits, fowls and developing chicken embryos. However, only a partial sign for developing cycle of the organism was observed in the peritoneal fluid. The schizogony of protozoa was not detected in any organs of the inoculated animals. 2. In the animals inoculated, the parasites of I-III types, relative to the process of time in the host, have changed their shapes into IV-VI types and then VI-X types however, the type X was not restored to the type I. In guinea pigs, more grape type was detected then X type in number, and in process of time, the shape was changed from round to elliptic, and the pointed end of the organism appeared in flesh color. 3. None of the organisms were found from the blood stream of inoculated animals excepts only a few piroplasma as were found in the experimental mice shortly after inoculation or i.v. route, No protozoa was also found in the organs of the animals autopsied. 4. The form change of protozoa was more clear in these animals than that in susceptible host. The most piroplasmas have changed their forms into VI-X types 30 hours after inoculation and were dispersed from R.B.C. Tracing of piroplasrna beyond this stage was not attempted. 5. The small type piroplasma comprises in I. X types and it is further believed that VII-X types which were freed from one cell proceeded to invade others. The from of development resembles to the small type of piroplasma which is prevailing in Japan.

  • 피부흡충인 Entobdella hippoglossi가 숙주인 Atlantic halibut의 피부에 기생할 때 기생 부위별 밀도와 숙주의 피부 점액세포 밀도를 조사하여 E. hippoglossi의 기생이 피부점액 세포의 밀도와 관계가 있는가를 조사하였다. 숙주인 Atlantic halibut 복측과 배측에 기생하는 E. hippoglossi의 밀도는 비슷하였으나 기생충의 크기는 복측이 배측보다 더 컸으며, 배측내에서는 배측의 머리부분에 기생하는 E. hippoglossi의 밀도가 배측의 중앙부나 꼬리부분에서보다 높았다. 숙주의 점액세포 크기, 밀도 그리고 표피 점액층의 두께는 대체로 기생충의 밀도와 비례하였고, 숙주내에서 기생충의 선택적 기생은 점액세포의 밀도와 관계가 있었다.

Serine Proteases of Parasitic Helminths

  • Serine proteases form one of the most important families of enzymes and perform significant functions in a broad range of biological processes, such as intra- and extracellular protein metabolism, digestion, blood coagulation, regulation of development, and fertilization. A number of serine proteases have been identified in parasitic helminths that have putative roles in parasite development and nutrition, host tissues and cell invasion, anticoagulation, and immune evasion. In this review, we described the serine proteases that have been identified in parasitic helminths, including nematodes (Trichinella spiralis, T. pseudospiralis, Trichuris muris, Anisakis simplex, Ascaris suum, Onchocerca volvulus, O. lienalis, Brugia malayi, Ancylostoma caninum, and Steinernema carpocapsae), cestodes (Spirometra mansoni, Echinococcus granulosus, and Schistocephalus solidus), and trematodes (Fasciola hepatica, F. gigantica, and Schistosoma mansoni). Moreover, the possible biological functions of these serine proteases in the endogenous biological phenomena of these parasites and in the host-parasite interaction were also discussed.

Inhibitory Activity of Oak Pyroligneous Liquor against Coleosporium Plectranthi, an Obligate Parasite Responsible for the Rust Disease on Perilla Leaf

  • BACKGROUND: Coleosporium plectranthi, an obligate parasite, which is responsible for the rust disease of Perilla frutescens, a plant in Korea, commonly known as Perilla. All rusts are obligate parasites, meaning that they require a living host to complete their life cycle. They generally do not kill the host plant but can severely reduce growth and yield. Food and feed spoilage fungi cause great economic losses worldwide. It is estimated that between 5 and 10% of the world food production is wasted due to fungal deterioration. Rust disease of Perilla is highly frequent and is widely spread in Korea. The present study was designed to investigate a novel media for the urediniospore germination in vitro and anti-rust activity as well as GC-MS analysis of oak pyroligneous liquor. METHOD AND RESULTS: Urediniospores were collected from the infected leaf of Perilla. Spore suspension was made and the suspension was inoculated in the 2% water agar media with proper humidity, then they were incubated at $26^{\circ}C$ for 56 hrs. The GC-MS analysis of the oak pyroligneous liquor was also done to check the chemical composition. GC-MS analysis of the wood vinegar was found 15 compounds, among them o-mthoxyphenol (25.93%), 2,6-dimethoxyphenol (16.06%), 4-methylenecyclohexanone (10.69%), 2,3-dihydroxytoluene (7.84%), levoglucosane (6.14%) and propanoic acid (5.32%) were the major components. Different concentration of the oak pyroligneous liquor was used, and spore inhibition was recorded on the basis of spore counting. The best results were noted at the concentration of 50% solution where 31.8% spores were inhibited. CONCLUSION: On the basis of the chemical composition of the oak pyroligneous liquor and the activity recorded we can use it as an anti-rust agent.

Influence of abiotic factors on seasonal incidence of pests of tasar Silkworm Antheraea mylitta D.

  • Rearing of tropical tasar silkworm, Antheraea mylitta Drury is mainly conducted in outdoor on Terminalia tomentosa W. & A. a nature grown primary host plant available in forest and also on raised primary host plant Terminalia arjuna Bedd. Temperature, relative humidity and rainfall are the main environmental factors for occurrence of pests (parasites and predators) of tasar silkworm during I, II and III crop rearing in the tropical tasar producing zones. The present study was aimed to study the influence of abiotic factors on prevalence of tasar silkworm pests. The study was conducted at different agro-climatic regions viz., Central Tasar Research &Training Institute, Ranchi, Jharkhand, Regional Extension Centre, Katghora, Chattisgarh and Regional Extension Centre, Hatgamaria during 2010-13 covering 3 seed crop and 6 commercial crops. Data on incidence of tropical tasar silkworm endo-parasitoids like Uzi Fly, Blepharipa zebina Walker and Ichneumon fly (Yellow Fly), Xathopimpla pedator, Fabricius and Predators such as Stink bug (Eocanthecona furcellata Wolf), Reduviid bug (Sycanus collaris Fabricius) and Wasp (Vespa orientalis Linnaeus) was recorded Weekly. The meteorological data was collected daily. Data was collected from 4 different agro-climatic zones of tasar growing areas. Analysis of the data revealed a significant negative correlation between abiotic factors and incidence of ichneumon fly and uzi fly. Based on the 3 years data on prevalence of pests region-wise pest calendars and prediction models were developed.

Infection characteristics of Metagonimus species (Digenea: Heterophyidae) metacercariae in fish from major rivers of Korea

  • This article analyzed the infection characteristics of metacercariae of Metagonimus spp. (MsMc) in fish from 9 major water systems in Korea. A total of 19,568 fish in 87 species were examined over a period of 10 years (2011-2020). MsMc were detected in fish from all 44 survey areas in 9 water systems. Most of the surveyed sites showed very low and low infection levels (66.7%), while 33.3% of the areas, such as Tamjin-gang and Seomjin-gang, revealed moderate and high infection levels. High endemicity depends on the abundance of susceptible fish species, especially sweet smelt (Plecoglosus altivelis). The susceptibility index (SI) with MsMc in index fish, Zacco spp., was very low and low levels in 62.0%, moderate in 28.0%, and high in 10.0% regions. The SI was highest in the following order: Yeongam-cheon (283.8), Hoeng-cheon (192.3), Togyo-jeosuji (131.2), Deokcheon-gang (119.1), and Joyang-gang (106.3). The recent infection status of MsMc in P. altivelis was analyzed by the survey localities. In addition, except for P. altivelis, 9 fish species were highly infected with MsMc in some survey areas, including Zacco platypus, Z. koreanus, Z. temminckii, Opsariichthys uncirostris, Rhynchocypris oxycephalus, Carassius auratus, Acheilognathus rhombeus, Onchorhynchus masou, and Tribolodon hakonensis. In Korea, 74 fish species (15 families) are collectively listed as second intermediate hosts of Metagonimus spp. This review provides several novel characteristics of MsMc infection and clarifies the fish species of second intermediate host of Metagonimus spp. in this country.

한강납줄개의 조개유생 보육에 따른 작은말조개 내 산란 특성 (Spawning Characteristics of Hangang Bitterling, Rhodeus pseudosericeus (Pisces: Acheilognathinae) in the Host Mussel with Glochidia)

  • 본 연구는 2017년 4~6월까지 흑천 일대에서 한강납줄개의 조개 유생보육에 따른 작은말조개 내 산란 특성에 관한 연구를 실시하였다. 조사기간 동안 확인된 조개는 327개체이었고, 글로키디아 유생을 보육한 조개 중 한강납줄개의 산란이 확인된 조개는 34개체(14.1%)로 나타났다. 아가미 위치에 따른 글로키디아 유생을 보육한 조개와 보육하지 않은 조개에서 확인된 알과 자어수는 왼쪽 외반새 5.0±3.58개(range, 1~8) vs. 2.8±2.13개 (1~9), 왼쪽 내반새 1.5±0.96개 (1~3) vs. 1.7± 0.82개(1~3), 오른쪽 내반새 1.4±0.51개(1~2) vs. 2.1±1.85개(1~6), 오른쪽 외반새 6.4±4.82개(1~20) vs. 3.0±1.86개(1~6)로 나타났다. 아가미 위치에 따른 글로키디아 유생을 보육한 조개와 보육하지 않은 조개에서 확인된 알과 자어의 출현빈도는 왼쪽 외반새 82.6% vs. 73.4%, 왼쪽 내반새 17.4% vs. 9.4%, 오른쪽 내반새 26.1% vs. 15.6%, 오른쪽 외반새 58.7% vs. 67.2%로 나타났다. 글로키디아 유생을 보육한 조개와 보육하지 않은 조개에서 확인된 알과 자어수는 8.5±6.43개(1~23) vs. 4.5±3.38개(1~14)였다. 한강납줄개 알과 자어수 그리고 출현빈도는 글로키디아 유생을 보육한 조개와 외반새에서 높은 것으로 나타났다.

Plerocercoids of Nybelinia surmenicola (Cestoda: Tentacularidae) in Squids, Todarodes pacificus, from East Sea, the Republic of Korea

  • A visceral helminth of the squid, Todarodes pacificus, is reported from the East Sea, the Republic of Korea. Total 39 squid samples were purchased from a fish market in Jumunjin-eup, Gangneung-si (City) from August 2014 to July 2015 and were examined for helminth parasites with naked eyes and under a stereomicroscope after opening the abdominal cavity with a pair of scissors. Whitish larval worms were mainly found in the stomach and abdominal cavity of the squid. They were detected in 25 (64.1%) out of 39 squids examined, and the infection density was 7 larvae per infected squid. Spatula-shaped larvae were $8.2{\times}2.0mm$ in average size, round to slightly flattened anteriorly, with round hatching posteriorly, and had characteristic 4 tentacles with numerous hooklets in the scolex. The larvae were identified as the plerocercoid stage of Nybelinia surmenicola by their morphological features. This finding represents a new host record and the first report of N. surmenicola infection in T. pacificus squids from the east coast of Korea.

한국 남해안산 농어에 기생한 흡충류 2종 (Two digenean parasites infestations of sea bass, Lateolabrax japonicus(Cuvier), form the Korean southern sea)

  • 한국 남해안에서 채집된 농어의 장에서 흡충류 2종-Tergestia laticollis, Biovarium cryptocotyle-이 검출되었다. T. laticollis는 구흡반의 주위로 거꾸로된 왕관모양의 두엽들과 인두주변에 6쌍의 판자모양 구조물 및 충란의 크기 등에 의해 다른 종들과 구분되어졌으며, B. cryptocotyle의 가장 특징적인 점은 난소가 2분지된 점이었다. 이 두 종은 우리나라에서 처음 기록되는 미기록종들이었으며, 농어는 T.laticollis의 새로운 종숙주임이 금번 연구를 통해 밝혀졌다.
