Species A rotaviruses (RVAs) replicate and assemble their immature particles within electron dense compartments known as viroplasms, where lipid droplets (LDs) interact with the viroplasm and facilitate viral replication. Despite the importance of LD formation in the life cycle of RVAs, the upstream molecules modulating LD formation remain unclear. This study aimed to find out the role of sterol regulatory element-binding proteins (SREBPs) in reprogramming of LD formation after RVA infection. Here, we demonstrate that RVA infection reprograms the sterol regulatory element-binding proteins (SREBPs)-dependent lipogenic pathways in virus-infected cells, and that both SREBP-1 and -2 transactivated genes, which are involved in fatty acid and cholesterol biosynthesis, are essential for LD formation. Our results showed that pharmacological inhibition of SREBPs using AM580 and betulin and inhibition of their downstream cholesterol biosynthesis (simvastatin for HMG-CoA reductase) and fatty acid enzymes (TOFA) negatively modulated the intracellular triacylglycerides and cholesterol levels and their resulting LD and viroplasm formations. Interestingly, pharmacological inhibition of SREBPs significantly reduced RVA protein synthesis, genome replication and progeny production. This study identified SREBPs-mediated lipogenic reprogramming in RVA-infected host cells, which facilitates virus replication through LD formation and its interaction with viroplasms, suggesting that SREBPs can be a potential target for the development of efficient and affordable therapeutics against RVA infection.
This experiments was performed to determine the effects of Gangzitongmaekeum(降脂通脈飮 : GTE) on antioxidation activity and hyperlipidemia induced by hypercholesterolemic diet in mice. The results obtained were as follows : 1. GTE showed a safety in cytotoxicity and toxicity of human fibroblast cells and liver. 2. GTE showed DPPH scavenging activity, superoxide dismutase (SOD)-like activity and inhibitory effect on reactive oxygen species (ROS). 3. GTE treated mice showed body and liver weight decrease, compared to the hyperlipidemia-induced control group. 4. GTE decreased total cholesterol and LDL cholesterol levels significantly, but HDL cholesterol levels not significantly. 5. GTE decreased triglyceride levels significantly. 6. Glucose levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. 7. Albumin levels in GTE treated mice were similar with the hyperlipidemia-induced control group. 8. The lipophagy in liver compared with the control group tended to be decreased in GTE treated mice. In the change of aorta, the cell was regular and boundary of vessel wall was clear, compared to the hyperlipidemia-induced control group. 9. TBARS levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. 10. The change of SOD and catalase activity significantly increased compared to the hyperlipidemia-induced control group. 11. ACAT mRNA level and HMG-CoA reductase mRNA levels in GTE treated mice significantly decreased compared to the hyperlipidemia-induced control group. Conclusion : These results suggest that Gangzitongmaekeum is effective in antioxidation activity and dietary hyperlipidemia-induced mice.

This study examined the effect of hesperidin supplementation with an ethanol diet on lipid and antioxidant metabolism in rats. Male Sprague-Dawley rats were divided into two groups (n=10), and were assigned to one of two dietary categories: $E_8$, ethanol diet (50 g/L) for 8 wks; $E_8H_4$, ethanol diet for the first 4 wks and hesperidin (0.02%, w/w) supplemented ethanol diet for the last 4 wks. The plasma and hepatic lipids, hepatic cholesterol regulating enzyme activity, hepatic antioxidant enzyme activity and lipid peroxidation were determined. Supplementation with hesperidin for the last 4 wks during the 8 wks period of the ethanol diet, significantly increased the ADH activity. In conjunction with the chronic administration of ethanol, hesperidin supplementation resulted in a significant decrease in the hepatic cholesterol and triglyceride concentrations compared to the $E_8$ group. The hepatic HMG-CoA reductase and ACAT activities were significantly lower in the hesperidin-supplemented group. When comparing hepatic antioxidant enzyme activities, SOD, GSH-Px, and G6PD activities and GSH level were significantly higher in the $E_8H_4$ group than in the E8 group. Plasma TBARS levels were significantly lower in rats fed ethanol with hesperidin compared to the rats fed only ethanol; however, the hepatic TBARS levels were not significantly different between the groups. Accordingly, the additional hesperidin supplement with an ethanol diet might be effective for improving the hepatic lipid metabolism and antioxidant defense system.
The objective of this study was to investigate the effects of supplementation of an antiobese functional formula (FC-GT) on body weight and lipid metabolism in rats fed a high-fat diet. Three groups of male Sprague-Dawley rats were fed different diets for 6 weeks: normal control (NC), high-fat (HF), and high-fat supplemented with powdered antiobese functional formula (FC-GT) (5% wt/wt) groups. Although body weight was not significantly different among the groups, relative weights of epididymal and perirenal white adipose tissues were significantly lower in the FC-GT group than in the HF group. FC-GT supplementation significantly lowered the plasma total cholesterol and triglyceride concentrations, whereas it elevated the ratio of HDL-C/total-C and improved the atherogenic index. Hepatic cholesterol and triglyceride concentrations were significantly lowered in the FC-GT group compared to the HF group. The accumulation of hepatic lipid droplets and the epididymal white adipocyte size of the FC-GT group were diminished compared to the HF group. Hepatic HMG-CoA reductase activity was significantly lower in the FC-GT group than in the HF group. Plasma GPT activity was significantly lowered in the FC-GT group compared to the HF group. Additionally, fecal weight was significantly increased in the FC-GT group than in the HF group. In addition, contents of fecal triglyceride and cholesterol were significantly higher in the FC-GT group compared to the other groups. The antioxidant activities of hepatic SOD, CAT, and GR were significantly increased in the FC-GT group compared to the HF group. Hepatic mARS and plasma mARS levels were significantly lowered in the FC-GT group compared to the NC group. Accordingly, we conclude that supplementation of FC-GT improves plasma and hepatic lipid levels in high-fat fed rats.

This study was purposed to investigate the effect of onion or beet on plasma and liver lipids, erythrocyte Na efflux channels and platelet aggregation in simvastatin (SIM) treated hypercholesterolemic rats. Forty Sprague Dawley rats were divided into four groups and fed 0.5% cholesterol based diets containing 2 mg/kg BW simvastatin or simvastatin with 5% onion or beet powder. Plasma total cholesterol was significantly increased in SIM group compared with the control (p<0.01), and the elevated plasma total cholesterol of SIM group was significantly decreased in SIM-onion and SIM-beet groups (p<0.05). HDL-cholesterol in SIM-beet group was significantly increased compared with other groups (p<0.05). Platelet aggregation in both the maximum and initial slope was significantly decreased in SIM group compared with SIM-onion group (p<0.05). Na-K ATPase was significantly decreased in SIM group compared with the control, SIM-onion and SIM-beet groups (p<0.05). Na passive leak was significantly increased in all groups treated with SIM compared with the control (p<0.05). The total Na efflux was decreased in SIM group and increased in SIM-onion group and the difference between these two groups was significant (p<0.05). There was no difference in intracellular Na among groups. In present study, simvastatin, a HMG CoA reductase inhibitor at dose of 2mg/kg BW/day rather increased plasma total cholesterol in rats, inferring that the action mechanism of simvastatin on cholesterol metabolism differ between rat and human. Onion and beet play favorable roles in cardiovascular system by restoring the reduced Na efflux through Na-K ATPase and Na-K cotransport in SIM treated rats.
일반사료, 고콜레스테롤사료 또는 포화지방함량이 높은 사료를 섭취한 흰쥐에서 항혈전작용과 혈액중성지방 및 콜레스테롤 수준에 관한 $\beta$-cyclodextrin($\beta$CD)의 급여효과를 조사하였다. 출혈시간은 일반사료, 고콜레스테롤 및 고지방사료 섭취군 모두에서 $\beta$CD 첨가구가 대조구에 비해서 각각 293%, 157% 및 218% 까지 유의적으로 연장되었다(p<0.05). 전혈응고시간은 일반사료, 고콜레스테롤 및 고지방사료섭취군에서 $\beta$CD 첨가구가 대조구와 비교할때 각각 202%, 168% 및 211%까지 유의적으로 높았다(p<0.05). $\beta$CD 첨가사료는 흰쥐 혈액내 총지질, 중성지방, 총콜레스테롤 및 저밀도지질단백질 콜레스테롤 함량을 현저하게 낮추는 원인이 되었다. 혈액내 총지질 함량은 일반사료, 고콜레스테롤 및 고지방사료 섭취군에서 $\beta$CD첨가구가 대조구에 비해서 각각 70%, 82% 및 87%까지 유의적으로 낮아졌다(p<0.05). 혈액내 중성지방함량은 일반사료, 고콜레스테롤 및 고지방사료 섭취군에서 $\beta$CD 첨가구가 대조구와 비교할 때 각각 89%, 43% 및 59%까지 유의적인 감소를 나타냈다(p<0.05). 혈액내 총콜레스테롤 함량은 일반사료, 고콜레스테롤 및 고지방사료 섭취군에서 $\beta$CD 첨가구가 대조구에 비해서 각각 28%, 62% 및 36%까지 유의적으로 낮아졌다(p<0.05). 혈액내 저밀도 지질단백질 콜레스테롤 함량은 일반사료, 고콜레스테롤 및 고지방사료섭취군에서 $\beta$CD 첨가구가 대조구에 비해서 각각 39%, 54% 및 25%까지 유의적인 감소를 나타냈다(p<0.05). 혈액내 총담즙산 함량은 일반사료, 고콜레스테롤 및 고지방사료섭취군에서 $\beta$CD 첨가구가 대조구에 비해서 각각 66%, 95% 및 97% 까지 유의적인 증가를 보였다(p<0.05). 일반사료 섭취군에서 $\beta$CD 첨가구의 HMG-CoA reductase 활성도는 대조구와 비교할 때 41%까지 유의적으로 낮아졌으며 분을 통한 스테로이드의 배설량은 167%까지 유의적인 증가를 나타냈다(p< 0.05). 이 결과는 $\beta$CD가 사람의 심장혈관계질환과 비만을 예방하는데 도움이 될 수 있는 혈액내 지질, 중성지방 및 콜레스테롤의 감소효과 그리고 항혈전작용에 관한 생리활성 기능을 갖는다는 것을 시사해 주고 있다.
기존 연구에서 주로 사용하는 기타 재래종 검정콩 종피추출물 (콩단백질 혹은 총 안토시안)의 효과를 연구하는 경우와 달리, 본 연구에서는 재래종 서리태보다 C3G와 D3G 안토시안이 2배 이상 풍부한 검정콩 CJ-3호 종피추출물을 이용하였다. STZ로 유도된 당뇨쥐에 4주간 검정콩 CJ-3호 종피추출물을 처치한 결과 비록 체중감소, 혈당강하, 인슐린저항성 완화 등의 효과는 미비하지만 근육량이 유의적으로 증가하였는데 이는 일반적으로 항비만 식이요법에 따라 나타나는 근육소모를 보완하는 중요한 효과이다. STZ유도 당뇨 쥐에서 억제된 글리코겐이용률이 검정콩 CJ-3호 종피추출물에 의하여 증가한 것은 포도당이 에너지원으로 사용되면서 지방조직의 지질이용률을 억제하여 혈중 TG를 개선시켰고 간에서 HMG-CoA reductase 활성을 조절함으로써 혈청 TC를 낮추는데 기여하였다. 4주간의 투여기간을 연장한다면 향후 당뇨에서 발생하는 인슐린저항성 및 당화적 산화성억제에 따른 지질의 과산화물의 생성을 억제할 수 있는 가능성도 기대할 수 있겠다. 또한 검정콩 CJ-3호는 병 저항성이 높고 종실특성이 우수한 녹자엽 검정콩으로 생산성이 높아서 기능성에 대한 반복 실험을 확인한다면 기능성식품시장의 산업화도 가능하다. 따라서 C3G와 D3G 안토시안이 풍부한 기능성 식품의 경우 당뇨, 비만 및 암 등 난치병질환에서 나타나는 혈중지질 개선을 위한 임상적 효과 또한 기대해 볼 수 있겠다.
본 연구는 고지방 (열량의 37%를 지방으로 대체)을 급여한 마우스의 체중과 체내 지질함량에 미치는 보리순의 영향을 규명하고자 하였다. 4주령의 ICR 마우스 (n = 32)를 1주일간 적응시킨 후 정상식이를 급여한 정상군, 고지방을 급여한 고지방대조군, 고지방-보리순분말군과 고지방-보리순열수추출물군으로 나누어 8주간 사육하였다. 보리순은 사람이 하루에 3잔의 차를 마시는 양을 고려하여 보리순 1%수준이 섭취되도록 분말과 열수추출물을 각각 식이에 첨가 조제하여 8주간 급여하였다. 식이섭취량은 보리순분말군이 고지방대조군에 비하여 유의적으로 낮았으나 보리순열수추출물은 식이섭취량에 영향을 미치지 않았다. 보리순분말과 보리순열수추출물은 고지방식이로 유도된 비만마우스의 체중을 효과적으로 낮추었으며 특히, 내장지방무게의 대표적 지표인 부고환지방조직 무게를 유적으로 낮추었다. 이는 보리순이 식이섭취 억제에 의존적으로 체중감소 효과를 나타내지 않음을 제시한다. 혈장의 총 콜레스테롤 함량은 보리순분말과 열수추출물 급여시 고지방 대조군에 비해 각각 25%와 20%의 감소효과를 보였으며 중성지질 함량은 보리순분말군과 열수추출물군 모두 28%와 43%의 유의적인 감소를 보였다. 또한 보리순분말과 보리순열수추출물은 혈장 중의 총 콜레스테롤에 대한 HDL-콜레스테롤 비(HTR)를 고지방대조군에 비하여 유의적으로 높였다. 간조직과 지방조직의 트리글리세리드 함량은 정상군에 비해 고지방대조군이 각각 1.3배, 1.2배의 증가를 보였으나 보리순분말과 열수추출물 급여시 모두 간조직과 지방조직의 트리글리세리드 함량이 정상군과 유사한 수준으로 개선되었다. 신장에서의 트리글리세리드 함량은 고지방식이로 증가하는 경향을 보였고 심장에서는 1.4배의 유의적인 증가를 나타내었으며, 보리순열수추출군에서만 유의적인 개선효과를 볼 수 있었다. 간조직과 신장의 콜레스테롤 함량은 실험군간에 차이가 없었으나, 심장 중의 콜레스테롤 함량은 고지방대조군이 정상군보다 유의적으로 높았다. 보리순열수추출물의 경우 심장의 콜레스테롤 함량을 유의적으로 낮추었다. 변중의 콜레스테롤 함량은 고지방 대조군이 정상군에 비하여 낮은 반면, 트리글리세리드 함량은 약 6배 높았다. 한편, 보리순말과 열수추출물 급여는 고지방 대조군에 비하여 변으로의 트리글리세리드 배설을 유의적으로 증가시켰으나 콜레스테롤 함량에는 영향을 미치지 않았다. 간조직 중 FAS활성은 정상군에 비해 고지방대조군에서 1.7배 높아진 반면, ${\beta}$-oxidation은 유의적으로 낮았다. 보리순분말과 열수추출물은 지방산 합성효소의 활성을 정상군 수준으로 개선하였으나 지방산 산화에는 영향을 미치지 않았다. 콜레스테롤 합성에 관련하는 HMG-CoA reductase와 ACAT 활성은 각각 정상군보다 고지방대조군에서 각각 2.1배, 1.5배씩 높았으나 보리순분말과 열수추출물 급여시 유의적으로 낮았다. 이와 같이 보리순분말과 열수추출물은 고지방 식이로 비만을 유도한 마우스의 간조직에서 지방산과 콜레스테롤 합성을 저해함으로써 지질개선에 효과적인 것으로 나타났다.

This study examined the effects of Lactobacillus acidophilus ATCC 43121 (LAB) on cholesterol metabolism in hypercholesterolemia-induced rats. Four treatment groups of rats (n=9) were fed experimental diets: normal diet, normal $diet+LAB(2{\times}10^6\;CFU/day)$, hypercholesterol diet (0.5% cholesterol, w/w), and hypercholesterol diet+LAB. Body weight, feed intake, and feed efficiency did not differ among the four groups. Supplementation with LAB reduced total serum cholesterol (25%) and VLDL+IDL+LDL cholesterol (42%) in hypercholesterol diet groups, although hepatic tissue cholesterol and lipid contents were not changed. In the normal diet group, cholesterol synthesis (HMG-CoA reductase expression), absorption (LDL receptor expression), and excretion via bile acids (cholesterol $7{\alpha}-hydroxylase$ expression) were increased by supplementation with LAB, and increased cholesterol absorption and decreased excretion were found in the hypercholesterol diet group. Total fecal acid sterols excretion was increased by supplementation with LAB. With proportional changes in both normal and hypercholesterol diet groups, primary bile acids (cholic and chenodeoxycholic acids) were reduced, and secondary bile acids (deoxycholic and lithocholic acids) were increased. Fecal neutral sterol excretion was not changed by LAB. In this experiment, the increase in insoluble bile acid (lithocholic acid) reduced blood cholesterol level in rats fed hypercholesterol diets supplemented with LAB. Thus, in the rat, L. acidophilus ATCC 43121 is more likely to affect deconjugation and dehydroxylation during cholesterol metabolism than the assimilation of cholesterol into cell membranes.

제 5 장 계약 해지 및 이용 제한

제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
