Dried fruits of Amomum villosum Lour. have been used an korean medicine to treat digestive diseases for a long time. It has been reported that Amomum villosum extracts(AVE) have effects for diabetes and steatosis in experimental models. But we did not find the report about the cholesterol synthesis inhibition effects of AVE. The objective of this study is to clarify the inhibitory effect of AVE against oleic acid and glucose-induced hypercholesterolemia in HepG2 cells. The results show that AVE had a significant inhibitory effect against oleic acid and glucose-induced cholesterol accumulation. Those effects seem to be caused by inhibition of AVE on oleic acid and glucose-induced decrease of HMG CoA reductase, which is the rate-limiting enzyme for cholesterol biosynthesis in liver. It is believed that the results of this study can provide basic data for the drug and functional food development of hypercholesterolemia treatments.
Interaction of dietary Magnesium, Calcium and Polyunsaturated fatty acid(vegetable oils)on 3-Hydroxy-3-Methylglutaryl Co-A reductase activity was studied for a period of 30 days using isocalories and isonitrogenous as a basal diet . The subject rabbits were divided into 18 feeding groups. The results are summarized as follows: 1. The ratio of ${\alpha}_1-lipoprotein$ par lipalbumin is 0.34 for control group, 0.38 the highest group fed 0.1 Mg(II) 10ml plus perilla oil and basal diet, the lowest 0.25 group fed 0.1M Mg(II) 5ml plus sesame oil and basal diet. 2. The ratio ratio of ${\alpha}_2-lipoprotein$ per lipalbumin is 0.64 for control group. 0.95 as the highest for the group fed 0.1M Ca(II) 15ml plus sesame oil and basal diet, 0.1M Ca(II) 5ml plus perilla oil and basal diet. 3. The ratio of ${\beta}-lipoprotein$ per lipalbumin is 0.71 for control group, the highest 0.81 for the groups fed 0.1M Mg(II) 10ml plus sesame oil and basal diet, the lowest 0.37 for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 4. In serum triglyceride, control group was 129.5mg%, the highest 155.4mg% for the group fed 0.1M Ca(II) 5ml plus sesame oil and basal diet, the lowest 85.7mg% for the group fed 0.1M Mg(II) 10ml plus soybean oil and basal diet. 5. In serum cholesterol, control group was 96.7mg%, the highest 152.5mg% for the group fed 0.1M Ca(II) 10ml plus sesame oil and basal diet, the lowest 80.5mg% for the group fed 0.1M Mg(II) 15ml plus soybean oil and basal diet. 6. In case of HMG-CoA reductase activity, control group was 0.95, the highest 0.98 for the group fed 0.1M Ca(II) plus soybean oil and basal diet. 7. Interaction between metal(II) ions and polyunsaturated fatty acid(vegetable oil) are soybean oil>sesame oil>perilla oil, for Mg(II). soybean oil>perilla oil>sesame oil, for Ca(II). Therefore, it is invetigated that the interaction between metal ion and polyunsaturated fatty acid is the higher, the cholesterol level is the lower, and HMG-CoA reductase activity is increased.

Synthesized Ile-Ala-Val-Pro (IAVP) peptide, which has the highest hypocholesterolemic effect among a number of synthesized derivatives of Ile-Ala-Val-Pro-Gly-Glu-Val-Ala (IAVPGEVA) isolated from 11S globulin of soy protein by pepsin digestion, was selected for investigation in the present study. Using a recombinant Syrian hamster 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), we studied in detail the inhibition of this enzyme by IAVP and compared the action of this peptide to that of lovastatin, a known competitive inhibitor of this enzyme. The concentration of IAVP required for 50% inhibition ($IC_{50}$) of HMGR activity in given experimental conditions was $340\;{\mu}M$. Kinetic analysis revealed that the studied peptide is a competitive inhibitor of HMGR with respect to both 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) and nicotinamide adenine dinucleotide phosphate (NADPH), with an equilibrium constant of inhibitor binding ($K_i\;=\;[E][I]/[EI]$) of $61{\pm}1.2\;{\mu}M$ and $157{\pm}4.4\;{\mu}M$, respectively. At the same conditions, $K_i$ and $IC_{50}$ for lovastatin were $2.2{\pm}0.1\;nM$ and 12.5 nM, respectively. Thus, the given peptide interacts with HMGR as a bisubstrate, consequently blocking access of both substrates to the active sites. The achieved results suggest the design of new peptide sequences having a higher relative affinity to binding sites of this enzyme and an enhancement of their hypocholesterolemic properties.
Hypercholesterolemia is one of the primary risk factors for cardiovascular diseases. The use of lactobacilli probiotics to reduce blood cholesterol levels have been extensively reported. However, more information is needed to evaluate the possible mechanisms involved and to identify possible targets for further therapeutic development. In this study, strains of lactobacilli were screened based on the ability to assimilate cholesterol, and prevention of cholesterol accumulation in hepatic (HepG2) and intestinal (HT-29) cells. Cell free supernatant (CFS) from Lactobacillus plantarum DR7 showed a higher ability to assimilate cholesterol, reduction in cholesterol accumulation in both HepG2 and HT-29 cells, accompanied by reduced mRNA expression of HMG-CoA reductase (HMGCR) in HepG2 (p<0.05), compared to other lactobacilli. The reduction of HMGCR expression was also diminished in the presence of an AMPK inhibitor (Compound C), suggesting that L. plantarum DR7 exerted its effect via the AMPK pathway, typically via the phosphorylation of AMPK instead of the AMPK mRNA expression in HepG2 (p<0.05). Altogether, our present study illustrated that lactobacilli could exert cholesterol lowering properties along the AMPK pathway, specifically via phosphorylation of AMPK that led to reduced expression of HMGCR.
The aim of the present study was to investigate the hypocholesterolemic effect and potential of tyramine derivatives from Lycii Cortex Radicis (LCR), the root bark of lycium (Lycium chenese Miller) in reducing lipid peroxidation. The activities of enzymes, hepatic 3-hydroxy 3-methylglutaryl (HMG) CoA reductase and acyl-CoA:cholesterol acyltransferase (ACAT) and LDL oxidation were measured in vitro and animal experiments were also performed by feeding LCR extracts to rats. The test compounds employed for in vitro study were trans-N-p-coumaroyltyramine (CT) and trans-N-feruloyltyramine (FT), LCR components, N-(p-coumaroyl)serotonin (CS) and N-feruloylserotonin (FS) from safflower seeds, ferulic acid (FA) and 10-gingerol. It was observed that FT and FS at the concentration of 1.2 mg/mL inhibited liver microsomal HMG CoA reductase activity by ~40%, but no inhibition of activity was seen in the cases of CT, CS, FA and 10-gingerol. Whereas, ACAT activity was inhibited ~50% by FT and CT, 34-43% by FS and CS and ~80% by 10-gingerol at the concentration of 1 mg/mL. A significant delay in LDL oxidation was induced by CT, FT, and 10-gingerol. For the animal experiment, five groups of Sprague-Dawley male rats were fed high fat diets containing no test material (HF-control), 1 and 2% of LCR ethanol extract (LCR1 and LCR2), and 1% of extracts from safflower seed (Sat) and ginger (Gin). The results indicated that total cholesterol level was significantly lower in Saf, LCR2 and Gin groups, and HDL cholesterol level was lower only in Gin group when compared with HF-control group; while there was no difference in the serum triglyceride levels among the five experimental groups. The level of liver cholesterol was significantly lower in LCR1 and LCR2 groups than HF-control Serum levels of TBARS were significantly lower only in LCR2 group when compared with HF-control group. From the observed results, we concluded that LCR can be utilized as a hypocholesterolemic ingredient in combination with ginger, especially for functional foods.
Objectives: We performed this study to evaluate the antioxidative and hypolipidemic effect of Artemisia iwayomogi (韓茵蔯), Curcuma longa L. (鬱金) and Raphanus sativus L. (蘿?子) (ACR). Method: We enriched Artemisiae Capillaris, Curcumae Longae and Raphani Semen compound with alcohol. ACR extract is treated to HepG2 cell. Cell groups are devided into 3 groups: normal, control and ACR treated group. We measured polyphenol, flavonoids, DPPH and ABTS radical scavenging activity, ROS, glutathione, GSH peroxidase, GSH reductase, SOD, catalase, free fatty acid, lipid peroxidation and suppression of ACAT1 and HMG-CoA reductase expression on mRNA level. Results: 1. ACR contained polyphenol and flavonoids and increased GSH significantly in HepG2 cell. 2. ACR increased GPx, GR, and catalase activity significantly in HepG2 cell. 3. ACR increased DPPH and ABTS radical scavenging activity significantly in HepG2 cell and decreased ROS. 4. ACR decreased free fatty acid and MDA significantly in HepG2 cell. 5. ACR suppressed ACAT1 and HMG-CoA reductase expression on mRNA level in HepG2 cell. Conclusion: This study suggests that ACR has antioxidative and hypolipidemic effect and might be effective in prevention and treatment of dyslipidemia.
본 연구에서는 Bacillus subtilis NUC1균주로 발효한 탈지대두 grits 발효물인 FD와 탈지대두 grits에 녹두를 2.5, 5, 10%로 각각 첨가하여 발효한 FDM을 각각 제조하여 in vitro 상에서의 콜레스테롤 개선능을 검색하였다. FD와 FDM군의 콜레스테롤 흡착능을 살펴본 결과, 모든 군에서 70% 이상의 흡착능을 보였다. 특히 2.5% 녹두를 첨가하여 발효시킨 FDM군(2.5% FDM)은 90%의 가장 높은 흡착능을 보였다. 2.5% FDM군은 42%의 HMG-CoA reductase 저해 활성을 보였고, 또한 HepG2 세포를 이용하여 측정한 세포 내의 콜레스테롤 함량과 apolipoprotein AI, CIII의 개선효과에서도 가장 우수한 개선효과를 보였다. 따라서 2.5% 녹두를 첨가하여 발효시킨 FDM군은 고콜레스테롤 예방에 도움을 줄 것으로 생각된다.
본 연구는 고콜레스테롤 식이를 섭취한 랫드에서 파리유충 에탄올추출물(Ethanolic extract of fly maggot, EM)의 경구투여가 혈액 지질감소에 미치는 영향을 조사하였다. Sprague-Dawley 수컷 랫드를 이용하여 4 처리구(EM 투여량; 대조군=0, 5.0, 7.0, 9.0 mg/100 g 체중)로 구분해서 6주 동안 진행하였다. EM 투여군은 대조군과 비교할 때 혈청 중성지방, 총콜레스테롤, LDL-C가 유의하게 낮았다(p<0.05). HMG-CoA reductase activity는 대조군과 비교할 때 EM 투여군에서 낮았으나 총스테롤, 중성스테롤 및 담즙산 배설량은 EM 투여군에서 유의하게 높았다(p<0.05). EM의 혈액 콜레스테롤 감소와 관련한 생물학적 작용기작을 규명하기 위해서 고콜레스테롤 식이를 섭취한 랫드에서 유도된 생체유전자 sterol response element binding proteins (SREBPs) 및 the peroxisome proliferator-activated receptors ($PPAR{\alpha}$) 발현을 측정하였다. EM은 고콜레스테롤 식이를 공급받은 랫드의 간에서 SREBP-$1{\alpha}$, SREBP-2 mRNA 발현을 억압함과 동시에 $PPAR{\alpha}$ mRNA 발현을 촉진시키는 것으로 나타났다(p<0.05). 본 연구의 결과는 파리유충 에탄올추출물이 고콜레스테롤 식이를 섭취한 랫드에서 지질대사와 관련한 생화학적 매개변수 및 유전자발현 조절을 통하여 혈액 콜레스테롤을 낮춘다는 새로운 사실을 발견하였다.
본 연구는 차세대 랫드에서 혈액 지질 감소에 관한 식이 내 오메가 6와 3 지방산 비율의 생화학적 메카니즘을 조사하였다. 실험처리군은 오메가 6:오메가 3비율 0 (대조구), 1:1, 8:1, 19:1로 구분하였다. 혈액 중성지방, 총콜레스테롤, 저밀도지질단백질(LDL-C), aspartate aminotransferase (AST), alanine aminotransferase (ALT), glucose의 수준은 오메가 6:오메가 3 비율 1:1 그룹에서 가장 낮았다. 혈액 고밀도지질단백질(HDL-C), 인지질의 수준은 오메가 6:오메가 3 비율 1:1 그룹이 가장 높았다. HMG-CoA reductase activity는 전체 저리구 가운데 오메가 6:오메가 3 비율 1:1, 8:1 그룹에서 가장 강하게 억제되었으나 분을 통한 스테롤의 배설량은 촉진되었다. 혈액 오메가 6:오메가 3 비율은 섭취한 식이 내 오메가 6:오메가 3 비율 증가에 따른 농도 의존적으로 감소하였다. 본 연구결과는 부모 세대로부터 차세대 랫드에 이르기까지 오메가 6:오메가 3 비율이 8:1 이하로 낮아진 식이를 섭취하면 영양소 대사작용 활성화 메카니즘에 의한 혈액 중 유해지질 감소, 간 기능 유지 및 성장을 자극한다는 새로운 사실을 나타낸다.
8주령의 F1B golden Syrian 햄스터에게 동맥경화 유발식이와 함께 녹차 추출물을 각각 500 혹은 1,000 mg/kg b.w.의 양을 매일 경구투여하면서 6주간 사육하였을 때 햄스터 체내의 지질대사와 대동맥 내에서의 지방의 축적 정도에 미치는 영향을 조사하였다. 실험 결과 녹차추출물의 경구투여는 동맥경화유발식이를 섭취하는 햄스터의 혈중 중성지방과 총콜레스테롤치를 농도의존적으로 감소시켰고, 대동맥궁내에서의 지방의 축적을 예방하였다. 특히 녹차추출물 1,000 mg/kg b.w. 투여는 동맥경화유발식이를 섭취한 햄스터에서 간장내의 LDL receptor mRNA level을 증가시키는 경향을 보였다. 이러한 결과로 볼 때 녹차 추출물의 투여는 동맥경화유발식이를 섭취한 햄스터의 혈중 콜레스테롤치를 감소시키고 LDL receptor의 발현을 증가시킴으로써 동맥경화를 예방할 수 있음을 보여주었다.
