In the course of screening for microbes with nematicidal activity, we found that Enterobacter asburiae HK169 displayed promising nematicidal activity against the root-knot nematode Meloidogyne incognita, along with plant growth-promoting properties. Soil drenching of a culture of HK169 reduced gall formation by 66% while also increasing root and shoot weights by 251% and 160%, respectively, compared with an untreated control. The cell-free culture filtrate of the HK169 culture killed all juveniles of M. incognita within 48 h. In addition, the nematicidal activity of the culture filtrate was dramatically reduced by a protease inhibitor, suggesting that proteolytic enzymes contribute to the nematicidal activity of HK169. In order to obtain genomic information about the HK169 isolate related to its nematicidal and plant growth-promoting activities, we sequenced and analyzed the whole genome of the HK169 isolate, and the resulting information provided evidence that the HK169 isolate has nematicidal and plant growth-promoting activities. Taken together, these observations enable the future application of E. asburiae HK169 as a biocontrol agent for nematode control and promote our understanding of the beneficial interactions between E. asburiae HK169 and plants.
To idenifty effect of Bojungbangam-tang kakambang on Cisplatin-Induced G2/M Phase Arrest in Human Renal Proximal Tubular HK-2 Cells. Cytotoxicity of cisplatin was detected in HK-2 cells and the value of IC50 is about $25\;{\mu}M$. The treatment of cisplatin to HK-2 showed the G2/M phase cell cycle arrest. The ethanol extract of Bojungbangam-tang kakambang (EBTKB), a new herbal prescription composed of ten crude herbs, inhibited cisplatin-induced G2/M phase arrest in HK-2 cells. EBTKB increased G0/G1 peak in cisplatin-treated HK-2 cells. p53, p21 and p27 expression were increased in cisplatin-treated HK-2 cells. Inhibitory effect of EBTKB on cisplatin-induced G2/M phase arrest was accomplished through inhibition of p53, p21 and p27 expression. Also, reduced CDK2 and cyclin A expression by cisplatin were increased by EBTKB, but cyclin E was not changed. Reduction of ERK activation and increment of p38 activation by cisplatin were increased ERK activation and decreased p38 activation by EBTKB. Cisplatin had no effect on JNK activation, but EBTKB increased JNK activation. These results can suggest that EBTKB inhibits cisplatin-induced G2/M phase arrest in HK-2 cell through reduction of p53-dependent p21 and p27 protein, ERK activation and p38 inactivation.
The principal objective of this study was to determine the optimal conditions for the production of biosurfactant by the indigenous bacterium, Pseudoalteromonas sp. HK-3, originating from oil-spilled areas. The relationship between total biosurfactant production and the factors affecting biosurfactant production were evaluated by statistical analysis using SPSS software. The effects of various supplemental carbon sources (e.g., glucose, dextrose, mannitol, citrate, acetate) on the maximal production of biosurfactant by the test culture of Pseudoalteromonas sp. HK-3 was then evaluated. As a result, mannitol was found in this study to be the best supplemental carbon source for the production of biosurfactant. A spot inoculation of crude cultural liquid containing the HK-3 cells generated the largest clear zone, whereas only small clear zones appeared around the spots inoculated with either supernatant only or cell pellets following centrifugation. Our results demonstrated that the HK-3 test culture supplemented with 2% mannitol at an initial pH of 6 generated the maximal amount of biosurfactant within 72 h of incubation.
본 연구는 신생아 태변에서 분리된 Lactococcus lactis HK-9의 다양한 생리적 특성과 항균활성을 조사하기 위하여 실시하였다. 균주 HK-9을 MRS 배지에서 배양하였고, 형태 및 생리학적 특성에 대하여 조사하였으며, BIOLOG 시험과 16SrRNA 염기서열 분석을 통해 균주를 동정하였다. 그 결과 Lactococcus lactis으로 동정되었고, L. lactis HK-9로 명명하였으며, 이 균주의 계통수 분석을 통해 GenBank에 [GU936712]로 등록하였다. 배양기간에 따른 L. lactis HK-9의 생장과 중간 대사산물로서 lactic acid와 acetic acid의 생산 및 pH의 변화를 조사하였으며, 이들 유기산의 생성은 HPLC를 통하여 확인하였다. 유기산의 생성은 L. lactis HK-9의 생장에 따라 증가하는 것을 확인하였으며, 배양 60시간이 지난 후 생성된 lactic acid와 acetic acid의 농도는 각각 495.6 mM, 104.3 mM이었다. 배양 초기의 pH는 7.0이었으며 배양 기간 동안 4.1로 감소하였다. 다양한 그람양성 세균과 그람음성 세균을 대상으로 농축된 HK-9 배양상등액의 항균력을 조사하였으며, 그 결과 항균범위가 그람양성 세균에서만 나타나는 것이 관찰되었다. 농축된 배양상등액을 protease를 처리한 후 항균활성은 소멸하였으며, 상등액의 박테리오신 추정 분자의 분자량은 tricine-SDS-PAGE를 통하여 약 4 kDa으로 확인되었다.
This study aimed to determine the immune-stimulating effects of heat-killed Lactobacillus plantarum Ln1 (HK-Ln1) through the production of nitric oxide (NO) and pro-inflammatory cytokine achieved by inducing NF-κB and mitogen-activated protein kinase (MAPK)-signaling pathways in macrophages. HK-Ln1 showed higher NO and cytokine production compared to control (nonstimulated lipopolysaccharide); in addition, the expression of inducible nitric oxide synthase (iNOS) was induced through HK-Ln1treatment. The phosphorylation of IκB-α and p65 increased following treatment by HK-Ln1, which implicates IκB-α degradation and the translocation of p65 to nucleus. In addition, the phosphorylation of MAPKs, ERK 1/2, JNK, and p38 was induced following HK-Ln1 treatment.
대부분의 비뉴턴 유체 모델의 경우 격자볼쯔만 법을 이용한 3차원 유동해석에 효율적으로 적용하기 어려운 문제점을 가지고 있다. 이 연구에서는 이를 해결하기 위해 개발된 격자볼쯔만법 전용 비뉴턴 유체 모델인 Hydro-Kinetic 모델을 3차원 격자볼쯔만법 해석에 적용할 수 있도록 수정하고 이를 3차원 급축소 및 급확대 유동에 적용하여 얻은 결과를 통해서 강한 전단유동장에서 HK모델 유체의 거동을 분석하였다. HK모델은 변형률과 완화시간과의 관계를 나타낸 모델로 강한 전단유동장에서 국소적으로 큰 변형률이 발생하는 경우 완화시간이 급격하게 감소하여 해석이 불안정해지는 경향이 있어 격자볼쯔만법 해석안정성을 확보하기 위해서는 HK모델의 인수 ${\Gamma}$와 완화시간을 일정한 구간에서 변화하도록 제한 할 필요가 있다.
HK shiitake mushroom mycelium (HKSMM: 14% β-glucan)은 식품의약품안전처의 개별인정형 간 건강기능식품 원료이다. HKSMM의 50% ethanol 추출물 (HKSMM50)에 대한 면역증강 효과를 Concanavalin A (ConA)로 활성화한 human T lymphocyte Jurkat cells에서 연구하였다. Active hexose correlated compound (AHCC)는 positive control로 사용하였다. ConA로 활성화한 Jurkat cells에 HKSMM50 (0, 25, 50, 100 ㎍ g/ml) 및 AHCC (100 ㎍ g/ml)를 처리하고 3시간 또는 6시간 배양하였다. Jurkat cells의 cytosol과 nucleus에 함유된 transcription factor인 nuclear factor of activated T cells (NFAT) 함량은 Western blotting으로 측정하였다. Interleukin-2 (IL-2)와 interferon-gamma (IFN-γ) 함량 및 cyclooxygenase-2 (COX-2) 활성은 enzyme-linked immunosorbent assay (ELISA) kit로 분석하였다. HKSMM50은 cytosolic NFAT protein 함량은 낮추었고, nuclear NFAT protein함량은 증가시켰다. IL-2와 IFN-γ 함량은 증가되었고, COX-2 활성과 apoptosis는 억제되었다. AHCC 효과는 HKSSM50의 효과와 유사하였다. 이와 같은 결과는 HKSMM50가 ConA로 활성화된 Jurkat cells에서 NFAT protein을 활성화시켜, IL-2와 IFN-γ 함량을 증가시켰음을 의미한다. 또한 HKSMM50은 COX-2 활성과 apoptosis를 억제하였다. 따라서 이와 같은 결과는 HKSMM이 면역증진을 위한 건강기능식품 원료로 사용할 수 있음을 의미한다.
Although previous studies on hexokinase (HK) II indicate both the N- and C-terminal halves are catalytically active, we show in this study the N-terminal half is significantly more catalytic than the C-terminal half in addition to having a significantly higher $K_m$ for ATP and Glu. Furthermore, truncated forms of intact HK II lacking its first N-terminal 18 amino acids ($\Delta$18) and a truncated N-terminal half lacking its first 18 amino acids ($\Delta$18N) have higher catalytic activity than other mutants tested. Similar results were obtained by PET-scan analysis using $^{18}F-FDG$. Our results collectively suggest that each domain of HK II possesses enzyme activity, unlike HK I, with the N-terminal half showing higher enzyme activity than the C-terminal half.
The incidence of stress-related type 2 diabetes (stress-T2D), which is aggravated by physiological stress, is increasing annually. The effects of Lactobacillus, a key component of probiotics, have been widely studied in diabetes; however, studies on the effects of postbiotics are still limited. Here, we aimed to examine the mechanism through which heat-killed Lactiplantibacillus plantarum LRCC5314 (HK-LRCC5314) alleviates stress-T2D in a cold-induced stress-T2D C57BL/6 mouse model. HK-LRCC5314 markedly decreased body weight gain, adipose tissue (neck, subcutaneous, and epididymal) weight, and fasting glucose levels. In the adipose tissue, mRNA expression levels of stress-T2D associated factors (NPY, Y2R, GLUT4, adiponectin, and leptin) and pro-inflammatory factors (TNF-α, IL-6, and CCL-2) were also altered. Furthermore, HK-LRCC5314 increased the abundance of Barnesiella, Alistipes, and butyrate-producing bacteria, including Akkermansia, in feces and decreased the abundance of Ruminococcus, Dorea, and Clostridium. Thus, these findings suggest that HK-LRCC5314 exerts protective effects against stress-T2D via gut microbiome modulation, suggesting its potential as a supplement for managing stress-T2D.

Recently, we have reported that 3-hydrogenkwadaphnin (3-HK), a diterpene ester isolated from Dendrostellera lessertii (Thymealeaceae), is very effective against leukemia cell lines without any detectable effects on normal cells (Moosavi et al., 2005b). In this study, we report that 3-HK induces $G_1$ cell-cycle arrest, differentiation and apoptosis in APL NB4 cell line. Indeed, the drug between 24 to 96 h induced 7-65% growth inhibition of NB4 cells. Cell viability was also decreased by 2-55% between 24 to 96 h treatments with the drug, respectively. These effects of the drug were also dose-dependent. According to flow cytomtry results, 3-HK (15 nM) induced a significant G1-arrest up to 24 h which was consequently followed with appearance of sub-$G_1$ peak at 72 to 96 h. Hoechst 33258 staining and DNA fragmentation assays confirmed the occurrence of apoptosis among the treated cells. On the other hand, NBT reducing assay, Wright-Giemsa staining, phagocytic activity and expression of cell surface markers (CD11b and CD14) confirmed that the inhibition of proliferation is associated with differentiation especially toward macrophage-like morphology. Interestingly, 3-HK at 5 and 10 nM enhanced the effects of all-trans retinoic acid (ATRA) in NB4 cells. Based on these results, 3-HK might become an ideal candidate for treatment of APL patients pending full exploration of its biological functions.
