연구 배경 : 허파혈관주위세포는 허파미세혈관에서 혈액공기 장벽을 이루고 있는 중요한 세포이다. 이 세포는 생리학적으로 혈류와 혈관의 투과성을 조절하는 기능이 있다. 본 연구는 급성폐손상/급성호흡곤란증후군에서 혈관 과투과성 및 개형에 혈관주위세포의 변화가 중요한 역할을 할 것으로 보고 시작하게 되었다. 흰쥐로 부터 일차 배양한 허파혈관주위세포에 저산소 상태를 만들었을 때, 세포의 생존능에 미치는 영향과 저산소증에 의해 유도되는 유전자의 발현을 살펴보았다. 방 법 : 흰쥐로부터 허파혈관주위세포를 일차 배양 및 계대 배양하였다. 광학 현미경 및 세포 면역 화학 염색으로 세포를 확인하였다. 2% $O_2$의 세포 배양기와 $200{\mu}M$$CoCl_2$를 처리하였다. 세포의 증식은 tryphan blue 염색 후 세포수를 세는 방법을 택하였다. 유전자 발현은 역전사 중합 효소 연쇄반응을 이용하였다. 결 과 : 1. 흰쥐로부터 허파혈관주위세포를 성공적으로 일차 배양 및 계대 배양할 수 있었다. 2. 2% $O_2$나 $CoCl_2$에서 혈관주위세포는 24시간, 36시간, 48시간에 증식이 억제되었다. 3. 허파혈관주위세포에 저산소 상태의 자극을 주면 VEGF와 smad-2의 발현이 현저하게 증가하였다. 4. 허파혈관주위세포의 HIF$1{\alpha}$, COX-2는 저산소상태에서 VEGF, smad-2에 비해 발현의 변화가 현저하지 않았다. 결 론 : 허파혈관주위세포를 일차 배양함으로써 허파꽈리혈관장벽의 연구를 위한 한 모델을 만들었고, 저산소 상태에서 증식의 억제와 유전자 발현의 변화를 살펴봄으로써 허파혈관손상의 기전을 규명하는데 향후 도움이 될 것으로 보인다.
Objective: This study investigated the effects of different dietary energy sources on early postmortem muscle metabolism of finishing pigs. Methods: Seventy-two barrow ($Duroc{\times}Landrace{\times}Yorkshire$, DLY) pigs ($65.0{\pm}2.0kg$) were allotted to three iso-energetic and iso-nitrogenous diets: A (44.1% starch, 5.9% crude fat, and 12.6% neutral detergent fibre [NDF]), B (37.6% starch, 9.5% crude fat, and 15.4% NDF) or C (30.9% starch, 14.3% crude fat, and 17.8% NDF). After the duration of 28-day feeding experiment, 24 pigs (eight per treatment) were slaughtered and the M. longissimus lumborum (LL) samples at 45 min postmortem were collected. Results: Compared with diet A, diet C resulted in greater adenosine triphosphate and decreased phosphocreatine (PCr) concentrations, greater activity of creatine kinase and reduced percentage bound activities of hexokinase (HK), and pyruvate kinase (PK) in LL muscles (p<0.05). Moreover, diet C decreased the phosphor-AKT level and increased the hydroxy-hypoxia-inducible $factor-1{\alpha}$ ($HIF-1{\alpha}$) level, as well as decreased the bound protein expressions of HK II, PKM2, and lactate dehydrogenase A (p<0.05). Conclusion: Diet C with the lowest level of starch and the highest levels of fat and NDF could enhance the PCr utilization and attenuate glycolysis early postmortem in LL muscle of finishing pigs.
In the neurovascular unit, the neuronal and vascular systems communicate with each other. O2 and nutrients, reaching endothelial cells (ECs) through the blood stream, spread into neighboring cells, such as neural stem cells, and neurons. The proper function of neural circuits in adults requires sufficient O2 and glucose for their metabolic demands through angiogenesis. In a central nervous system (CNS) injury, such as glioma, Parkinson's disease, and Alzheimer's disease, damaged ECs can contribute to tissue hypoxia and to the consequent disruption of neuronal functions and accelerated neurodegeneration. This review discusses the current evidence regarding the contribution of oxygen deprivation to CNS injury, with an emphasis on hypoxia-inducible factor (HIF)-mediated pathways and Notch signaling. Additionally, it focuses on adult neurological functions and angiogenesis, as well as pathological conditions in the CNS. Furthermore, the functional interplay between HIFs and Notch is demonstrated in pathophysiological conditions.
Since hypoxia-inducible factor-lalpha (HIF-lalpha) and the arylhydrocarbon receptor (AhR) shared the AhR nuclear translocator (Arnt) for hypoxia- and AhR-mediated signaling, respectively, it was possible to establish the hypothesis that hypoxia could regulate Cyplal expression. In order to understand the mechanism of Cyplal gene expression, we demonstrated here that hypoxic agents such as cobalt chloride, desferrioxarnine, and picolinic acid reduced the TCDD induced Cyplal promoter activity based on the determination of luciferase activity in Hepa I cells transfected with pmCypla1-Luc. Also cobalt chloride inhibited the TCDD stimulated Cyplal mRNA level as well as EROD activities in both Hepa I and MCF-7 cells. Hypoxic agents such as cobalt chloride, picolinic acid, and desferrioxamine showed inhibition of luciferase activity that was induced by lnM TCDD treatment with dose dependent manner. Concomitant treatment of 150${\mu}$M ferrous sulfate with 1∼100${\mu}$M desferrioxarnine or 1∼100${\mu}$M picolinic acid recovered from the hypoxic agents-inhibited luciferase activity that was stimulated by TCDD. Reciprocally, the hypoxic agents down regulated TCDD induced Cyplal mRNA level and CYP1A1 enzyme activity in Hepa I cells.
Objectives : In this study, we made an effort to investigate the protective mechanism of Ukgan-san (UGS) extracts on hypoxia-induced C6 glial cell death. Methods : The cell viability was assessed by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide (MMT) assay and cell morphological changes were analysed with microscope after staining with crystal violet (CV). Reactive oxygen species (ROS) formation was assessed by flow cytometer after staining with 2'7'-dichlorofluorescein diacetate (DCF-DA). We also analyzed expression of hypoxia-inducible factor-1 alpha (HIF-$1{\alpha}$) and p53, processing of procaspase-3 and procyclic acidic repetitive protein (PARP) by western blot method. Results : We estimated the elevated cell viability by UGS extract on $CoCl_2$-induced C6 glial cells. UGS attenuated $CoCl_2$-induced ROS formation in C6 glial cells and also showed a protective activity compared to antioxidants and exhibited abrogation of LDH-released by $CoCl_2$. UGS suppressed the typical apoptotic cell death markers, caspase-3 and PARP activation. UGS inhibited $CoCl_2$-induced HIF-1${\alpha}$ expression which is known as a major regulator for hypoxia-induced cell death, and suppressed p53 expression. Conclusions : These results suggest that UGS extract contains protective constituents for hypoxia-induced C6 glial cell death.

KPLA-012, a benzopyranyl 1,2,3-triazole compound, is considered a potent $HIF-1{\alpha}$ inhibitor based on the chemical library screening, and is known to exhibit anti-angiogenetic and anti-tumor progressive effects. The aim of this study was to investigate the pharmacokinetic properties of KPLA-012 in ICR mice and to investigate in vitro characteristics including the intestinal absorption, distribution, metabolism, and excretion of KPLA-012. The oral bioavailability of KPLA-012 was 33.3% in mice. The pharmacokinetics of KPLA-012 changed in a metabolism-dependent manner, which was evident by the low recovery of parent KPLA-012 from urine and feces and metabolic instability in the liver microsomes. However, KPLA-012 exhibited moderate permeability in Caco-2 cells ($3.1{\times}10^{-6}cm/s$) and the metabolic stability increased in humans compared to that in mice (% remaining after 1 h; 47.4% in humans vs 14.8% in mice). Overall, the results suggest that KPLA-012 might have more effective pharmacokinetic properties in humans than in mice although further studies on its metabolism are necessary.
Hypoxia, a widespread occurrence observed in various malignant tumors, results from rapid tumor growth that outpaces the oxygen supply. Tumor hypoxia precipitates several effects on tumor biology; these include activating angiogenesis, intensifying invasiveness, enhancing the survival of tumor cells, suppressing anti-tumor immunity, and fostering resistance to therapy. Aligned with the findings that correlate CMGC kinases with the regulation of Hypoxia-Inducible Factor (HIF), a pivotal modulator, reports also indicate that hypoxia governs the activity of CMGC kinases, including DYRK1 kinases. Prolyl hydroxylation of DYRK1 kinases by PHD1 constitutes a novel mechanism of kinase maturation and activation. This modification "primes" DYRK1 kinases for subsequent tyrosine autophosphorylation, a vital step in their activation cascade. This mechanism adds a layer of intricacy to comprehending the regulation of CMGC kinases, and underscores the complex interplay between distinct post-translational modifications in harmonizing precise kinase activity. Overall, hypoxia assumes a substantial role in cancer progression, influencing diverse aspects of tumor biology that include angiogenesis, invasiveness, cell survival, and resistance to treatment. CMGC kinases are deeply entwined in its regulation. To fathom the molecular mechanisms underpinning hypoxia's impact on cancer cells, comprehending how hypoxia and prolyl hydroxylation govern the activity of CMGC kinases, including DYRK1 kinases, becomes imperative. This insight may pave the way for pioneering therapeutic approaches that target the hypoxic tumor microenvironment and its associated challenges.
재생냉각 시스템의 냉각제로 사용하는 탄화수소계열 연료인 Jet A-1의 냉각특성과 강제대류 열전달 특성을 파악하기 위해 가열된 직선 튜브를 제작하였다. 냉각제의 입, 출구온도와 튜브의 외벽 온도를 측정하여 강제대류 열전달 특성을 고찰하였다. 결과를 통하여 Jet A-1을 사용한 경우에는 일정 유동조건에 대하여 burnout이 발생하는 최초의 벽 온도를 결정함으로써 냉각제가 수용할 수 있는 최대 열유속을 확인하였다. 냉각제의 유속과 입구온도가 대류열전달 특성에 직접적인 영향을 주는 인자임을 확인하였다.
본 연구는 인간 폐암세포의 저산소 상태를 재현하기 위한 $CoCl_2$의 처리 조건을 최적화 하였고, 최적화 된 저산소 상태에서 인간 폐암세포의 암화 과정 및 기전을 규명하였다. 인간 폐암세포, A549와 H460에 500 ${\mu}M$$CoCl_2$를 24시간 처리하였을 때 저산소 상태의 대표적인 전사인자, HIF-$1{\alpha}$의 발현이 증가함을 확인하였고 인간 폐암세포들의 성장에는 전혀 영향을 미치지 않음을 확인하였다. 또한 $CoCl_2$를 처리한 인간 폐암 세포에서 상피-중간엽 이행(epithelial-to-mesenchymal-like transition)의 대표적인 마커인 E-cadherin 발현의 감소와 ${\alpha}$-SMA의 증가를 확인하였고, 세포-세포 간 junction 부위가 깨어짐을 E-cadherin 형광염색 실험을 통하여 확인하였다. 더 나아가 $CoCl_2$를 처리한 인간 폐암 세포에서 상피-중간엽 이행의 분자적 기전을 밝히기 위해 세포벽에 존재하는 인테그린(integrin)의 발현을 웨스턴 블랏팅과 FACS분석을 통하여 알아본 결과, $CoCl_2$를 처리한 인간 폐암세포에서 인테그린 ${\beta}3$발현의 증가를 확인하였다. 뿐만 아니라, $CoCl_2$를 처리한 인간 폐암세포에서 인테그린 ${\beta}3$의 하부 신호전달 물질인 국소부착 카이네이즈(FAK)의 인산화가 증가함을 확인하였다. 상기의 결과로서, 국소부착 카이네이즈의 인산화를 저해함으로써 인간 폐암세포가 악성세포로 전이되는 것을 저해할 수 있을 것으로 기대 되어진다.
Genome engineering with clustered regularly interspaced short palindromic repeats (CRISPR) system can be used as a tool to correct pathological mutations or modulate gene expression levels associated with pathogenesis of human diseases. Owing to well-established local administration methods including intravitreal and subretinal injection, it is relatively easy to administer therapeutic genome engineering machinery to ocular tissues for treating retinal diseases. In this context, we have investigated the potential of in vivo genome engineering as a therapeutic approach in the form of ribonucleoprotein or CRISPR packaged in viral vectors. Major issues in therapeutic application of genome engineering include specificity and efficacy according to types of CRISPR system. In addition to previous platforms based on ribonucleoprotein and CRISPR-associated protein 9 derived from Campylobacter jejuni, we evaluated the therapeutic effects of a CRISPR RNA-guided endonuclease derived from Lachnospiraceae bacterium ND2006 (LbCpf1) in regulating pathological angiogenesis in an animal model of wet-type age-related macular degeneration. LbCpf1 targeting Vegfa or Hif1a effectively disrupted the expression of genes in ocular tissues, resulting in suppression of choroidal neovascularization. It was also notable that there were no significant off-target effects in vivo.
