Molecular mechanisms of hederagenin in bone formation (Hederagenin의 뼈 형성 관련 작용 기전 연구)

  • Purpose: Osteoporosis is characterized by structural deterioration of the bone tissue because of the loss of osteoblastic activity or the increase in osteoclastic activity, resulting in bone fragility and an increased risk of fractures. Hederagenin (Hed) is a pentacyclic triterpenoid saponin isolated from Dipsaci Radix, the dried root of Dipsacus asper Wall. Dipsaci Radix has been used in Korean herbal medicine to treat bone fractures. In this study, we attempted to demonstrate the potential anti-osteoporotic effect of Hed by examining its effect on osteoblast differentiation in MC3T3-E1 cells. Methods: Osteoblastic MC3T3-E1 cells were cultured in 0, 1, and 10 ㎍/mL Hed for 3 and 7 days. The activity of alkaline phosphatase (ALP), bone nodule formation and level of expression of bone-related genes and proteins were measured in MC3T3-E1 cells exposed to Hed. The western blot test was used to detect the activation of the bone morphogenetic protein-2 (BMP2)/ Suppressor of Mothers against Decapentaplegic (SMAD)1 pathway. Results: Hed significantly increased the proliferation of MC3T3-E1 cells. Intracellular ALP activity was significantly increased in the 1 ㎍/mL Hed-treated group. Hed significantly increased the concentration of calcified nodules. Furthermore, Hed significantly upregulated the expression of genes and proteins associated with osteoblast proliferation and differentiation, such as Runt-related transcription factor 2 (Runx2), ALP, osteopontin (OPN), and type I procollagen (ProCOL1). Induction of osteoblast differentiation by Hed was associated with increased BMP2. In addition, Hed induced osteoblast differentiation by increasing the activity of SMAD1/5/8. These results suggest that Hed has the potential to prevent osteoporosis by promoting osteoblastogenesis in osteoblastic MC3T3-E1 cells via the modulation of the BMP2/SMAD1 pathway. Conclusion: The results presented in this study indicate that Hed isolated from Dipsaci Radix has the potential to be developed as a healthcare food and functional material possessing anti-osteoporosis effects.

  • This study was aimed to develop a new formula for herbal medicine-safety classification in terms of evidence-based medicine. Recently, human equivalent dose(HED)-based therapeutic index was developed for herbal medicine-safety classification by transforming $LD_{50}$ to HED. However, the use of the $ED_{50}$ and $LD_{50}$ to derive the therapeutic index may be misleading as to safety, depending on the slope of the dose-response curves for therapeutic and lethal effects. To overcome this deficiency, HED-based MOS(Margin of Safety)was developed and suggested in this study. The HED-based MOS developed by using $LD_1$, changing to ALD(approximate lethal dose), and $ED_{99}$. The HED-based MOS seems to be more useful and safer than HED-based therapeutic index since its values for several herbal medicines are basically two times less than the values from HED-based therapeutic index. Thus, HED-based MOS can be a good example of Evidence-based approach for herbal medicine-safety classification.

Risk Factors for Heavy Episodic Drinking among Korean Adults: Differences by Gender and Age (성인의 폭음 관련요인: 성별, 연령별 차이)

  • Purpose: Most research on risky drinking among adults in Korea has focused on problem drinking and high-risk drinking. This paper examines the nature of heavy episodic drinking_(HED) among adults and seeks to identify the risk factors for HED by analyzing differences by gender and age. Method: Participants were 3,886 adults aged 19-64 years from the database of the fifth Korean National Health and Nutritional Examination Survey (KNHANES-V, 2010). Data were analyzed by conducting the Rao_Scott chi-square test, an ANCOVA, and logistic analysis with SAS 9.2. Results: Individuals between 19 and 29 years of age had the highest HED among all age groups. Individuals between 40 and 49 (male adults) and those between 30 and 39 (female adults) showed HED associated with stress, suicidal ideation, smoking, and other health behavior-related variables. Risk factors that affected HED in male adults were age, educational status, and current smoking, while for female adults, the factors were age, educational status, employment, marital status, and current smoking. Conclusions: It is necessary to develop appropriate intervention programs that abate negative emotions and lessen the risk of HED in adults with consideration to gender and age differences.

Hed1 Promotes Meiotic Crossover Formation in Saccharomyces cerevisiae

  • Homologous recombination occurs between homologous chromosomes and is significantly involved in programmed double-strand break (DSB) repair. Activation of two recombinases, Rad51 and Dmc1, is essential for an interhomolog bias during meiosis. Rad51 participates in both mitotic and meiotic recombination, and its strand exchange activity is regulated by an inhibitory factor during meiosis. Thus, activities of Rad51 and Dmc1 are coordinated to promote homolog bias. It has been reported that Hed1, a meiosis-specific protein in budding yeast, regulates Rad51-dependent recombination activity. Here, we investigated the role of Hed1 in meiotic recombination by ectopic expression of the protein after pre-meiotic replication in Saccharomyces cerevisiae. DNA physical analysis revealed that the overexpression of Hed1 delays the DSB-to-joint molecule (JM) transition and promotes interhomolog JM formation. The study indicates a possible role of Hed1 in controlling the strand exchange activity of Rad51 and, eventually, meiotic crossover formation.

A study of the hybrid electric drive generating mode in naval ships (함정용 하이브리드 전기추진 시스템 발전기 모드 적용에 대한 연구)

  • Because of recent interests in energy conservation and prevention of environmental pollution, research related to these topics is increasing. The U.S. Navy has started to study the HED (hybrid electric drive) system in order to improve the COGAG propulsion system's fuel efficiency in AEGIS destroyers. (This ship's fuel consumption is 40% of the total fuel consumption of the U.S. Navy.) In addition, the Korean Navy is considering applications of the HED system in AEGIS destroyers. The purpose of this study is to analyze the U.S.A.'s HED system and to simulate its generating mode energy saving rate using LabVIEW. The results confirmed that the fuel savings are about 700 kg/h.

A Study on Hybrid Electric Drive System for the AEGIS Destroyer (이지스 구축함용 HED 시스템에 관한 연구)

  • Arleigh Burke(DDG-51) and Sejong warship are AEGIS destroyer of US Navy and Korea Navy. These are designed to make more than 30knots by applying a COGAG(COmbined Gas turbine And Gas turbine) system. However, the gas turbine(LM2500) in this system has a low SFC (Specific Fuel Consumption) when the warship operated low speed. So, many kinds of companies are researching the HED(Hybrid Electric Drive) system to improve this problem. The purpose of this paper is to analyze the HED system and simulate by Sejong warship data. Serveral methods were used for that purpose. More specifically, the equipment modeling are employed for regression analysis by LabVIEW. As a result, it was found that the warship installed HED system could cut their fuel bills by as much as about 80,000,000won per year.

Effects of Antibiotic and Yeast Supplemental High Energy Diet on Growth Performance, Blood Characteristics and Carcass Trait in Broilers (고에너지 사료 내 항생제와 효모제의 첨가가 육계의 생산성, 혈액 성상 및 도체 특성에 미치는 영향)

  • This study was conducted to investigate the effects of antibiotic and yeast supplemental high energy diet on growth performance, blood characteristics and carcass trait in broilers. Total of four hundred-eighty broilers were randomly allocated into three treatments with eight replications for five weeks. Dietary treatments included 1) CON (control; basal diet), 2) HED (high energy diet) and 3) YD (HED; yeast added to HED instead of virginiamycin, Sacchromyces cerevisiae, $15{\times}10^{10}$). During whole period, weight gain had high tendency in HED treatment. However, there were not significant among treatments (P>0.05). Feed intake was higher in YD treatment than others. However, there were not significant among each treatments (P>0.05). Feed/Gain ratio was significantly lower in HED treatment than others (P<0.05). In blood characteristics, RBC, WBC and lymphocyte were not significant (P>0.05) among treatments. Liver weigh, LW/BW ratio, leg meat weigh, LMW/BW ratio, breast meat weigh, BMW/BW ratio, abdominal fat weigh and AF/BW ratio were not significant (P>0.05). However, body weight was improved (P<0.05) in HED treatment. In conclusion, this experiment is shown that HED treatment affects Feed/Gain ratio and body weight at final period in broilers.

Development of Dual Energy Radiation Detector (이중 에너지 방사선 검출기 개발)

  • In this paper, we are suggested development of dual-mode detector for dual-energy digital radiography. Design of dual-energy radiography module for commercial BIS (Baggage Inspection System) is used in the spectrum of the X-ray generator and detector for dual-mode features and radiological characteristics were analyzed. BIS suggestl on the image detector module being used to target X-ray tube to simulate X-ray spectrum and simulated spectrum to offer through the new radiographic characteristics of the detector modules were investigated. Using X-ray experiments with an increase in the thickness of the copper filter low energy detector (LED) and high-energy detector (HED) as the difference between the output signal increases. HED, especially in the size of the output signal decreases with increasing thickness of the copper filter was found.

  • Single exposure dual X-ray imaging can be used to separate soft and dense-material images for medical and industrial applications. This study keep focusing baggage inspection system(BIS) specifically. New detector modules for single exposure dual X-ray imaging are consisted of low energy detector (LED) and high energy detector (HED). First, the optimized thickness of copper filter coupled HED to separate low energy and high energy was simulated by the given X-ray energy (140 kVp, 1 mA) using Monte Carlo simulation codes, MCNPX. So as a result of simulation, the copper filter thickness is 0.7 mm. For the design of PIN photodiode, ATLAS device simulation tool was used. 16 channels PIN photodiode of 1.5 mm ${\times}$ 3.2 mm for Dual X-ray imaging detector was fabricated in the process of ETRI. And its dark current and quantum efficiency, terminal capacitance were measured. It was proven that the Lanex Fast B coupled HED were a sufficient candidate to replace the CsI(Tl) commerced in dual X-ray system, since these give a strong signal, overcoming system noise. Finally dual X-ray image was acquired through correction of the LED X-ray Image and the HED X-ray Image.

DENTAL MANAGEMENT OF ECTODERMAL DYSPLASIA : A CASE REPORT (외배엽 이형성증 환자의 치험례)

  • Ectodermal dysplasia is a genetic disease which shows various congenital dysplasias in tissues differentiated from the ectoderm. As the most common type of the ectodermal dysplasia, hypohidrotic ectodermal dysplasia(HED) shows dysplasia mainly in the hair, fingernails, teeth and the skin. Symptoms are more severe in males than in females and heterozygous females are usually normal showing no symptom. The treatment for these patients differ according to individuals, but since patients can easily become depressed socially and emotionally due to a decrease in mastication and speech function caused by multiple loss of teeth as well as some aesthetic problems, an early treatment is required. In a case, with a 10 years-old boy diagnosed with HED which shows partial edentia of the maxilla, and the edentia of the mandible in the pediatric dentistry department of the Chosun University Dental Hospital, a fabrication of denture resulted in the recovery of mastication and speech function and aesthetic improvement due to an increase of the face height.
