에탄올 유발 위점막 손상에 대한 황련 물 추출물의 방어효과 (Protective Effects of Coptidis Rhizoma on Ethanol-induced Gastric Ulcer in Mice)

  • The exposure of gastric mucosa to ethanol produces acute ulcers mediated by inflammatory processes, hemorrhagic erosions and increase of reactive oxygen species. The purpose of this study was to assess the effects of Coptidis Rhizoma(CR) aqueous extracts on hydrochloride (HCl)/ethanol induced gastric ulcer in mice as compared with rebamipide (30 mg/kg) and ranitidine (100 mg/kg). Stomach ulcers were induced by oral ingestion of HCl/ethanol. CR extracts (125, 250 and 500 mg/kg) were orally administered, once a day for 7 continuous days, and 1 hr after last 7th treatment of CR extracts stomach ulcers were induced. Effects of CR extracts on HCl/ethanol-induced gastric ulcer were evaluated based on gross and microscopic observations with anti-oxidant activities. All three different dosages of CR extract significantly decreased HCl/ethanol-induced gastric ulcer compared with the HCl/ethanol control mice. CR extracts also strengthened the antioxidative defense systems - decreased the level of lipid peroxidation but increased the level of catalase, superoxide dismutase and nitrate/nitrite compared with the HCl/ethanol control. The effects of CR extract 500 mg/kg were similar to that of 30 mg/kg rebamipide, and CR extract 250 mg/kg showed similar anti-ulcer effects as compared with ranitidine 100 mg/kg. These results suggest that the gastroprotective effects of CR extracts on mice ulcer models can be attributed to its ameliorating effect on oxidative damages.

염산-에탄올에 의해 유발된 흰쥐 위염에 대한 마치현의 억제 작용 (The Inhibitory Effects of Portulaca oleracea L. on HCl-ethanol Induced Gastritis in Rats)

  • Objectives : The objective of this study was to examine the effects of P. oleracea into the HCl-ethanol induced gastritis in rats, and to isolate and determine the chemical compounds from P. oleracea. Methods : The rats were orally administered with crude extract or fractions or isolated compounds of P. oleracea 30 mins before the induction of gastric lesion by oral administration of HCl-ethanol. The gastric lesional area was measured using pixel counting software. Then the chemical compounds from P. oleracea was isolated and determined by LC-MS and NMR. Results : The inhibition effect of oral administration of crude extract of P. oleracea at a dose of 500 mg/kg in HCl-ethanol induced gastritis was similar to cimetidine. Then, aqueous fraction at a dose of 240 mg/kg exhibited the effects similar to cimetidine. Then, the aqueous fraction was further separated by MPLC and yielded four sub fractions. Among those sub fractions, agent II at a dose of 40 mg/kg possessed the strongest effect in the HCl-ethanol induced gastritis. The water fraction yielded-Uridine, Adenosine, Guanosine, which were characterized by Mass, 1H-NMR, 13C-NMR. Conclusions : This study suggest that a P. oleracea and its compounds showed potent efficacy on the development of HCl-ethanol induced gastritis. Thus, P. olaracea can be a potential natural resource for the management of gastritis although the mechanism of action involved in the treatment remains to be explored.

  • Objectives : The purpose of this study is to evaluate whether or not a pretreatment with Portulaca oleracea has an antioxidant effect in HCl-ethanol induced gastric mucosal damage. Methods : We elucidated the level of reactive oxygen species (ROS), lipid peroxidation, and two important constituents of antioxidant defense such as superoxide dismutase (SOD), glutathione (GSH) in these effects. Results : The oral administration of crude extract from P. oleracea attenuated the gastritic lesion area, submucosal edema and hemorrhage, and mucosal necrosis induced by HCl-ethanol. The MDA levels of control group were higher than those in the rats given the P. oleracea pretreatment. While the GSH levels of control were decreased, the GSH activity on the gastric mucosal layer maintain normal level in rats given the Portulaca oleracea pretreatment before HCl-ethanol induced gastritis significantly increased. However, the SOD activites were not altered by P. oleracea. Conclusions : The administration of Portulaca oleracea have a protective antioxidant effect against the gastric lesion induced by HCl-ethanol and may therefore be a promising drug for gastritis and gastric ulcer.

Evaluation for Protective Effect of Rutin, a Natural Flavonoid, against HCl/Ethanol-Induced Gastric Lesions

  • In this study we investigated the protective effects of rutin, a natural plant flavonoid, on HCl/ethanol-induced gastric lesions in rats. Rutin showed the antioxidant activities, the acid-neutralizing capacities, and the inhibitory effects on the growth of Helicobacter pylori (H. pylori.), which are equivalent to control compounds. In addition, rutin significantly inhibited HCl/ethanol-induced gastric lesions. Antigastritic action of rutin may be associated with the antioxidant activities, acid-neutralizing capacities, anti-H. pylori action, and the stimulation of mucus secretion. From these results, we could suggest that rutin may be useful for the treatment and/or protection of gastritis.

흰쥐의 염산.에탄올 유발 위염 위조직에서 ginsenoside $Rb_1$의 항산화 효과 (Anti-oxidative Effect of Ginsenoside $Rb_1$ on the HCI.Ethanol-Induced Gastric Tissue in Rats)

  • In the previous study, we demonstrated that ginsenoside $Rb_1$ isolated from the butanol fraction of the head of Panax ginseng had significant gastroprotective activity on gastritis and gastric ulcer models in rats. It has been well established that drugs to have capacity of scavenging or inhibiting the generation of reactive oxygen radicals prevent the gastric mucosal injury. Ginsenoside $Rb_1$ was tested on HCl ethanol-induced gastritis in rats, DPPH-induced free radical scavenging effect, MDA assay, GSH activity, and SOD activity in gastric tissue. It showed significant inhibition in HCl ethanol-induced gastritis, and al~o significantly increase of GSH activated SOD. We speculate that the protective effect of ginsenoside $Rb_1$ against HCl ethanol-induced gastric mucosal damage is originated from the increase of GSH and the activation of SOD.

위염 및 Helicobacter pylori에 미치는 Amomi Semen Butanol 추출물의 영향 (Effects of Amomi Semen Butanol Fraction on Gastritis and Helicobacter pylori)

  • In a preliminary screening of plant extracts for the antigastritic and anti- Helicobacter pylori (H. pylori) actions in rats, the ethanol extract of Amomi Semen (AS) showed positive activity in HCl Ethanol-induced gastric lesions and H. pylori. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent butanol fraction significantly reduced HCl, Ethanol-induced gastric lesions at the oral dose of 350 mg/kg. Also butanol fraction has an inhibitory effect on the growth of H. pylori $(MIC=1.43\;{\mu}g/mL)$. In pylorus ligated rats, butanol fraction showed decrease in the volume of gastric secretion and acid output, of which effects were stronger in other fractions. We isolated 6 subfractions by column chromatography. The protective effects of 6 subfractions of Amomi Semen were also significant in the HCl, Ethanol induced gastric lesion model. These results might suggest that they had inhibitory action in gastric lesion through inhibition of gastric acid secretion. Butanol fraction of AS can be applied as treatment of H. pylori. Butano fractions and ethanol extract of AS was carried out or the development of a new gastroprotective supplementary product.

염산-에탄올에 의해 유발된 흰쥐 위염에 대한 생강 및 법제생강 추출물의 억제작용 (Inhibitory Effects of Ginger and Processed (Beopje) Ginger Extracts on HCl-ethanol Induced Gastritis in Rats)

  • 본 연구에서는 한의학 이론에 근거한 법제(포제)의 가공처리과정을 거친 생강과 일반생강의 위손상 억제효과를 알아보고자 하였다. Sprague-Dawley계 수컷 흰쥐에 HCl-ethanol로 급성위염을 유도하고, 이에 대한 위손상 억제효과를 검토하였다. 생강과 법제생강을 70% ethanol로 열수 추출 및 농축하여 실험에 사용하였고, 생강과 법제생강 투여군 모두 유의적인 위손상 억제효과가 있는 것으로 나타났다(p<0.05). 생강 저농도와 법제생강 저농도의 위손상 억제율은 40.2%, 64.9%, 생강 고농도와 법제생강 고농도의 위손상 억제율은 68.4%, 99.6%로 법제생강 투여군이 생강 투여군보다 위손상 억제효과가 증가되었다. 특히 법제생강 고농도의 위손상 억제율은 대조약물인 cimetidine의 74.9%보다 높은 99.6%를 나타내었다. 위액분비량은 생강과 법제생강을 투여한 군 모두 유의적으로 감소하였고, 법제생강 투여군의 위액분비량(0.32 mL, 2.69 mL)이 생강 투여군(2.66 mL, 3.23 mL)보다 유의적으로 감소되었다(p<0.05). 위액 pH는 법제생강 고농도가 유의성 있게 증가하였고, 유리산도는 cimetidine과 법제생강 고농도에서 유의성 있게 감소되었다(p<0.05). 생강과 법제생강의 투여 용량의 증가에 따라 위손상 억제효과가 증가하였으며, 특히 법제생강고농도는 대조약물인 cimetidine보다도 높은 위손상 억제효과를 나타내었다. 이상의 결과로 생강은 위손상 억제효과가 있음을 확인하였고, 법제의 가공과정에 의해 생강의 위손상 억제효과가 더 증가되었음을 확인할 수 있었다.

  • Alginate를 가열에 의해 저분자화하여 저분자화에 따른 위점막 보호작용을 검토하기 위해 랫드에 있어서 저분자 alginate인 HAG-10, HAG-50, HAG-100 및 alginate의 투여 가 $HCl{\cdot}ethanol$ 용액과 침수스트레스에 의해 유발되는 위손상과 위궤양의 억제효과에 미치는 영향을 광학현미경으로 점막의 형태를 관찰하면서 그 억제율과 조직학적 변화를 측정하였다. $HCl{\cdot}ethanol$ 용액과 침수스트레스에 의해 유발되는 위손상과 위궤양에 대한 억제효과에 미치는 저분자 alginate의 영향을 검토한 결과, 그 억제효과는 alginate에서 가장 높았고, 다음으로 HAG-50과 HAG-100 및 HAG-10의 순으로 HAG-50, HAG-100 및 alginate에서 현저한 효과를 보였으나, HAG-10은 아무런 효과를 나타내지 않았다. 위손상과 위궤양점막을 광학현미경으로 관찰해 볼 때, 대조군과 HAG-10에서는 위점막 전체에 걸친 출혈과 부식에 의한 응고괴사가 진행되었고 점막 (粘膜)과 점막근판 (粘膜筋板) 및 근육층 (筋肉層)에 부종의 발생과 조직의 파괴가 일어났으나 HAG-50, HAG-100 및 alginate에서는 가벼운 병변이 진행되어 위점막층에 약간의 출혈만 관찰되어 위손상억제 및 위궤양 억제효과를 뚜렷히 나타내었다.

  • 본 실험 결과, EF 열수 추출물이 항산화 활성뿐만 아니라, MAPK 신호 경로의 활성을 조절하고 전 염증성 사이토카인의 수준을 낮추는 것으로 나타났으며, 특히 EF 열수 추출물의 낮은 농도보다 높은 농도에서 더 뛰어난 효과를 나타내었다. 이는 EF 열수 추출물이 NADPH 산화효소, 항산화 단백질 및 염증성 단백질 발현을 조절하여 위 조직 내 급성 위염을 완화시킬 수 있음을 시사한다.

  • 본 연구는 고구마와 참마 추출 혼합물의 항산화능을 평가하였으며, 에탄올로 유도된 급성 위염 동물모델에서 HCl/ethanol로 유도된 위염 억제 효과 여부를 검증하였다. 총 폴리페놀 및 총 플라보노이드 함량, DPPH 및 ABTS 라디칼 소거활성은 고구마나 참마의 단독 사용보다 추출 혼합물에서 높았다. 알코올성 급성 위염에 있어서도 단독 사용보다 추출 혼합물이 임상적 행동 점수 및 위액량, 위 점막 손상면적, TNF-α 수치 등에서 유의적인 감소를 확인하였다. 이러한 결과는 고구마와 참마 추출 혼합물이 항산화능이 뛰어나며 위 점막 보호 효과가 있음을 확인할 수 있었다. 따라서 고구마와 참마 추출 혼합물은 알코올성 급성 위염에 있어 위 보호효과를 기대할 수 있으며, 지역 소재의 기능성 발굴에 가능성이 있음을 시사한다.
