Studies on the Effect of the Extract of Eugenia Flos on Gastritis and Gastric Lesion (급만성 위염 및 위손상에 관한 정향 추출물의 효과)

  • This study was perfonned to investigate effectiveness eness on the gastritis and gastric lesion with the methanol extract of the flower buds of Eugenio caryophyllata. The extract was fractionated with hexane, chIorofonn, ethyl acetate, butanol, followed by bioassay Oil antigastritis. The ethyl acetate and the buthanol fraction reduced significantly HCI.ethanol induced gastric lesion at the dose of 165 and 215 mg/kg, p.o., respectively. These results may indicate that remarkably.effective are ethyl acetate and butanol fractions in HCI-ethanol induced gastric lesion. Howeever, the fractions didn't exhibit any inhibition of gastric secretion and acid output. The buthanol fraction reduced significantly the acetic acid induced ulcer at a daily dose of 215 mg/Kg, p.o., given for 10 days. These result showed considerable inhibit of acetic acid induced ulcer without inhibition of indomethacin induced gastric lesion. The methanol extract showed low acute toxicity with minimum lethal dose of more than 3000 mg/kg, p.o. in mice. In conclusion, Eugenia F10s exhibited antigastric activity which might be attributable to inhibition of gastric secretion. It is indicated that activie component may be present in the buthanol fraction.

Antigastritic and Antiulcer Actions of the Fraction of Taheebo (Taheebo 분획물의 의염 및 위궤양에 대한 효과)

  • In a preliminary screening of plant extracts for the antigastritic and antiulcer actions in rats, the methanol extracts of Taheebo showed positive activity in HCI . ethanol-induced gastric lesion. Among the systematic fractions of hexane, chloroform, butanol and water, the most potent $H_2O$ fraction reduced significantly HCI . ethanol-induced gastric lesion at the oral dose of 300 mg/kg. In pylorus ligated rats chloroform and butanol fraction showed decreases in the volume of gastric secretion and acid output of which effects were stronger in chloroform fraction. Further assays with hexane butanol and $H_2O$ fraction disclosed that it significantly suppressed the aspirin-induced ulcer. The butanol fraction reduced significantly acetic acid induced ulcer at the dose of 400 mg/kg. The butanol and $H_2O$ fraction reduced the malondialdehyde level in HCI . ethanol-induced gastric lesion. In pylorus ligated rats, chloroform and butanol fraction reduced the malondialdehyde level and in aspirin-induced ulcer, chloroform fraction reduced that levle. These results might suggest that the butanol and $H_2O$ fraction of Taheebo had inhibitory action in gastric lesion and ulceration through inhibition of gastric acid secretion and the decrease malondialdehyde level.

  • Alnus japonica cortex has been used as antidiarrhea, antihemorrhage and the remedy of indigestion. This study was performed to investigate the effectiveness of the methanol extracts of the Alnus japonica cortex on the gastric lesion and ulcer. The methanol extract was fractionated with hexane, chloroform and butanol, followed by bioassay on antigastritic and antiulcer activity. The methanol extract showed low acute toxicity with minimum lethal dose of more than 5000 mg/kg, p.o. in mice. The chloroform and the butanol fraction reduced gastric lesion in HCI. ethanol induced gastritic model. On gastric secretion in pylorus ligated rat, the hexane and chloroform fraction decreased the volume and acidity. The butanol fraction had significant inhibitory effects on aspirin and Shay's ulcer. The butanol fraction showed a tendency to inhibit the decrease of mucin secretion due to ingestion of absolute ethanol.

Antigastritic and Anti-ulcerative Effect of P020701 (생약복합제 P020701의 항위염 및 항궤양 효과)

  • Present study was carried out for development of a new supplementary product with gastroprotective effect. Natural Products mentioned that have GI protective property in Dongeuibogam and reports were evaluated anti-bacterial activity against Helicobacter pylori, then five heres were selected. The material used for the test were water extract of Alpinia oxyphlla (AO), Astragalus membranaceus (AM), Cinnamomum loureirii(CL), Citrus aurantium(CA), Amomum villosum(AV). They were tested individually on HCI ethanol-induced gastric lesion in rats, AV CL, AO showed the most significant effectiveness, respectively. Then two mixture different in their content ratio (P020701-1,-2) and complex with P020701-1 (CP) were made, and tested on HCI·ethanol, indomethacin-induced gastric lesion, aspirin-ligature, Shay ulcer and gastric secretion. P020701-1, -2 and CP showed significant effect on HCI ethanol, indomethacin-induced gastric lesion, and Shay ulcer. It can be regarded that the antigasuitic and anti-ulcerative effects of P020701- 1, -2 and CP are originated from reduction of total acid output identified by gastric secretion test.

The Influence of Methanol Extract of Ulmus davidiana var. japonica Cortex on Gastric Erosion and Ulcer and Paw Edema in Rats (느릅나무 수피 엑스가 흰쥐의 위염, 위궤양 및 발부종에 미치는 영향)

  • The bark of Ulmus davidiana var. japonica(Ulmaceae) has been described in traditional books to have diuretic and laxative actions, to ameliorate edematous disease, and to be used in insomnia and gastointestinal disorders. Thus this study was carried out to elucidate some of the pharmacological activities of the plant extracts. The results obtained in tills experiment indicated that its methanol extract elicited remarkable inhibition of HCI.ethanol induced gastric lesion, Shay ulceration and gastric secretion. However, it showed no anti-inflammatory action. The acute toxicity of the extract was low, that is, the minimum lethal dose was more than 2000 mg/kg by oral administration in mice. The systematic fractionation of the methanol extract by hexane, ether, ethylacetate and butanol resulted in potent prevention of gastric erosion in butanol and water fractions.

  • In a preliminary screening of the plant extracts for the antigastritic action in rats, the extract of Aralia elata(Araliaceae) showed positive activity in HCI plus ethanol induced gastric lesion. Systematic fractions with hexane, chloroform, ethyl acetate and butanol resulted in the most patent activity with the butanol fraction: This butanol fraction at the oral dose of 200 mg/kg exhibited significant inhibition of absolute alcohol induced gastric lesion which was more potent than 100 mg/kg of cimetidine and had significant stimulation of mucus secretion. The butanol fraction showed significant decreases in the ulcer indices of Shay ulcers and inhibition of gastric juice secretion with acid output in pylorus-ligated stomachs of rats. It also suppressed the acetic acid induced gastric ulcer. These results might suggest that the butanol fraction had inhibitory action in gastric lesion and ulceration through inhibition of gastric acid secretion and stimulation of mucin secretion in the stomachs of rats.

  • In order to investigate the effect of BODUOPAE-SAN(BOS), experiments were performed on analgesic effect induced by acetic acid, duration of hypnosis induced by pentobarbital-Na in mice and gastric ucler in Shay rats and indomethacin induced gastric ulcer, HCI-ethanol induced gastrci lesion in rats and gastric juice secretion in Shay rats. The results are as followings. 1. BOS showed significantly an analgesic effect induced by acetic acid. 2. BOS prolonged the duration of hypnosis induced by pentobarbital-Na in rats 3. BOS rised the spontaneous motility of isolatied ileum of mice temporarily. 4. BOS depressed the gastric motility of rats significantly. 5. BOS showed an anti-ulcer effect in Shay rats and indometacine-induced ulcer rats significantly. 6. BOS reduced the ulcer index of the HCl-ethanol induced gastric lesion in rats. 7. BOS reduced the gastric juice secretion in Shay rats. From above results think BOS is more effective in comparison to the OPAESAN alone on gastrointestinal disorder.

  • In the previous study, we demonstrated that ginsenoside $Rb_1$ isolated from the butanol fraction of the head of Panax ginseng had significant gastroprotective activity on gastritis and gastric ulcer models in rats. It has been well established that drugs to have capacity of scavenging or inhibiting the generation of reactive oxygen radicals prevent the gastric mucosal injury. Ginsenoside $Rb_1$ was tested on HCl ethanol-induced gastritis in rats, DPPH-induced free radical scavenging effect, MDA assay, GSH activity, and SOD activity in gastric tissue. It showed significant inhibition in HCl ethanol-induced gastritis, and al~o significantly increase of GSH activated SOD. We speculate that the protective effect of ginsenoside $Rb_1$ against HCl ethanol-induced gastric mucosal damage is originated from the increase of GSH and the activation of SOD.

  • An extract of Alnus japonica (Betulaceae) cortex has been traditionally used for purifying blood, and curing feces containing blood, enteritis, diarrhea, alcoholism and cut wounds. In the preliminary test was carried out for determining whether it has the novel pharmacological activity, the butanol fraction showed significant inhibitory effect on carrageenan-induced paw edema as an acute inflammation, HCI.ethanol-induced gastric lesion and aspirin-ligation gastric ulcer. Ginsenoside $Rb_1$: the Anti-Ulcer Constituent from the Head of Panax ginseng

  • We previously reported that the butanol (BuOH) fraction of the head of Panax ginseng exhibited gastroprotective activity in peptic and chronic ulcer models. In order to identify the active constituent, an activity-guided isolation of the BuOH faction was conducted with a HCI$.$ethanol-induced gastric lesion model. The BuOH fraction was passed through a silica-gel column using a chloroform-methanol gradient solvent system, and six fractions (frs. 1-6) were obtained. The active fr. 5 was further separated by silica-gel column, to yield 6 subfractions (subfrs. a-f). Subfr. d was composed of ginsenosides Re, Rc and $Rb_1$. The most active constituent was ginsenoside $Rb_1$ ($GRb_1$), a protopanaxadiol glycoside, which was investigated for its anti-ulcer effect. Gastric injury induced by HCI$.$ethanol, indomethacin and pyloric ligation (Shay ulcer) was apparently reduced with oral $GRb_1$ doses of 150 and 300 mg/kg. $GRb_1$ at these dosage significantly increased the amount of mucus secretion in an ethanol-induced model. The anti-ulcer effects were consistent with the result of histological examination. These results suggest that the major active constituent in the head of Panax ginseng is $GRb_1$ and that anti-ulcer effect is produced through an increase in mucus secretion.
