헤더-채널 분기관의 채널 돌출길이가 2상 유동 분배에 미치는 영향에 대한 연구 (Study on Effect of Channel Intrusion Depth on the Two-Phase Flow Distribution at Header-Channel Junction)

  • The main objective of this work is to experimentally investigate the effect of angle variation and intrusion depth of channels on the distribution of two-phase flow at header-channel junctions. The dimensions of the header and the channels in cross-section were fixed at $16mm{\times}16mm$ and $12mm{\times}1.8mm$, respectively. Air and water were used as the test fluids. Two different header-channel positions were tested : a vertical header with horizontal channels (case VM-HC) and a horizontal header with horizontal channels (case HM-HC). In all cases, the intrusion depths of the channels are 0 mm, 2 mm, and 4 mm. For the case of the intrusion depth of VM-HC, the flow distribution became more uniform. However, the intrusion depth negatively affected the flow distribution for the case of HM-HC because liquid separation delay occurred.

당뇨병성 족부질환자에 대한 가정간호서비스의 비용-효과분석 (Cost-effectiveness Analysis of Home Care Services for Patients with Diabetic Foot)

  • Purpose: This study was a retrospective survey to examine economic feasibility of home care services for patients with diabetic foot. Methods: The participants were 33 patients in the home care services (HC) group and 27 in the non-home care services (non-HC) group, all of whom were discharged early after inpatient treatment. Data were collected from medical records. Direct medical costs were calculated using medical fee payment data. Cost-effectiveness ratio was calculated using direct medical costs paid by the patient and the insurer until complete cure of the diabetic foot. Effectiveness was the time required for a complete cure. Direct medical costs included fees for hospitalization, emergency care, home care, ambulatory fees, and hospitalization or ambulatory fees at other medical institutions. Results: Mean for direct medical costs was 11,118,773 won per person in the HC group, and 16,005,883 won in the non-HC group. The difference between the groups was statistically significant (p=.042). Analysis of the results for cost-effectiveness ratio showed 91,891 won per day in the HC patients, and 109,629 won per day in the non-HC patients. Conclusion: Result shows that the cost-effectiveness ratio is lower HC patients than non-HC patients, that indicates home care services are economically feasible.

고지방식이로 고지혈증이 유도된 흰쥐에서 황화된 수용성 β-glucan의 항고지혈증 효과 (Antihyperlipidemic Activities of a Chemically Engineered Sulfated Mushroom β-glucan on High Fat Dietary-induced Hyperlipidemia in Sprague-Dawley Rats)

  • 본 연구는 영지버섯 균사체로부터 얻은 수용성 베타글루칸(SBG)이 고지방식이로 유도한 흰쥐의 항고지혈 효과 및 혈중 지질수준에 미치는 영향을 조사하였다. 5주령의 수컷 흰쥐에 고지방식이를 2주간 급여하여 비만을 유도하고, 그 후 2주 동안 다음과 같은 5가지 group의 식이를 급여하였다: 1) 일반사료 식이군(NC), 2) 고지방사료 식이군(HC), 3) 고지방사료 및 고농도(200 mg/kg) SBG 식이군(HC-HSBG) 4) 고지방사료 및 저농도(20 mg/kg) SBG 식이군(HC-LSBG), 5) 고지방사료 및 로바스타틴(lovastatin) 식이군(HC-Lov). 실험 결과, 식이 효율(Food efficiency ratio)은 고지방식이를 급여한 군들에서 일반사료 식이군에 비하여 증가하였으나 통계적 유의성은 없었다. 혈액학적 결과에서 고농도 및 저농도의 SBG와 로바스타틴 투여군은 고지방식이만 급여한 군(HC)보다 호중구와 중성구 수치가 유의성 있게 증가하였다. 또한 혈청 내 총콜레스테롤, 중성지질 및 LDL-C는 고농도 및 저농도 SBG 투여군 모두에서 유의성 있게 감소되었다. 이 같은 결과는 화학적으로 황화된 수용성 베타글루칸(SBG)이 흰쥐에서 혈중 콜레스테롤 수치를 낮추어 항고지혈증 효과를 나타내는 것을 의미한다.

Effects of a traditional Chinese medicine formula and its extraction on muscle fiber characteristics in finishing pigs, porcine cell proliferation and isoforms of myosin heavy chain gene expression in myocytes

  • Objective: This study evaluated the effects of a traditional Chinese medicine formula (TCMF) on muscle fiber characteristics in finishing pigs and the effects of the formula's extract (distilled water, ethyl acetate and petroleum ether extraction) on porcine cell proliferation and isoforms of myosin heavy chain (MyHC) gene expression in myocytes. Methods: In a completely randomized design, ninety pigs were assigned to three diets with five replications per treatment and six pigs per pen. The diets included the basal diet (control group), TCMF1 (basal diet+2.5 g/kg TCMF) and TCMF2 (basal diet+5 g/kg TCMF). The psoas major muscle was obtained from pigs at the end of the experiment. Muscle fiber characteristics in the psoas major muscle were analyzed using myosin ATPase staining. Cell proliferation was measured using 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) dye and cytometry. Isoforms of MyHC gene expression were detected by real-time quantitative polymerase chain reaction. Results: The final body weight and carcass weight of finishing pigs were increased by TCMF1 (p<0.05), while the psoas major muscle cross-sectional area was increased by TCMF (p<0.05). The cross-sectional area and diameter of psoas major muscle fiber Ι, IIA, and IIB were increased by TCMF2 (p<0.05). The cross-sectional area and fiber diameter of psoas major muscle fiber IIA and IIB were increased by diet supplementation with TCMF1 (p<0.05). Psoas major muscle fiber IIA and IIB fiber density from the pigs fed the TCMF1 diet and the type IIB fiber density from the pigs fed the TCMF2 diet were lower compared to pigs fed the control diet (p<0.05). Pigs fed TCMF2 had a higher composition of type Ι fiber and a lower percentage of type IIB fiber in the psoas major muscle (p<0.05). The expression levels of MyHC Ι, MyHC IIa, and MyHC IIx mRNA increased and the amount of MyHC IIb mRNA decreased in the psoas major muscle from TCMF2, whereas MyHC Ι and MyHC IIx mRNA increased in the psoas major muscle from TCMF1 (p<0.05). Peroxisome proliferator-activated receptor ${\gamma}$ $coactivator-1{\alpha}$ and CaN mRNA expression in the psoas major muscle were up-regulated by TCMF (p<0.05). Porcine skeletal muscle satellite cell proliferation was promoted by $4{\mu}g/mL$ and $20{\mu}g/mL$ TCMF water extraction (p<0.05). Both $1{\mu}g/mL$ and $5{\mu}g/mL$ of TCMF water extraction increased MyHC IIa, MyHC IIb, and MyHC IIx mRNA expression in porcine myocytes (p<0.05), while MyHC Ι mRNA expression in porcine myocytes was decreased by $5{\mu}g/mL$ TCMF water extraction (p<0.05). Porcine myocyte MyHC Ι and MyHC IIx mRNA expression were increased, and MyHC IIa and MyHC IIb mRNA expression were down-regulated by $5{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). MyHC Ι and MyHC IIa mRNA expression in porcine myocytes were increased, and the MyHC IIb mRNA expression was decreased by $1{\mu}g/mL$ TCMF ethyl acetate extraction (p<0.05). Four isoforms of MyHC mRNA expression in porcine myocytes were reduced by $5{\mu}g/mL$ TCMF petroleum ether extraction (p<0.05). MyHC IIa mRNA expression in porcine myocytes increased and MyHC IIb mRNA expression decreased by $1{\mu}g/mL$ in a TCMF petroleum ether extraction (p<0.05). Conclusion: These results indicated that TCMF amplified the psoas major muscle cross-sectional area through changing muscle fiber characteristics in finishing pigs. This effect was confirmed as TCMF extraction promoted porcine cell proliferation and affected isoforms of MyHC gene expression in myocytes.

MDS-HC 2.0을 이용한 일부 독거노인의 건강문제 조사연구 (A Study the Health Problems of the Living-alone Elderly Using MDS-HC 2.0)

  • Purpose: This study used MDS-HC 2.0 to identify health problems in the elderly who live alone. Method: The subjects of this study were 100 living-alone elders aged over 65 in Daegu city. This study was performed through home visiting interviews during the period from the 1st of September to the 30th of September 2005. Analysis in this study was made using SPSS Version 11.0. Results: Frequent health problems in the living-alone elderly were: preventive heath measure (90), health promotion (80), social function (78), visual function (75), depression and anxiety (61), pain and communication disorder (60), cognition (55), environmental assessment (48) and oral health (45) in order of frequency. Informal services were provided to 73% of the subjects for emotional support. IADL services were provided to 57% of the subjects, and ADL services to 66%. Conclusion: Nursing intervention programs based on health problems from MDS-HC 2.0 should be provided in order to cope with individual health problems and living-alone elders needs. The results of this study suggest that MDS-HC 2.0 is applicable to help decide nursing interventions for the elderly who live alone in community.

  • Monoclonal antibodies (MoAbs) are a highly diversified class of proteins with major research and commercial applications such as diagnostics and therapeutics. Currently, the dominant method for producing MoAbs is through the hybridoma technique. However, this technique is slow, tedious, labor intensive, and expensive. The production of MoAbs in cultured transgenic plant cells can offer some advantages over that in the over that in the mammalian systems. The media to cultivate plant cells are well defined and inexpensive. Contamination by bacteria or fungi is easily monitored in plant tissue cultures. Furthermore, these contaminants are usually not potent pathogens to human beings. In our interdisciplinary research efforts, heavy chain monoclonal antibody (HC MAb) was inserted into Ti plasmid vector and transferred into A. tumefaciens for the transformation in tobacco cells. It was found that 76% of the transformants produced HC MAb. The presence of HC MAb in the cell membrane fraction indicated that the signal peptide was functional and efficient. The change of the HC MAb concentration during a batch culture followed a similar trend as dry cell concentration, indicating that the production of HC MAb was growth related. The long-term repeated subcultures of 11 cell lines showed that there was no obvious trend of neither the decrease nor the increase of the productivity with the repeated subcultures.

돼지 $\beta$-Casein을 이용한 EGFP 발현 Knock-in 벡터의 구축 및 발현 검증 (Construction and Expression Analysis of Knock-in Vector for EGFP Expression in the Porcine $\beta$-Casein Gene Locus)

  • 본 연구는 돼지 $\beta$-casein 유전자 위치에서 EGFP가 발현될 수 있는 knock-in 벡터를 구축하기 위하여 실시되었다. 돼지의 $\beta$-casein 유전자를 이용하여 knock-in 벡터를 구축하기 위해 돼지의 태아 섬유아세포로부터 $\beta$-casein 유전자를 동정하였고 EGFP, SV4O polyA signal을 동정하였다. Knock-in 벡터는 5' 상동 영역 약 5 kb와 3' 상동 영역 약 2.7 kb로 구성되어있으며, positive selection marker로 $neo^{r}$ 유전자를, negative selection marker로 DT-A 유전자를 사용하였다. 구축된 knock-in 벡터로부터 EGFP의 발현을 확인하기 위하여 생쥐 유선 세포인 HC11 세포에 knock-in 벡터를 도입하였다. 그 결과 EGFP의 발현을 HC11 세포에서 확인하였다. 이와 같은 결과로서 이 block-in 벡터는 knock-in 형질전환 돼지를 생산하는데 사용될 수 있을 것으로 생각된다.

Monascus pilosus로 발효시킨 후발효 녹차가 고지방 식이 마우스의 체중과 혈청 지방함량 및 간 조직 항산화계 효소활성에 미치는 영향 (Dietary Effects of Post-fermented Green Tea by Monascus pilosus on the Body Weight, Serum Lipid Profiles and the Activities of Hepatic Antioxidative Enzymes in Mouse Fed a High Fat Diet)

  • Monascus pilosus로 발효시킨 후 발효 녹차의 첨가식이가 고지방식이 ICR 마우스의 체중과 혈청 지방함량 및 간 조직 항산화계 효소활성에 미치는 영향을 조사하였다. 정상대조군(NC), 고지방식이대조군(HC), 비발효녹차 분말을 2% 첨가한 고지방식이군(CHA) 및 M. pilosus로 발효시킨 후발효녹차 분말을 2% 첨가한 고지방식이군(mCHA)으로 구분하여 7주간 사육하였다. 체중은 CHA군과 mCHA군간의 유의적인 차이가 없으나 HC군에 비하여 각각 24.15 및 29.66%가 감소되었으며 식이섭취량에는 차이가 없었다. 체중 당 간 및 신장의 중량은 CHA군 및 mCHA군이 HC군에 비해 증가하였다. 체중 당 부고환주위지방의 무게는 mCHA군에서 유의하게 감소하였다. 혈청 TG와 의총콜레스테롤 함량은 mCHA군에서 NC군 수준으로 감소하였다. CHA군과 mCHA군의 HDL-cholesterol 함량은 HC군에 비하여 96.55 및 119.32%가 높았다. CHA군과 mCHA군의 CHA군 및 mCHA군의 LDL-cholesterol 함량은 각각 0.67 및 0.41을 나타내었다. 간 조직의 GSH 함량은 HC군에 비하여 CHA군이 11.24%, mCHA군은 23.68%가 높았다. LPO 함량은 CHA군과 mCHA군이 HC군에 비하여 43.70 및 45.38%가 낮았다. CHA군과 mCHA군의 xanthine oxidoreductase D type 효소활성은 HC군에 비하여 각각 18.52 및 33.33%가 감소하였으며 mCHA군은 NC군 수준으로 높았다. XOD O type 효소활성은 CHA군과 mCHA군이 HC군에 비하여 각각 16.67 및 33.33%가 높았다. superoxide dismutase 활성은 CHA군과 mCHA군이 HC군에 비하여 유의차는 없었다. mCHA군의 glutathione S-transferase 활성은 HC군에 비하여 유의적으로 높았으며 특히, mCHA군의 glutathione peroxidase 활성은 HC군의 2.55배, CHA군의 1.5배로 높았으며 NC군과 대등하였다. CHA군과 mCHA군의 ALT 활성 HC군에 비하여 현저하게 낮았다. 간 조직의 현미경 관찰결과, CHA군과 mCHA군에서는 간 세포 내 축적된 지방의 크기가 작은 포말형으로 변화되고, 이로 인해 간 세포판과 동양혈관 형성이 간 소엽단위로 뚜렷하게 구분되었다. 이상의 실험결과로 보아 M. pilosus로 발효시킨 후발효 녹차는 고지방식이에 의해 유발되는 비만과 고지혈증 및 지방간을 예방 혹은 경감시켜줄 수 있는 유효한 가능성을 시사하였다.

  • 1990년 1월부터 1993년 12월까지 영남대학교 의과대학 부속병원에서 출생한 적정체중아(appropriate for gestational age infants) 207명을 대상으로 중앙 상완위 및 두위를 측정하여 관찰한 결과 재태 연령의 증가에 따라 중앙 상완위(y=0.3181x-2.2069, r=0.81 p<0.001)와 중앙 상완위 대두위 비(y=0.0049x+0.1128, r=0.62, p<0.001)는 직선적인 증가를 보였으며 이를 기초로 작성된 재태 연령에 대한 표준 곡선은 향후 신생아의 발육 상태를 평가하는 참고치로서 활용될 수 있을 것으로 생각된다.

  • Six monoclonal antibodies to human cardiac troponin I (hcTnI) were produced to eventually develop an immunosensor for acute myocardial infarction (AMI). For the characterization of these antibodies, a set of 11 different peptides covering selected ranges of the complete amino acid sequence of hcTnI was prepared and used for epitope mapping. Such analysis allowed to select an appropriate pair of antibodies that can form a sandwich type of immune complexes and was consequently used for an immunoassay.
