Protective Immunity of Pichia pastoris-Expressed Recombinant Envelope Protein of Japanese Encephalitis Virus

  • Japanese encephalitis virus (JEV) envelope (E) protein holds great promise for use in the development of a recombinant vaccine. Purified recombinant E (rE) protein may be useful for numerous clinical applications; however, there are limitations in using the Escherichia coli expression system for producing high-quality rE protein. Therefore, in this study, the yeast expression system was used to generate the rE protein. For protein production using the yeast system, the full-length JEV E gene was cloned into Pichia pastoris. SDS-PAGE and immunoblotting analysis demonstrated that the rE protein had a molecular mass of 58 kDa and was glycosylated. The predicted size of the mature unmodified E protein is 53 kDa, suggesting that post-translational modifications resulted in the higher molecular mass. The rE protein was purified to greater than 95% purity using combined ammonium sulfate precipitation and a SP-Sepharose Fast Flow column. This purified rE protein was evaluated for immunogenicity and protective efficacy in mice. The survival rates of mice immunized with the rE protein were significantly increased over that of Hyphantria cunea nuclear polyhedrosis virus E protein (HcE). Our results indicate that the rE protein expressed in the P. pastoris expression system holds great promise for use in the development of a subunit vaccine against JEV.

The Influences of Visual and Hearing Impairment on Activities of Daily Living for the Community Dwelling Elderly (재가노인의 시청각기능장애가 일상생활수행능력에 미치는 영향)

  • Purpose: The aim of this study is to identify the influence of visual and hearing impairment on the activities of daily living of community dwelling elderly. Methods: Data were collected by home visiting interviewers from 452 older people aged 65 years or older living in community. Resident Assessment Instrument MDS-HC(2.0version) was used for data collection. Data analysis for descriptive statistics, Chi-square test and multiple regression was made by SAS 6.2 Results: 34.7% of the subject had hearing impairment and 64.3% had visual impairment Among IADL. one half of them were dependent in ordinary house work and meal preparation. In the case of ADL. 13.9% of subjects were dependent in bathing and 8.9% in personal hygiene. There was significant difference in IADL performance by visual and hearing impairment On the other hand, ADL performance showed the significant difference. only in the case of hearing impairment. As the result of input of visual and hearing impairment in the process of regression. variances were increased from 3% to 11%. Conclusions: Large proportions of older people living in the community have visual and hearing impairment. It could be confirmed that hearing and vision were significant factors influencing on IADL performance of older people. Intervention and support policy for elderly needs to focus on improvement of visual and hearing impairment.

  • The expression of a recombinant human lactoferrin is reported in mouse HC11 mammary epithelial cells. Expression of human lactoferrin (hLF) was achieved by placing its cDNA under the control of the bovine ${\beta}$-casein gene. To improve the hLF expression level in a cell culture system, two artificial introns were also introduced to construct expression vectors. One intron was a hybrid-splice signal consisting of bovine ${\beta}$-casein intron 1 and rabbit ${\beta}$-globin intron II. The other intron was a DNA fragment spanning intron 8 of the bovine ${\beta}$-casein gene. The hybrid intron moderately elevated hLF expression, whereas intron 8 alone did not express any detectable amount of hLF as judged by Northem and Western blot analyses. When the two introns were used together they contributed to a synergistic elevation of hLF expression. These data indicate that artificial introns on both sides of the hLF cDNA were necessary to increase expression of cDNA.

Productivity and Nutritive Values of Different Fractions of Oil Palm (Elaeis guineensis) Frond

  • Productivity, nutrient contents, in vitro gas production and in sacco degradability of different fractions and whole OPF were determined to assess the feeding value of OPF as a ruminant feed. An in vivo digestibility trial was also carried out using goat. Freshly harvested OPF was randomly collected, partitioned and weighed. An OPF from 21 years older palm weighed 13.4 kg and the annual fresh matter yield of petiole, leaflet and midrib was 46.5, 11.8 and 3.4 ton/ha, respectively. Leaflet contained 439, 926, 698, 501, 168, 196, 748 and 52 (g/kg) of dry matter (DM), organic matter (OM), neutral detergent fiber (NDF), acid detergent fiber (ADF), cellulose (CE), hemicellulose (HC), total carbohydrate (TC) and non fiber carbohydrate (NFC), respectively. Petiole contained lower (p<0.01) DM, CP and EE contents than leaflet. Organic matter, CE and TC contents were higher (p<0.01) in petiole compared to leaflet. Silica and lignin contents were highest (p<0.01) in midrib followed by leaflet, whole OPF and least in petiole. The Ca, P, Na, K and Mg contents (g/100 g DM) of leaflet were 0.529, 0.182, 0.039, 0.876, and 0.168, respectively. In vitro DM digestibility (g/100 g) at 48 h of leaflet, petiole and midrib was 32.7, 38.7 and 30.2, respectively. The in sacco DM degradation (g/100 g) at 48 h of leaflet was higher than that of whole OPF, petiole and midrib. The in vivo digestibility of DM, OM, CP and ADF of whole OPF was 52, 56, 43 and 26%, respectively. It can be concluded that leaflet is the most nutritious fraction of OPF and midrib is the least. The nutrient content and digestibility of the whole OPF showed that OPF could be an alternative roughage source for ruminant diets.

Clinical Prediction Based on HPV DNA Testing by Hybrid Capture 2 (HC2) in Combination with Liquid-based Cytology (LBC)

  • Primary screening by HPV DNA testing is an effective method for reducing cervical cancer and has proven more sensitive than cytology. To advance this approach, many molecular methods have been developed. Hybrid capture 2 provides semi-quantitative results in ratios of relative light units and positive cutoff values (RLU/PC). Twenty-five thousand and five patients were included in this study to analyze the correlation between the ratio of RLU/PC and stage of cervical dysplasia. The results show that the RLU/PC ratios ranged from 0-3500 while almost normal cases, ASC-US and ASC-H, had values below 200. Of those samples negative for cytology markers, 94.6% were normal and their RLU/PC ratios were less than 4. With an RLU/PC ratio greater than 4 and less than or equal to 300, the percentages in all age groups were normal 53.6%, LSIL 20.2%, ASC-US 17.2%, HSIL 6.13%, ASC-H 2.72%, and AGC 0.11%, respectively. In contrast, 64.0% of samples with a RLU/PC ratio greater than 300 and less than or equal to 3500 were LSIL. These results should contribute to cost effective cervical cancer management strategies. Further studies of associations with particular HPV genotypes would be useful to predict the risk of progression to cancer.

  • Three minerals in orchard grass (Dactylis glomerata Linn.) grown under different potassium (K20) levels in water culture, potassium(K), magnesium(Mg) and calcium(Ca) were divided into water, alcohol, and acid soluble fractions to know the reason of different lea¬ching rate. The results were as follows; (1) The percentage of K in each fraction to total K was not affected by the fertilization levels, and it was 90-93, 4-6, and 2~3% in water, methanol and 0.5% HCI soluble fractions, respectively. (2) About 800/c of the extractive Mg was water soluble, and 14-22% of total Mg were extracted by O. 5% HC! solution. (3) The percentage of water, O. 5% HCI- and 1 M HCI-soluble Ca to total Ca were affected by the fertilization levels, and were 26-46%, 44-54%, and 8-22%, respectively. From the above facts, it is known that the solubility in water was the highest in K and followed by Mg and Ca. While the solubility in HCI solution was opposite to that in water.

  • Porous glass ceramics composed of Li$_2$O.-Ti $O_2$-P$_2$ $O_{5}$ -CaO were prepared by melting and 2 step heat treatment for nucleation at 61$0^{\circ}C$ and crystallization at 78$0^{\circ}C$. subsequently $\beta$-Ca$_3$(P $O_4$)$_2$crystal phase was selectively leached out in 1N-HC1 solution for 3 days, leaving Li $Ti_2$(P $O_4$)$_3$crystal phases. prepared porous glass ceramics were immersed in 1M AgN $O_3$solution for ion exchange. Staphylococcus aureus and salmonella typhi were used in this study. It was found taht the resultant porous glas ceramics show excellent bacteriostatic properties.

  • This paper deals with the development of Laser Doppler Vibrometer (LDV) that can mea~ure the tlequency and amphlude of the ultrasonic vibralion. Hc-Ne laser (632.8 om) is used as a light source, and Michelson interferometer in winch frequency of the objective beam is shIfted by Bragg cell IS adopted The frequency modulated signal centered at 40 MHz flom the PIN diode IS amplified. down-col1vel1ed to 2.5 MHz, filtered and digiLized. The voltage output that is proportional to the velocity of the vibratwg surface is obtawed using digItal PLL. A microprocessor is used to extract the frequcncy aud amplitude of the vibratIOn from the voltage output. It is found that the developed LDV can measure up to 300 kHz vibratIOn and the mlillmUITI measurable amplitude is I nm at 300 kHz. We believe thatlhis LDV can be used to measure the vibratIOn of the heavy electric maclllnery and micro-size structures. tures.

A Study on the Environmental Carrying Capacity Assessment of Chongju City (도시 환경용량평가에 관한 연구 -청주시를 사례로-)

  • The purpose of this study is to assess the environmental carrying capacity of Chongju City for the environmental management and the urban growth management. The urban environmental carrying capacity assessment of the city by the index of ecological footprint(EF), shows that the ecosystem of the city has been overloaded and most of the deficiencies has come from outside of the city. The EF index, the area of land per capita required for production and consumption in the city, was 1.731 ha per capita in 1989 and 1.901 ha per capita in 1999. On the other side, the ecologically productive land is 0.0175 ha per capita. It means that every citizen owes 1.88 ha per capita to the ecosystem in 1999. The land consumption of the city has increased by 0.1705 ha per capita during the last 10 years. The capacity of infrastructure and the service supply estimated by the Onishi model does not exceed the demand of the city in 1999. But the rapidly increasing population and fast urban growth need the expansion of the capacity. The water supply capacity of the city appears to be sufficient in 1999, but the water supply demand will increase in the future. The capacity of sewage treatment facilities seems to be sufficient, but the higher level of sewage treatment facilities should be adopted for the improvement of water quality as the generation of sewage will increase and its characteristics will also make the wastewater treatment difficult. Due to the decrease of solid waste generated, the land fill capacity for solid waste disposal is not insufficient at present, but the capacity will be saturated in the near future. Therefore, the scientific management system of solid wastes should be introduced. The air quality of the city meets both the national air quality standard and WHO recommendation standard, but the strong regulation and control of automobile emission gas such as CO, $CO_2$, NOx and HC is required for clean air.

Influence of toothbrush abrasion and surface treatments on the color and translucency of resin infiltrated hybrid ceramics

  • PURPOSE. The study compared the color change, lightness, and translucency of hybrid resin ceramics exposed to toothbrush abrasion and surface treatment. MATERIALS AND METHODS. Four hybrid ceramics [Lava Ultimate (LU), Vita Enamic (EN), Shofu HC (SH), and Crystal Ultra (CU)] were compared with a glass-ceramic (Vita Mark II) control. One hundred and twenty specimen blocks were prepared using a precision saw machine. Specimens in each material were divided into four subgroups based on the surface treatment (polishing or staining) and a storage medium (water or citric acid). Simulated tooth brushing with a mixture of 100 RDA (radioactive abrasives) with 0.3 ml distilled water was used for 3650 cycles (7300 strokes) for each specimen. Measurements for the color change, lightness, and translucency were measured after toothbrushing using a spectrophotometer. Statistical analysis compared outcomes using paired t-test, ANOVA, and Tukey post hoc test. RESULTS. The maximum color change was identified in SH (stained acid) [1.44 (0.40)], whereas the lowest was identified in EN (polished water) [0.66 (0.16)] material. The maximum and minimum loss of surface translucency was observed in SH (polished water) [12.3 (0.52)] and EN (stained acid) [6.5 (0.55)] specimens, respectively. Lastly, loss of lightness was the highest in VM (polished acid) [69 (0.95)], whereas the lowest was observed in CU (stained water) [56.7 (0.86)]. CONCLUSION. The comparison presented a significant effect of toothbrush abrasion on translucency and lightness of the hybrid resin ceramics. Color change was not significantly influenced irrespective of the storage medium employed. Surface staining demonstrated the preservation and stability of color and optical properties under the influence of toothbrush abrasion and chemical trauma.
