The ${\beta}$-Lactoglobulin (BLG) gene expression is differentially regulated during development of the mammary tissues. Such differential regulation of the BLG gene expression can be reiterated in the cultured mammary HC11 cells. In the growing non-confluent HC11 cells, the BLG promoter activity was shown to be partially repressed by the upstream regulatory sequence. The repression was gradually diminished and switched to activation as the cells grew confluent. The differential regulation of the BLG promoter was controlled by the 5'-regulatory sequence located at the upstream of 205 bp. Electromobility shift assay showed that nuclear extract from HC11 cells differentially bound on the regulatory sequence, depending on the cell confluency, which was in accordance with the differential transcriptional activity. DNase I foot-print assay, however, revealed that all nuclear extracts presented the same foot-prints, regardless of confluency of HC11 cells. These results suggest that differential regulation BLG gene expression by the 5'-regulatory sequence may be accomplished by competitive and/or cooperative binding of differential regulatory factors on the same regulatory element.

This paper presents a new evaporative system monitoring method using a virtual HC sensor for an automotive on-board diagnosis. A development was made at providing mathematical expressions from the lambda control information to estimate the HC mass flow purged into the intake manifold from the canister for implementing a virtual HC sensor. The change of the lambda averagevalue reflected the influence of the additional fuel from purging results the sensor estimation of the purged HC amount. Based on this virtual HC sensor, a new evaporative system monitoring method was proposed comparing the amount of purged HC amount with the amount of the HC gas evaporated from the fuel tank and absorbed into the canister. Finally, the method was validated with a simulation using the data logged from the retail car.

The large and rapid changes of glucose utilization in lactating mammary tissue in response to changes in nutritional state must be largely related by external signal of insulin. This also must be related with the quantity and composition of the diet in vivo. To characterize the mode of gut extract protein with insulin, in vitro experiment was conducted with HC11 cells. The gut extract protein has not only the same effect as insulin alone but also the synergistic effect with insulin in 2-Deoxy[3H] glucose uptake. Although the gut extract did not modulates glucose uptake via increasing the rate of translation of the GLUT1 protein, northern blot analysis indicated that the gut extract protein increased the expression of GLUT1 mRNA by a threefold and also there was a dose-dependent increase in the expression of GLUT1 mRNA. The gut extract protein is therefore shown to be capable of modulating glucose uptake by transcription level with insulin in HC 11 cells.

락토페리신은 다양한 생리활성을 나타내는 락토페린(약 80kD)에서 유래된 항균성펩타이드 분획물(5kD)이다. 마우스HC11유선상피세포에서 인체 락토페리신의 발현은 bovine beta-casein을 promotor로 하고 인체 락토페리신 cDNA를 삽입하여 제작한 pBL1-cin발현벡타를 이용하였다. 이 발현벡타를 이용하여 인체 락토페리신 발현여부를 RT-PCR, northern blot, dot blot분석을 통하여 확인하였다. pBL1-cin 발현백타를 HC11세포에 transfection 하여 얻은 RNA를 이용하여 RT-PCR를 한 결과 150bp의 크기로 확인되었고 Northern blot 분석결과는 약 2.3 kb의 크기로 확인되었다. 인체 락토페린 polyclonal항체를 이용하여 dot blot한 결과 인체 락토페리신이 분비됨을 확인하였다.

Background: Efficient gene editing technology is needed for successful knock-in. Homologous recombination (HR) is a major double-strand break repair pathway that can be utilized for accurately inserting foreign genes into the genome. HR occurs during the S/G2 phase, and the DNA mismatch repair (MMR) pathway is inextricably linked to HR to maintain HR fidelity. This study was conducted to investigate the effect of inhibiting MMR-related genes using CdCl₂, an MMR-related gene inhibitor, on HR efficiency in HC11 cells. Methods: The mRNA and protein expression levels of MMR-related genes (Msh2, Msh3, Msh6, Mlh1, Pms2), the HR-related gene Rad51, and the NHEJ-related gene DNA Ligase IV were assessed in HC11 cells treated with 10 μM of CdCl₂ for 48 hours. In addition, HC11 cells were transfected with a CRISPR/sgRNA expression vector and a knock-in vector targeting Exon3 of the mouse-beta casein locus, and treated with 10 μM cadmium for 48 hours. The knock-in efficiency was monitored through PCR. Results: The treatment of HC11 cells with a high-dose of CdCl₂ decreased the mRNA expression of the HR-related gene Rad51 in HC11 cells. In addition, the inhibition of MMR-related genes through CdCl₂ treatment did not lead to an increase in knock-in efficiency. Conclusions: The inhibition of MMR-related gene expression through high-dose CdCl₂ treatment reduces the expression of the HR-related gene Rad51, which is active during recombination. Therefore, it was determined that CdCl₂ is an inappropriate compound for improving HR efficiency.

Calculations are presented for the molecule HC2N and its geometrical isomers. The structures, harmonic frequencies and dipole moments are reported. The potential energy surface of the [H,C,C,N] system is investigated in detail, and the transition states, intermediate complexes, and the energies of barrier for the isomerization and dissociation reactions are computed in order to determine the reaction paths and to estimate the stability of the isomers. The barriers of isomerization among HCCN, HCNC and HNCC are computed to be rather large and dissociations of these molecules are highly endothermic, indicating that these molecules are kinetically stable. The association reactions HC + CN→HCCN, HC + NC→HCNC, and HN + CC →HNCC are barrierless and very exothermic, suggesting that they may be considered as efficient means of producing the HCCN and the isomers in the laboratory and in interstellar space.

Engine-out HC emissions were investigated during cold and hot start. The tests were conducted according to engine cooling temperatures which were controlled by simulated coolant temperatures of cold and hot start, on a 1.5L, 4-cylinder, 16 valve, multipoint-port-fuel-injection gasoline engine. Real time engine-out HC emissions were measured at a exhaust port and cylinder head using Fast Response Flame Ionization Detector(FRFID). Unburned hydrocarbons emitted at the cold coolant temperature were much higher than those of the hot coolant temperatures. And the main source of the high HC emission was confirmed as misfire at cold coolant temperature. In addition, the effect of intake valve timing on engine-out HC emissions was investigated. The results obtained indicate that optimized intake phasing provides the potential for start-up engine-out HC emissions reduction.

본 논문의 연구에서는 IEEE 802.11e HCCA에서 멀티미디어와 같은 트래픽에 대해 보다 안정적인 전송을 위하여 일정 시간 동안 채널을 사용할 수 있도록 HC (Hyper Coordinator)가 허가함에 있어서 IEEE 802.11 PHY (physical layer)의 모듈레이션 변화에 따른 동적인 대응이 필요함을 논하고, 문제 해결을 위한 방안을 제시하였다. IEEE 802.11e의 HC는 멀티미디어와 같은 트래픽의 전송을 위하여 BSS (Basic Service Set) 안에 있는 무선 단말기와 TSPEC (Traffic Specification)을 이용하여 CAP (Controlled Access Phase) 채널을 설정하고 관리한다. 그러나 HC가 TSPEC을 설정할 당시와는 달리 무선 단말기들은 이동하거나 물리적으로 채널 환경의 변화로 IEEE 802.11 PHY의 모듈레이터 변경으로 설정된 TXOP 듀레이션 (duration)이 적절하지 않게 된다. 이와 같은 문제점을 밝히고 해결 방안을 아래와 같이 제안한다. 실험을 위해서는 NS-2와 유사하지만 보다 세련된 시뮬레이터를 활용하였다.

To characterize the mode of growth factors and gut regulatory peptides with insulin, in vitro experiment was conducted with HC11 cells. All the growth factor alone and the combinations of growth factors significantly (p<0.05) increased in glucose uptake. Insulin, EGF and IGF-1 exhibited a stimulation of glucose uptake for at least 24 h. Furthermore, the highest (p<0.05) synergistic effect was shown in EGF plus IGF-1 and the second synergistic effect in insulin plus EGF while no synergistic effect was found between insulin and IGF-1. However, the gut regulatory peptides neither potentiated nor inhibited the action of insulin on glucose uptake. Although growth factors did not modulates glucose uptake via increasing the rate of translation of the GLUT1 protein, RT-PCR analysis indicated that the growth factors significantly (p<0.05) increased the expression of GLUT1. The growth factors are therefore shown to be capable of modulating glucose uptake by transcription level with insulin in HC 11 cells.

We have observed the 10-9 transitions of $HC_3N$ and its $^{13}C$ substitutes ($H^{13}CCCN,\;HC^{13}CCN$, and $HCC^{13}CN$), and the vibration ally excited 12-11 ($v_r=1$) $HC_3N$ transition toward the Sgr B2 molecular cloud. The observed $HC_3N$ emission shows an elongated shape around the Principal Cloud ($\~$4.5 pc in R.A. $\times$ 7.4 pc in Decl.). The optically thin $H^{13}CCCN$ line peaks around the (N) core and we derive the total column density $N(H^{13}CCCN) = 4 {\times}10^{13} cm^{-2}$ at this position. Toward the 2' N cloud which shows the peculiar chemistry, the $HC_3N$ lines show enhancements compared to the extended envelope. The shocks of the 2' N may have resulted in the enhancement of $HC_3N$. The hot component of $HC_3N$ is strongly concentrated around the (N) core and its HPW is $\~$0.9 pc in diameter. We derive the lower limit of the abundance ratio $N(HC_3N)/N(H^{13}CCCN)$ to be larger than 40 in most regions except the (M) and (N) cores. The fractionation processes of $^{13}C $at this region may not be as effective as previously reported.
