펜톤시약을 이용한 이온교환수지의 직접분해 (The Direct Dissolution of Ion-Exchange Resin by Fenton's Reagent)

  • 양이온교환수지인 IRN-77을 직접 분해 처리하기 위하여 Fonton시약을 적용하였으며, 분해반응의 특징으로써 반응의 효율 및 안전을 위해 수지를 먼저 건조시키고 $FeSO_4$ 용액을 수지에 완전히 흡수시킨 후 $H_2O_2$를 첨가하는 방법을 적용하였다. 수지 분해반응의 특성은 반응이 개시되기까지 반응유도시간이 필요하였으며, 반응유도시간은 $FeSO_4$의 농도가 낮을수록 또한 $H_2O_2$의 초기 첨가량이 적을수록 길었다. 단위량의 수지를 분해하는데 적절한 반응조건으로서 $FeSO_4$의 농도는 0.9 M 및 15% $H_2O_2$의 용액의 첨가량은 수지량에 대해 6-7배 비율로 나타났으며, 반응유도시간을 포함하여 1.5시간 이내에 완전 분해가 가능하였다. $H_2O_2$의 첨가방법은 반응 초기 및 반응개시 후로 나누어 첨가하므로서 $H_2O_2$의 분해효율 및 첨가량을 최소화하였다. 가열효과로서 분해반응 개시 전에 비교적 낮은 온도인 $50^{\circ}C$ 정도로 가열하면 반응유도시간이 5분 이내로 단축되었으며, 수지의 양을 5g 및 10g 으로 증가시킨 결과, $H_2O_2$의 첨가비율을 9-10배 정도로 증가시키면 완전분해가 가능하였다.

Effect of Genistein on Antioxidative Defence System and Membrane Fluidity in Chick Skeletal Muscle Cells

  • This study was conducted to investigate the protective effect of genistein on the antioxidative defence system and membrane fluidity in chick skeletal muscle cells after supplementation with 0, 20, 40, and $80{\mu}mol/L$ genistein in $50{\mu}mol/L$ $FeSO_4/H_2O_2$ treated cells for 24 h. Genistein supplementation recovered the decreased activity of total superoxide dismutase induced by $FeSO_4/H_2O_2$, significantly increased glutathione peroxidase activity (p<0.05) and decreased malondialdehyde production (p<0.05). The treatment of 80 mol/L genistein in $FeSO_4/H_2O_2$ treated cells decreased the secretion of creatine kinase (p<0.05). Fluorescence polarization values and microviscosities observed with $FeSO_4/H_2O_2$ treated cells were significantly higher than those observed with no $FeSO_4/H_2O_2$ treated cells. The addition of $80{\mu}mol/L$ genistein improved the increased fluorescence polarization value (p<0.05) caused by $FeSO_4/H_2O_2$ treatment. The microviscosity value was significantly decreased by adding genistein (p<0.05). In conclusion, genistein protected skeletal muscle cells from oxidative damage by improving antioxidative status and membrane fluidity.

알칼리 침전제에 의해 제조된 아연 중간생성물 및 산화아연 결정화 (Synthesis of Zn-intermediate from alkali agents and its transformation to ZnO crystallinity)

  • ZnO는 출발물질인 황산아연과 알칼리 침전제인 NaOH와 Na2CO3에 의해 생성된 아연 중간생성물의 변이 거동과 결정화 조건에 따라 제조하였다. ZnO 결정화를 위해 아연 중간생성물인 Zn4(OH)6SO4·H2O, Zn5(OH)6(CO3)2·H2O를 각각 400℃, 800℃에서 1시간 하소하였고, 하소 온도는 열중량 분석을 기반으로 하였다. 아연 중간생성물인 Zn4(OH)6SO4·H2O은 400℃에서 Zn4(OH)6SO4·H2O, ZnO 결정상이 혼재됨을 확인하였고, 800℃에서 완전히 열분해되어 순수한 ZnO만 형성되었다. Na2CO3와 반응하여 생성된 아연 중간생성물인 Zn5(OH)6(CO3)2·H2O는 400℃ 이상에서 완전한 ZnO의 결정상을 확인하였다. Na2CO3와 반응을 통해 상대적으로 낮은 하소 온도에서 나노 입자의 ZnO를 합성할 수 있었다.

Pseudomonas sp. P2에 의한 Polychlorinated Biphenyls(PCBs) 분해에 대한 영양원의 영향 (Effects of Nutritional Sources on Degradation of Polychlorinated Biphenyls (PCBs) by Pseudomonas sp. P2)

  • 유일한 탄소원으로서 Biphenyl을 포함하고 있는 액체배지에서 Pseudomonas sp. P2의 생육에 미치는 영양원의 효과에 관해 연구하였다. Pseudomonas sp. P2의 특성을 조사하기 위한, 배양시간은 균의 생육 유선에서 대수성장기 말인 100시간으로 하였다. PCBs를 분해하는 Pseudomonas sp. P2의 생육을 위한 최적 조성은 1000 mg/L $NH_4NO_3$, 1000 mg/L KH_2PO_4$, 100 mg/L MgSO_4$.$7H_2O$, 30 mg/L $CaCl_2$.$2H_2O$, 200 mg/L NaCl, 10mg/L $FeSO_4$.$7H_2O$였다. Pseudomonas sp. P2에 의한 100시간 배양후 절연유 내에 있는 PCBs의 분해률은 500mg/L에서 59.3%, 1000mg/L에서 57.6%, 1500mg/L에서 51.4%, 2000mg/L에서는 48.775 였다.

  • The objective of this work was to study the properties of purification of two liquid phase for exclusion of impurities in each phase. The experiments for process variables were carried out in the temperature range($H_{2}SO_{4}$ phase: $413{\sim}513$ K, $HI_{x}$ phase: $353{\sim}453$ K) and in the $N_{2}$ flow rate range($H_{2}SO_{4}$, $HI_{x}$ phase: $50{\sim}200$ mL/min). As the results, it is appeared that the principles of $H_{2}SO_{4}$ phase purification was due to stripping, evaporation and reverse bunsen reaction and $HI_{x}$ phase purification was due to stripping and reverse bunsen reaction. In purification of $H_{2}SO_{4}$ phase, the concentration rate of $H_{2}SO_{4}$ phase was controled by temperature but the temperature had few effects on yield of $H_{2}SO_{4}$. In purification of $HI_{x}$ phase, we observed products of side reactions($H_{2}S$, S) over 433 K. The purity of $HI_{x}$ phase was increased with increasing $N_{2}$ flow rate because impurites were decreased with increasing conversion of reverse reaction.

The Viability Change of Yeast Cell Responding to Trehalose Accumulation and Maintaining Neutral Trehalase Activity under Extracellular pH Acidified by $H_2SO_4$

  • Saccharomyces cerevisiae KNU5377 (KNU5377) and S. cerevisiae ATCC24858 (ATCC24858) were exposed to $H_2SO_4$ as a stress, which was added at various concentrations to a YPD media. The growth of KNU5377 was reduced to approximately 60% in the YPD media containing 40 nm sulfuric acid when compared to the non-stressed condition. When their growth was monitored during an overnight culture, two strains, KNU5377 and ATCC24858, could not grow when exposed to over 50 mM of sulfuric acid. After a short exposure to this acid for 1 h, KNU5377 exhibited stronger resistance against $H_2SO_4$ than ATCC24858. The neutral trehalase activity of KNU5377 unchanged despite under various concentrations of $H_2SO_4$. In contrast, It at of ATCC24858 was much low at higher $H_2SO_4$concentrations. Trehalose, a non-reducing disaccharide, was maximally accumulated after a short exposure to 60 nm $H_2SO_4$ for KNU5377, but it was reduced under more severe stressful conditions. These results suggest that KNU5377 should modulate the trehalose concentrations under the severe stress condition of high sulfuric acid concentrations. The most highly induced protein in the KNU5377 exposed to sulfuric acid was found to be an approximately 23 kDa protein, which was revealed to be the 605 large subunit ribosomal protein, Ll3 by FASTA search results.

  • To precipitate the complex gels of the pH 6, 7, 8, 9 included in a flux and an aluminum hydroxides gel, an aqueous solution of a mixture of $Na_2CO_3\;and\;Na_2PO_4{\cdot}12H_2O$ was added with stirring in an aqueous solution of a mixture of $Al_2(SO_4){_3}{\cdot}18H_2O,\;Na_2SO_4\;and\;K_2SO_4$, and then the complex gels were aged in 20 h at $90^{\circ}C$. As the hydrolysis pH changed, it had an effect on the physical properties such as the crystal structure, crystal morphology and a phase transition temperature of the AlO(OH) gel, and also on the crystal structure, crystal morphology, particle size and particle size distribution of the ${\alpha}-Al_2O_3$ platelets prepared by molten-salt precipitation. Also, in this study, the complex gels were crystallized at $1,200^{\circ}C$ and thereafter dried at $110^{\circ}C$, and then it was investigated to effect of the hydrolysis pH on the crystal structure, morphology and particle size distribution of the ${\alpha}-Al_2O_3$ platelets crystals using XRD, DTA, SEM and particle size analyzer.

SI 열화학 수소 제조 공정에서 분젠 반응을 통한 상 분리 특성 (Phase Separation Characteristics via Bunsen Reaction in Sulfur-Iodine Thermochemical Hydrogen Production Process)

  • The Sulfur-iodine(SI) thermochemical cycle is one of the most promising methods for massive hydrogen production. For the purpose of continuous operation of SI cycle, phase separation characteristics into two liquid phases ($H_2SO_4$-rich phase and $HI_x$-rich phase) were directly investigated via Bunsen reaction. The experiments for Bunsen reaction were carried out in the temperature range, from 298 to 333 K, and in the $I_2/H_2O$ molar ratio of $0.109{\sim}0.297$ under a continuous flow of $SO_2$ gas. As the results, solubility of $SO_2$, decreased with increasing the temperature, had considerable influence on the global composition in the Bunsen reaction system. The amounts of impurity in each phase(HI and $I_2$ in $H_2SO_4$-rich phase and $H_2SO_4$ in $HI_x$-rich phase) were decreased with increasing $H_2SO_4$ molar ratio and temperature. To control the amounts of impurity in $HI_x$-rich phase, temperature is a factor more important than $I_2/H2_O$ molar ratio. On the other hand, the affinity between $HI_x$ and $H_2O$ was increased with increasing $I_2/H2_O$molar ratio.

황-요오드 수소 제조 공정의 분젠 반응 부분에서 $O_2$의 역할 (The Role of Oxygen in Bunsen Reaction Section of Sulfur-Iodine Hydrogen Production Process)

  • The Sulfur-Iodine (SI) thermochemical hydrogen production process of a closed cycle consists of three sections, which are so called the Bunsen reaction section, the $H_2SO_4$ decomposition section and the HI decomposition section. To identify the role of oxygen that can be supplied to the Bunsen reaction section via the $H_2SO_4$ decomposition section, Bunsen reactions with a $SO_2,\;SO_2-O_2$ mixture and $SO_2-N_2$ mixture as feed gases were carried out using a stirred reactor in the presence of $I_2/H_2O$ mixture. As the results, the amounts of $I_2$ unreacted under the feed of mixture gases were higher than those under the feed of $SO_2$ gas only, and the amount of HI produced was relatively decreased. The results of Bunsen reaction using $SO_2-O_2$ mixture were similar to those using $SO_2-N_2$ mixture. It may be concluded that an oxygen in $SO_2-O_2$ mixture has a role as a carrier gas like a nitrogen in $SO_2-N_2$ mixture. The effects of oxygen were decreased with increasing temperature and decreasing oxygen content in $SO_2-O_2$ mixture.

태안지역 강우의 화학성분 특성 (Volume-Weighted Ion Cocentration of Rainwater in Taean Area)

  • 태안지역 강우의 화학적 특성을 알아보기 위하여 2000년 영농기 (4월${\sim}$10월) 동안 태안지역에 내린 강우의 화학적 성분조성과 강우량 가중평균 (volume-weighted mean) 이온농도 변화를 조사한 결과는 다음과 같다. 조사기간중 강우 pH는 편서풍의 영향을 받는 기간인 4월과 5월이 6월${\sim}$9월에 비해 높게 나타나 계절적 특성을 보였다. 강우시료에 대한 pH별 강우분포는 pH 5.6 이상이 42.1%로 가장 많았으며, pH $5.6{\sim}5.0$$4.5{\sim}5.0$ 범위의 강우가 각각 21.1%와 31.6%를 차지하였다. 빗물의 조성은 $SO_4$가 음이온의 50% 이상을 차지하였고 전체 양이온 함량의 약 70%가 $NH_4\;^+$$Ca_2\;^+$로 구성되어 있다. 월별 강우 pH는 측정치 $(pH_{obs})$는 6월에서 9월 사이에 낮은 반면, 이론치 $(pH_{the})$는 같은 기간에 증가하여 대기중의 알카리성 이온들에 의한 중화력이 다른 기간에 비해 낮았다. 또한 강우중 비해염 sulfate는 총sulfate 함량의 약 97%이었다. 강우의 산성도 기여율 $(nss-SO_4\;^{2-}/NO_3\;^-)$은 평균 2.0으로 비해염 $SO_4\;^{2-}$에 의한 기여율이 $NO_3\;^-$ 보다 2배 높았다.
