The Changes of Growth Patterns and the Production of Brain-Derived Neurotrophic Factors (BDNFs) in Perfusion Cultivation of Human Neuroblastoma Cells

  • It was shown that brain-derived neurotrophic factors (BDNFs) secreted from human neuroblastoma cells can significantly improve the growth of the neurites of PC12 nerve cells. The addition of purified BDNFs elongated the neurites of PC 12 nerve cells two to three times more than the case where the addition was not made. The perfusion rate strongly affected the change of the size of human neuroblastoma cells because the cell size decreased as the perfusion rate increased. This could also influence the productivity of BDNF from the cells. It is also important to note that the BDNF production was decreased when the cell size was reduced. BDNF production rate also decreased at a fast perfusion rate in a smaller cell size. At the relatively fast perfusion rate of 18 ml/h, the ratio of apoptotic to necrotic cells dramatically decreased, which possibly caused the decrease of BDNF production. It has been proven that the secretion of BDNF from human neuroblastoma cells was a partially growth-related process by yielding 6.2$\times l0^{-8}/g$ of BDNF/cell/h of growth related parameter and $0.48{\times}l0^{-9}/g$ of BDNF/cell/h of nongrowth-related parameter in a growth kinetic model. In addition, it was also found that the perfusion rate played a very important role in controlling the cell death mechanism.

행성상 성운 NGC 7009, NGC 7027의 모델 연구

  • 행성상 성운 NGC 7009와 NGC 7027의 모델을 연구하기 위해 1998년 8월 14일부터 16일 까지 3일간의 Keck I HIRES의 분광 관측 자료를 이용하였다. 사용한 파장 영역은 $3250{\AA}{\sim}8725{\AA}$이고, H I, He I, He II, C II, C III, N II, N III, [N II], [O I], [O II], [O III], [S II], [S III], [Cl III], [Cl IV], [Ar III], [Ne III] 등의 방출선을 얻었다. 본 연구에서는 이러한 방출선의 선윤곽(line profile)을 IRAF와 StarLink/Dipso를 이용해 분석하여 성분을 구하였는데 H I, He I, He II, [O III], [C III], [N II] 등 대부분은 2개로 분리되었고, [S II], [Ar III]의 경우에는 3개로 분리되었다. 후자의 경우 구각(shell)의 단순한 팽창뿐만 아니라 다른 제3의 방향으로 팽창하고 있는 성분이 존재함을 의미한다. 비교적 S/N가 양호한 선들의 FWHM(Full Width at Half Maximum)을 측정하여 팽창속도를 구한 후 이 선들이 생성에 기여한 지역의 온도 또는 이온화 에너지 (IP)에 대한 팽창속도의 변화를 조사하였다. 또한 두 행성상 성운의 물리적 특성과 화학 원소 함량을 결정하기위해 광이온 모델 Tlusty Code와 CLOUDY를 사용하여 NGC 7009의 선행연구 Hyung & Aller(1995), Kwitter & Henry(1998)와 비교하였고, NGC 7027은 Zhang & Liu(2005), Middlemass(1990)와 비교하였다. NGC 7009과 NGC 7027의 수소 밀도($N_H$)를 $8,000cm^{-3}$으로 가정했을 때, NGC 7009의 중심별의 온도는 77,000K이고, 구각(shell)의 크기는 0.031~0.044pc이며 장축에 대해서만 결과를 나타내었다. NGC 7027의 경우, 중심별의 온도는 180,000K이고, 구각(shell)의 크기는 0.015~0.02pc으로 판단된다.

Methanolic Extract from Sea Cucumber, Holothuria scabra, Induces Apoptosis and Suppresses Metastasis of PC3 Prostate Cancer Cells Modulated by MAPK Signaling Pathway

  • Sea cucumber, Holothuria scabra, is a well-known traditional Asian medicine that has been used for suppressing inflammation, promoting wound healing, and improving immunity. Moreover, previous studies demonstrated that the extract from H. scabra contains many bioactive compounds with potent inhibitory effect on tumor cell survival and progression. However, the effect of the methanolic extract from the body wall of H. scabra (BWMT) on human prostate cancer cells has not yet been investigated. In this study, we aimed to investigate the effects and underlying mechanism of BWMT on prostate cancer cell viability and metastasis. BWMT was obtained by maceration with methanol. The effect of BWMT on cell viability was assessed by MTT and colony formation assays. The intracellular ROS accumulation was evaluated using a DCFH-DA fluorescence probe. Hoechst 33342 staining and Annexin V-FITC/PI staining were used to examine the apoptotic-inducing effect of the extract. A transwell migration assay was performed to determine the anti-metastasis effect. BWMT significantly reduced cell viability and triggered cellular apoptosis by accumulating intracellular ROS resulting in the upregulation of JNK and p38 signaling pathways. In addition, BWMT also inhibited the invasion of PC3 cells by downregulating MMP-2/-9 expression via the ERK pathway. Consequently, our study provides BWMT from H. scabra as a putative therapeutic agent that could be applicable against prostate cancer progression.

Protective effects of red ginseng treated with gold nanoparticles against H2O2-induced oxidative stress in neuronal PC-12 cells (금 나노입자를 처리한 홍삼의 산화스트레스 완화 및 PC-12 신경세포 보호)

  • Red ginseng prepared from fresh 6-year-old ginseng treated with colloidal gold nanoparticles was extracted using hot water to evaluate its total phenolic and flavonoid contents, antioxidant capacity, and neuroprotective effects. Water extract of red ginseng treated with gold nanoparticles (WERGGN) had total phenolic and total flavonoid contents of 212.2 mg gallic acid equivalents/$^{\circ}Bx$ and 3.5 mg catechin equivalents/$^{\circ}Bx$, respectively. The antioxidant capacities of WERGGN measured using ABTS, DPPH, and ORAC assays were 272.3, 141.2, and 868.4 mg vitamin C equivalents/$^{\circ}Bx$, respectively. The WERGGN showed protective effects on the viability of neuron-like PC-12 cells against oxidative stress induced by hydrogen peroxide in a dose-dependent manner, partly because of a reduction in intracellular oxidative stress. Acetylcholinesterase and butyrylcholinesterase, which degrade the neurotransmitter acetylcholine to terminate neurotransmission, were inhibited by treatment with WERGGN. These results suggest that WERGGN is useful as a functional material to decrease oxidative stress and neuronal damage.

Effects of Opuntia ficus-indica var. saboten Ripe Fruits on Protection of Neuronal PC-12 Cells and Cholinesterase Inhibition (백년초의 PC-12 신경세포 보호 및 콜린가수분해효소(cholinesterase) 저해 효과)

  • Oxidative stress caused by reactive oxygen species is ascribed to many neurodegenerative diseases like Alzheimer's disease. Phenolic antioxidants can reduce the oxidative stress. In this study, ripe fruits of Opuntia ficus-indica var. saboten (OFS) were extracted using 80% (v/v) aqueous ethanol. Total phenolic and flavonoid contents of the OFS fruits (100 g) were 409.9 mg gallic acid equivalents and 72.2 mg catechin equivalents, respectively. The OFS fruits had antioxidant capacity at 381.2, 298.2, and 3,219.9 mg vitamin C equivalents/100 g in ABTS, DPPH, and ORAC assays, respectively. The OFS fruits showed protective effects on PC-12 cells against oxidative stress in a dose-dependent manner, partly due to decrease of intracellular oxidative stress. Furthermore, the OFS fruits inhibited both acetylcholinesterase and butyrylcholinesterase. Consequently, these results suggest that the OFS fruits might be served as a source of functional materials to reduce oxidative stress in neuronal cells and to inhibit cholinesterases.

ALMA Observations of a Massive-star-forming Infrared Dark Cloud Core MSXDC G053.11+00.05 MM1

  • We present the ALMA observations of the infrared dark cloud (IRDC) core MSXDC G053.11+00.05 MM1 at the distance of 1.7 kpc. While the core was first identified at 1.2 mm with a mass of 124 Msun, recent near- and mid-infrared observations have revealed a parsec-scale molecular hydrogen (H2 1-0 S(1) at 2.12 micron) outflow and two early class young stellar objects (YSOs) at the center of the core, one of which is likely massive (M > 8 Msun). From the ALMA Band 7 observations with a resolution of 0.5", we have found a dust filament of < 0.1 pc in which five dense cores are embedded in the 870 micron continuum. The brightest core is consistent with one of the two previously-detected YSOs, but the other four are newly discovered implying their very deeply embedded status. We have also detected several molecular line emission including H13CO+ and C17O as well as 13CO outflow with complicated morphology. At the brightest core, the methanol line (CH3OH) shows velocity gradients, which may support the existence of a circumstellar disk around a high-mass protostar. Based on the derived properties of the dense cores, we discuss their association with the two YSOs and H2 outflow detected in infrared and high-mass star-formation process occurring in IRDC cores.

  • Recently, KARI developed in-house meteorological sensor processing system named MESIS for GOES GVAR 5-CH Imager for better KOMPSAT EOC mission operation. MESIS consists of antenna system, receiver, serial telemetry card, processing and mapping software, and 2 NT PC systems. This paper shows system requirement, system design, characteristic and test results of processing system. System operation concept and sample image are also provided. Implemented system was proven to be fully operational through lots of operations covering from RF signal reception to web publishing.

NEW DIGITAL H$\alpha$ OBSERVATION BY SOLAR FLARE TELESCOPE AT BOAO

  • Recently, we have set up a new digital CCD camera system, MicroMax YHS-1300 manufactured by Roper Scientific for Ha observation by Solar Flare Telescope at Bohyunsan Optical Astronomy Observatory. It has a 12 bit dynamic range, a pixel number of 1300$\times$1030, a thermoelectric cooler, and an electric shutter. Its readout speed is about 3 frames per second and the dark current is about 0.05 e-/p/s at $-10^{\circ}C$. We have made a system performance test by confirming the system linearity, system gain, and system noise that its specification requires. We have also developed a data acquisition software which connects a digital camera con-troller to a PC and acquires H$\alpha$ images via Microsoft Visual C++ 6.0 under Windows 98. Comparisons of high quality H$\alpha$ images of AR 9169 and AR 9283 obtained from SOFT with the corresponding images from Learmonth Solar Observatory in Australia confirm that our H$\alpha$ digital observational system is performed properly. Finally, we present a set of H$\alpha$ images taken from a two ribbon flare occurred in AR 9283.

  • This paper is to investigate a defect for thermal barrier coating layers by acoustic emission method in 4-point bending test. The two-layer thermal barrier coating is composed of $150\mu{m}\;CoNiCrAlY\;bond\;coating\;by\;vacuum\;plasma\;spray(VPS)\;process\;and\;250\mu{m}\;ZrO_2-8wt%Y_2O_3$ ceramic coating layer by air plasma spray(APS) process on Inconel-718. The specimen prepared by cyclic thermal test(500, 1000, 2000cycle) at $1050^{\circ}C$ The AE monitoring system is composed of PICO type sensor, a wide band pre-amplifier(40dB), PC and AE DSP(16/32 PAC) board. The AE event, amplitude, Cumulative energy and count of coating specimens is evaluated according to cyclic thermal test.

Evaluation of base shield plates effectiveness in reducing the drag of a rough circular cylinder in a cross flow

  • An experimental investigation has been conducted to determine the effectiveness of base shield plates in reducing the drag of a rough circular cylinder in a cross flow at Reynolds numbers in the range $3{\times}10^4{\leq}Re{\leq}10.5{\times}10^4$. Three model configurations were investigated and compared: a plane cylinder (PC), a cylinder with a splitter plate (MC1) and a cylinder fitted with base shield plates (MC2). Each configuration was studied in the sub and supercritical flow regimes. The chord of the plates, L, ranged from 0.22 to 1.50D and the cavity width, G, between the plates was in the range from 0 to 0.93D. It is recognized that base shield plates can be employed more effectively than splitter plates to reduce the aerodynamic drag of circular cylinders in both the sub- and supercritical flow regimes. For subcritical flow regime, one can get 53% and 24% drag reductions for the MC2 and MC1 models with L/D=1.0, respectively, compared with the PC model. For supercritical flow regime however, the corresponding drag reductions are 38% and 7%.
