Effect of Chungganhaeju-hwan in Ethanol-induced Neuronal Cell Damage (청간해주환(淸肝解酒丸)의 알코올 유도 뇌신경세포 손상에 대한 보호 효과)

  • Objectives : In this study, we evaluated the effect of Chungganhaeju-hwan(CGHJH) on hydrogen peroxide($H_2O_2$)-induced and ethanol(EtOH)-induced neuronal damage in vitro and in vivo, respectively. Methods:We carried out the anti-oxidant effects of CGHJH against hydrogen peroxide($H_2O_2$)-induced toxicity in HT22 and PC12 cells using thiazolyl blue tetrazolium bromide. Then, to investigate the protective effect on CGHJH against EtOH-induced memory impairment and hippocampal cell damage in male ICR mice, we performed novel object recognition test(NORT), and analysed the brain tissues after immunohistochemistry and western blotting. Results:CGHJH showed protective effect from $H_2O_2$-induced cell toxicity at doses of $1\sim100{\mu}g$/mL in both HT22 and PC12 cells. CGHJH had also recovery effect from EtOH-induced memory impairment in ICR mice from NORT and it protected hippocampal cells against EtOH toxicity in the result of cresyl violet and NeuN immunoreactivity. Conclusion : These results demonstrate that CGHJH has protective effect in neuronal cells against $H_2O_2$ and EtOH toxicities and this effect could be a main role of recovery effect on EtOH-induced memory loss.

Improving the Color Gamut of a Liquid-crystal Display by Using a Bandpass Filter

  • To improve the color gamut of a liquid-crystal display (LCD), we propose a bandpass filter that is added to the backlight unit to optimize the backlight spectrum. The bandpass filter can only transmit red, green and blue light in the visible range, while reflecting the unwanted light. We study the optical properties of the bandpass filter using the transfer-matrix method, and the effect of the bandpass filter on the color gamuts of LCDs is also investigated. When a bandpass filter based on a 5-layer configuration comprising low and high refractive indices ((HL)2H) is used in phosphor-converted white-light-emitting diode (pc-WLED), K2SiF6:Mn4+ (KSF-LED), and quantum-dot (QD) backlights, the color gamuts of the LCDs improve from 72% to 95.3% of NTSC, from 92% to 106.7% of NTSC, and from 104.3% to 112.2% of NTSC respectively. When the incident angle of light increases to 30°, the color gamuts of LCDs with pc-WLED and KSF-LED backlights decrease by 2.9% and 1% respectively. For the QD backlight, the color gamut almost does not change. When the (HL)2H structure is coated on the diffusion film, the color gamut can be improved to 92.6% of NTSC (pc-WLED), 105.6% of NTSC (KSF-LED), and 111.9% of NTSC (QD). The diffusion film has no obvious effect on the color gamut. The results have an important potential application in wide-color-gamut LCDs.

Fast temporal detection of intracellular hydrogen peroxide by HyPer

  • HyPer is the genetically encoded biosensor of intracellular hydrogen peroxide ($H_2O_2$), the most stable of the reactive oxygen species (ROS) generated by living cells. HyPer has a high sensitivity and specificity for detecting intracellular $H_2O_2$ by confocal laser microscopy. However, it was not known whether high speed ratiometric imaging of $H_2O_2$ by HyPer is possible. We thus investigated the sensitivity of HyPer in detecting changes to the intracellular $H_2O_2$ levels in HEK293 and PC12 cells using a microfluorometer imaging system. Increase in the HyPer ratio were clearly evident on stimulations of more than $100{\mu}M$ $H_2O_2$ and fast changes in the HyPer ratio were observed on ratiometric fluorescent images after $H_2O_2$ treatment. These results suggest that HyPer is a potent biosensor of the fast temporal production of intracellular $H_2O_2$.

  • The recent surge in consumption of traditional liquor related to health, the results of analyzing the chemical characteristics and antioxidant activity by creating Kwaha-ju and Baekhwha-chun, which use Godubab as a method of treating glutinous and normal grains, are as follows. Kwaha-ju is mainly made of glutinous rice, 5.21~12.52% alcohol content, 1.07~2.47g/L in reducing sugar content, 3.22~3.77% pH, Baikhwha-chun alcohol content was 3~5%, except for some experimental groups, and pH was low. As a result of the spectrum analysis of traditional liquor, 50% of the experimental group in Baikhwa-chun is located in the negative area of PC1 such as overload, and some amino acids such as lactic acid, isoleucine, and choline are the main ingredients of glucose, xylose, and maltose. Therefore, the chemical properties and antioxidant activity of Kwaha-ju and Baikhwa-chun, which show similar composition ratio of traditional liquor materials, and use them as basic data to identify and manufacture traditional liquor fermentation through mutual analysis.

Effects of Ginsenosides $Rg_3$ and $Rh_2$ OH the Proliferation of Prostate Cancer Cells

  • Ginseng has an anti-cancer effect in several cancer models. This study was to characterize active constituents of ginseng and their effects on proliferation of prostate cancer cell lines, LNCaP and PC3. Cell proliferation was measured by $[^3H]$thymidine incorporation, the intracellular calcium concentration by a dual-wavelength spectrophotometer system, effects on mite-gen-activated protein (MAP) kinases by Western blotting, and cell attachment and morphologic changes were observed under a microscope. Among 11 ginsenosides tested, ginsenosides $Rg_3\;and\;Rh_2$ inhibited the proliferation of prostate cancer cells. $EC_{50}s\;of\;Rg_3\;and\;Rh_2$ on PC3 cells were $8.4{\mu}M\;and\;5.5{\mu}M$, respectively, and $14.1{\mu}M\;and\;4.4{\mu}M$ on LNCaP cells, respectively. Both ginsenosides induced cell detachment and modulated three modules of MAP kinases activities differently in LNCaP and PC3 cells. These results suggest that ginsenosides $Rg_3\;and\;Rh_2$-induced cell detachment and inhibition of the proliferation of prostate cancer cells may be associated with modulation of three modules of MAP kinases.

Selective Cytotoxicity of a Novel Platinum(II) Coordination Complex on Human Bladder Cancer Cell Lines and Normal Kidney Cells

  • We have synthesized a novel platinum(II) coordination complex containing cis-1,2-diaminocyclohexane (DACH) as a carrier ligand and 1,2-dichloroethane (DCE) as a leaving group. In addition, nitrate was added to improve the water-solubility. A new series of [Pt(cis-DACH)(DCE)] $2NO_3(PC)$ was evaluated for its cytotoxic activity on T-24 and J-82 human bladder carcinoma cells and normal primary cultured kidney cells. PC has demonstrated high levels of cytotoxicity against T-24 and J-82 cells. The cytotoxicity of PC against rabbit proximal renal tubular cells, human renal cortical cells and human renal cortical tissues, determined using the MTT assaying technique, the $[^3H]-thymidine$ uptake and glucose consumption tests, was found to be quite less than those of cisplatin. Based on these results, this novel platinum(II) coordination complex appears to be better for improving antitumor activities with low nephrotoxicity and is a valuable lead in the development of new clinically available anticancer chemotherapeutic agents.

Synthesis of Nanocrystalline ZnFe2O4 by Polymerized Complex Method for its Visible Light Photocatalytic Application: An Efficient Photo-oxidant

  • Nanocrystalline Zn$Fe_2O_4$ oxide-semiconductor with spinel structure was synthesized by the polymerized complex (PC) method and investigated for its photocatalytic and photoelectric properties. The observation of a highly pure phase and a lower crystallization temperature in Zn$Fe_2O_4$ made by PC method is in total contrast to that was observed in Zn$Fe_2O_4$ prepared by the conventional solid-state reaction (SSR) method. The band gap of the nanocrystalline Zn$Fe_2O_4$ determined by UV-DRS was 1.90 eV (653 nm). The photocatalytic activity of Zn$Fe_2O_4$ prepared by PC method as investigated by the photo-decomposition of isopropyl alcohol (IPA) under visible light (${\geq}$ 420 nm) was much higher than that of the Zn$Fe_2O_4$ prepared by SSR as well as Ti$O_{2-x}N_x$. High photocatalytic activity of Zn$Fe_2O_4$ prepared by PC method was mainly due to its surface area, crystallinity and the dispersity of platinum metal over Zn$Fe_2O_4$.

Comparative Effects of Phytase Derived from Escherichia coli and Aspergillus niger in Sixty Eight-week-old Laying Hens Fed Corn-soy Diet

  • Two hundred and sixteen 68-week-old Hy-Line brown laying hens were used in a 6-week feeding trial to compare the efficacy of phytases Optiphos (OPT) and Natuphos (NAT), which were isolated from Escherichia coli and Aspergillus niger, respectively. Hens were randomly allotted into six treatments with six replications (six layers in three adjacent cages) per treatment according to their initial BW. The hens were then subjected to one of the following dietary treatments: i) Positive Control (PC; available phosphorus (AP) 0.4%); ii) Negative control (NC; AP 0.2%); iii) NAT1 (NC+250 FTU/kg NAT); iv) NAT2 (NC+500 FTU/kg NAT); v) OPT1 (NC+250 FTU/kg OPT); vi) OPT2 (NC+500 FTU/kg OPT). Feed intake, egg production, egg quality, apparent nutrient digestibility and serum P and Ca concentration were evaluated to compare the effect of the two phytases. Feed intake and eggshell thickness were not affected by the treatments. Superior effects (p<0.05) of OPT were only observed in egg production and egg weight compared with NAT. Characteristics such as eggshell breaking strength, apparent digestibility of N, Ca and P and serum P concentration were equally increased with the supplementation of both phytases (p<0.05), where no significant difference was observed in those characteristics between PC and phytase supplementation at 500 FTU/kg. Equally effective improvements (p<0.05) were also observed in egg production and DM digestibility, where no improvements were observed (p<0.05) between the PC group and the groups with phytase supplementation at 500 FTU/kg. Equal increases in the serum Ca level were observed when the groups with phytase supplementation were compared to the PC group. Overall, the results of this study suggest that NAT and OPT are equally effective at liberating phytate-bound complexes when included in 0.2% available phosphorus diets for 68-week laying hens; either source of phytase can be fed to commercial 68-week laying hens at 500 FTU/kg to correct the negative effects associated with a 0.2% available phosphorus diet. In conclusion, either source of phytase can be fed to commercial first cycle laying hens at 500 FTU/kg to effectively replace inorganic phosphorus when economically justified.

Molecular Characterization of Ischemia-Responsive Protein 94 (irp94) Response to Unfolded Protein Responses in the Neuron

  • The ischemia-responsive 94 gene (irp94) encoding a 94 kDa endoplasmic reticulum resident protein was investigated its molecular properties associated with unfoled protein responses. First, the expression of irp94 mRNA was tested after the reperfusion of the transient forebrain ischemia induction at the central nervous system in three Mongolian gerbils. Second, irp94 expression in PC12 cells, which are derived from transplantable rat pheochromocytoma cultured in the DMEM media, was tested at transcriptional and translational levels. The half life of irp94 mRNA was also determined In PC12 cells. Last, the changes of irp94 mRNA expression were investigated by the addition of various ER stress inducible chemicals (A23187, BFA, tunicamycin, DTT and $H_2O_2$) and proteasome inhibitors, and heat shock. High level expression of irp94 mRNA was detected after 3 hours reperfusion in the both sites of the cerebral cortex and hippocampus of the gerbil brain. The main regulation of irp94 mRNA expression in PC 12 cells was determined at the transcriptional level. The half life of irp94 mRNA in PC12 cells was approximately 5 hours after the initial translation. The remarkable expression of irp94 mRNA was detected by the treatment of tunicamycin, which blocks glycosylation of newly synthesized polypeptides, and $H_2O_2$, which induces apoptosis. When PC12 cells were treated with the cytosol proteasome inhibitors such as ALLN (N-acetyl-leucyl-norleucinal) and MG 132 (methylguanidine), irp94 mRNA expression was increased. These results indicate that expression of irp94 was induced by ER stress including oxidation condition and glycosylation blocking in proteins. Expression of irp94 was increased when the cells were chased after heat shock, suggesting that irp94 may be involved in recovery rather than protection against ER stresses. In addition, irp94 expression was remarkably increased when cytosol proteasomes were inhibited by ALLN and MG 132, suggesting that irp94 plays an important role for maintaining the ERAD (endoplasmic reticulum associated degradation) function.
