Sphingoshine-1-Phosphate Enhances Meiotic Maturation and Further Embryonic Development in Pigs

  • Sphingosine-1-phosphate (S1P) has a many function involved proliferation, differentiation and survival of many cells. In this study, to investigate whether S1P improve the developmental competence of porcine embryos, 50 nM of S1P were supplemented during in vitro maturation (with EGF or without EGF) medium and/or in vitro culture (IVC) medium. Addition of S1P was significantly increased the rate of oocytes reaching metaphase II (MII) compared to the control (83.5 vs. 64.1%) in without EGF medium, but not with EGF medium (89.5 vs. 84.6%). When treated with $1{\mu}M$ of N1N-dimethylsphingosine (DMS), a sphingosine kinase inhibitor which is blocked endogenous generation of S1P, the meiotic progression rates to MII stage (without EGF: 45.2 and with EGF: 66.7%) were significantly decreased and degeneration rates (without EGF: 51.2 and with EGF: 30.1%) were increased in both medium compared to control group during IVM periods. Also, the rates of blastocyst formation was significantly increased in the S1P treated group compared to control group (29.0 vs. 19.2%) of EGF supplemented medium, whereas there were no effect in the EGF free medium (9.0 vs. 10.5%). After 12 h IVM, the phosphorylation of ERK1 and ERK2, which is major signaling pathway of MAP kinase, were increased in the S1P group than that of control or DMS group. When supplemented of S1P during IVC, the rates of blastocyst formation and total cell number (30.2% and 40.6) were significantly increased in S1P-treated group compared with control (20.1% and 32.5), DMS (12.3% and 25.1), and S1P plus DMS group (24.7% and 33.6). The percentage of apoptosis nuclei in the S1P group was significantly decreased than other groups. Also, the rates of blastocyst formation (26.7 vs. 14%) and total cell number (42.8 vs. 32.5) were significantly increased in the S1P group than those of control group when S1P added during the entire IVM and IVC periods. Taken together, our results indicate that S1P supplementation in IVM and/or IVC medium affects beneficial effect of meiotic maturation and subsequent developmental competence of porcine embryos.

청소년의 성폭력 태도유형 관련요인 (A Study of Factors Influencing Adolescent Attitudes toward Sexual Violence)

  • The purpose of this study is to afford basic data for planning a program to strategically prevent adolescent sexual violence by clarifying the concept of sexual violence in adolescence and investigating factors influencing the adolescent attitudes towards sexual violence. Subjects of the study were 288 adolescents between the ages of 14 to 21, living in K city in Chungnam Province. The data was collected for this study between May 30 and June 13, 2001. For this study, the following materials were used as instruments: the types of attitudes toward sexual violence developed by Nam, S. Y.(2000), the antisocial personality by Min, B. K., Choi, S. J., & Lee, G. H.(1978), the self-esteem developed by Rosenberg(1965) and interpreted by Jeon, B. J.(1974), the family support developed by Cobb(1976), the stereotyped sex role and the attitude on sexuality by Kim, E. J.(1997). The data was analyzed, using SPSS 7.5 for Windows Program, descriptive statistics, t-test, ANOVA, and discriminant analysis. The results of this study were as follows: 1. Adolescents of the first type, ones blaming social morals, amounted to 34.7%(100 subjects), the second type, ones blaming the victims, amounted to 22.3%(64 subjects), the third type, ones claiming reform of consciousness, amounted to 25.3%(73 subjects), and the fourth type, ones coping actively, amounted to 17.7%(51 subjects). 2. Attitudes toward sexual violence were influenced a great deal by the sex of the subject. 3. The study revealed that various types of the attitudes toward sexual violence was significantly different from one another, related to antisocial personality(F=2.80, P=.04). Especially the first type and the third type were most different from each other. The latter showed the antisocial personality strongly, compared with the former. 4. Discriminant analysis revealed that the most powerful predictors of attitudes toward sexual violence were the sex of the subject and whether or not they had an antisocial personality. 5. The degree of possibility of identifying a risk group of adolescents was 45.9% and the possibility of identifying a non-risk group was 73.3%. The hit ratio was 67.8%. In conclusion, the results of the study lead us to understand the type of adolescent attitudes toward sexual violence. The results will be useful to furnish a guideline for nursing intervention and education programs to prevent sexual violence in adolescence.

Percoll에 의한 미니돼지 정액내 세균 제거가 정자 성상과 수정란 분할에 미치는 영향 (Effect of Bacteria Eliminated Sperm by Percoll Method on Sperm Quality and Embryo Cleavage in Miniature Pig)

  • The objectives of this study was to evaluate the efficiency of the bacteria eliminated sperm by percoll gradient method on sperm quality and embryo cleavage in vitro in pig. The semen of miniature pig collected by gloved-hand method pre-warmed ($37^{\circ}C$) in thermos bottle, and separated by 65% percoll. Analysis of sperm ability was estimated by examining viability, capacitation and acrosome reaction using chlortetracycline (CTC) and the abnormality. Also, fertility of sperm was monitored with cleavage rate of embryo after IVF using separated and un-separated sperm by percoll. The result, viability of separated sperm was significantly(p<0.05) higher($83.6{\pm}$2.0 vs $59.0{\pm}4.4%$) than un-separated sperm. The results of CTC analysis showed the percentage of F- and B-patterned separated sperm was higher in separated that than un-separated sperm. On the contrary, the percentage of AR-patterned form unseparaed sperm was significantly(p<0.05) higher($13.6{\pm}0.8$ vs $8.1{\pm}0.6%$) than separated sperm. Also, abnormality of un-separated sperm was significantly(p<0.05) higher($2.2{\pm}0.4$ vs $16.8{\pm}2.8%$) than separated sperm. However, the cleavage rates of embryo using separated sperm by percoll and un-separated sperm had not significantly difference on 2 cell stage(9.25 vs 11.88%), 4 cell stage(26.76 vs 24.51%) and >4 cell stage(63.99 vs 63.61%) at 48h of IVF. Therefore, the sperm separated by percoll method showed improvement in sperm quality than un-separated sperm in miniature pig.

Streptococcus thermophilus와 Lactobacillus casei를 이용한 미강 첨가 발효유의 품질특성에 관한 연구 (Quality Characteristics of Yogurt prepared with Rice Bran Streptococcus thermophilus and Lactobacillus casei)

  • 본 실험은 쌀겨를 발효식품에 이용하고자 상업용 균주인 Streptococcus thermophilus(ST-body1), Lactobacillus casei(LC-10)을 이용하여 발효유를 제조하고, 발효시간 경과에 따라 적정산도, 유산균수, 유기산 측정, 유리아미노산 측정, 관능검사를 측정하였다. ST-body1은 대조구와 처리구 모두 발효 3시간째에 급격하게 산도가 증가하였다. 하지만 LC-10은 산도의 증가폭이 크지 않아 24시간 발효를 하여도 산도가 0.5%를 넘지 못하였다. 균주의 생균수는 대조군과 실험군 모두 배양 24시간 동안 뚜렷한 경향 없이 $10^6{\sim}10^9CFU/mL$ 사이로 존재하였다. 유기산 및 유리아미노산 측정 결과, ST-body1 생장 및 생육에는 glutamic acid가 LC-10 생장 및 생육에는 aspartic acid가 밀접한 관계를 보였으며, LC-10은 발효 시 methionine이 20~30시간 경에 감소하다가 증가하는 것으로 보아 methionine을 발효 시 이용하고, 재생산하는 것으로 추측된다. 관능검사 결과, 선호도는 ST-body1이 미강의 존재 유무에 관계없이 우수하였다.

Nutritional Constraints and Possibilities for Pig Production on Smallholders Farms in Central Vietnam

  • This study aimed to evaluate the nutritional situation of pigs kept in three ecological zones of central Vietnam: Upland, Lowland and Coastal Area. An interview-based questionnaire was made and surveys were conducted in 27 villages and data were collected from 1,200 participating households. The current study showed that amounts of feed and crude protein content in the diets for fattening pigs and sows are deficient for all three regions. Amounts of feed as DM (kg/d) fed to growing pigs of 20-50 kg BW was deficient by 0.54 kg (29%) in Lowland, 0.53 kg (28.6%) in the Coastal area and 0.42 kg (22.4%) in Upland. The deficiency in CP in the diets of growing pigs in this period (20-50 kg) was largest at 20.7 g/d (62.1%) in Lowland, following by 22.1 g/d (66.4%) in Coastal and 23.2 g/d (69.7%) in Upland. Amount of feed as DM (kg/d) fed to growing pigs of 50-90 kg BW had a deficiency of 1.26 (48.9%), 1.25 (51.2%) and 1.14 (51.5%) kg/d in Lowland, Coastal and Upland, respectively. The deficiencies in crude protein in the growing diet during this period in Lowland, Coastal and Upland regions were 27 g/d (68.3%), 29 g/d (71.9%) and 30 g/d (74.6%), respectively. The deficiency in DM intake (kg/d) of pregnant sows in the Lowland area was 0.3 kg (15%), 0.33 kg (16%) in the Coastal area and 0.47 kg (23.5%) in the Upland area. Crude protein content in the diet of pregnant sows raised in Lowland was 8 g/d (32.0%) deficient, in the Coastal region the deficiency was 11 g/d (42.7%) and in Upland this deficiency was 15 g/d (61.2%). The deficiency in DM intake (kg/d) of lactating sows raised in Lowland was 1.47 kg (31.1%), in the Coastal area this was 1.69 kg (39.2%) and in Upland it was most deficient at 2.46 kg (57.1%). The lack of crude protein content in the diets of sows raised in Lowland was 45 g/d (63.4%), in the Coastal region it was 46 g/d (65%), and in Upland it was 55 g/d (78.9%). The low input of feed in these areas is especially due to low quality and to the insufficient intake of nutrients by the pig. As a result, production and income of farmers are low.

연안 양식장 퇴적물의 비료화를 위한 알카리 안정화 공법의 적용 (Application of Alkaline Stabilization Processes for Organic Fertilizer of Coastal Sediments)

  • 알카리 안정화방법에 의거하여 양식장 퇴적물을 농업용 유기질 비료로 사용하기 위한 기초연구로서 제조과정에 따른 비료의 유효성분 및 중금속농도를 분석하고, 비료의 유해성 여부를 평가하였다. 퇴적물과 축산분뇨 혼합율 및 첨가제의 양을 달리하여 비료를 제조한 결과 생석회 $30{\%}$ 첨가시 건중량비 1 : 4 이었을 때가 최적의 반응조건 이었다. 또한, 알카리 안정화제로서 수산화 마그네슘도 영양분의 보강제 측면에서 사용될 수 있음을 보여 주었다. 제조된 퇴적물 조비료는 질소 및 칼륨 함량을 기준으로 할 때 중급 내지 고급 비료로 평가되었으며, 칼슘과 마그네슘의 함량은 고급 비료에 속하였다. 이상의 결과들은 알카리 안정화 방법으로 양식장 퇴적물을 유기질 비료로 전환 생산할 수 있음을 제시한다. 또한 알카리 안정화에 의해 제조된 비료내의 중금속 및 병원성균에 의한 유해성은 없는 것으로 나타났다.

Characteristics of proteolytic microorganisms and their effects on proteolysis in total mixed ration silages of soybean curd residue

  • Objective: The objective of this study was to isolate proteolytic microorganisms and evaluate their effects on proteolysis in total mixed ration (TMR) silages of soybean curd residue. Methods: TMRs were formulated with soybean curd residue, alfalfa or Leymus chinensis hay, corn meal, soybean meal, a vitamin-mineral supplement, and salt in a ratio of 25.0: 40.0:30.0:4.0:0.5:0.5, respectively, on a basis of dry matter. The microbial proteinases during ensiling were characterized, the dominate strains associated with proteolysis were identified, and their enzymatic characterization were evaluated in alfalfa (A-TMR) and Leymus chinensis (L-TMR) TMR silages containing soybean curd residue. Results: Both A-TMR and L-TMR silages were well preserved, with low pH and high lactic acid concentrations. The aerobic bacteria and yeast counts in both TMR silages decreased to about 105 cfu/g fresh matter (FM) and below the detection limit, respectively. The lactic acid bacteria count increased to 109 cfu/g FM. The total microbial proteinases activities reached their maximums during the early ensiling stage and then reduced in both TMR silages with fermentation prolonged. Metalloproteinase was the main proteinase when the total proteinases activities reached their maximums, and when ensiling terminated, metallo and serine proteinases played equally important parts in proteolysis in both TMR silages. Strains in the genera Curtobacterium and Paenibacillus were identified as the most dominant proteolytic bacteria in A-TMR and L-TMR, respectively, and both their proteinases were mainly with metalloproteinase characteristics. In the latter ensiling phase, Enterococcus faecium strains became the major sources of proteolytic enzymes in both TMR silages. Their proteinases were mainly of metallo and serine proteinases classes in this experiment. Conclusion: Proteolytic aerobic bacteria were substituted by proteolytic lactic acid bacteria during ensiling, and the microbial serine and metallo proteinases in these strains played leading roles in proteolysis in TMR silages.

란탄족 원소의 여러자리 리간드 착물에 관한 연구 (제 1 보) 물아닌 용액에서 용액열량계에 의한 열역학적 함수결정 (Studies on the Complexes of Lanthanide ion with Multidentate Ligand (I). Determination of Thermodynamic Parameters with Solution Calorimetric Method in Nonaqueous Solvents)

  • 아세트니트릴 용액과 메탄올 용액에서 가벼운 란탄족원소인 $La^{3+},\; Ce^{3+},\;Eu^{3+}$ 이온과 여러자리 리간드로 크라운 에테르류, diaza 거대고리류와 podand 형 리간드인 diamine ether의 착물형성에 대한 Log K, ΔH, ΔS값을 용액열량계로 결정하였다. 메탄올 용액에서는 금속이온반경과 거대고리 리간드 동공의 비에 따라서 선택적으로 안정한 착물이 형성되며 리간드 주게원자의 영향은 없었다. 아세트니트릴 용액에서는 발열반응으로 엔탈피 주도반응이었으며 거대고리 리간드의 주게원자가 산소인 경우는 최적크기에 따르는 선택적인 착물의 안정도를 나타냈으나 질소원자로 치환되면 공유결합성의 증가로 더욱 안정한 착물이 형성되었다. Diaza[2,2] 착물의 경우 $Eu^{3+}Ce^{3+}$ 순으로 안정도가 증가하였으며 이를 최적크기개념과 금속이온의 전하밀도 증가순으로 설명하였다. Diamine ether 착물의 경우는 macrocyclic effect가 없음으로 작은 안정도상수 값을 나타냈다.

사료내 Zeolite 첨가가 비육돈의 생산성과 도체특성에 미치는 영향 (Effect of Dietary Zeolite Treated on the Performance and Carcass Characteristics in Finishing Pigs)

  • 본 연구는 비육돈에 대한 황토의 주성분인 zeolite의 첨가효과를 구명하기 위하여 증체량, 사료요구율, 육질, 악취제거 및 경제성 등을 조사하였으며 생후 93일령의 비육돈 120두(4처리$\time$3반복$\time$10두)를 공시하여 출하시까지 70일간 사양시험을 실시하였다. 일당증체량은 비육돈 전기의 경우 zeolite 4% 첨가구가 1.00kg으로서 대조구의 0.8kg에 비하여 약 25%로 크게 향상되었다(P<0.05). 그러나 비육돈 후기의 일당 증체량은 대조구(0.78kg)를 포함한 모든 처리구에서 0.74-0.83kg으로서 유의한 차이는 없었다. 사료요구율은 시험 전기간 동안 대조구에서는 3.43으로서 가장 높았으며, 그 다음은 zeolite 1%구와 2%로서 각각 3.24와 3.13이었으며 zeolite 3% 첨가구는 3.00으로서 약 7.4%의 개선효과가 나타났다(P<0.05). 시험 전기간 동안의 $NH_3$ 가스 발생량은 zeolite 4% 첨가구가 대조구에 비하여 약 33.9-53.9%의 개선효과가 나타났다(P<0.05). 도체중과 등지방 두께는 대조구와 zeolite 첨가구 간에 차이가 없었으며, A등급 출현율은 zeolite 4% 첨가구가 60.00%로서 대조구의 36.67%에 비하여 높게 나타났다. 경제성 분석 결과, 증체 kg당 사료비는 zeolite 4% 첨가구는 910원, 2% 첨가구는 919원, 1% 첨가구는 941원 및 대조구는 982원으로서 zeolite를 4% 첨가함으로써 개선효과가 크게 나타났다. 이상의 결과, 비육돈에 대하여 zeolite를 4% 수준으로 첨가․급여하면 사료요구율, 육질 A등급 출현율, 분내 암모니아가스 발생량 및 경제성면에서 효과적인 것으로 사료된다.

위암조직에서 종양관련항원 TAG-72와 CEA의 농도 및 분포 (Concentration and Distribution of Tumor Associated Antigens, TAG-72 and CEA, in Stomach Cancer)

  • 악성종양의 진단 및 치료에 있어서 특정 종양에 대한 항체를 이용하는 연구가 활발히 진행되고 있다. 단세포군항체를 이용한 방사면역신티그라피로 암의 조기 진단 및 영상화가 가능하고 나아가 방사면역치료는 암의 선택적 치료에 도움이 될 수 있다. 위암은 우리나라에서 가장 흔한 악성종양으로 방사면역신티그라피와 방사면역치료법이 새로운 방법으로 모색되고 있다. 이러한 진단 및 치료법의 성공여부를 결정하는 중요한 인자의 하나가 종양조직내에서 종양관련 항원의 농도와 분포이다. 따라서 본 연구에서는 단세포군 항체를 이용한 방사면역학적 방법의 임상 이용을 위한 기초 연구의 일환으로 in vitro quantitative autoradiography를 이용하여 종양 관련항원인 TAG-72와 CEA의 위암조직내 농도 및 분포를 측정하였다. 33예의 위암조직에서 얻은 동결절편을 $1.3\sim83.3$ nmol/liter의 $^{125}I$ 표지 항 TAG-72 단세포군 항체 B-72.3과 항 CEA 단세포군 항체 CEA-79로 반응시킨 후 이 표본들의 자가방사법 디지탈 영상을 H & E 염색과 immunoperoxidase염색 표본과 비교하였으며, 특정 단세포군 항체의 결합에 대한 컴퓨터 분석으로 조직내 항원의 농도와 분포를 측정하였다. TAG-72는 25예(75.7%)의 조직에서 검출되었으며 그 농도는 $8.4\sim525.3$ pmol/gram이었고, CEA는 32예 (96.9%)에서 검출되었으며 그 농도는 $8.8\sim592.9$ pmol/gram 이었다. CEA의 위암 조직내 발현농도는 중앙치가 101.7 pmol/gram 으로 TAG-72의 중앙치인 27.9 pmol/gram 보다 높았다. TAG-72의 조직내 분포는 41.4%에서 병변 부위의 암세포 분포와 일치하였고, CEA의 분포는 병변 부위의 80.5%에서 암세포와 일치하는 소견을 나타내었다. TAG-72의 농도는 점액성 선종(mucinous adenocarcinoma)과 점액함유 선종(mucin containing adenocarcinoma)에서 다른 선종보다 더 높았다. CEA의 농도는 위암의 병리학적 종류에 따른 유의한 차이가 없었다. 이상의 결과로 위암조직에서 TAG-72와 CEA항원이 다양하게 발현됨을 알 수 있었고 CEA는 TAG-72 보다 더 빈번하게 균일한 분포로 발현하는 것으로 나타났다.
