This study describes an efficient and stable droplet vitrification following cryopreservation of strawberry shoot tip (Fragaria × ananassa Duch.) accessions 'Massey' and 'MDUS3816'. The shoot tips were precultured in Murashige and Skoog (MS) liquid medium supplemented with sucrose (0.3-0.7M). Precultured explants were osmoprotected with loading solution (LS, C4) containing 17.5% glycerol and 17.5% sucrose for 40 min and exposed to dehydration solution (B1) containing 50% glycerol and 50% sucrose for 40 min at 25oC. Subsequently, the explants were transferred onto droplets containing 2.5 µL PVS3 on sterilized aluminum foils (4 cm× 0.5 cm) prior to direct immersion in liquid nitrogen (LN) for 1 h. The highest regrowth rate (%) in both the cultivars was obtained when the shoot tips were precultured with 0.3M sucrose for 30 h + 0.5M sucrose for 16 h at 25oC. The cryopreserved shoots tips exhibited 57.8 % recovery rate by culturing in NH4NO3-free MS medium supplemented with 3% sucrose, 1.0 g/L casein, 1.0mg/L GA3, and 0.5 mg/L BA for 5 weeks and in MS medium supplemented with 0.5 mg/L GA3 for 8 weeks. Variation was not observed in both of ploidy analysis and morphological investigation on plantlets of two accessions cryopreserved under variable preculture conditions.
The experiment was carried out to investigate the properties of bark extractives form Larix epilepsies and to evacuate their suitability as a bonding agent. The yield and reactivity were measured to examine the influence of temperature and time and the effect of carbonation and sulfonation. To define the possibility of practical application as wood adhesives the viscosity and gelation time were measured at 33% concentration. The results obtained were summarized as follows : 1. As the both yield and reactivity were high, extraction for 2 hours at $80^{\circ}C$ was the optimal temperature and time. 2. The highest effect achieved at 1% $Na_2CO_3$ about carbonation and 1% $Na_2SO_3$ : $NaHSO_3$ and 0.25% $Na_2SO_3$ about sulfonation. The sulfonation of 0.25% $Na_2SO_3$ increased the yield and reactivity most highly. 3. By using hot water as extraction liquid the yield was 17.2%, while the addition of 1% and 5% NaOH to the extraction liquid increased the yield to 38.6% and 44.6%, respectively. 4. Hot water extracts showed the highest reactivity(68.8%). The addition of 1% and 5% NaOH led to decrease in reactivity(49.3% and 25.8%, respectively). 5. At 33% concentration of the extracts the viscosity appeared very variable. Significantly high values of viscosity was measured in 1% NaOH solution, while very low values appeared for 5% NaOH solution. 6. The shortest gelation time was determined at pH 7 to 10 and the highest at pH 4. The use of paraformaldehyde resulted in gelation times longer than those of 37% formaldehyde solution. 7. Except the sulfonation extracts of hot water and 1% NaOH, the other extracts were found unsuitable due to high viscosity(1% NaOH extracts, sulfonation extracts) or to curing inability(5% NaOH extracts, sulfonation extracts of 5% NaOH). 8. From the three extract solutions which appeared to be suitable for use as bonding agents the hot water extracts and the sulfonation extracts of hot water were superior in extract reactivity, while the sulfonation extracts of 1% NaOH exceeded the other two extracts in extract yield.

This study mechanically measured physical properties of jelly made by adding kappa carrageenan and agar-agar to the sipjeondaebo extract, varying the addition ratio. The sipjeondaebo extract used in the study showed 4.95 pH, 7.0 Brix$^{\circ}$, and high turbidity of more than 3.0 NTU. The sugar content was fixed at 25.2 Brix$^{\circ}$ when isomaltooligosaccharide was added in a 25-percent concentration. When the jelly was mechanically measured after it became hard in the refrigerator, the measurement results did not show any significant difference in cohesiveness between different addition ratios of agar-agar to kappa carrageenan. In the measurement of chewiness and gumminess, the jelly show a significantly decreasing tendency in both the two properties as the increase in the addition rate of agar-agar was made. Hardness was also measured lower as the more addition of agar-agar was made, and its the highest level was shown when there was the addition of kappa carrageenan only. Springiness was the highest when the ratio of kappa carrageenan to agar-agar was 0.7%:0.3%, while it was measured the lowest when agar-agar was added in a 1.0% concentration. In the measurement of the jelly's physical properties, the results showed the highest level of appearance when the ratio of kappa carrageenan to agar-agar was 0.7% but indicated that there was no significant difference in flavor. The overall Quality was evaluated to maintain the highest level at the kappa carrageenan to agar-agar ratio of 0.7%:0.3%, followed by a 0.5%:0.5% addition ratio.

Experimental culture of Halocynthia roretzi was carried out near coast by Wolsung from January to August 1997 to investigate accumulation of heavy metals (Cd, Pb and Cu) by H. roretzi, Water temperature, salinity, pH and Suspended Particulate Matter (SPM) were $10.7\sim25.2^{\circ}C$, $33.0\sim34.6\%_{\circ},\;8.25\sim8.49\;and\;3.84\ksim7.92\;mg/L$, respectively, The heavy metal accumulation in H. roretzi was in the order of Cu>Pb>Cd. Heavy metal concentrations in the organs showed following order; Gill${\geq}Viscera{\gg}$Gonad>Muscle>Mantle for Cd, $Gill{\geq}Viscera{\gg}$ Gonad> Muscle$ {\geq}$Mantle for Pb and Viscera>Gill${\gg}$ Gonad>Muscle>Mantle for Cu.

The adsorption and desorption of herbicides such as napropamide and pendimethalin was studied in three kinds of soil. sandy loam. silty clay and loam. The results of batch tests performed with various shaking time, pH, organic matter content and temperature in soil were summarized as follows. The shaking times reached to the equilibrium of the adsorption and desorption for napropamide and pendimethalin in soil were 12 and 6 hours. respectively. For each soil. the adsorption rates of napropamide were 23.35%. 31.57% and 25.95%, the desorption rates of them were 18.42%, 13.42% and 15.89%, respectively. And the adsorption rates of pendimethalin were 59.61%, 77.26% and 64.02%, and the desorption rates of them were 3.23%, 2.93% and 3.07%, respectively. The adsorption isotherms with the Freundlich equation showed better consistency than those with the Langmuir one. The adsorption was affected by the organic matter content when it exceed 2.0%. But if the organic matter content is below 2.0%, it was affected by the clay content. When the organic matter content is 0.95~7.45%, the adsorption coefficients ($K_{fa}$) of napropamide and pendimethalin were 1.17~2.50 and 4.74~16.08 and the desorption coefficients($K_{fd}$) of them were 5.33~34.06 and 24.25~134.00, respectively. Because of the physical adsorption between herbicide molecules and soil surface, little effect of pH variation of soils was appeared for the adsorption and desorption. Because of the solubility of herbicide is related to the temperature, the adsorption rate was decreased and the desorption rate was increased with the temperature increase, respectively.
This study was carried out to reveal the influence of discharge change on matters and stream water quality between pH, EC and dissolved matters obtained by observation of long-term and short-term on stream water quality and separated runoff components from stream water by using HYCYMODEL. From January in 1998 to September in 1999, it was estimated by relationships of character of water quality and discharge for the experimental watershed in Mt. Palgong. The results were summarized as follows : 1. Annual average pH values of stream water in 1998 and in 1999 were 6.48(6.22~6.89) and 6.52(5.75~7.18), respectively. The observed annual average pH values were maintaining identical values in general, but pH values decreased continuously during the four months after thinning in the experimental watershed. So thinning is suspected of the major cause for the decrease. 2. Annual average EC values of stream water in 1998 and in 1999 were $26.69(17.95{\sim}33.5){\mu}S/cm$ and $25.19(17.5{\sim}33.8){\mu}S/cm$, respectively. The observed annual average EC values were maintaining identical values in general. 3. As a result of the comparison of average dissolved ions of rainfall and stream water, $Na^+$, $Mg^{2+}$, $Ca^{2+}$, $Cl^-$, $NO_3{^-}$, and $SO_4{^{2-}}$ showed minus values between incomings and outgoings. $Na^+$ and $NO_3{^-}$ among the dissolved ions of stream water showed the highest concentration out of cations and anions respectively. 4. By the change of pH value in stream water due to rainfall events, pH value decreased with increasing runoff as pH value increased before and after peak flow. 5. By the change of EC value in stream water due to rainfall events, EC value decreased with increasing runoff of first rainfall as EC value changed with runoff before and after peak flow. 6. As the runoff increased, the concentration of $Na^+$, $Ca^{2+}$, $K^+$, total cation, $Cl^-$, and $SO_4{^{2-}}$ in stream water lowered. On the other hand, the runoff decreased, their concentration in stream water tended to get high. But in terms of $NO_3{^-}$ and total anion, they turned out vice versa. $Mg^{2+}$ produced no reaction. 7. The base flow among runoff components separated by using HYCYMODEL influenced greatly on pH, EC, concentration of cation and anion.
This study was conducted to investigate the physicochemical characteristics and biological activities of water and 30%, 50%, 70%, 100% ethanol extracts from Artemisia Argyi H. and fermented Artemisia Argyi H. The yield was the highest in the 30% ethanol extract with Argyi H. at 29.74%. Total phenol and flavonoid contents were the highest in 70% ethanol extract with Argyi H. at 72.25 mg/g and 33.34 mg/g, respectively. The antioxidant activities of all extracts were significantly increased in a dose dependent manner. The 70% ethanol extract from Argyi H. show the highest level of DPPH, ABTS radical scavenging activity and bleaching inhibition activity in ${\beta}$-carotene linoleic acid system. Tyrosinase inhibition activity was also higher in the 70% ethanol extract, and the lowest in the 100% ethanol extract with Argyi H. at 50.01% and 11.44% at $500{\mu}g/ml$ concentration, respectively. At $250{\mu}g/ml$ concentration, the xanthin oxidase inhibition activity of water extract was more than 60%, and it was higher than the extracts. These results suggest that the 70% ethanol extract of Artemisia Argyi H. has a high rate of biological activities and can be useful to develop functional food ingredients.
The compost dextrose(CD) medium was selected a an optimum liquid culture media for Portobella(Agaricus brunescens). The optimal conditions for the mycelial growth of Agaricus brunescens were 25 and pH 7.0. Sucrose and soluble starch were favorable to mycelial growth. Malt extract and ammonium nitrate appeared to be more favorable than the other kinds of nitrogen source. The optimum C/N ratio as about 10.When the collected Portobella trains were compared with growth characteristics, KME 49007 appeared the highest of $44.6kg/3.3m^2$.

The effect of culture temperature on the production of soluble form of B. macerans cyclodextrin glucanotransferase (CGTase) in recombinant E. coli was investigated. E. coli cell was cotransformed with two plasmids (pTCGT1 and pGroll) in which the cgt and groEL/ES genes are under the control of T7 promoter and pzt-1 promoter, respectively. When tetracycline (10 ng/ml) and IPTG (l mM) were added as inducers at the early-exponential phase (2 h) and mid-exponential phase (3h), respectively, the solubilization of the inclusion body CGTase was greatly dependent on the temperature of the culture. At low culture temperature of $25^\circ{C}$, 2- or 3-fold higher activity and specific activity were obtained over $37^\circ{C}$. SDS-PAGE analysis revealed that about 62% of CGTase in the total CGTase protein was found in the soluble fraction by applying overexpression of GroEL/ES chaperone and by cultivation of E. coli at $25^\circ{C}$, whereas 33% of CGTase was detected in the soluble fraction at $37^\circ{C}$. Therefore, the expression of GroEL/ES and cultivation at $25^\circ{C}$ greatly enhanced the soluble production of CGTase in E. coli.
Boar semen extender Kp (Hankook Life-Science, Korea) was newly formulated by authors. This study was carried out to investigate the optimal concentrations of EDTA, Tris and citrate buffers in the Kp extender (Basic Kp) on the pH change during storage. And then the motility of boar sperm with the Kp pH stabilized (Modified Kp) was compared with those of commercial products imported into Korea such as BTS (Mini-tube. Germany; BTSg), BTS (Tri-bio, USA; BTSa) and Modena (SGI, USA). The pH values of all extenders were increased gradually with the storage days. Especially, the initial pH of Basic Kp was higher than that of BTSg, BTSa and Modena, and also higher than physiological pH of boar sperm (6.8∼7.5). When Basic Kp was added with various concentrations of EDTA (0, 0.63, 1.25 & 2.37g/L), Tris (0, 0.18, 0.35, 0.71 & 1.42g/L) and Citrate (0, 0.75, 0.81, 1.00, 1.25 & 1.50g/L) buffers for pH down-regulation and stabilization of pH, the group added with 1.25g EDTA, 1.42g Tris and 1.00g Citrate well maintained the neutral range of pH during storage (6.88 at day1 to 7.33 at day6 in Modified Kp), Especially, the concentrations of the buffers added in Modified Kp were lower, until 1/2∼1/4 ranges, than those in Modena and other extenders. The motility of frozen-thawed boar sperm diluted with Modified Kp was significantly higher than that of Basic Kp, BTSg, BTSa and Modena (87.0% vs. 55.0∼71.0% at day1; 13.3% vs. 0∼6.3% at day6), Conclusively, Modified Kp in this experiment was kept the favorable physiological conditions in spite of low concentrations of the buffers and motility of frozen-thawed boar sperm was obtained better than that of Basic Kp and other commercial products such as BTSg, BTSa and Modena.
