  • In 1975, a trial for channel catfish spawning was made for the first time in Korea, by stocking 11 pairs of brood fish in a $470m^2$ earthen pond provided with 5 cans of 35 cm in diameter with depths of about 60 cm as spawning nests. The same trial and another pen cage method combined with pituitary injection were practiced in 1976. All these trials failed without any spawning. On the other hand 4 females and 1 male were put in a recycling water system indoor aquarium for exhibition purpose it late spring of 1976. After removing 3 females which were injured from being bitten by the male, remaining one pair of the catfish spawned successfully in July and the most eggs developed normally. The water in the recycling system showed the pH range of 7.2 to 7.4,and mean water temperature of every 10 day period of a month gradually increased from 19.3 to 23.6C until spawning. The outdoor .ponds where the spawning was failed received water from the same origin as the indoor recycling system, and the mean temperature of every ten day period ranged Iron 17.2 to $29.8^{\circ}C$ in 1975 and 19.7 to $28.4^{\circ}C$ in 1976 these being considered well falling in the optimum range for catfish spawning, but pH ranged from 7.3 to 9.5 under the influence of photosynthesis of algae and weeds in the ponds. The high pH value may suppress the spawning of catfish as pointed out by Nelson in 1957.

  • Aspergillus ochraceus biofilm, developed on an inert support, can produce tannase in Khanna medium containing 1.5% (w/v) tannic acid as the carbon source, at an initial pH of 5.0, for 72 h at 28℃. Addition of 0.1% (w/v) yeast extract increased enzyme production. The enzyme in the crude filtrate exhibited the highest activity at 30℃ and pH 6.0. At 50℃, the half-life (T50) was 60 min and it was 260 min at pH 6.0. In general, addition of detergents and surfactants did not affect tannase activity significantly. Tannase has potential applications in various biotechnological processes such as the production of propyl gallate and in the treatment of tannin-rich effluents. The content of tannins and total phenolic compounds in effluents from leather treatment was reduced by 56-83% and 47-64%, respectively, after 2 h of enzyme treatment. The content of tannins and total phenolic compounds in the sorghum flour treated for 120 h with tannase were reduced by 61% and 17%, respectively. Interestingly, the same A. ochraceus biofilm was able to produce tannase for three sequential fermentative process. In conclusion, fungal biofilm is an interesting alternative to produce high levels of tannase with biotechnological potential to be applied in different industrial sectors.

  • Shotcrete, rockbolt, and steel rib are installed to support ground after tunnel was excavated. These are important supports for NATM applied tunnels. Recently, lattice girder is increasingly used because it is easily installed. In this study, we developed a new lattice girder by increasing contacting area between spider and rod. To verify the effect of the new lattice girder, the 3-point and 4-point flexural strength tests were carried out for LG-$50{\times}20{\times}30$, LG-$70{\times}20{\times}30$, LG-$95{\times}22{\times}32$. As a result, in case of contacting area, strength of new SGS lattice girder is 17.95% higher than that of original lattice girder. In case of weakness point, strength of new SGS lattice girder is 19.37% higher than that of original lattice girder.

  • Background: To compare the effects of adding two different doses of dexamethasone on the duration and quality of the fascia iliaca block in patients undergoing proximal femoral fracture surgery. Methods: A total of 60 patients (age 18-70 years) undergoing proximal femoral nailing surgery under spinal anesthesia were given fascia iliaca block after random assignment to one of the two groups: Group H received an injection of levobupivacaine (0.5%) 28 ml with 2 ml (8 mg) dexamethasone, and Group L received an injection of levobupivacaine (0.5%) 28 ml with dexamethasone 1 ml (4 mg) with 1 ml normal saline. Assessment of the duration of analgesia and the total tramadol requirement over 48 hours were noted after a successful block. Results: The duration of analgesia was found to be significantly longer in Group H ($17.02{\pm}0.45h$) than in the Group L patients ($14.29{\pm}0.45h$) with a p-value of 0.000. Postoperative analgesic requirement (amount of tramadol in mg) was significantly higher in Group L (Q2: 200.0; IQR: 100.0, 200.0) as compared to Group H (Q2: 100.0; IQR: 100.0, 200.0) with a p-value of 0.034. No patient showed any sign of neurotoxicity. Conclusions: Dexamethasone, in a dose of 8 mg, is superior to 4 mg when used as an adjuvant with levobupivacaine in the FIB. Though both prolonged analgesia and were effective in reducing oral/intravenous analgesics, 8 mg dexamethasone can be recommended as a more efficacious adjuvant to local anesthetics in the FIB.

  • Dry corn gluten meal of 70% protein content was enzymatically hydrolyzed by alkaline protease in a pH-state reactor. Such process variables as temperature, pH, and enzyme-to-substrate ratio were varied, and at each condition degree of hydrolysis was monitored and calculated. The ultimate degree of hydrolysis, which ranged between 25 and 28% based on gluten protein mass, was not significantly affected by the process variables. However, $50^{\circ}C$ and pH 9-10 appeared optimum. Kinetic analysis indicated enzyme deactivation was negligible during the hydrolysis, and the experimental data were near perfectly fitted to the model kinetic equation which was modified after neglecting enzyme deactivation term. The enzyme reaction was 1$100\times$ scaled up and basically the same hydrolysis performance was resulted. Amino acid analysis showed the hydrolyzate was relatively rich in glutamine/glutamic acid, leucine, and alanine at 19.6, 16.1, and 12.3 mole %, respectively.

  • A freshwater cyanobacterium, Phormidium autumnale KNUA026, was isolated from puddles of icy water in Gyeongsan City, South Korea and its potential as a biofuel feedstock was investigated. Maximal growth was obtained when the culture was incubated at $25^{\circ}C$ and around pH 9.0. The total lipid content of the isolate was approximately 14.0% of dry weight and it was found that strain KNUA026 was able to autotrophically synthesize heptadecane ($C_{17}H_{36}$) which can be directly used as fuel without requiring a transesterification step. As this benthic cyanobacterium was capable of forming thick mats, it could be easily harvested by gravitational settling and this property may reduce the cost of production in commercial applications. Hence, P. autumnale KNUA026 appears to be a promising resource for use in the production of microalgae-based biofuels.

  • The effects of carbon, nitrogen sources and culture conditions on glucoamylase production from Aspergillus sp. were investigated. Among tested carbon sources, soluble starch was most effective for the production of the enzyme, and the level of concentration for the optimal enzyme production was found to be 5%. For nitrogen sources, yeast extract was best for the enzyme production, with the level of 0.1%. The enzyme was maximally produced by cultivating the organism at medium of initial pH 6.0, and temperature of 28$^{\circ}C$. Wheat bran was most suitable for the enzyme production from the organism in solid state culture.

  • This study analyzed the antibacterial activity of a Rubus coreanum (Bokbunja) ethanol extract. The antimicrobial activity was determined by disc diffusion, minimum inhibitory concentration (MIC), and growth inhibition methods with seven kinds of bacteria related to foodborne illness (Bacillus cereus, Escherichia coli, Escherichia coli O157:H7, Listeria monocytogenes, Pseudomonas aeruginosa, Staphylococcus aureus, and Salmonella typhimurium). In the results, disc diffusions of the ethanol extract from R. coreanum (9.8-17.5 mm at $4,000{\mu}g/disc$) clearly showed the antimicrobial activity of the extract against all tested microorganisms. Rubus coreanum promoted an inhibitory effect as follows: E. coli O157:H7 > P. aeruginosa > L. monocytogenes > E. coli > S. aureus > B. cereus ${\geq}$ S. typhimurium. In the MIC test, R. coreanum showed high antimicrobial effect against L. monocytogenes at 500 ppm. Moreover, the R. coreanum ethanol extract showed strong growth inhibition against microorganisms, similar to the MIC results. These results show that a R. coreanum ethanol extract has powerful antimicrobial activity against all tested microorganisms, suggesting that R. coreanum will be useful as a potential natural preservative.
