염욕의 pH와 매염제의 변화에 따른 코치닐의 염색성 연구 I -면직물을 중심으로- (A Study on Cochineal Dyeing by Various Mordants and pH Conditions I -Treatment on Cotton Fabric-)

  • In this study, cotton fabric specimens were pre-mordanted using Sn, Al, Cu, Cr, and Fe, and subsequently dyed using cochineal, maintaining the pH of the dye bath constant using pH buffer solutions of 4, 5, 6, 7, and 8. In the cases of Al, Cr, and Sn mordanting, the dye-uptake was well accomplished in the acidic range of pH 4 and 5. However, the dye-uptake was not accomplished in the range over pH 6. In the cases of Cu and Fe mordanting, however, the pH value did not affect the dye-uptake and the dyeability in the alkaline range was not decreased remarkably. In the case of non-mordant, the dye-uptake was not accomplished at all, and the Cu and Fe mordanting gave rise to the best dyeability. In the cases of non-mordanting and Al, Cu mordanting, there was no change in the $\lambda_{max}$, and in the cases of Sn and Cr mordanting, the $\lambda_{max}$ moved toward the shorter wavelength. In the case of Fe mordanting only, the $\lambda_{max}$ moved gradually toward the longer wavelength, and due to the pH effect deep-dyeing effect was revealed.

Effect of pH on the Vascular Tone and $^{45}Ca$ Uptake in the Aorta of Spontaneously Hypertensive Rats

  • The effect of extracellular and intracellular pH on vascular tone and $^{45}Ca$ uptake were investigated in aortic strips and dispersed single aortic smooth muscle cells of spontaneously hypertensive rats (SHR) and aged-matched Wistar-Kyoto rats (WKY). The contraction produced by a change of extracellular pH (pHo) in the range of $6.5{\sim}8.3$ was estimated by comparison with the level of vascular tone at pH 7.4. Contraction was induced below pHo 6.5 in WKY, pHo 7.1 in SHR, and over pHo 8.0 on both strains. The amplitude of contraction induced by high pHo (over pHo 7.7) was similar in SHR and WKY, but that induced by low pHo (below pHo 7.1) in SHR was greater than that in WKY. Either high pHo- or low pHo-induced contractions in WKY and SHR were not induced in the Ca-free Tyrode's solution and were induced by the addition of Ca. $^{45}Ca$ uptake increased progressively as pHo was increased from 6.8 to 8.1 in the single aortic smooth muscle cells of WKY and SHR. $NH_4Cl$ induced a gradually developing contraction in a dose-dependent manner $(5\;mM{\sim}30\;mM)$ and the removal of $NH_4Cl$ induced transient contraction was followed by profound relaxation in the aortic rings of both strains. The contractions induced by $NH_4Cl$ or by the removal of $NH_4Cl$ in SHR were significantly greater than that in WKY. These contractions were not induced in Ca-free Tyrode's solution. $^{45}Ca$ uptake was increased by $NH_4Cl$ (20 mM) and was not changed by the removal of $NH_4Cl$ (20 mM) in the aortic strips of WKY and SHR. As a summary of above results, the vascular tone of SHR was more sensitive to the change pHi and pHo than that of WKY. The contractions induced by change of extracellular or intracellular pH depended on extracellular Ca in the aorta of SHR nnd WKY. However, the Ca uptake was in accord with the changes of contraction but increase in contraction by low pH was not accompanied by an increase in Ca uptake in both strains.

다시마(Saccharina japonica)의 생장에 따른 영양염 및 CO2 흡수율과 광합성 특성 변화 (Variations in Nutrients & CO2 Uptake Rates and Photosynthetic Characteristics of Saccharina japonica from the South Coast of Korea)

  • 국내 해조류 양식종인 다시마(Saccharina japonica (Laminaria japonica Areschoug))의 영양염 및 무기탄소 흡수율과 광합성 특성을 살펴보기 위해, 부산시 기장군 일광해역 양식장에서 2011년 l월부터 5월동안 시료를 채집하여 실내 배양하였다. 배양 시료의 엽장과 엽폭 범위는 각각 46~288 cm, 4.8~22.0 cm였으며, 이들의 용존산소 평균 생산율은 $6.9{\pm}5.8{\mu}mol\;g^{-1}$ fresh weight(FW) $h^{-1}$, 질산염과 인산염 흡수율은 각각 $175.6{\pm}161.1\;nmol\;g^{-1}\;FW\;h^{-1}$, $12.7{\pm}10.1\;nmol\;g^{-1}\;FW\;h^{-1}$ 그리고 용존무기탄소 흡수율은 $8.9{\pm}7.9{\mu}mol\;g^{-1}\;FW\;h^{-1}$이었다. 다시마 엽장에 따른 이들 성분의 생산/흡수율은 로그함수 관계를 보여, 엽장 100~150 cm 이상부터는 다시마의 광합성 및 생장율이 다소 둔화되는 것으로 나타났다. 용존 산소 생산율과 질산염, 인산염, $TCO_2$ 흡수율은 각각 음의 선형관계를 보였으며, 특히 용존산소 생산율과 $TCO_2$ 흡수율은 높은 상관관계 ($r^2$=0.94)를 나타내어 서로에 대한 간접 지표가 될 수 있음을 의미한다. 다시마의 최대양지수율($F_v/F_m$)과 용존산소 및 무기탄소 생산/흡수율사이에 선형관계가 나타났으며, 양자수율이 높을수록 활발한 광합성작용으로 용존산소 생산이 활발함을 증명하였다. 광합성 형광특성 중 최대상대전자전달률은 비교적 엽체가 생장할수록 증가하였으며, 엽체의 부착부에 비해 상부 말단으로 갈수록 증가하였다. 이는 다시마가 생장할수록 광합성활성 조직 비(단위 면적당 습중량)가 증가하여 나타난 결과로 사료된다. 기장해역에서 다시마에 의한 연간 무기탄소 흡수량은 $1.0\sim1.7{\times}10^3C$ ton으로 추정되며, 이는 부산시 이산화탄소 배출량의 0.02~0.03%에 해당한다. 따라서 해조류에 의한 연안의 탄소 및 물질순환과 지구시스템에서의 역할을 파악하기 위해 추가적인 연구가 필요할 것으로 생각된다.

Evidence for Sulfite Proton Symport in Saccharomyces cerevisiae

  • The kinetics of sulfite uptake were examined in a wild-type laboratory strain of Saccharomyces cerevisiae to determine if carrier-mediated sulfite uptake involved a proton symport, as previous studies on sulfite uptake have suggested both an active process and facilitated diffusion. Accumulation of intracellular sulfite was initially rapid and linear up to 50 sec. Uptake was saturable at final concentrations equal to or greater than 3 mM sulfite, and increased 2-fold in the presence of 2% glucose. Uptake was significantly reduced in cells pretreated with 100-500 $\mu$M carbonyl cyanide mchlorophenylhydrazone (CCCP) or 2,4-dinitrophenol (DNP), both of which dissipate proton gradients. Uptake was also significantly inhibited in the presence of 1 mM arsenate, an inhibitor of ATP synthesis. Extracellular alkalization was observed in cells incubated with 1-2 mM sulfite in a weak tartrate buffer at pH 3.5 and 4.5. These findings suggest that the bisulfite ion, $HSO_3^-$, an anionic form of sulfite, is taken up by a carrier-mediated proton symport. A met16 sull sul2 mutant, impaired in both sulfite formation and sulfate uptake, was found able to grow on a medium with sulfite as the sole Sulfur source, indicating that the sulfate transporters Sul1p and Sul2p are not required for sulfite uptake.

Functional Expression of Choline Transporter-Like Protein 1 in LNCaP Prostate Cancer Cells: A Novel Molecular Target

  • Prostate cancer is one of the most common cancers in men. Choline PET or PET/CT has been used to visualize prostate cancer, and high levels of choline accumulation have been observed in tumors. However, the uptake system for choline and the functional expression of choline transporters in prostate cancer are not completely understood. In this study, the molecular and functional aspects of choline uptake were investigated in the LNCaP prostate cancer cell line along with the correlations between choline uptake and cell viability in drug-treated cells. Choline transporter-like protein 1 (CTL1) and CTL2 mRNA were highly expressed in LNCaP cells. CTL1 and CTL2 were located in the plasma membrane and mitochondria, respectively. [3H]Choline uptake was mediated by a single Na+-independent, intermediate-affinity transport system in the LNCaP cells. The anticancer drugs, flutamide and bicalutamide, inhibited cell viability and [3H]choline uptake in a concentration-dependent manner. The correlations between the effects of these drugs on cell viability and [3H]choline uptake were significant. Caspase-3/7 activity was significantly increased by both flutamide and bicalutamide. Furthermore, these drugs decreased CTL1 expression in the prostate cancer cell line. These results suggest that CTL1 is functionally expressed in prostate cancer cells and are also involved in abnormal proliferation. Identification of this CTL1-mediated choline transport system in prostate cancer cells provides a potential new therapeutic target for the treatment of this disease.

이매패류 3종의 크기 및 수온에 따른 여수율과 노닐페놀 흡수율의 관계 (Relationship of the Clearance Rate and Nonylphenol Uptake Rate of Three Bivalve Species with Different Size Classes and Temperatures)

  • 본 연구에서는 두 개의 크기집단의 바지락(Ruditapes philippinarum), 재첩(Corbicula japonica), 홍합(Mytilus edulis)을 세 개의 수온 조건(5, 13 그리고 $18^{\circ}C$)에서 용존 노닐페놀에 노출시킴으로써 개체 크기와 수온 변화가 여수율과 용존 노닐페놀 흡수율에 미치는 영향을 조사하였다. 이를 통해 이매패류의 여수율과 유기화합물질 흡수과정의 관련성에 대해 평가하고자 하였다. 단위무게당 여수율($CR_{wt}$)은 모든 개체 크기와 온도 범위에서 홍합(0.3-4.9 L $g^{-1}\;h^{-1}$)이 가장 높았으며, 재첩(0.44-1.98 L $g^{-1}\;h^{-1}$), 바지락(0.08-0.6 L $g^{-1}\;h^{-1}$) 순으로 나타났다. 한편 단위무게당 노닐페놀의 흡수율은 종 간 뚜렷한 차이를 보이지 않았다. 단위무게당 여수율과 노닐페놀 흡수율은 일반적으로 개체의 크기가 증가함에 따라 감소하고, 온도가 증가함에 따라 증가하는 경향을 보였다. 세 종 모두에서 노닐페놀 흡수율은 여수율과 높은 상관관계를 보였다. 한편 노닐페놀 흡수율은 같은 여수율 범위에서 홍합, 재첩, 바지락 순으로 증가하였다. 여수율과 노닐페놀 흡수율의 높은 상관관계로부터 여수율이 이매패류의 용존 유기화합물 흡수에 중요한 역할을 한다는 것을 추정할 수 있다.

  • Phenyl styryl ketone ligands 1-5 containing different functional groups were synthesized and attached to the aminated poly (styrene-divinvlbenzene) copolymer through diazotization. The coupling of the ligand with polymer was confirmed by FT-IR. The variation of the uptakes of Pb (II) or Cu (II) ions loaded in the prepared polymers (6-10) was examined at different pH values. An increase of metal uptake in the polymers was observed with the increase in pH value in aqueous solution.

갈색해조류에 의한 카드뮴의 흡착 및 탈착 특성 (Characteristics of Cadmium Biosorption and Desorption by Brown Marine Algae)

  • The biosorption and desorption of Cd were carried out using brown marine algae, known as the good biosorbent of heavy metals. The content of alginate bound to light metals could be changed by the physical and the chemical pretreatment of Sargassum fluitans biomass. The Cd uptake was independent of the alginate content. In case of protonated biomass, Cd uptake was the lowest because the alginic acid of biomass was dissolved to cadmium solution during the biosorption. The maximum Cd uptake of Sargassum biomass was ranged from 79 mg/g to 139 mg/g. In case of raw biomass, the higher the alginate content of biomass, the higher was the Cd uptake. 100% of Cd and light metals sorbed in the biomass were eluted at 0.1N HCI(pH 1.1). However, the elution efficiency in $CaCl_2$ and $Ca{(NO_3)}_2$solution was varied by the concentration, the solid to liquid ratio and the pH of calcium solution. The distribution coefficient between Cd and protons in the desorption solution at pH ranged from 1.6 to 2.9 was observed on the constant stoichometric coefficient(1.3).

정량화한 $^{99m}Tc$-DMSA 신섭취율의 공식표 (Formal Charts for Qnantified $^{99m}Tc$-DMSA Renal Uptake Rates)

  • To Assessment of the quantified renal uptake rates in every $^{99m}Tc$-DMSA scan tests of patients is actually difficult because of time consumption and complicated calculations required to measure the correct dose of the infused radionulide and radiation decay, the adjustment for the depth of kidney and the subtraction of background count. We've formulated two regression models for the quantified renal uptake rates[I] from the simple renal uptake rates[H] with a square shaped ROI (Region-Of-Interest) in 25 cases (Group 1) and with a kidney shaped ROI in 37 cases (Group 2), respectively. The regression model for the Group 1 was $[I]_1$=0.885 $[H]_1$-4.575 (P<0.005), and for the Group 2 was $[I]_2$=0.591 $[H]_2$-2.105 (p<0.005). The formal charts were clinically convenient to evaluate the individual renal functions in patients with $^{99m}Tc$-DMSA renal scan.

새로운 천연염재로서 메타쉐콰이어 열매의 활용(1): 면섬유 염색을 중심으로 (Utilization of Metasequoia(Metasequoia glyptostroboides) Cone as a New Natural Dye Resource(1): Dyeing of Cotton Fiber)

  • The aim of this study was to investigate the possibility utilizing Metasequoia(Metasequoia glyptostroboides) cone as a new natural dye resource. Dyeing onto cotton fiber was carried out to study the effect of dyeing conditions and mordanting effect on dye uptake, color change, and colorfastness. FT-IR analysis supported that hydrolyzable tannins were contained in the extracted colorant. Metasequoia cone colorant showed low affinity to cotton fiber and maximum dye uptake was obtained at pH 3.5 showing YR Munsell color. Mordanting improved dye uptake regardless of mordant type, especially Fe($C_5H_{10}FeO_6$) mordant was effective as much as 2 times higher dye uptake comparing with un-mordanted sample. The color of dyed fabric with mordanting showed YR Munsell color except of the Fe($FeSO_4{\cdot}7H_2O$) mordanted sample showing Y Munsell color. Colorfastness to rubbing and washing was relatively good, whereas lightfastness of the dyed fabrics was above grade 3/4 except that the dyed samples with Fe mordanting showed grade 2. It is necessary to apply Metasequoia cone colorant onto other fibers, especially protein fibers, for evaluating its efficacy as a new natural dye resource.
