Statistical Optimization for Improved Production of Cyclosporin A in Solid-State Fermentation

  • This work evaluates the effect of different amino acids on production of Cyclosporin (CyA) production in solid-state fermentation that was previously optimized for different fermentation parameters by one factor at-a-time for the maximum production of CyA by Tolypocladium inflatum MTCC557. Based on the Plackett-Burman design, glycerol, ammonium sulfate, $FeCl_3$, and inoculum size were selected for further optimization by response surface methodology (RSM). After identifying effective nutrients, RSM was used to develop mathematical model equations, study responses, and establish the optimum concentrations of the key nutrients for higher CyA production. It was observed that supplementation of medium containing (% w/w) glycerol, 1.53; ammonium sulfate, 0.95; $FeCl_3$, 0.18; and inoculum size 6.4 ml/5g yielded a maximum of 7,106 mg/kg as compared with 6,480 mg CyA/kg substrate using one factor at-a-time. In the second step, the effect of amino acids on the production of CyA was studied. Addition of $_L$-valine and $_L$-leucine in combination after 20 h of fermentation resulted in maximum production of 8,166 mg/kg.

  • This study is to investigate the possibility of using activated carbon prepared from Iraqi date-pits (ADP) which are produced from palm trees (Phoenix dactylifera L.) as low-cost reactive material in the permeable reactive barrier (PRB) for treating lead (Pb+2) from the contaminated groundwater, and then compare the results experimentally with other common reactive materials such as commercial activated carbon (CAC), zeolite pellets (ZP). Factors influencing sorption such as contact time, initial pH of the solution, sorbent dosage, agitation speed, and initial lead concentration has been studied. Two isotherm models were used for the description of sorption data (Langmuir and Freundlich). The maximum lead sorption capacities were measured for ADP, CAC, and ZP and were found to be 24.5, 12.125, and 4.45 mg/g, respectively. The kinetic data were analyzed using various kinetic models particularly pseudo-first-order, pseudo-second-order, and intraparticle diffusion. COMSOL Multiphysics 3.5a depend on finite element procedure was applied to formulate transmit of lead (Pb+2) in the two-dimensional numerical (2D) model under an equilibrium condition. The numerical solution shows that the contaminant plume is hindered by PRB.

A Study on Pressure Characteristic in Various Inner Structure of Valves (밸브 운동부 구조 변화에 따른 압력특성에 관한 연구)

  • In general, the control valves are essential components in hydraulic systems. Structural changes within the valves remain a challenge because many parameters of valve tend to interact in terms of static and dynamic performance. Therefore, the valve characteristics is applied directly to the stability of hydraulic system. Inner structure of the valve which is used mainly in the industries is made up poppet type and spool type. This paper made a description of the method for numerical analysis and modeling of the valve with a built-in moving part of four-type. Based on the physical parameters of the valves, a numerical model of objected valve is developed using the bond graph method. It is to verified the results that the moving part of four-type has an effect on pressure and flow characteristics. Also, It is analyzed the results which has an effect on response characteristic by angular of poppet valve face and inertia variation of the valve with a built-in moving part. In the results, it is confirmed that the rising and settling time vary with the shape of moving part in valve.

Flux pinning and critical current density in $TiO_2$-doped $MgB_2$ superconductor

  • $MgB_2$ doped with $TiO_2$ was prepared by the in-situ solid state reaction to study the effects of $TiO_2$ dopant on the flux pinning behavior of $MgB_2$ superconductor. From the field-cooled and the zero-field-cooled temperature dependences of magnetization, the realms of vortex-glass and vortex-liquid states of $TiO_2$-doped $MgB_2$ were determined in the H-T diagram (the temperature dependence of upper critical magnetic field and irreversibility line). The critical current density was estimated from the width of hysteresis loops in the framework of Beam's model at different temperatures. The results indicate that nano-scale $TiO_2$ inclusions play a role of the effective pinning centers and lead to the enhanced upper critical field and critical current density. It is suggested that the grain-boundary pinning mechanism is realized in $TiO_2$-doped $MgB_2$ superconductor.

  • The use of biogas as an energy source reduces the chance of possible emission of two greenhouse gases, $CH_4$ and $CO_2$, into the atmosphere at the same time. Its nature of being a reproducible energy source makes its use even more attractive. This research if for the hydrogen production through the steam reforming of the biogas. The biogas utilized 3D-IR matrix burner in which the surface combustion is applied. The nickel catalyst was used inside a reformer. Parametric screening studies were achieved as Steam/Carbon ratio, biogas component ratio, Space velocity and Reformer temperature. When the condition of Steam/Carbon ratio, $CH_4/CO_2$ ratio, Space velocity and Refomer temperature were 3.25, 60%:40%, 19.32L/$g{\cdot}hr$ and $700^{\circ}C$ respectively, the hydrogen concentration and methane conversion rate were showed maximum values. Under the condition mentioned above, $H_2$ concentration was 73.9% and methane conversion rate was 98.9%.

  • In designing high efficiency electrical machines, calculation of iron loss is very important. And it is reported that in the induction motor and in the T-joint of 3 phase transformer, there occurred rotational magnetic field and much iron loss is generated owing to this field. In this paper, rotational power loss in the electrical machine under rotational magnetic field is discussed. Until now, loss analysis is based on the magnetic properties under alternating field. And with this one dimensional magnetic propertis, it is difficult to express iron loss under rotational field. In this paper, we used two dimensional magnetic property data for the numerical calculation of rotational power loss. We used finite element method for calculation and the analysis model is two dimensional magnetic property measurement system. We used permeability tensor instead of scalar permeability to present two dimensional magnetic properties. And in this case, we cannot uniquely define energy functional because of the asymmetry of the permeability tensor, so Galerkin method is used for finite element analysis.

  • When Deterministic Design Optimization (DDO) methods are used, deterministic optimum designs are frequently pushed to the design constraint boundary, leaving little or no room for tolerances (or uncertainties) in design, manufacture, and operating processes. In the Reliability-Based Design Optimization (RBDO) model for robust system design, the mean values of uncertain system variables are usually used as design variables, and the cost is optimized subject to prescribed probabilistic constraints as defined by a nonlinear mathematical programming problem. Therefore, a RBDO solution that reduces the structural weight in uncritical regions does not only provide an improved design but also a higher level of confidence in the design. In this work, we seek to improve the quality of RBDO processes using efficient optimization techniques with object of improving the resulting objective function and satisfying the required constraints. Our recent RBDO developments show its efficiency and applicability in this context. So we present some recent structural engineering applications demonstrate the efficiency of these developed RBDO methods.

  • Titanium alloy sheets have excellent specific strength and corrosion resistance as well as good performance at high temperature. Recently, titanium alloys are widely employed not only aerospace parts but also bio prothesis and motorcycle. However, due to the low formability and large spring back at room temperature, titanium alloy sheets were usually formed by slow forming or hot forming with heating die and specimen. In the sheet metal forming area, FE simulation technique to optimize forming process is widely used. To achieve high accuracy FE simulation results, Identification of material properties and deformation characteristic such as yield function are very important. In this study, uniaxial tensile and biaxial tensile test of Ti-6Al-4V alloy sheet with thickness of 1.0mm were performed at elevated temperature of 873k. Biaxial tensile tests with cruciform specimen were performed until the specimen was breakdown to characterize the yield locus of Ti-6Al-4V alloy sheet. The experimental results for yield locus are compared with the theoretical predictions based on Von Mises, Hill, Logan-Hosford, and Balat's model. Among these Logan-Hosford's yield criterion well predicts the experimental results.

Estimation of Nitrogen and Sulfur Dry Deposition over the Watershed of Lake Paldang (팔당호 유역에 대한 질소와 황의 건식 침적량 추정)

  • Lake Paldang is a main resource of drinking water for 20 million people in the greater Seoul area. Dry deposition amounts of nitrogen and sulfur were estimated for three typical days in each season over the watershed of Lake Paldang. Models- 3/CMAQ (Community Multiscale Air Quality) and MM5 (Mesoscale Model) were used to predict air quality and meteorology, respectively. Aerosols as well as gaseous pollutants were considered. Nitrogen was mainly deposited in the form of HNO, while most of sulfur was deposited in the form of SO$_2$. Contribution of secondary pollutants was the largest in fall since they were transported from the greater Seoul area. However, contribution of secondarily-formed particulate pollutants to the nitrogen deposition was the largest in winter because semi-volatile ammonium nitrate favors lower temperature. Annual deposition amounts of nitrogen and sulfur were 37% and 26% of their emission amounts, respectively, over the watershed of Lake Paldang. Higher value of the nitrogen deposition showed a more influence of pollutants emitted in the greater Seoul area.

Tidal Dwarf Galaxies around a Post-Merger Galaxy, NGC 4922

  • One possible channel for the formation of dwarf galaxies involves birth in the tidal tails of interacting galaxies. We report the detection of a bright UV tidal tail and several young tidal dwarf galaxy candidates in the post-merger galaxy NGC 4922 in the Coma cluster. Based on a two-component population model (combining young and old stellar populations), we find that its light predominantly comes from young stars (a few Myr old). The Galaxy Evolution Explorer (GALEX) ultraviolet data played a critical role in the parameter (age and mass) estimation. Our stellar mass estimates of the tidal dwarf galaxy candidates are ~10^{6-7} M_sun, typical for dwarf galaxies.
