A Study on the Effect of Herbal-acupuncture with Carthami Flos at Joksamni($ST_{36}$) on Collagen-induced Arthritis in Mice (족삼리(足三里) 홍화약침(紅花藥鍼)처치가 Collagen으로 유발한 생쥐의 관절염 모델에 미치는 영향)

  • Objectives : The purpose of this study is to observe the effects of Carthami Flos herbal-acupuncture (CF-HA) at Joksamni($ST_{36}$) on arthritis in mice induced by Collagen II. Methods : The author performed several experimental items, including arthritis evaluation, change in weight, spleen size and stenosis rate, change in cytokine level, IgG, IgM and anti-collagen II, change of immunocyte count and histological change of the CIA mouse joint. Conclusions are as follows: Results : 1. In the CF-HA, the arthritis index and rate and the incidence of arthritis were decreased as the experiment proceeded. 2. In the CF-HA, spleen swell and stenosis, joint edema and change were decreased. 3. In the CF-HA, the level of $IL-1{\beta}$, IL-6, $TNF-{\alpha}$ and $IFN-{\gamma}$ in blood serum were significantly decreased. 4. In the CF-HA, the level of IgG, IgM and anti-collagen II were decreased. 5. In the CF- HA, $IFN-{\gamma}$, $IFN-{\gamma}/IL-4$, IL-10 of the culture fluid was decreased. 6. In the CF-HA, the cell rate of $CD3e^+$ and $CD45R^+$, $CD4^+$ and $CD8^+$, $CD4^+/CD25^+$ in spleen was similar to the cell rate of the normal group. 7. In the CF-HA, the cell rate of $CD4^+/CD25^+$, $CD45R^+/CD69^+$ in a lymph node was decreased as in the normal group. 8. In the CF-HA, the cell rate of $CD3^+/CD69^+$, $CD11b^+/Gr-1^+$ in joints was decreased as in the normal group. 9. In the CF-HA, the cartilage destruction and the inflammation cell growth in the H&E stain were decreased. The collagen fiber in the M&T stain were less destructed, therefore the result was similar to the normal group. Conclusions : These results suggest that CH-HA at $ST_{36}$ has an effect in controlling immune reaction and suppressing inflammation in rheumatoid arthritis therefore, the continuous flow of the following study is expected.

  • Perichondrium is generally used for reconstruction of airway and successful regeneration of cartilage framework using perichondrium are reported by several authors. It has many advantages for airway reconstruction. It can maintain the stable framework and it has higher flexibility so it's easy to design according to the shape of defects. It resist strongly against infection process. Its airtightness and rapid mucosalization enables to predict good postoperative recovery and results. To investigate the differences in cartilage regeneration between avascular and vascularized perichondrial flap, this study was designed with vascularized flaps composed of vascularized perichondrium and central auricular artery and vein. Morphologic study was performed to determine the fate of the grafted perichondrium at regular intervals using light microscopy with H & E stain. Chondrogenic potential and formation of cartilaginous plate of experimental group was superior than in the control group. Grafted perichondrium was fed by consistent feeding vessel. At 6 weeks, the regenerated cartilage could hardly be distinguished form the normal cartilage framework but in control group, regenerated cartilaginous tissue was generally immature in same period. In conclusion, this study indicated that consistent vasculature to grafed tissue is the essential factor for successful reconstruction of cartilaginous framework.

  • The authors have studied cytomorphologically on the gian cells appearing in the giant cell lesions which had been collected from the biopsies at the department of oral pathology, college of dentistry, Seoul National University. The results are as follows: 1. We can classify two types of giant cells and large sized giant cells which have foamy, large nuclei and prominent nucleoli, and small sized giant cells which have small, round and homogenous stained cytoplasm. 2. We can not see the phagocytosed materials, only can see the foamy microvacuoles in the cytoplasm of giant cells. 3. We can not see the mitotic figures, nore fused figures in giant cells. 4. Osteoclasts in the periphery of bone tissue reveal large and pale stained with H-E stain, giant cells on the granulation tissue and chronic inflammatory tissue reveal deeply stained with hematoxylin and prominent nuclei, but smaller than osteoclasts.

  • Benign tracheal tumors are less common than malignant tumors, but they are relatively more important because they are operable and curable. Neurilemmoma is originated from Schwann cells of nerve sheath, which are characterized by benign, solitary, and encapsulated mass. Tracheal neurilemmomas are extremely rare and reported only 21 cases in the world literature. Recently, we experienced a case of neurilemmoma which arose from the trachea of a 48-year-old female patient who complained of progressive dyspnea. The tumor mass was removed successfully through bronchoscopic and tracheal fissure approach. The final pathological diagnosis viewed under a microscope after H&E stain was a neurilemmoma in which Antoni type A and type B both existed.

  • An electrospun Poly (lactice-co-glycolide acid) (PLGA) and Gelatin nanofiber tube was fabricated for potential intestinal stent application. Mechanical properties of tube were evaluated by tensile strength and burst strength tests. Physical and chemical properties were evaluated by contact angle measurement, swelling rates and porosity measurements. Biodegradability was investigated by immersion in simulated body fluid (SBF). Biocompatibility was investigated in vitro by cytotoxicity and proliferation studies by MTT assay, confocal microscopy and western blot using IEC-18 (Rat intestinal epithelial cell). After intestinal stent was implanted into rat bowel for periods from 7 to 10days, it was then analyzed using micro-computed tomography (Micro CT) and X-ray techniques. Futhermore, histological analysis was performed by hematoxylin-eosin (H&E) stain.

A Comparative Study About the Effect of Nonablative Laser and Thermal energy on Rabbit Knee Joint Capsular Properties (비융해성 LASER 및 열에너지의 가토 슬관절막에 대한 영향 비교)

  • Purpose: To evaluate the histologic effect(LM and EM findings)of nonablative LASER and thermal energy on knee joint capsule of rabbit. Material and Methods: The nonablative LASER and thermal energy was applied to the rabits(average age 36 weeks, weight 5 ㎏). There were divided into 4 groups with 6 rabbits in each. The group I received 6 watts of LASER, group Ⅱ 12 watts of LASER, group Ⅲ 60° of thermal energy, and group Ⅳ 70° of thermal energy. The histologic study included H-E, Massons trichrome stain and electron microscopy at immediate, 3 weeks and 6 weeks after operation. Results: The histologic finding in immediate after operation was shown a fibrous degeneration of collagen on all groups and related to the energy level. The histologic finding after 3 weeks showed fibrosis and this fibrosis related the level of energy. Especially the group IV was shown flattening of capsule and deep fibrosis. The histologic finding after 6 weeks was shown marked recovery of collagen arrangement and capillary proliferation in group Ⅰ,Ⅱ and Ⅲ. But in the group Ⅳ not recovered. Conclusion: The nonablative LASER or thermal energy can cause degeneration, fibrosis and contracture of joint capsular collagen.

Apoptosis in experimentally infected chicks with Salmonella gallinarum

  • This experiment was performed to investigate apoptosis during undergoing patho-genesis of Salmonella gallinarum(SG)-infected chicks. 16 days old, 49 chicks was infected with SG (10$\^$6/-10$\^$8/ CFU/㎖) experimentally, they were autopsied to remove liver, spleen, intestine and lung at 1, 6, 12hr, 1, 2, 4 and 7 day post infection(PI) respectively, for H-E and TUNEL staining. Grossly, white foci in the liver and enlarged spleen were seen on 4 day PI and coppery bronze liver, dark-red discolored intestine, green-yellowish discolored and enlarged spleen was observed on 7 day PI. Histopathologically, multi focal necrosis in the liver, follicle hyperplasia in the spleen and inflammatory cells infiltration in the intestine were shown from 2 day PI and more severely observed on 4 day and 7 day PI. In TUNEL analysis, apoptotic cells reached a maximum at 6hr PI in the liver and intestine and at 12hr PI in the spleen, and then decreased the levels of controls by 7 day PI.

FREE SKIN GRAFTING WITH FIBRIN ABHESIVE - CLINICAL AND HISTOPATHOLOGIC REVIEWS - (조직 접착제를 이용한 유리 피부 이식술 - 임상적, 조직병리학적 고찰 -)

  • A fibrin adhesive have been widely used in oral and maxillofacial surgery for microvascular anastomosis, autogenous chip bone grafts, many kinds of soft tissue surgery (vestibuloplasty, bleeding control after extraction, primary healing by covering of suture of a gum after the extirpation of large cysts). There are two principal components in adhesive systems biologically: lyophilized human fibrinogen and bovine thrombin. The fibrinogen component contains coagulation factor XIII and enhance the initial wound healing, which polymerizes soluble fibrin monomers into an insoluble clot. The thrombin is dissolved in a solution of calcium chloride to provide the second component. We applied fibrin adhesive, Beriplast (Behring, Behringwerke AG, D-3350, Marburg, FRD), to 4 patients for fixation of free skin grafting donors who had facial scar around eye, nose, mouth corner which received from accidents, or burn. We have experienced initial accelerated graft fixation between donor and recipient sites with no additional fixation. And It's made easy bleeding control and easy manipulation during operation. But two cases showed partial hypertrophic scar engrowth in above 3 months follow up, but no significant. Histopathological reviews in general were showed similar scar findings such as abundant collagen bundles in H&E, M/T stain, but slight positive signs in elastic and collagen antibody immunopathologic findings in hypertrophic scar cases.

  • The purpose of this study was to investigate the effects of irradiation on the rat submandibular gland which composed of the epithelial growth factors. The experimental animals were the Sprague Dawley strain rats, which were the rats as the non-irradiated control group and the rats as the experimental group which were divided into groups as the experimental duration of 1 hour, 3 hours, 6 hours, 12 hours, 1 day, 3 days, 1 week, 2 weeks, 4 weeks. The experimental animals were singly irradiated at a dose of 8 Gray gamma ray to their head and neck region by the Co-60 teletherapy unit and sacrificed after each experimental duration. The specimens were stained with H-E and Azan stain and examined light microscopically. The results of this study were obtained as follows. 1. The all of mucous and serous acini in submandibular gland showed similar pattern of changes in structure according to the lapse of time. 2. The acinic cells started to change after 1 hour, and repaired after 12hours with mitosis and proliferation of the cells between acini. The changes were marked after 1day, and repaired gradually in course of time. 3. The duct were dilatated irregularly, and the outline of the eosinophilic stained ductal cells changed indistinctly.

EXPERIMENTAL STUDY ON THE EFFECTS OF CYCLOPHOSPHAMIDE ON THE HEALING OF THE ORAL INCISED WOUND (항암제 Cyclophosphamide가 구강창상 치유에 미치는 영향에 관한 실험적 연구)

  • To assure that cyclophosphamide has been considered something that must be effective to retard wound healing, the Author has studied clinically and histopathologically. All rabbits were incised on the mucogingival junction of anterior teeth in mandible about 1 Cm. in length with mucosa and periosteum layer. The 21 rabbits within 6 months old, weighing about 1.5-1.8 kg were divided into a experimental group and a control group. 1) Group 1 (Experimental group)-After 14 rabbits dieted as the control group were injected with Endoxan (=Cyclophosphamide) in dosage of 30mg per 1kg-body weight intramuscularly, the surgery was performed. 2) Group 2 (Control group)-the surgery in this group was performed without Endoxan. The rabbits were sacrificed on the 1st, 3rd, 5th, 7th, 10the, 14th day after surgery and microscopic slides were made with H-E stain. The results might be summarized as follows; 1) Cyclophosphamide effected to retard healing process on the incised wound of oral mucosa and connective tissue. 2) Control wounds were healed by the 7th day after surgery. 3) Comparison between control and experimental wound did not show significant differences 14th day after surgery.
