Several methods have been used in the detection of Helicobacter pylori (H. pylori) which was believed to be a pathogenic organism causing chronic gastritis, benign peptic ulcer, gastric carcinoma or malignant lymphoma. Even though several methods were introduced for detection of H. pylori in stomach, there were controversies in their sensitivities and specificities. This experiment were designed to study the comparative analysis of staining methods (hematoxylin and eosin (H&E), Giemsa, Warthin-Starry and immunohistochemical stain) to dectect H. pylori in the gastric mucosa. The results were as follows. Average density score of H. pylori classified by Genta were 2.29 in Warthin-Starry stain, 2.19 in Giemsa stain, 1.34 in immunohistochemical stain and 0.98 in H&E stain. By comparison between inflammatory degree by Sydney system and result of Warthin-Starry stain, the detection rate and densities of H. pylori were increased from mild (61.5% and 0.8), moderate (90.4 and 2.1), and severe (100% and 3.2). From the above findings, Warthin-Starry stain is useful method for detection of H. pylori in gastric mucosa.
This study is a report about a specific patient whose primary stomach adenocarcinoma metastasized to uterine cervix adenocarcinoma. A thirty-nine year old female patient was initially diagnosed as having metastatic adenocarcinoma in the supraclavicular lymph node. Upon further examination, she was diagnosed with stomach adenocarcinoma. 8 months later, a cervix punch biopsy was performed. The stains used for examination were H&E stain, PAS stain, Alcian blue stain, Mucicarmine stain, Papanicolaou's (Pap.) stain, and as immunohistochemical stains, cytokeratin 7 and 20 were done. In the H&E stain, the tumor cells showed prominent and eccentric nuclei, thin nuclear membrane in abundant mucous cytoplasm, and cylinder shape. In the PAS stain, intracytoplasmic mucin vacuoles were stained with pink, and in Alcian blue and Mucicarmine stains, intracytoplasmic mucin vacuoles were stained with blue and red. As in the above results, she was diagnosed with undifferentiated adenocarcinoma. As found on the cytologic smear preparation of the uterine cervix stained by Papanicolaou's stains, the background was relatively clear, the number of malignant cells was relatively low, and large and eccentric nuclei in abundant cytoplasm were observed. Upon observing the tissue preparation of the uterine cervix biopsy by H&E stain, a clear background, large and eccentric nuclei, and a signet ring cell types were observed, and the number of malignant cells were fewer than in the primary uterine cervix adenocarcinoma. The vacuoles in cytoplasm were observed. The nuclear membrane and chromatin were thick and very rough, and upon observation by cytokeratin 7 and 20 of immunohistochemical stain, the tumor cells indicated a positive rate of 70% and 20%, respectively. According to these results, also she was diagnosed with metastasized uterine cervix adenocarcinoma. In summary of the results of pathologic findings on stomach biopsy and cytologic, histopathologic, and immunohistochemical finding on uterine cervix biopsy, the adenocarcinoma of her uterine cervix could assert the adenocarcinoma of signet ring cell type that was metastasized from the primary undifferentiated adenocarcinoma in stomach.
The demonstrations of collagenous fiber in mandibul ar bone formation were analized histochemically, using Carson's Mallary-Heidenhein stain, McManus' PAS reaction and H-E stain The stain ablity of osteoid increased with formation of bone tissue. In the surrounding areas of osteoblast and osteocyte were showed slight activity.
Background: Modified toluidine blue staining (MTBs) is a simple, inexpensive and time saving method to detect H. pylori in gastric biopsy specimens. As a metachromatic stain, it simultaneously highlights intestinal metaplasia, a gastric cancer precancerous lesion. The aim of this study was to assess the reliability of MTBs compared with hematoxylin-eosin (H&E) for H. pylori detection using immunoperoxidase staining as the gold standard. This technique would be beneficial for a routine diagnosis and confirmation of H. pylori eradication in developing countries where endoscopic-based approaches are dominant. Materials and Methods: Esophagogastroduodenoscopy with triple site gastric biopsies was undertaken in 207 dyspeptic patients at Thammasat University Hospital, Thailand between 1997 and 1999. H&E, MTBs and immunoperoxidase staining were applied to each specimen. The presence or absence of H. pylori with each stain was interpreted separately and the sensitivity, specificity, positive and negative predictive values of H&E and MTBs were calculated. Results: A total of 282 specimens from 207 patients were evaluated. Using immunoperoxidase staining, organisms were positive in 117 specimens (41%). MTBs proved almost equally sensitive as immunoperoxidase (99%) and significantly more sensitive than H&E (85%). It has comparable specificity (96% vs 96%), PPV (95% vs 94%), and NPV (99% vs 90%) to H&E, using immunoperoxidase staining as gold standard. MTBs compared with immunoperoxidase staining, is cheaper (2 USD vs 12 USD) and faster (20 min vs 16 hrs) compared to immunoperoxidase staining. Conclusions: MTBs is effective, economical and easy to use in daily practice for the detection of H. pylori in gastric biopsy specimens. In addition to saving time in evaluating H. pylori associated gastritis, with a high sensitivity and ability to demonstrate intestinal metaplasia, the technique may have a role in confirmation of H. pylori eradication for gastric cancer prevention in a developing country setting.
Discoloration sensitivities of woods grown in this country haven't reported yet. Therefore we examined discoloration sensitivities of domestic wood specimens to iron (0.1 %, $FeCl_3.6H_2O$), alkali (pH 12.0, NaOH). acid (pH 1.0, $C_2H_2O_4$) and exposing to sunlight (40 hrs), Thirty-six hardwood species were collected and examined. All specimens were prepared from heartwoods of the collected species. But the specimens of 4 Betula species were divided into sapwoods and heartwoods. By iron stain, the color differences (${\Delta}E$) of 21 wood specimens including one Betula sapwood showed above 12.0, which means strong discoloration sensitivities, and of 3 specimens including one Betula sapwood showed below 2.5, which means weak discolorations. The most strong iron discoloration species was Jungkukgulpi-namu (Pterocarya stenoptera). By alkali stain, the color differences (${\Delta}E$) of 3 wood specimens showed above 9.0, which means strong discoloration sensitivities, and of 18 wood specimens including 4 Berula sapwoods showed below 2.5, which means weak discolorations. By acid stain, the color differences (${\Delta}E$) of 6 wood specimens showed above 10.0 which means strong discoloration sensitivities, and of 12 wood specimens including one Betula sapwoods showed below 2.5, which means weak discolorations. By exposing to sunlight, the color differences (${\Delta}E$) of 31 wood specimens including one Betula sapwoods showed below 6.5, which means, strong discoloration sensitivities, and of only one specimens showed below 2.5, which means weak discoloration. The most strong discoloration species by exposing to sunlight was Guirung-namu (Prunus padus). In general, it was shown that hardwoods grown in Korea were most subject to change of color by exposing to sunlight and next were by iron stain. Domestic hardwoods showed some differences in discoloration sensitivities from domestic softwoods previously reported.
Diabetes mellitus revealed a chronic disorder of lipid, carbohydrate and protein metabolism characterized by insulin deficiency, and a striking tendency toward development of atherosclerosis, microangiopathy, nephropathy, neuropathy and recently cardiomyopathy etc. The mechanism of heart failure in patients with diabetic cardiomyopathy is not clear but diabetic cardiomyopathy usually occurs in persons with long standing diabetes. After diabetes induced in made Sprague- Dawley strain rats by injection of streptozotocin(60mg/kg), cardiac tissue with hematoxylin-eosin and Masson's trichrome stain was examined at 3 days, 1, 2, 4, 6 weeks later under light microscope. The results were obtained as follows : 1. In H&E stain of control group, myocardiac cells were shorter than skeletal muscle cell, which was branched out and connected each other at terminal with striation, intercalated disk and nucleus at center of cell. 2. In MT stain of control group, a few of collagen fibrile were seen at periva scular interstium, but wasn't seen between skeletal muscle fiber, and cardiac muscle was seen in various size. 3. In MT stain of experimental group, increased collagen fiber deposition at perivascular interstiums were seen periodically. 4. In MT stain of experimental group, increased collagen fiber deposition at interstitial matrix between perimyocardiac cells were seen at 3 day, 4 weeks and 6 weeks after DM induction. 5. In H&E stain of experimental group, partial degeneration of myocardiac cells was seen after 4 weeks of DM induction. From above results, streptozotocin induced diabetes mellitus increased collagen around perivascular and between intercellular matrix in heart.
Objectives: Burn is a severe dermal injury caused by heat. We were to investigated the effects of acupuncture or pharmacopunture treatment for the histologic and morphologic recovery on the mouse with the 3rd grade burn skin. Methods : We divided into 3 groups. One was a control group(n=3) that was not treated any treatments. Another was a acupuncture group(n=3) that was treated only acupuncture. The other was a pharmacopuncture group(n=3) that was treated only pharmacopuncture. We made a 3rd grade burned skin with the stainless steel heating apparatus. We made a treatment for the mice for a week(2 times for a day, totally 14 times treatments). We observed a dermal morphologic recovery on the mice and a histopathological photographs of the burn skin and subcutaneous tissue with H&E stain, Masson's trichrome stain, and VEGF, FGF and c-kit immunohistochemical stain. Results : The pharmacopuncture group were a better morphologic recovery than control group and acupuncture group. And the pharmacopuncture group were a better histopathological recovery than control group and acupuncture group on the burn skin and subcutaneous tissue with H&E stain, Masson's trichrome stain. And the pharmacopuncture group were a better histopathological recovery than control group and acupuncture group on the burn skin and subcutaneous tissue with VEGF, FGF and c-kit immunohistochemical stain. Conclusions : We suggest that the pharmacopuncture treatment is a better histologic and morphologic recovery than the no treatment or the acupuncture group with the burned mouse skin.
An experimental study was performed to study the tissue reactions to tissue conditioners in rabbits. Thirty rabbits were divided into three groups, 10, Hydro-cast group, 10, Coe-comfort group, and 10, heat-cured resin group. Tissue conditioners and heat-cured resin were embedded in the oral mucosa of rabbits. The tissue specimens were removed on 3rd, 7th, 14h, 21st and 28th day after embedding and examined under microscope after staining them with H-E stain, Van Gieson's stain, Masson's trichrome stain and PAS reaction. The results were as follows :
1. Tissue reactions to tissue conditioners were somewhat different from each other in the early stage, but, with the increase of the embedding period, the fibrous capsule was thickened in both. These tissue reactions were similar to those to heat-cured resin.
2. Newly formd fibrous components were stained deep-red with Van Gieson's stain and dark-green with Masson's trichrome stain. But their stainability was decreased as collagenous fibers became matured.
3. Newly formed fibrous components showed intense PAS reactivity, but PAS reactivity was reduced as the connective tissue capsule became completed.

The purpose of this study was to establish a method for in vitro culture of C parvum isolated in Korea by determination of suitable cell model to complete development of this parasite. The result obtained were summerized as follows: 1. To determine the most suitable cell line, six types of cell line were examined by microscopy. All cell lines were infected with C parvum and showed the highest infection score in HmLu cells. 2. The staining methods including DMSO-modified acid-fast(A-F) stain, hematoxylin-eosin(H & E) stain and immunofluorescence antibody(IFA) stain were applied to examine the infection of C parvum in cell culture. These staining methods were possible to examine the infection of C parvum in cell culture. The most sensitive one was IFA staining technique. 3. Developmental stages of C parvum in HmLu cell were observed. After the initial 8 hour incubation period, some trophozoites were observed. The meronts and gametes were appeared at 24-48 hour post inoculation(PI), and oocysts were observed firstly at 48-72 hour PI. 4. In H & E stain, the parasite appeared as basophilic within parasitophorous vacuole membrane(PVM) and lying in cytoplasm at near the nucleus of the host cells. It was able to distinguish the type I, type II meronts and gametes. 5. In DMSO-modified acid-fast stain, specific stained parasites were appeared firstly after 48 hour PI. The parasites were showed with different degrees of staining bright red color within PVM. 6. The endogenous stages of parasites in HmLu cell recovered at 48, 96, 120 and 144 hour after inoculation were reacted with rabbit immunized serum in immunofluorescence antibody and avidin-biotin complex peroxidase staining technique.
The purpose of this study was to evaluate the effect of orthodontic force on periodontal cellular activity by immunoperoxidase stain of epidermal growth factor, one of the tissue hormone. And supplementarily, to investigate of the changes of periodontal structures, periodontium was stained by H-E, Masson's Trichrome, P. A. S. stain after orthodontic force application. The experimental animals were four young adult dogs of average 8 month old. The fixed orthodontic appliance was cemented on mandibular right 4th premolar and 1st molar of each animal as experimental site. Mandibular left 4th premolar area of the same animal was used as control. The appliance consist of two silver crown soldered with 0.030' tube, $0.018\times0.022'$ S.S. sectional arch wire, and 0.009' open coil spring for manifestating of orthodontic force for bodily tooth movement of mandibular 4th premolar toward mesial direction. Experimental group was sacrificed at 1, 2, 3, 5 weeks from beginning of the experiment, and was investigated immunohistochemically and bistochemically by several staining methods. Findings were as follows: 1. The degree of EGF staining in control group was highest in epithelium of periodontium, and osteoclasts, osteoblasts and fibroblasts around the capillary were stained at higher level in periodontium. Generally, control group shows positive distribution of EGF all around the periodontal area. 2. The degree of EGF staining in control and 5 week group were similar, and did not show the significant different level between tension and pressure side. 3. All of 1, 2, 3 week group showed the same staining degree and distribution of EGF, and the tension side was more positive reaction of EGF stain than the pressure side. 4. The features of collagen fiber and periodontal fiber arrangement observed by H-E, Masson's Trichrome and P. A. S. stain revealed that oblique periodontal fibers were strectched in tension side, compressed in pressure side of all experimental group. Some fiber group in pressure side of 5 week group recovered the regular arrangement along the capillaries.
