The Relative Effectiveness of Liquid Methionine Hydroxy Analogue Compared to DL-methionine in Broilers

  • The present experiment was conducted to assess the efficacy of liquid methionine hydroxy analogue-free acid (MHA-FA) in comparison to DL-methionine (DL-Met) in broilers. 567 day-old Avian chicks were divided into 7 treatments with 5 replicates of 16 birds each. During the 35d (7-42 d) experimental periods chicks were given two basal diets. From 7 to 21d of age, a starting basal diet containing 19.5% protein and 0.33% methionine was supplemented with two graded levels of DL-Met (0.070 and 0.160%) or four levels of MHA-FA (0.118, 0.143, 0.221 and 0.268%). From 22 to 42d of age DL-Met (0.050 and 0.080%) or MHA-FA (0.071, 0.074, 0.112 and 0.140%) were added to a finishing basal diet with 18.0% protein and 0.28% methionine. Chicks fed on supplemental DL-Met or MHA-FA had significantly higher (p<0.05) body weight gain and feed conversion ratio (FCR) than the control group from 7-21d of age. During the finishing phase (22-42 d), body weight and weight gain of chicks in DL-Met or MHA-FA treatments were similar to those in the control, but FCR was improved (p<0.05) with supplementation of DL-Met or MHA-FA. Breast yield was higher (p<0.05) on DL-Met or MHA-FA supplemented than un-supplemented diets. The thigh meat yields emanating from diets with DL-Met or MHA-FA were lower (p<0.05) than that in control. Abdominal fat was also higher in broilers fed the control diet than in DL-Met or MHA-FA supplemented treatments. Methionine requirement of broilers was calculated to be 0.44 and 0.35% and cystine requirement was 0.35 and 0.31% for the starting (7-21 d) and finishing phase (22-42 d), respectively. The efficacy of MHA-FA in comparison to DL-Met for weight gain was 64 and 85% and for FCR was 55 and 60% at 7-21 and 22-42 d of age, respectively, while it was 74, 72, 52 and 48% for breast yield, thigh meat production, body energy content and energy deposition ratio at 42 d of age, respectively. In conclusion, in practical diet formulation for broiler chicks the average bioavailability of MHA-FA relative to DL-Met could be considered as 60 and 73% for 7 to 21d and 22 to 42 d of age, respectively.

${Ni_{1+x}}{Ti_x}{Fe_{2-2x}}O_4$계의 $\M"{o}ssbauer$ 스펙트럼 연구 (A Study on Mossbauer Spectra of the ${Ni_{1+x}}{Ti_x}{Fe_{2-2x}}O_4$ System)

  • $Ni_{1+x}/Ti_{x}Fe_{2-2x}O_4$계 시료중 스피넬의 단일상만 존재하는 x값이 0~0.7인 시료에 대한 Mossbauer 스펙트럼을 측정 분석하였다. $0{\leqq}x{\leqq}0.3$인 시료에서는 두 site의 $Fe^{3+}$ 이온에 의한 두 개의 6선 흡수선이 나타났고, $0.4{\leqq}x{\leqq}0.6$인 시료에서는 두개의 6선 흡수선과 2선 흡수선이 공존하였으며, x=0.7인 시료는 2선 흡수선만 나타났다. x값의 증가에 따라 6선 흡수선의 A-site 공명 흡수 면적이 B-site에 비해 상대적으로 커지고, 2선 홀수선의 공명 흡수면적도 6선 흡수선에 비해 커지는 것을 알 수 있었다. Mossbauer parameter중 이성질체 이동치 (I.S.)는 A-site의 경우에는 x값의 증가에 따라 커지는 반면 B-site는 큰 영향을 주지 않았고, 사중극자 분열치 (Q.S.)의 변화는 볼 수 없었으며, 두 Site의 미세자기장 ($H_{hf}$)은 X값의 증가에 따라 모두 감소하였다. 이러한 실험 결과로부터 시료의 금속 양이온 분포식을 구하였고, X값의 증가는 A-Site의 $Fe^{3+}-O^{2-}$ 결합의 공유 결합성을 약화시키는 반면 B-Site의 $Fe^{3+}O^{2-}$ 결합에는 큰 영향을 주지 않으며, 또한 X값의 증가에 따라 $Fe^{3+}\;_A-O^{2-}\;-Fe^{3+}\;_B$ 결합의 수가 감소하여 A-B 초교환 상호 작용이 약화되는 것을 알 수 있었다.

3${\beta}$-Hydroxy-12-oleanen-28-oic Acid 유도체들의 PTP-1B저해활성에 대한 CoMSIA분석 (CoMSIA Analysis on The Inhibition Activity of PTP-1B with 3${\beta}$-Hydroxy-12-oleanen-28-oic Acid Analogues)

  • 기질 화합물로써 3${\beta}$-Hydroxy-12-oleanen-28-oic acid 유도체(1-30)들과 그들의 protein tyrosine phosphatase(PTP)-1B 저해활성에 관한 비교분자 유사성 지수분석(CoMSIA)보델을 유도하였다. QSAR 모델의 통계 값은 CoMFA>CoMSIA${\geq}$HQSAR>2D-QSAR 모델의 순서로 양호하였다. 최적화된 CoMSIA F1 모델은 grid 3.0${\AA}$과 field fit 정렬조건에서 가장 족은 예측성과 상관성($r^2_{cf}$=0.754 및 $r^2_{ncv}$=0.976)을 나타내었다. 저해 활성에 관한 CoMSIA상의 기여비율(%)은 수소결합 받게장(48.9%), 입체장(25.8%) 및 소수성장(25.4%)의 순서이었다. 그러므로 기질 화합물의 PTP-1B에 대한 저해활성은 $R_4$-치환기의 수소결합 받게 장(A)에 의존적이었다. 등고도 분석 결과로부터 $R_1$-치환기는 수소결합 받게장이 작고 $R_3$-치환기는 입체장이 작으며 그리고 $R_4$-치환기는 수소결합 받게장, 소수성 및 입체장이 큰 치환기가 저해활성을 증가시킬 것으로 예측되었다.

Effects of Anion Supplementation on Growth Performance, Nutrient Digestibility, Meat Quality and Fecal Noxious Gas Content in Growing-finishing Pigs

  • Forty-eight ((Duroc${\times}$Yorkshire)${\times}$Landrace) pigs with an average initial body weight (BW) of $48.47{\pm}1.13\;kg$ were used in a 12-week growth trial to investigate the influence of Anion (silicate) supplementation on growth performance, nutrient digestibility, meat quality and fecal noxious gas content in growing-finishing pigs. Pigs were allotted into three dietary treatments in a randomized complete block design according to sex and initial BW. Each dietary treatment consisted of four replications with four pigs per pen. Dietary treatments included: i) CON (basal diet), ii) HCI (basal diet+3 g/kg Anion), iii) HCII (basal diet+6 g/kg Anion). No significant difference (p>0.05) was detected for average daily gain (ADG), average daily feed intake (ADFI) and gain/feed ratio (G/F) throughout the experiment, although dietary supplementation of Anion numerically increased these characteristics compared with CON. The dietary HCI group significantly (p<0.05) increased the coefficient of total tract apparent digestibility (CTTAD) of dry matter (DM), nitrogen (N) and energy compared with the CON group (p<0.05). No significant difference was observed in meat quality except that meat firmness was linearly (p<0.05) increased by the Anion supplementation, while an increased tendency in meat color and a decreased tendency in 2-thiobarbituric acid reactive substances (TBARS) was also observed (p<0.10). Anion supplementation linearly (p<0.05) decreased the fecal $NH_3$ compared with the CON group. However, dietary Anion supplementation at 3 g/kg decreased the $H_2S$ concentration compared with CON, while no significant difference was detected in the HCII group, although the $H_2S$ emission was numerically decreased compared with CON. In conclusion, supplementation of the diet with 3 g/kg Anion was found to exert a beneficial effect on nutrient digestibility and meat quality of growing-finishing pigs, and concomitantly decreased the noxious gas emission without negative effect on growth performance.

The First Photometric Study of NSVS 1461538: A New W-subtype Contact Binary with a Low Mass Ratio and Moderate Fill-out Factor

  • New multiband BVRI light curves of NSVS 1461538 were obtained as a byproduct during the photometric observations of our program star PV Cas for three years from 2011 to 2013. The light curves indicate characteristics of a typical W-subtype W UMa eclipsing system, displaying a flat bottom at primary eclipse and the O'Connell effect, rather than those of an Algol/b Lyrae eclipsing variable classified by the northern sky variability survey (NSVS). A total of 35 times of minimum lights were determined from our observations (20 timings) and the SuperWASP measurements (15 ones). A period study with all the timings shows that the orbital period may vary in a sinusoidal manner with a period of about 5.6 yr and a small semi-amplitude of about 0.008 day. The cyclical period variation can be interpreted as a light-time effect due to a tertiary body with a minimum mass of 0.71 M. Simultaneous analysis of the multiband light curves using the 2003 version of the Wilson-Devinney binary model shows that NSVS 1461538 is a genuine W-subtype W UMa contact binary with the hotter primary component being less massive and the system shows a low mass ratio of q(mc/mh)=3.51, a high orbital inclination of 88.7°, a moderate fill-out factor of 30 %, and a temperature difference of ΔT=412 K. The O'Connell effect can be similarly explained by cool spots on either the hotter primary star or the cool secondary star. A small third-light corresponding to about 5 % and 2 % of the total systemic light in the B and V bandpasses, respectively, supports the third-body hypothesis proposed by the period study. Preliminary absolute dimensions of the system were derived and used to look into its evolutionary status with other W UMa binaries in the mass-radius and mass-luminosity diagrams. A possible evolution scenario of the system was also discussed in the context of the mass vs mass ratio diagram.

Effects of Dietary Thiazolidinedione Supplementation on Growth Performance, Intramuscular Fat and Related Genes mRNA Abundance in the Longissimus Dorsi Muscle of Finishing Pigs

  • The objective of this study was to investigate the effect of dietary supplementation with thiazolidinedione (TZD) on growth performance and meat quality of finishing pigs. In Experiment 1, 80 castrated finishing pigs (Large White${\times}$Landrace, BW = 54.34 kg) were randomly assigned to 2 treatments with 5 replicates of 8 pigs each. The experimental pigs in the 2 groups were respectively fed with a diet with or without a TZD supplementation (15 mg/kg). In Experiment 2, 80 castrated finishing pigs (Large White${\times}$Landrace, BW = 71.46 kg) were divided into 2 treatments as designed in Experiment 1, moreover, carcass evaluations were performed. The results from Experiment 1 showed that TZD supplementation could significantly decreased the average daily feed intake (ADFI) (p<0.05) during 0 to 28 d, without impairing the average daily gain (ADG) (p>0.05). In Experiment 2, the ADG was significantly increased by TZD supplementation during 14 to 28 d and 0 to 28 d (p<0.05) and the feed:gain ratio (F:G) was significantly decreased by TZD supplementation during 0 to 28 d (p<0.05). Compared with the control group, TZD group had significantly higher serum triglyceride (TG) concentration at 28h and serum high-density lipoprotein (HDL) levels at 14 d (p<0.05). Moreover, there was an apparent improvement in the marbling score (p<0.10) and intramuscular fat (IMF) content (p<0.10) of the longissimus dorsi muscle in pigs treated by TZD supplementation. Real-time RT-PCR analyses demonstrated that pigs of TZD group had higher mRNA abundance of $PPAR{\gamma}$ coactivator 1 (PGC-1) (p<0.05) and fatty acid-binding protein 3 (FABP3) (p<0.05) than pigs of control group. Taken together, these results suggested that dietary TZD supplementation could improve growth performance and increase the IMF content of finishing pigs through regulating the serum parameters and genes mRNA abundance involved in fat metabolism.

SO2 노출된 4개 수종의 엽내 광색소 함량 및 SOD 활성 변화 (Changes of Photosynthetic Pigment Contents and SOD Activity in the Leaves of Four Tree Species Exposed to SO2)

  • 목본 식물을 대상으로 SO$_2$에 의해 나타나는 생리적인 반응을 구명하기 위하여 소나무, 오갈피나무, 현사시, 상수리나무를 대상으로 SO$_2$를 500 ppb, 800 ppb로 하루에 8시간씩 7일간 처리하여 잎의 광색소 함량과 SOD 활성을 비교 분석한 결과는 다음과 같았다. SO$_2$의 처리 농도가 증가할수록 4개 수목의 엽 내 엽록소 함량은 감소하였으며, 엽록소 a와 엽록소 b, 카로티노이드 함량의 변화는 수종별, 처리별로 다른 경향을 보였다. 오갈피나무와 상수리나무의 엽록소 b와 a의 비는 SO$_2$는 500 ppb 처리구에서는 증가하다가 800ppb 처리구에서는 감소하였다. 즉 500ppb에서는 엽록소 a가 파괴되며, 800ppb에서는 엽록소 b도 파괴될 수 있음을 보여 주었으며, SO$_2$에 대한 민감성은 엽록소 a가 엽록소 b보다 높은 것으로 나타났다. 4개 수종의 잎 SOD활성은 수종별, 처리별 큰 차이를 나타냈다. 오갈피나무와 상수리나무는 500 ppb 처리구에서는 SOD 활성이 증가하다가 더 높은 농도에서는 활성이 감소하였으며, 소나무와 현사시의 경우는 500ppb 처리와 800ppb 처리에서 높은 SOD활성을 유지하여 내성을 보인다. 그러나 광색소와 SOD 활성을 기준으로 판단해 볼 때, SO$_2$에 대한 저항성은 현사시가 가장 높은 것으로 판단된다.

Forsythia suspensa Extract Has the Potential to Substitute Antibiotic in Broiler Chicken

  • Two experiments were conducted to investigate the potential for Forsythia suspensa extract (FSE) to substitute for antibiotic in broiler chicken. First, a well-diffusion assay procedure and a 2-fold dilution method were used to determine the bacteriostatic activity of FSE on Escherichia coli K88, staphylococcus aureus, and salmonella was assayed. An inhibitory effect of FSE was observed on the growth of these bacteria. This effect seems to be dose depended, which disappeared after 25.00, 12.50, 1.56 mg/ml. Second, a 42-d trial with 252 broiler chickens (d 1, $38.7{\pm}1.1$ g BW) was conducted to evaluate the effect of dietary supplementation of FSE in broiler chicken. The feeding program consisted of a starter diet from d 1 to 21 and a finisher diet from d 22 to 42. Dietary treatments included were: i) NC: negative control fed a corn-soybean meal based diet; ii) PC: positive control group fed based diet with chlortetracycline; and iii) FC: a test group fed with 100 mg FSE/kg diet. In this study growth performance did not differ among treatments during the starter period. However, dietary supplemental chlortetracycline and FSE increased (p<0.05) average daily gain (ADG), average daily feed intake (ADFI) compared with NC during the finisher and overall phase. Apparent digestibility of calcium on d 21, digestibility of energy and calcium on d 42 of FC was greater (p<0.05) than NC. Moreover, cecal Escherichia coli counts for birds from FC were lower (p<0.05) than NC. Dietary FSE supplementation also improved (p<0.05) villus height and villus height to crypt depth ratios in both duodenum and ileum and decreased (p<0.05) crypt depth in the duodenum. Duodenum villus height and villus height to crypt depth ratio in both duodenum and ileum from the FC group were also greater (p<0.05). Serum growth hormone and IGF-1 were not influenced by different treatments. Apparently, FSE has the potential to substitute for antibiotic in broiler chicken.

Deletion of the Lmna Gene Induces Growth Delay and Serum Biochemical Changes in C57BL/6 Mice

  • The A-type lamin deficient mouse line ($Lmna^{-/-}$) has become one of the most frequently used models for providing insights into many different aspects of A-type lamin function. To elucidate the function of Lmna in the growth and metabolism of mice, tissue growth and blood biochemistry were monitored in Lmna-deficient mice, heterozygous ($Lmna^{+/-}$) and wide-type ($Lmna^{+/+}$) backcrossed to C57BL/6 background. At 4 weeks after birth, the weight of various organs of the $Lmna^{-/-}$, $Lmna^{+/-}$ and $Lmna^{+/+}$ mice was measured. A panel of biochemical analyses consisting of 15 serological tests was examined. The results showed that Lmna deficient mice had significantly decreased body weight and increased the ratio of organ to body weight in most of tissues. Compared with $Lmna^{+/+}$ and $Lmna^{+/-}$ mice, $Lmna^{-/-}$ mice exhibited lower levels of ALP (alkaline phosphatase), Chol (cholesterol), CR (creatinine), GLU (glucose), HDL (high-density lipoprotein cholesterol) and higher levels of ALT (alanine aminotransferase) (p<0.05). $Lmna^{-/-}$ mice displayed higher AST (aspartate aminotransferase) values and lower LDL (lowdensity lipoprotein cholesterol), CK-MB (creatine kinase-MB) levels than $Lmna^{+/+}$ mice (p<0.05). There were no significant differences among the three groups of mice with respect to BUN (blood urea nitrogen), CK (creatine kinase), Cyc C (cystatin C), TP (total protein), TG (triacylglycerols) and UA (uric acid) levels (p>0.05). These changes of serological parameters may provide an experimental basis for the elucidation of Lmna gene functions.

Effects of Immune Stress on Performance Parameters, Intestinal Enzyme Activity and mRNA Expression of Intestinal Transporters in Broiler Chickens

  • Immune stress is the loss of immune homeostasis caused by external forces. The purpose of this experiment was to investigate the effects of immune stress on the growth performance, small intestinal enzymes and peristalsis rate, and mRNA expression of nutrient transporters in broiler chickens. Four hundred and thirty-two 1-d-old broilers (Cobb500) were randomly assigned to four groups for treatment; each group included nine cages with 12 birds per cage. Group 1 = no vaccine (NV); Group 2 = conventional vaccine (CV); group 3 = lipopolysaccharide (LPS)+conventional vaccine (LPS); group 4 = cyclophosphamide (CYP)+conventional vaccine (CYP). The results demonstrated that immune stress by LPS and CYP reduced body weight gain (BWG), feed intake (FI), small intestine peristalsis rate and sIgA content in small intestinal digesta (p<0.05). However, the feed conversion ratio (FCR) remained unchanged during the feeding period. LPS and CYP increased intestinal enzyme activity, relative expression of SGLT-1, CaBP-D28k and L-FABP mRNAs (p<0.05). LPS and CYP injection had a negative effect on the growth performance of healthy broiler chickens. The present study demonstrated that NV and CV could improve growth performance while enzyme activity in small intestine and relative expression of nutrient transporter mRNA of NV and CV were decreased in the conditions of a controlled rational feeding environment. It is generally recommended that broilers only need to be vaccinated for the diseases to which they might be exposed.
