Effect of feed restriction on the maintenance energy requirement of broiler breeders

  • Objective: This study aimed to evaluate the effect of the ad libitum and restricted feeding regimen on fasting heat production (FHP) and body composition. Methods: Twelve Hubbard broilers breeders were selected with the same body weight and submitted in two feeding regimes: Restricted (T1) with feed intake of 150 g/bird/d and ad libitum (T2). The birds were randomly distributed on the treatments in two runs with three replications per treatment (per run). The birds were adapted to the feed regimens for ten days. After that, they were allocated in the open-circuit chambers and kept for three days for adaptation. On the last day, oxygen consumption (VO2) and carbon dioxide production (VCO2) were measured by 30 h under fasting. The respiratory quotient (RQ) was calculated as the VCO2/VO2 ratio, and the heat production (HP) was obtained using the Brower equation (1985). The FHP was estimated throughout the plateau of HP 12 hours after the feed deprivation. The body composition was analyzed by dual-energy X-ray absorptiometry scanning at the end of each period. Data were analyzed for one-way analysis of variance using the Minitab software. Results: The daily feed intake was 30 g higher to T2 (p<0.01) than the T1. Also, the birds of the T2 had significatively (p<0.05) more oxygen consumption (+3.1 L/kg0.75/d) and CO2 production (+2.2 L/kg0.75/d). That resulted in a higher FHP 359±14 kJ/kg0.75/d for T2 than T1 296±17.23 kJ/kg0.75/d. In contrast, the RQ was not different between treatments, with an average of 0.77 for the fasting condition. In addition, protein and fat composition were not affected by the treatment, while a tendency (p<0.1) was shown to higher bone mineral content on the T1. Conclusion: The birds under ad libitum feeding had a higher maintenance energy requirement but their body composition was not affected compared to restricted feeding.

Antifungal and carboxylesterase-producing bacteria applied into corn silage still affected the fermented total mixed ration

  • Objective: This study investigated the effects of corn silage as a source of microbial inoculant containing antifungal and carboxylesterase-producing bacteria on fermentation, aerobic stability, and nutrient digestibility of fermented total mixed ration (FTMR) with different energy levels. Methods: Corn silage was used as a bacterial source by ensiling for 72 d with an inoculant mixture of Lactobacillus brevis 5M2 and L. buchneri 6M1 at a 1:1 ratio. The corn silage without or with inoculant (CON vs MIX) was mixed with the other ingredients to formulate for low and high energy diets (LOW vs HIGH) for Hanwoo steers. All diets were ensiled into 20 L mini silo (5 kg) for 40 d in quadruplicate. Results: The MIX diets had lower (p<0.05) acid detergent fiber with higher (p<0.05) in vitro digestibilities of dry matter and neutral detergent fiber compared to the CON diets. In terms of fermentation characteristics, the MIX diets had higher (p<0.05) acetate than the CON diets. The MIX diets had extended (p<0.05) lactic acid bacteria growth at 4 to 7 d of aerobic exposure and showed lower (p<0.05) yeast growth at 7 d of aerobic exposure than the CON diets. In terms of rumen fermentation, the MIX diets had higher (p<0.05) total fermentable fraction and total volatile fatty acid, with lower (p<0.05) pH than those of CON diets. The interaction (p = 0.036) between inoculant and diet level was only found in the immediately fermentable fraction, which inoculant was only effective on LOW diets. Conclusion: Application of corn silage with inoculant on FTMR presented an antifungal effect by inhibiting yeast at aerobic exposure and a carboxylesterase effect by improving nutrient digestibility. It also indicated that fermented feedstuffs could be used as microbial source for FTMR. Generally, the interaction between inoculant and diet level had less effect on this FTMR study.

양배추즙 및 양배추 혼합즙의 인체위암세포(AGS) 성장 억제효과와 HCl-Ethanol로 유발된 흰쥐의 항위염 효과 (Inhibitory Effects of Cabbage Juice and Cabbage-Mixed Juice on the Growth of AGS Human Gastric Cancer Cells and on HCl-Ethanol Induced Gastritis in Rats)

  • 본 연구에서는 양배추즙과 양배추 혼합즙의 항위염 효과를 검토하고자 in vitro에서 AGS 인체위암세포의 성장 억제율, 염증인자 발현, 항산화력, 제산력 결과를 바탕으로 SD-rat에 HCl-ethanol로 급성위염을 유도하여 이에 대한 위 손상 억제효과를 평가하였다. 인체위암세포(AGS) 성장 억제율을 조사한 결과 클로렐라(Ch)가 20%, 케일(K)이 21%, 양배추(C) 42% 순으로 성장 억제율이 상승하였고, 양배추에 클로렐라나 케일을 7:3 비율로 혼합하였을 때 양배추-클로렐라(CCh) 57%, 양배추-케일(CK) 65%로 성장 억제율이 상승하는 경향을 나타내었다. 염증인자 발현 또한 양배추(C) 단일로 처리했을 때보다 양배추-클로렐라 혼합(CCh), 양배추-케일 혼합(CK)에서 염증인자 발현이 유의적으로 감소되는 경향을 나타냈다(p<0.05). DPPH radical 소거능에서도 2 mg 농도에서 양배추(C) 61.7%, 양배추-클로렐라(CCh) 87.3%, 양배추-케일(CK) 88.1%로 높은 항산화력을 나타냈으며, 제산력 실험에서도 양배추 고농도(CH) 34.8%, 양배추-클로렐라 고농도(CChH) 40.1%, 양배추-케일 고농도(CKH) 44.9%로 양성대조약물인 cimetidine 41.8%와 비슷하거나 더 높은 제산력을 나타냈다. 이를 바탕으로 HCl-ethanol 위염모델에서 위 손상에 대한 영향을 평가한 결과, 양배추를 기본으로 한 모든 저농도(CL, CChL, CKL), 고농도(CH, CChH, CKH) 투여군에서 보호 효과가 나타났으며 양배추 단일 고농도(CH) 46%보다 클로렐라와 케일을 혼합한 양배추-클로렐라 고농도(CChH) 71%, 양배추-케일 고농도(CKH) 74%에서 그 보호 효과가 유의적으로 높아져 양성 대조약물과 비슷한 보호 효과를 나타내었다(p<0.05). 위액분비량과 위액 pH 또한 모든 투여군에서 보호 효과가 유의적으로 나타났으며(p<0.05), 특히 양배추-케일 혼합즙(CKH)이 대조약물보다 더 높은 보호 효과를 나타내었다. 이상의 결과에서 양배추즙에 클로렐라나 케일을 7:3 비율로 혼합하였을 때 위염예방효과가 시판 대조약물만큼 크게 상승하여 장기간 섭취 시 뛰어난 항위염 효과를 기대할 수 있겠다.

액체-입자 Swirling 유동층에서 유동입자 흐름 및 열전달 특성 (Characteristics of Particle Flow and Heat Transfer in Liquid-Particle Swirling Fluidized Beds)

  • 직경이 0.102 m이고 높이가 2.5 m인 액체-입자 swirling(나선)흐름 유동층에서 유동 입자의 흐름 및 열전달 특성을 고찰하였다. 액체유속($U_L$), 유동 입자의 크기($d_p$) 그리고 연속상인 액체의 나선 유도흐름 액체량의 비($R_s$)가 유동층 내 유동입자의 체류량 유동층 내부 열원과 유동층간의 총괄 열전달 계수에 미치는 영향을 검토하였다. 액체-입자 나선흐름 유동층에서 입자 체류량은 입자의 크기와 나선유도흐름 액체량의 비가 증가함에 따라서 증가하였으나, 액체유속의 증가에 따라서는 감소하였다. 유동층 내부에서 나선 유도 흐름 액체량의 비가 0.1~0.3인 경우에 유동 입자의 국부체류량은 유동층 중심부에서 큰 값을 나타내었으나, $R_s$의 값이 0.5일 때는 반경 방향 입자 체류량은 거의 균일한 분포를 보이며, $R_s$의 값이 0.7일 때는 유동층 중심부의 입자 체류량이 상대적으로 감소하는 경향을 나타내었다. 유동층 내부열원과 유동층간의 열전달 특성은 열원 표면과 유동층간의 온도차 요동 자료의 위상공간 투영과 kolmogorov 엔트로피 해석으로 고찰할 수 있었으며, 나선 유도 흐름 액체량의 비($R_s$)가 0.1에서 0.5까지 증가할수록 온도차 요동 자료의 위상공간 투영은 점점 안정되고 규칙성이 증대되는 상태를 나타내고, kolmogorov 엔트로피 값은 감소하는 경향을 나타내었다. 열원 표면과 유동층간의 온도차 요동 자료의 kolmogorov 엔트로피 값은 액체의 유속이 증가함에 따라 최대값을 나타내었다. 열원과 유동층간의 총괄 열전달 계수는 액체유속, 층공극률, 나선 유도 흐름 액체량의 비가 증가함에 따라서 최대값을 나타내었으며, 유동 입자의 크기가 증가함에 따라 증가하였다. 내부 열원과 유동층간의 총괄 열전달 계수가 최대값을 나타낼 때의 액체의 유속 조건에서 온도차 요동자료의 kolmogorov 엔트로피의 값도 최대값을 나타내었다. 액체-입자 나선흐름 유동층에서 입자 체류량과 열전달 계수를 무차원군의 상관식으로 나타낼 수 있었다.

브랜드화된 돈육의 품질 특성 (Quality Properties of Brand Pork)

  • 시중에 유통 중인 브랜드 돈육의 품질 특성을 조사하기 위해 3개 브랜드(Y처리구 : 재래돼지 $\times$멧돼지, 맥강과 한약제 급여 ; J처리구 : LY$\times$D, 무항생제, 생균제 급여; M처리구 : LY$\times$D) 돈육 등심을 구매하여 품질 특성을 분석하였다. 수분과 조회분 함량은 Y처리구가 J와 M 처리구에 비해 높았지만, 조지방 함량은 J처리구가 Y와 M처리구에 비해 현저하게 낮았다(p<0.05). Y처리구의 미경산돈이 거세돈보다 수분과 조지방 함량이 현저하게 높았다(p<0.05) Y처리구의 미경산돈의 pH는 J와 M처리구의 미경산돈에 비해 현저하게 높았다(p<0.05) 돈육의 L(명도)값과 $W^*$(백색도)값은 Y처리구가 J와 M처리구에 비해 현저하게 낮았다(p<0.05) 조직 특성 중 탄력성은 J처리구가 Y와 M처리구에 비해 현저하게 높았다(p<0.05). 관능적 점수는 Y처리구가 J와 M처리구에 비해 높았고, M처리구의 미경산돈은 거세돈에 비해 다즙성이 현저하게 높았다(p<0.05). 지방산 조성 중 linoleic acid와 arachidonic acid 함량은 Y처리구가 J와 M 처리구에 비해 높았지만 palmitic acid, pamitoleic acid, stearic acid 및 oleic acid 함량은 낮았다. 포화지방산 함량은 Y처리구가 J와 M처리구에 비해 낮았지만, 불포화지방산, 필수지방산, 불포화지방산/포화지방산 비, 필수지방산/포화지방산 비 및 필수지방산/불포화 지방산 비는 가장 높은 함량을 나타내었다. 성별 간에는 M처리구에서 거세돈이 미경산돈에 비해 stearic acid 함량이 현저하게 높았다(p<0.05). 이상의 결과를 종합해 볼 때 재래돼지$\times$멧돼지를 교잡하여 맥강과 한약재를 급여한 Y처리구가 J와 M처리구에 비해 육의 이화학 특성과 관능적 특성에서 우수한 것으로 나타났다.

  • 동력경운깅가 일반 경사지에서 견인주행하는 경우 견인주행성능과 주행특성을 구명하기 위하여 토양의 종류 및 상태는 일정하게 하고 지면의 기하학적 조건과 견인주행속도 및 견인하중을 변수로 하여 외부동력전달계의 시점인 좌우차륜과 토양간에 발생하는 차륜구동력 및 굴름정항과 Engine에서 구동륜까지 내부전달계를 통하여 전달된 동력의 이론치와 실험치와의 부합여부를 검정하고 부가적으로 동력경운기가 경사지기계화의 동력기로써의 가능성여부와 문제점을 찾으려한다.

발아 고단백 콩의 Lactobacillus brevis 젖산발효에 의한 가바와 이소플라본 함량 및 라디칼 소거활성의 비교 (Comparison of γ-aminobutyric acid and isoflavone aglycone contents, to radical scavenging activities of high-protein soybean sprouting by lactic acid fermentation with Lactobacillus brevis)

  • 본 연구는 발아된 고단백 콩(high protein soybeans, HPSs)으로부터 가바(GABA)와 비배당체 이소플라본(isoflavone aglycones)이 증가된 콩-분말 요구르트(SPY)를 제조하였다. 이를 위해 L. brevis로 발효 증자 고단백콩 발아체(FSHPS, 0-4 cm)를 72시간 발효하였다. 발효 고단백콩(FHPS), 0 cm 발효 증자 고단백콩 발아체(FSHPS-0), 1cm 발효 증자 고단백콩 발아체(FSHPS-1), 2cm 발효 증자 고단백콩 발아체(FSHPS-2) 및 4 cm 발효 증자 고단백콩 발아체(FSHPS-4)의 총 유리아미노산 함량은 각 79.53, 489.93, 877.55, 780.53 및 979.97 mg 이었다. 발효되지 않은 고단백콩 발아체(UFSHPS-1, 1 cm) 및 FSHPS-1의 글루탐산(GA) 및 GABA 함량이 각각 최고 100.31 mg/100 mL 및 101.60 mg/100 mL로 관찰되었다. 또한 FSHPS-1에서 가장 높은 DPPH(63.28%) 및 ABTS(73.28%) 라디칼 소거능을 보였다. 그러나 FSHPS-4에서 isoflavone aglycone 비율이 81.63%로 가장 높았다. 특히, FSHPS-1은 높은 가바 함량과 기능적 특성을 나타내어 두유 산업에 응용할 수 있을 것이다.

Mammary Performance of First Lactation Bali Cows (Bibos banteng) Fed Grass-Legume Based Diets in Relation to the Role of Glucose

  • A study of mammary function in relation to glucose metabolism of first lactation Bali cows on grass-legume diets was carried out using 12 primiparous cows (initial BW $263.79{\pm}21.66kg$) for 16 weeks starting immediately post calving. The animals were randomly allocated into 4 dietary treatment groups R1, R2, R3 and R4, receiving from the last 2 months of pregnancy onwards, rations based on a mixture of locally available grass and legume feed ad libitum. On a DM basis R1 contained 70% elephant grass (PP, Penicetum purpureum) plus 30% Gliricidia sepia leaves (GS), R2 was 30% PP plus 25% GS supplemented with 55% Hibiscus tilliacius leaves (HT, defaunating effect), R3 and R4 were 22.5% PP+41.25% GS+11.25% HT+25% concentrate, with R4 supplemented with zinc-diacetate. TDN, CP and zinc contents of the diets were 58.2%, 12.05% and 18.3 mg/kg respectively for R1, 65.05%, 16.9% and 25.6 mg/kg respectively for R2, 66.03%, 16.71% and 29.02 mg/kg respectively for R3 and 66.03%, 16.71% and 60.47 mg/kg respectively for R4. Milk production and body weights were monitored, an energy and protein balance trial conducted, overall glucose kinetics parameters assessed, mammary blood flow (MBF) and metabolite arteriovenous differences (${\Delta}AVs$) measured to get uptake data and mammary performance relationships. Parameters of glucose kinetics at peak lactation or during dry condition were not affected by ration quality. Glucose pool size, space of distribution and flux increased by 61.77, 62.26 and 82.08%, respectively, during lactation compared to the dry period. Mean glucose flux of lactating Bali cows was $5.52mg/min.kgBW^{0.807}$ which resembles the range of values of temperate dairy cows. Calculation showed that glucose requirements for maintenance, milk lactose and fat-glycerol synthesis, and the formation of NADPH reached 461.69 g for a yield of 1 kg/d or equal to 320.62 mg/min, which was less than the average glucose flux of lactating Bali cows of 481.35 mg/min. Mammary blood flow (MBF) values ranged from 56 to 83 l/h for the different treatments and the ratio MBF per kg milk produced improved from av. 1540 l/kg for R1 to av. 967 l/kg for R4 treated cows. Mammary glucose uptake ranged from 6.27 to 12.03 g/h or 120 to 140 g/kg milk. Glucose uptake was mass-wise 2 to 4 times the amount secreted as lactose, which indicated values less than the calculated mammary glucose needs and that little lactose was synthesized. The excess glucose taken-up was used for other metabolic processes. Linear relationships between metabolite ${\Delta}AVs$ and arterial blood plasma concentration [A] showed that in Bali cows triglycerides (TG), phenylalanine (Phe) and tyrosine (Tyr) have high coefficients of determination, i.e. 0.77, 0.81 and 0.69, respectively. For glucose, the relationship is quadratic with an $R^2$ value of 0.49. It was concluded that lactose synthesis was inadequate, which led to a speculation that milk yield could be improved by increased lactose synthesis.

Variability in Drug Interaction According to Genetic Polymorphisms in Drug Metabolizing Enzymes

  • There are significant differences in the extent of drug interactions between subjects. The influence of the genetic make up of drug metabolizing enzyme activities (CYP3A5, CYP2C19 and UDP-glucuronosyl transferase) on the pharmacokinetic drug interaction potential were studied in vivo. Nineteen healthy volunteers were grouped with regard to the $CYP3A5^{*}3$ allele, into homozygous wild-type (CYP3A5^{*}1/1^{*}1$, n=6), heterozygous $(CYP3A5^{*}1/^{*}3$, n=6), and homozygous variant-type $(CYP3A5^{*}3/^{*}3$, n=7) subject groups. The pharmacokinetic profile of intravenous midazolam was characterized before and after itraconazole administration (200 mg once daily for 4 days), and also following rifampin pretreatment (600 mg once daily for 10 days), with a washout period of 2 weeks in between. For omeprazole and moclobemide pharmacokinetic interaction study 16 healthy volunteers were recruited. The volunteer group comprised 8 extensive metabolizers and 8 poor metabolizers of CYP2C19, which was confirmed by genotyping. Subjects were randomly allocated into two sequence groups, and a single-blind, placebo-controlled, two-period crossover study was performed. In study I, a placebo was orally administered for 7 days. On the eighth morning, 300 mg of moclobemide and 40 mg of placebo were coadministered with 200 mL of water, and a pharmacokinetic study was performed. During study n, 40 mg of omeprazole was given each morning instead of placebo, and pharmacokinetic studies were performed on the first and eighth day with 300 mg of moclobemide coadministration. In the UGT study pharmacokinetics and dynamics of 2 mg intravenous lorazepam were evaluated before and after rifampin pretreatment (600 mg once daily for 10 days), with a washout period of 2 weeks in between. The subjective and objective pharmacodynamic tests were done before and 1, 2, 4, 6, 8, and 12 hrs after lorazepam administration. The pharmacokinetic profiles of midazolam and of its hydroxy metabolites did not show differences between the genotype groups under basal and induced metabolic conditions. However, during the inhibited metabolic state, the $CYP3A5^{*}3/^{*}3$ group showed a greater decrease in systemic clearance than the $CYP3A5^{*}1/^{*}1$ group $(8.5\pm3.8$ L/h/70 kg vs. $13.5\pm2.7$ L/h/70 kg, P=0.027). The 1'-hydroxymidazolam to midazolam AUC ratio was also significantly lower in the $CYP3A5^{*}3/^{*}3$,/TEX> group $(0.58\pm0.35,$ vs. $1.09\pm0.37$ for the homozygous wild-type group, P=0.026). The inhibition of moclo-bemide metabolism was significant in extensive metabolizers even after a single dose of omeprazole. After daily administration of omeprazole for 1 week, the pharmacokinetic parameters of moclobemide and its metabolites in extensive metabolizers changed to values similar to those in poor metabolizers. In poor meta-bolizers, no remarkable changes in the pharmacokinetic parameters were observed. The area under the time-effect curves of visual analog scale(VAS), choice reaction time, and continuous line tracking test results of lorazepam was reduced by 20%, 7%, 23% respectively in induced state, and in spite of large interindividual variablity, significant statistical difference was shown in VAS(repeated measures ANOVA, p=0.0027).

In the UGT study pharmacokinetics and dynamics of 2 mg intravenous lorazepam were evaluated before and after rifampin pretreatment (600 mg once daily for 10 days), with a washout period of 2 weeks in between. The subjective and objective pharmacodynamic tests were done before and 1, 2, 4, 6, 8, and 12 hrs after lorazepam administration. The pharmacokinetic profiles of midazolam and of its hydroxy metabolites did not show differences between the genotype groups under basal and induced metabolic conditions. However, during the inhibited metabolic state, the $CYP3A5^{*}3/^{*}3$ group showed a greater decrease in systemic clearance than the $CYP3A5^{*}1/^{*}1$ group $(8.5\pm3.8$ L/h/70 kg vs. $13.5\pm2.7$ L/h/70 kg, P=0.027). The 1'-hydroxymidazolam to midazolam AUC ratio was also significantly lower in the $CYP3A5^{*}3/^{*}3$,/TEX> group $(0.58\pm0.35,$ vs. $1.09\pm0.37$ for the homozygous wild-type group, P=0.026). The inhibition of moclo-bemide metabolism was significant in extensive metabolizers even after a single dose of omeprazole. After daily administration of omeprazole for 1 week, the pharmacokinetic parameters of moclobemide and its metabolites in extensive metabolizers changed to values similar to those in poor metabolizers. In poor meta-bolizers, no remarkable changes in the pharmacokinetic parameters were observed. The area under the time-effect curves of visual analog scale(VAS), choice reaction time, and continuous line tracking test results of lorazepam was reduced by 20%, 7%, 23% respectively in induced state, and in spite of large interindividual variablity, significant statistical difference was shown in VAS(repeated measures ANOVA, p=0.0027).
