The Influence of the Substituents on the Benzo Ring for Antioxidant Activity of 4-Methyl-2H-benzopyran-2-one Analogues (4-Methyl-2H-benzopyran-2-one 유도체들의 항산화 활성에 관한 Benzo 고리상 치환기들의 영향)

  • The influences on antioxidant activities of the substituents ($R_1-R_4$) on benzo ring in 4-Methyl-2H-benzopyran-2-one analogues (1-23) were discussed quantitatively using three dimensional quantitative structure-activity relationships (3D-QSARs: Comparative molecular field analyses (CoMFA) and Comparative molecular similarity indice analyses (CoMSIA)) methods. The statistical qualities of CoMSIA models were better than those of CoMFA models and the CoMSIA 2 model was optimized model ($q^2$=0.700 & $r^2$=0.979). Also, the contribution ratios (%) of the optimized CoMSIA 2 model were H-bond donor field 43.5%, electrostatic field 41.8% and steric field 14.7% so that the antioxidant activity exhibited a strong correlation with H-bond donor and electrostatic factor of molecules. From the analytical results of the CoMSIA contour maps, if the positive charge favor group and H-bond donor disfavor group were placed in the $R_1-R_4$ positions on the benzo ring, it was predicted that the groups would raised the antioxidant activity.

  • Purpose: Erythropoietin (EPO) has neuroprotective effects in many animal models of brain injury, including hypoxic-ischemic (HI) encephalopathy, trauma, and excitotoxicity. Current studies have demonstrated the neuroprotective effects of EPO, but limited data are available for the neonatal periods. Here in we investigated whether recombinant human EPO (rHuEPO) can protect the developing rat brain from HI injury via modulation of NMDA receptors. Methods: In an in vitro model, embryonic cortical neuronal cell cultures from Sprague-Dawley (SD) rats at 19-days gestation were established. The cultured cells were divided into five groups: normoxia (N), hypoxia (H), and 1, 10, and 100 IU/mL rHuEPO-treated (H+E1, H+ E10, and H+E100) groups. To estimate cell viability and growth, a 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay was done. In an in vivo model, left carotid artery ligation was performed on 7-day-old SD rat pups. The animals were divided into six groups; normoxia control (NC), normoxia Sham-operated (NS), hypoxia-ischemia only (H), hypoxia-ischemia+vehicle (HV), hypoxia-ischemia+rHuEPO before a HI injury (HE-B), and hypoxia-ischemia+rHuEPO after a HI injury (HE-A). The morphologic changes following brain injuries were noted using hematoxylin and eosin (H/E) staining. Real-time PCR using primers of subunits of NMDA receptors (NR1, NR2A, NR2B, NR2C and NR2D) mRNA were performed. Results: Cell viability in the H group was decreased to less than 60% of that in the N group. In the H+E1 and H+E10 groups, cell viability was increased to >80% of the N group, but cell viability in the H+E100 group did not recover. The percentage of the left hemisphere area compared the to the right hemisphere area were 98.9% in the NC group, 99.1% in the NS group, 57.1% in the H group, 57.0% in the HV group, 87.6% in the HE-B group, and 91.6% in the HE-A group. Real-time PCR analysis of the expressions of subunits of NMDA receptors mRNAs in the in vitro and in vivo neonatal HI brain injuries generally revealed that the expression in the H group was decreased compared to the N group and the expressions in the rHuEPO-treated groups was increased compared to the H group. Conclusion: rHuEPO has neuroprotective property in perinatal HI brain injury via modulation of N-methyl-D-aspartate receptors.

Low Resistivity Ohmic Ni/Si/Ni Contacts to N-Type 4H-SiC (낮은 접촉저항을 갖는 Ni/Si/Ni n형 4H-SiC의 오옴성 접합)

  • Characteristics of ohmic Ni/Si/Ni contacts to n-type 4H-SiC are investigated systematically. The ohmic contacts were formed by annealing Ni/Si/Ni sputtered sequentially The annealings were performed at 950℃ using RTP in vacuum ambient and N₂ ambient, respectively. The specific contact resistivity(p/sub c/), sheet resistance(R/sub s/), contact resistance (R/sub c/) transfer length(L/sub T/) were calculated from resistance(R/sub T/) versus contact spacing(d) measurements obtained from TLM(transmission line method) structure. While the resulting measurement values of sample annealed at vacuum ambient were p/sub c/ = 3.8×10/sup -5/Ω㎠, R/sub c/ = 4.9 Ω and R/sub T/ = 9.8 Ω, those of sample annealed at N₂ ambient were p/sub c/ = 2.29×10/sup -4/Ω㎠, R/sub c/ = 12.9 Ω and R/sub T/ = 25.8 Ω. The physical properties of contacts were examined using XRD 3nd AES. The results showed that nickel silicide was formed on SiC and Ni was migrated into SiC. This result indicates that Ni/Si/Ni ohmic contact would be useful in high performance electronic devices.

Decolorization of Poly R-478 Dye by Coriolus versicolor IFO 30388 (구름버섯(Coriolus versicolor IFO 30388)에 의한 Poly R-478 염료의 탈색)

  • Effects of nitrogen and carbon sources on the decolorization rate of poly R-478 dye by a white rot basidiomycete Coriorus versicolor IFO 30388 were examined. The fungus exhibited 87.2% of decolorization rate when it was cultured in the state of stationary in a nitrogen-limited medium (pH 4.5) which contained 2.0% glucose, 0.04% ammonium tartrate, 0.02% poly R-478 dye, 2% $KH_2PO_4$, 0.5% $MgSO_4{\cdot}7H_2O$, 0.1% $CaCl_2{\cdot}2H_2O$, 0.002% thiamine-HCl and 10 mM 2,2 dimethylsuccinate (sodium) at $28^{\circ}C$ for 10 days. Decolorization of the dye occurred in the presence of nitrogen source in the medium and decolorization rate increased rapidly after depletion of $NH_4^+$ from the medium.

  • KSTAR (Korea Superconducting Tokamak Advanced Research) 장치는 Tokamak 개념의 핵융합 연구 장치로서 플라즈마를 가두기 위한 자장을 발생하는 토로이달 자장(Toroidal Field, TF) 코일과 플라즈마 발생 및 형상 조정을 위한 폴로이달 자장 (Poloidal Filed, PF) 코일로 구성되며, 초전도 코일을 이용한다. TF코일의 전원장치로는 40 kA급의 안정된 직류 전원장치가 필요하며, PF 코일의 전원장치로는 빠른 전류상승 및 피드백 기능을 갖춘 정밀 대전류 전원을 필요로 한다. 또한 초전도 코일의 ??????치현상 발생 시 코일과 전원장치 보호를 위한 대전류 직류 차단시스템을 필요로 한다. KSTAR 장치의 설계에 의하면 상하 7쌍의 초전도 PF 코일에 약 1MA/sec급의 고속 전류구동을 운전 시나리오에 따라 인가하여 핵융합 연구를 위한 플라즈마를 생성한다. 본 논문은 TF 및 PF 코일에 대전류를 인가하기 위해서 개발된 전원장치 (Power Supply, PS)에 관한 연구이다.

A Study on the Removal Performance of PM10 by the Air-washer and the Ultrasonic Humidifier (자연 기화식 가습기와 초음파 가습기의 PM10 제거성능)

  • The purpose of this study is to evaluate the PM10 removal efficiencies of an Air-washer and an Ultrasonic humidifier by mock-up experiments. The concentrations of PM10 were measured at 5 minute intervals using $\beta$-Ray method. The average PM10 removal efficiency of the Air-washer was 79.8%, while the PM10 concentrations increased with the operation of Ultrasonic humidifier. In the case of operating the Air-washer, the R.H(Relative Humidity) level of the test room was maintained at around 45%, regardless of initial R.H condition. The R.H of the test room with Ultrasonic humidifier was influenced by the initial R.H condition.

New candidate for skin depigmentation: The inhibitory effect and cytotoxicity of small molecule compounds at in vitro cell culture

  • To obtain effective and safe topical depigmenting agents, we synthesized hydroxybenzoates, alkoxybenzoates, and 3,4,5-trimethoxycinnamate containing a thymol moiety and screened then for high-level inhibitory activity against melanin synthesis. Among them, 5-methyl-2-(methylethyl)phenyl (2Ε)-3-(3,4,5-trimethoxyphenyl)prop-2-enoate (Melasolv)$^{TM}$ 4h, showed the most potent depigmenting effect ($IC_{50}$/ = 10$\mu$M) with low cytotoxicity ($IC_{50}$/ = 200$\mu$M). To find the inhibition mechanism of our candidate, various in vitro tests were performed such as DPPH assay, tyrosinase activity in mushroom or in culture cell and expression of tyrosinase, TRP-l and TRP-2. The result of this study suggested that 4h inhibited melanin synthesis by reducing the expression of tyrosinase and TRP-l at the transcriptional level in melan-a melanocytes.s.

Profiles of microRNAs in Mice Follicles According to Gonadotropins during in vitro Culture (생쥐 난포의 체외배양 중 생식샘자극호르몬에 따른 미세리보핵산 발현 양상)

  • Objective: MicroRNAs (miR) are known to repress target genes at post-transcriptional level and play important roles in development and maturation of cell. However, the expression profiles of miR during ovarian follicle maturation have not been fully elucidated. Here, we designed this study to investigate the expression profiles of miR in oocytes and granulose cells (G-cells) after in vitro culture according to gonadotropins and adding hCG. Methods: Ovaries from 12-day-old mice (C57BL6) were removed and preantral follicles were isolated and cultured in $20\;{\mu}L$-drop of culture media with supplementation of either rFSH, rLH, or rFSH+rLH. After their full maturation, follicles were incubated with rhCG and rEGF. RNA was isolated from oocytes and G-cells, and real-time PCR were performed with primers of miR known to be expressed in the mouse ovary (mmu-miR-16, -miR-27a, -miR-126, -miR-721). Results: FSH+LH group showed the highest ovulation and MII rates among gonadotropin groups. The profiles of miRs in oocytes and G-cells differed according to gonadotropin groups and adding hCG. The profiles of miRs showed divergent changes between oocytes and G-cells. Conclusion: miR expression profiles are altered by gonadotropins and supplementation of hCG during in vitro maturation of murine follicles. Target gene study must be necessary to validate these findings.

Influences of Forest Management Practices on pH and Electrical Conductivity in the Throughfall and Stemflow with the Abies holophylla and Pinus koraiensis Dominant Watershed (전나무림, 잣나무림 유역에서 수관통과우와 수간유하수의 수소이온농도 및 전기전도도에 미치는 산림시업의 영향)

  • This research was conducted to evaluate the effect of forest management practices on pH and electrical conductivity to get fundamental information on water purification capacity after forest operation. Rainfall, throughfall and stemflow were sampled at the study sites which consist of Abies holophylla and Pinus koraiensis in Gwangreung Experimental Forest for S months from May to November 1999. Mean pH of the throughfall of the beginning of the event was higher in management (thinning and pruning) sites of Abies holophylla and Pinus koraiensis stands than nonmanagement site of Abies holophylla and Pinus koraiensis stands. In addition, pH of the throughfall of the total amount of the event showed similar trends which are higher pH in the management sites compared with the non- management sites. This result indicates that managements such as thinning and pruning improve tree butler capacity of rainfall pH. According to the linear regression results, pH of the throughfall of the total amount of the event in non-management sites = 0.735${\times}$pH of the throughfall of the beginning of the event in non-management sites+1.849 ($R^2\;=\;0.82$) and pH of the throughfall of the total amount of the event in management sites= 0.863${\times}$pH of the throughfall of the beginning of the event in management sites +1.0242 ($R^2\;=\;0.87$). In case of stemflow pH, pH of the sternflow of the total amount of the event in non-management sites = 0.53${\times}$pH of the stemflow of the beginning of the event in non- management sites+2.7709 ($R^2\;=\;0.64$) and pH of the stemflow of the total amount of the event in management sites = 0.5854${\times}$pH of the stemflow of the beginning of the event in management sites+2.7045 ($R^2\;=\;0.65$). Electrical conductivity (EC) of the throughfall of the beginning and total amount of the event was highest in non- management site in Abies holophylla, followed by management sites in fsies Abies holophylla, non-management site in Pinus koraiensis, and management sites in Pinus koraiensis stands, respectively. According to the linear regression results, EC of the throughfall of the total amount of the event in non-managementsites = 0.4045${\times}$EC of the throughfall of the beginning of the event in non-management sites+26.766 ($R^2\;=\;0.69$) and EC of the throughfall of the total amount of the event in management sites = 0.6002${\times}$EC of the throughfall of the beginning of the event in management sites+8.0184 ($R^2\;=\;0.54$). In case of stemflow EC, EC of thestemflow of the total amount of the event in non-management sites = 0.6298${\times}$EC of the stemflow of the beginning of the event in non-management sites+11.582 ($R^2\;=\;0.72$) and pH of the stemflow of the total amount of the event in management sites =0.602${\times}$pH of the stemflow of the beginning of the event in management sites+20.783($R^2\;=\;0.49$).

Identification of Pseudomonas aeruginosa Genes Crucial for Hydrogen Peroxide Resistance

  • An opportunistic human pathogen, Pseudomonas aeruginosa, contains the major catalase KatA, which is required to cope with oxidative and osmotic stresses. As an attempt to uncover the $H_2O_2$-dependent regulatory mechanism delineating katA gene expression, four prototrophic $H_2O_2$-sensitive mutants were isolated from about 1,500 TnphoA mutant clones of P. aeruginosa strain PA14. Arbitrary PCR and direct cloning of the transposon insertion sites revealed that one insertion is located within the katA coding region and two are within the coding region of oxyR, which is responsible for transcriptional activation of several antioxidant enzyme genes in response to oxidative challenges. The fourth insertion was within PA3815 (IscR), which encodes a homolog of the Escherichia coli iron-sulfur assembly regulator, IscR. The levels of catalase and SOD activities were significantly reduced in the iscR mutant, but not in the oxyR mutant, during the normal planktonic culture conditions. These results suggest that both IscR and OxyR are required for the optimal resistance to $H_2O_2$, which involves the expression of multiple antioxidant enzymes including KatA.
