Development of PCR-Based Sequence Characterized DNA Markers for the Identification and Detection, Genetic Diversity of Didymella bryoniae with Random Amplified polymorphic DNA(RAPD)

  • Gummy stem blight pathogen is very difficult not only to monitor the inoculum levels prior to host infection, and also it is destructive and hard to control in field condition. We have applied RAPD technique to elucidate the genetic diversity of the genomic DNA of Didymella bryoniae and also to generate specific diagnostic DNA probe useful for identification and detection. The 40 primers produced clear bands consistently from the genomic DNA of twenty isolates of Didymella bryoniae, and two hundred seventy-three amplified fragments were produced with 40 primers. The combined data from 273 bands was analyzed by a cluster analysis using UPGMA method with an arithmetic average program of NTSYS-PC (Version 1.80) to generate a dendrogram. At the distance level of 0.7, two major RAPD groups were differentiated among 20 strains. RAPD group (RG) I included 8 isolates from watermelon except one isolate from melon. RAPD group (RG) IV included 12 isolates from squash, cucumber, watermelon and melon.. In amplification experiment with SCAR specific primer RG1F-RG1R resulted in a single band of 650bp fragment only for 8 isolates out of 20 isolates that should be designated as RAPD Group 1. However, same set of experiment done with RGIIF-RGIIR did not result in any amplified product.. Our attempts to detect intraspecific diversity of ITS region of rDNA by amplifying ITS region and 17s rDNA region for 20 isolates and restriction digestion of amplified fragment with 12 enzymes did not reveal polymorphic band. In order to develop RAPD markers for RGIV specific primer, a candidate PCR fragment( ≒1.4kb) was purified and Southern hybridized to the amplified fragment RGIV isolates. MR Findings of Seizure-Related Cerebral Cortical Lesions during Periictal Period

  • Purpose: This study investigated the MRI, MR angiography (MRA) and MR perfusion findings of seizure-related cerebral cortical lesions during the periictal period. Materials and Methods: From a retrospective review of the institutional database between 2011 and 2014, a total of 21 patients were included in this study. Two radiologists assessed periictal MRI, including MRA and MR perfusion, in patients with seizure-related cortical lesions. The parameters examined include: location of cortical abnormality, multiplicity of the affected cortical region, cerebral vascular dilatation, perfusion abnormality and other parenchymal lesions. Results: All patients showed T2 hyperintense cerebral cortical lesions with accompanying diffusion restriction, either unilateral (18/21, 85.7%) or bilateral (3/21, 14.3%). Of the 21 patients enrolled, 10 (47.6%) had concurrent T2 hyperintense thalamic lesions, and 10 (47.6%) showed hippocampal involvement. Of the 17 patients (81%) who underwent MRA, 13 (76.5%) showed vascular dilatation with increased flow signal in the cerebral arteries of the affected cortical regions. On MR perfusion, all 5 patients showed cortical hyperperfusion, corresponding to the region of cortical abnormalities. Conclusion: Seizure-related cerebral cortical lesions are characterized by T2 and diffusion hyperintensities, with corresponding cerebral hyperperfusion and vascular dilatation. These findings can be helpful for making an accurate diagnosis in patients with seizure.

국내 문학관 건축의 유형과 공간.형태구성 특징에 관한 연구 - 경상도 지역을 중심으로 - (A Study on the Pattern of Domestic Literature Museum and the Space.Form Composition Characteristic - Focused on Gyeongsang-do region -)

  • This study considered the characteristic through the present state of domestic literature museum and grouping by type to help the understanding for domestic literature museum. And conducted a case study on Gyeongsang-do region literature museum to grasp the space form composition characteristic of literature museum. The result gained through these studies is as follows. First, grouping domestic literature museum by type, we can conduct the classification founded on location character, an exhibition writer, and the main body of erection and maintenance management. Second, the classification founded on location character of literature museum is able to be divided into the type of the house of writer's birth, a literary work, writing, and etc. Third, the classification founded on the number of exhibition writers can be divided into the type of independence, an individual pavilion, and integration. Fourthly, the classification founded on the main body of erection and management can be divided into the case in which a local self-governing body is wholly in charge of erection and management, a local government is in charge of erection but entrusts management to a corporate body, etc., a corporate body is in charge of erection and management, and a private person is in charge of erection and management. Fifthly, speaking of the characteristic by type of the Gyeongsang-do region literature museum, the classification founded on location has the type of the house of writer's birth the most, the classification founded on the number of exhibition writers has the type of independence the most, and the classification founded on the main body of erection and management has the most the type in which a local self-governing body is in charge of erection and management. Also, for the characteristic by space form, the case which expresses the character of Korean traditional architecture by form is many the most, and there are pieces of work to pursue shape beauty through the articulation of mass or molding manipulation and the change by space form through the proper combination of concreteness and abstraction as well.

경북 지역 소나무의 연륜생태학적 특성에 관한 연구 (The Dendrochronological Characteristic of Pinus densiflora in Gyeongbuk Region)

  • 본 논문은 경북지역 소나무의 연륜생장에 대하여 연륜생태학적 해석을 통하여 월별 기후인자(월별평균온도, 월별강수량)간의 관계를 기초통계량 및 상관분석을 실시하였다. 실측된 연륜생장곡선은 수령 및 비기후적인 요소를 제거하기 위하여 음지수생장곡선을 이용하여 표준화를 실시하였다. 경북지역 소나무 연륜생장과 기후인자간의 관계에서 당해연도 $2\~4$월까지의 월별평균온도가 정의 상관으로 영향을 미치고 있으며. 월별 강수의 경우 당해연도 $1\~3$월의 강수가 연륜생장에 정의 상관으로 영향을 미치는 것으로 나타났다. 이러한 결과로 미루어 경북지역 소나무의 연륜생장에 있어서 휴면기가 끝나고 생장이 시작되는 시점에서의 기후인자가 생장을 촉진시키는 요인인 것으로 판단된다.

닭의 대퇴부 골단조직의 세포증식에 대한 면역조직화학적 관찰 (Immunohistochemical observations of proliferating cells in distal epiphyseal tissue of chicken femurs)

  • The present study was focussed to assess the proliferating cells in the distal epiphyseal tissue of the chicken femur by immunohistochemical staining methods. Four chickens were administrated intraperitoneally by twice consecutive injections, 1 day interval with bromodeoxyuridine(Brdur, 0.05 mg/gm BW/time), and then were killed by exsanguination of jugular vein at 2 hours after last injection. Samples were taken from femur distal epiphyseas of chicken. Labeling indexes(LI) were calculated as the ratio of the number of anti-Brdur monoclonal antibody-labeled cells in the each tissue layers from basal layer of the integument to bone marrow. The overall LI were found to be $13.90{\pm}3.44%$, $30.03{\pm}7.52%$, $16.00{\pm}9.41%$, $0.00{\pm}0.00%$ and $60.03{\pm}13.39%$ at basal layer of integument, perichordrium, reseving zone in cartilage, hypertrophic zone in cartilage and bone marrow respectively. LI in proliferating zone of cartilage were found to be $36.99{\pm}7.59%$, $32.83{\pm}5.38%$ and $22.02{\pm}6.27%$ at reserving zone side region, middle region, and hypertrophic zone side region respectively. The tissue layers with higher LI were odered as bone marrow, reserving zone side region in proliferating zone, middle region in proliferating zone, perichondrium, hypertrophic zone side region in proliferating zone. reserving zone of cartilage and basal layer of integument. These data indicate that the overall LI in the each tissue layer of distal epiphyseas of the chicken femur were concluded to be higher than that in another tissue of adult birds but hypertrophic zone of cartlage were appeared to be not proliferating cells.

  • One of the most significant characteristics of JPEG 2000, the emerging still image standards, is the Region of Interest(ROI) coding. Until now, there have been many proposed ROI methods. However, all of these methods can not be applied completely in applications. In this paper, the ROI coding mechanisms in JPEG2000 is first outlined and the diversity of ROI coding methods is discussed in detail. Then a comparative selection about using a proper ROI method according to ROI applications' requirements is proposed. Finally the experimental results show that there are also several parameters that control the effectiveness of ROI coding, such as the codeblock size, ROI size, wavelet filter type, the number of wavelet decomposition levels, and ROI importance score.

Primary Salivary Gland Adenocarcinoma in a Dog

  • A 17-year-old neutered male Miniature Pinscher dog presented with a mass on the left side of the submandibular region. Fine needle aspiration revealed malignant epithelial cells from the salivary gland but no evidence of metastasis was found on radiography, ultrasonography, or computed tomography. The cervical mass was surgically resected, and the histopathological examination confirmed adenocarcinoma of the mandibular salivary gland. Seven months after the initial diagnosis, the dog is alive without any clinical signs. This report describes the clinical findings, cytology, diagnostic imaging, and histopathological characteristics of a mandibular salivary gland adenocarcinoma in a Miniature Pinscher dog.

  • Phytopathogenic Pectobacterium carotovorum subsp. carotovorum (Pcc) LY34 secretes multiple isozymes of the plant cell wall degrading enzyme endoglucanases. We have cloned a third cel gene encoding CMCase from Pcc LY34. The structural organization of the celC gene (AY188753) consisted of an open reading frame (ORP) of 1,116 bp encoding 371 amino acid residues with a signal peptide of 22 amino acids within the NH$_2$-terminal region of pre-CelC. The predicted amino acid sequence of CelC was similar to that of Peetobaeterium ehrysanthemi Cel8Y (AF282321). The CelC has the conserved region of the glycoside hydrolase family 8. The apparent molecular mass of CelC was calculated to be 39 kDa by CMC-SDS-PAGE. The cellulase­minus mutant of Pee LY34 was as virulent as the wild-type in pathogenicity tests on tubers of potato. The results suggest that the CelC of Pce LY34 is a minor factor for the pathogenesis of soft-rot.

Ultrasonographic and Computed Tomographic Features of Dermoid Cyst of the Neck in a Dog: A Case Report

  • A 13-year-old castrated male mixed-breed dog presented for a health screening with a small nodule on the left hindlimb, which was revealed to be a mast cell tumor. A CT scan was performed to evaluate the mast cell tumor, and it incidentally revealed a well-marginated, fluid-attenuating, non-contrast enhanced mass in the deep ventral region of the left lower neck. Ultrasonography confirmed a well-capsulated mass with a cranio-ventrally protruding lesion extending into sternohyoid muscles. The outer layer of the mass had a hypoechoic thick wall and the internal parenchyma contained hyperechoic foci and multiple hyperechoic parallel lines. The mass was surgically resected. Histopathological examination confirmed a dermoid cyst characterized by cornifying squamous epithelial cells, keratinaceous debris, and hair shaft fragments. The dog showed no signs of recurrence or additional abnormalities three months post-surgery. This report highlights the importance of considering dermoid cysts in the differential diagnosis of neck masses on imaging examinations using CT or ultrasonography.
