Bacillus sp.유래 $\beta$-Mannanase 정제 및 Guar Gum가수분해 올리고당의 Bifidobacterium spp.에 대한 증식활성 (Purification of Bacillus sp. $\beta$-Mannanase and the Growth Activity of Bifidobacterium spp. by Guar Gum Hydrolysates.)

  • DEAE-sephadex ion exchange column chromatography에 의해 Bacillus sp. 유래 $\beta$-mannanase 의정제를 수행하여 비활성 21.57 units/$m\ell$, 정제배율 95.33을 나타내었다. SDS-PAGE에 의한 단일밴드를 확인하였고, 분자량은 38.9 kDa으로 결정되었다. 정제효소에 의해 guar gum galactomannan을 가수분해하여 1차 activated carbon column chromatography 와 2차 Sephadex G-25 gel filtration에 의해 당가수분해물을 분리 회수하여 TLC 및 FACE 에 의해 주요 당가수분해물은 중합도 5와 7로 확인되었다. B. longum B. bifidum, B. infantis, B. adolescentis, B. animalis, and B. breve의 생육활성에 대한 중합도 5와 7의 영향을 검토하기 위하여 modified-MRS 배지상에 탄소원으로 중합도 5와 7을 대체하여 생육활성을 비교한 결과 B. longum 에서는 중합도 5 galactomannooligosaccharide를 탄소원으로 대체한 경우 Standard MRS와 배교하여 0.62배로 감소하였다. 또한 중합도 5의 올리고당이 중합도 7의 올리고당보다 생육활성에 크게 기여하는 것으로 나타났다.

두유박을 이용한 압출면의 제조 (Making of extruded noodles mixed with soymilk residue)

  • 압출면의 영양강화 목적으로 두유박을 제면 원료인 소맥분과 섞어 점도 특성을 고찰하고, 압출면을 제조한 후 조리시험, 조리면의 조직감 시험 및 관능검사를 통하여 알아본 결과 두유박 첨가 비율이 증가함에 따라 복합분의 점도, 조리면의 조직감 및 조리 품질은 감소하였다. 두유박 10% 첨가 압출면의 제면 특성을 향상시키기 위하여 각종 첨가제의 첨가효과를 보면, 첨가제의 첨가량이 증가함에 따라 점도는 증가하였으며, 첨가제중에서는 xanthan gum이 가장 효과적이었다. 조직감에 있어서는 Na-alginate 2.0%, Na-carboxymethyl cellulose 2.0%, guar gum 0.5%, xanthan gum 1.0%가 좋을 결과를 나타내었다. 관능검사도 이와 유사한 결과를 보였다.

Textural Properties and Structures of Wheat and Maize Starch-Gum Mixed Gels During Storage

  • Effects of commercial and lab-generated gellan gums on the textural properties, structure, and aging of wheat and maize starch gels were investigated using a rapid visco-analyzer (RVA), rheometer, scanning electron microscope (SEM), and X-ray diffractometer. Gellan and guar gums increased the peak and final viscosities, but xanthan gum and gum arabic reduced them. The maize starch had higher breakdown viscosity than the wheat starch, regardless of the type of gum. The hardness of all starch gels increased during storage, but their cohesiveness and springiness decreased. The degree of the gel hardness showed higher in maize starch than in wheat starch and the effect of gum addition had a difference with gum type. The wheat starch-guar and -gellan gum mixed gels showed higher elasticity and cohesiveness after storage. The starch-gellan gum mixed gels had dense and stable network structures, and were well maintained even after 7 days of storage. Most of the gums had anti-aging effect on X-ray diffraction pattern of starch gels.

대두분 첨가 압출면의 제면특성 (Making characteristics of extruded noodles mixed with soybean flour)

  • 압출면의 영양강화 목적으로 대두분을 소맥분과 섞어 점도 특성을 고찰하고, 압출면을 제조할 후 조리시험, 조리면의 조직감시험 및 관능정사 결과 대주분 첨가비율이 증가함에 따라 복합분의 점도, 조리면의 조직감 및 조리품질은 저하하였다. 대두분 10% 첨가 압출면에 각종 첨가제의 첨가효과를 보면, 첨가제의 첨가량이 증가함에 따라 점도는 증가하였으며, 첨가제 중에서는 xanthangum이 가장 효과적이었다. 조직감에 있어서는 Na-carboxymethyl cellulose 2.0%, guar gum 2.0%, xanthan gum 1.0%에서 밀가루면과 비슷한 값을 나타내었으며, 관능검사도 이와 유사한 결과를 보였다.

천연고분자 분해성 필름의 생체적합성 연구 (Biocompatibility of Biodegradable Films by Natural Polymers)

  • 최근에 생분해성 고분자를 이용한 의료용재료로의 응용에 대한 관심이 고조되고 있다. 본 연구에서는 천연고분자(xanthan, locust bean, guar gum, chitosan 및 algin)를 생체재료로 사용할 목적으로 용액주조법에 의하여 분해성 필름을 제조하였다. 천연고분자로부터 제조한 생체재료의 가능성을 피부이식제로서 생체적합성 측정에 의하여 평가하였다. 이들을 실험동물의 등부위에 피하 삽입하여 필름의 시간에 따른 구조적, 중량적변화와 랫드의 혈액학적 변화를 측정하여 생체적합성을 연구하였다. 랫드에 대한 테스트결과 locust bean과 guar gum은 삽입 24시간, 48시간 후 조직에서의 생체 이물 반응에 의한 생체 부적합성을 나타내었으며, 일부 천연고분자로부터 제조한 분해성 필름은 단기간의 이상적인 생체재료로서의 가능성을 나타내었다.

구연산과 구아검 첨가 냉면의 품질특성 (Quality Characteristics of Naengmyeon Noodle Containing Citric Acid and Guar Gum)

  • Purpose: The consumption of noodles has increased domestically. However, noodles with high carbohydrate content can cause an increase in blood glucose compared with other foods. Therefore, in this study, Naengmyeon with high resistant starch was prepared for decreasing blood glucose by the addition of 0.5% citric acid (CN), 1% guar gum (GN) or 0.5% citric acid and 1% guar gum (CGN), and then it was incubated in a refrigerator for 3 days, and stored in a freezer for 1 month. Methods: The quality characteristics of these Naengmyeon noodles was evaluated based on total starch, resistant starch, water absorption, cooking loss, turbidity, in vitro starch hydrolysis, and in vivo glucose response. Results: There was no significant difference in the total starch, cooking loss, and turbidity. The resistant starch of GN (1.70%) and CGN (1.84%) was significantly increased when compared with that in Naengmyeon with no additives (N) and CN. In terms of water absorption, CN (86.01%) was the lowest in samples, followed by GN (92.17%), N (94.20%), and CGN (99.16%). CGN with high resistant starch was the lowest in in vitro starch hydrolysis in samples. However, it had no effect on the in vivo glucose response. In vitro starch hydrolysis exhibited a significant positive correlation (r=0.533; p<0.01) with in vivo glucose response. Conclusion: Therefore, future studies are needed to establish the standard for resistant starch contents in processed carbohydrate foods for delaying the increase in blood glucose. If this standard is established, it might help to develop processed foods for diabetic patients.

Teff (Eragrostis tef) 분말을 이용한 Gluten-free 식빵의 품질 특성에 잔탄검과 구아검이 미치는 영향 (Effects of Xanthan Gum and Guar Gum on the Quality Characteristics of Gluten-free Bread using Teff (Eragrostis tef) Flour)

  • This study investigated quality characteristics and sensory properties of gluten-free bread using teff flour and various gums. The five samples used in this study were control (wheat flour), T (teff flour), TG (teff flour, guar gum), TX (teff flour, xanthan gum), and TGX (teff flour, guar gum, xanthan gum). Baking loss, moisture content, pH, salinity, brix, dough expansion, color value, texture property, and sensory property were measured for each sample. As a result, pH was the highest in TX at 6.11 and the lowest in the control at 5.77. Salinity was the highest in the control at 0.80% and lowest in TG at 0.04%. Brix was the lowest in TX at $1.10^{\circ}Bx$. While moisture content was the highest in the control at 43.58%, gluten-free breads using teff flour had no significant difference (p<0.05). Baking loss rate of bread was no significant difference among samples (p<0.05). Dough expansion rate by fermentation was the highest in the control at 108.89% and the lowest in T at 17.84%. L-value of crust had no significant difference (p<0.05). For L-value of crumb, the control was the highest at 64.34 and T was the lowest at 33.84. In texture properties analysis, hardness was the highest in TGX at 16.00 N and the lowest in the control at 2.87 N. There was no significant difference in springiness (p<0.05). Chewiness was the highest in TX and TGX. The result of sensory properties was that while there were no significant differences in color, salty taste, flavor, and overall acceptability, there was a significant difference in softness and sweetness (p<0.05). These results showed that use of teff flour and gums rather than using only teff flour for bread manufacturing can improve quality characteristics in dough expansion and texture properties. It is considered that xanthan gum or guar gum are appropriate on bread using teff for quality characteristics.

메밀빵 제조: 2. 활성 글루텐과 수용성 gum물질이 메밀빵 특성에 미치는 효과 (Development of Buckwheat Bread: 2, Effects of Vital Wheat Gluten and Water-Soluble Gums on Baking and Sensory Properties)

  • 메밀분 30%와 강력분 70%의 복합분으로 강력분만을 사용한 이스트 발효빵과 유사하도록 메밀빵을 만들기 위하여 활성 글루텐과 gum 물질을 첨가하였으며 복합분의 제빵 적성에 미치는 그들의 효과를 알아보기 위하여 첨가물별 메밀빵의 물리적, 관능적 검사를 통하여 메밀빵의 품질 특성을 비교하였다. 메밀에 결여된 글루텐의 기능성을 보완하기 위하여 활성 글루텐과 gum물질을 첨가한 결과 메밀빵의 부피와 무게가 모두 증가하였으며 그 효과는 xanthan gum>활성 글루텐>guar gum 순 이었고, 단독 사용시보다는 활성 글루텐과 gum물질을 같이 사용하였을 때 효과가 더욱 증대되어 대조구 밀빵과 유사한 부피를 나타내었다. 빵 껍질색은 guar gum의 첨가로 L, a값이 증가하여 밝아졌고 xanthan gum을 첨가한 경우에는 L, b값이 감소하여 상대적으로 어두운 색을 나타내었다. 빵 내부색은 활성 글루텐과 xanthan gum 첨가로 L, b값이 감소하여 어두워졌다. 관능검사 QDA 결과 메밀빵에 활성 글루텐이나 gum 물질을 첨가함으로써 빵 속의 부서짐성이 유의적으로 감소되었고 그 밖의 탄력성, 촉촉함, 깔깔함 등의 조직감이 개선되었다. Amylograph에서 호화는 밀가루보다 메밀가루가 낮은 온도에서 일찍 시작되었고 최고 점도는 밀가루보다 메밀가루가 현저히 높았다. 메밀가루 30%와 강력분 70%의 복합분에 활성 글루텐과 gum물질을 간이 첨가함으로써 호화 점도가 감소되고 제빵 적성이 개선되었다. 또한 100% 강력분의 호화 점도쪽으로 낮아질수록 관능검사에서 메밀랑은 부드럽고 촉촉하게 나타났으며 부서지는 정도도 감소하였다. 이러한 관점에서 첨가물에 따른 메밀빵의 저장기간 중 노화변화에 대한 연구가 수행되어져야 할 것으로 사려된다.

탄수화물의 급원과 식이섬유의 종류를 달리한 식이가 제 2형 당뇨 쥐의 당대사 및 지질대사에 미치는 영향 (Effect of Feeding with Different Source of Carbohydrate and Fiber on Carbohydrate and Lipid Metabolism in Type 2 Diabetic Rats)

  • This study was designed to evaluate the effects of fructose(F) or sucrose(S) and guar gum intake on carbohydrate and lipid metabolism in 15-week-old male Goto-Kakizaki(GK) rats. Fifty rats were randomly assigned to 5 groups which were different in carbohydrate(25% of carbohydrate) and fiber(5% w/w) sources. The carbohydrate(CHO) sources of each group were comstarch(control group, 100% of CHO), fructose with cellulose(F), fructose with guar gum(FG), sucrose with cellulose(S), and sucrose with guar gum(SG). Each group was fed exterimental diet for 4 weeks. We measured food intake, body weight gain, adipose tissues weight and organs weight. We conducted oral glucose tolerance test(OGTT) and measured plasma insulin concentration to examine carbohydrate metabolism. To evaluate lipid metabolism, we measured the lipid profile of plasma, liver and feces. Food intake and weight gain of FG or SG groups tended to be less than those of F or S groups. Perirenal and epididymal fat pad weights of SG group were significantly lower than those of S group and those of FG group tended to be lower than those of F group. In OGTT, blood glucose values of F or S groups were significantly higher than those of C group, and FG or SG groups tended to be lower than those of F or S groups during the experimental time. The area under the curve(AUC) of C group was significantly highest among the groups, AUC and plasma insulin concentration of FG or SG groups tended to be lower than those of F or S groups. Plasma and hepatic triglyceride (TG) of FG and SG groups were significantly lower than those of F and S groups, plasma and hepatic total lipid(TL) and total cholesterol(TC) of FG and SG groups tended to be lower than those of F and S groups. Fecal TL, TG and TC of FG or SG groups tended to be higher than those of F and S groups. In conclusion, intake of guar gum should improve carbohydrate and lipid metabolism in partial substitution of fructose or sucrose for cornstarch in GK rats.

Rheological and Pasting Properties of Naked Barley Flour as Modified by Guar, Xanthan, and Locust Bean Gums

  • To understand the effects of adding different gums (guar, xanthan, and locust bean gums) on naked barley flour (NBF), the rheological and pasting properties of NBF-gum mixtures were measured at different gum concentrations (0, 0.3, and 0.6% w/w). Steady shear rheological properties were determined by rheological parameters for power law and Casson models. All samples showed a clear trend of shear-thinning behavior (n=0.16~0.48) and had a non-Newtonian nature with yield stress. Consistency index, apparent viscosity, and yield stress values increased with an increase in gum concentration. Storage modulus values were more predominant than loss modulus values with all concentrations of gums. There is a more pronounced synergistic effect of elastic properties of NBF in the presence of xanthan gum. Rapid visco analyser pasting properties showed that the addition of gums resulted in a significant increase in the peak, breakdown, setback, and final viscosities, whereas the pasting temperature decreased.
