The Identification and Physiological Properties of Lactobacillus plantarum JK-01 Isolated from Kimchi (김치로부터 분리한 Lactobacillus plantarum JK-01의 동정 및 생리적 특성)

  • In order to identify probiotic microorganisms, 25 isolates of Lactobacillus sp. were selected from kimchi based on their growth rates, lactic acid production and salt tolerance. The isolate JK-01 was identified as Lactobacillus plantarum by the API kit and 16S rDNA analysis (99.9% of homology), and named as L. plantarum JK-01. The maximum number of L. plantarum JK-01 was reached at 18 hr fermentation in MRS broth and the pH gradually decreased to 4.5. L. plantarum JK-01 showed high enzyme activities for xylanase, amylase, protease, and phytase on MRS agar plates containing each substrate. L. plantarum JK-01 showed high resistance to acidic pH and bile salts, and grew well even at pH 2.0 and 1.0% bile salt. In particular, L. plantarum JK-01 showed high heat stability as shown by $3.3{\times}10^3$ CFU/mL at $60^{\circ}C$. The isolate showed remarkable antimicrobial activity against E. coli in MRS broth based on its disappearance after 18 hr and clear zone formation using a paper disk assay. These results suggest that L. plantarum JK-01 may be probiotic in nature.

Physiological Characteristics and GABA Production of Lactobacillus acidophilus RMK567 Isolated from Raw Milk (원유에서 분리한 Lactobacillus acidophilus RMK567의 GABA 생성 및 생리적 특성)

  • In order to develop a new starter for fermented milk, 2082 bacteria were isolated from raw milk. The strain that showed excellent acid forming and ${\gamma}$-aminobutyric acid (GABA) production ($711.40{\mu}g/g$ D.W) characteristics after incubation at $37^{\circ}C$ for 18 hr was selected and identified as Lactobacillus acidophilus by the result of API carbohydrate fermentation pattern and 16S rDNA sequence. L. acidophilus RMK567 was investigated for its physiological characteristics. RMK67 strain showed good GABA production compared with commercial lactic acid bacteria. The optimum growth temperature of L. acidophilus RMK567 was $40^{\circ}C$ and cultures took 15 hr to reach pH 4.3. L. acidophilus RMK567 showed higher sensitivity to penicillin-G, novobiocin, as compared to other 14 different antibiotics. However, it showed more resistance to kanamycin, neomycin, streptomycin. It showed higher leucine arylamidase and ${\beta}$-galactosidase activities compared to 16 other enzymes. It was comparatively tolerant to bile juice and able to survive at pH 2 for 3 hr. It showed resistence to Escherichia coli, Salmonella typhimurium and Staphylococcus aureus with rates of 29.2%, 39.1% and 51.4%, respectively. Based on these and previous results, L. acidophilus RMK567 could be an excellent starter culture for fermented milk with excellent GABA contents.

Crystallization Kinetics by Thermal Analysis (DTA) on Starting Glass Compositions for PDP(Plasma Display Panel) Rib (열분석에 의한 PDP 격벽용 출발유리조성의 결정화 특성 연구)

  • In order to overcome trade-off among compositions, process and properties of the glasses with high PbO-base composition for PDP Rib, we studied glass crystallization and crystallization kinetics by Differential Thermal Analysis(DTA). Glass powder was obtained through melting/cooling/grinding, with 3 wt%TiO2 addition for the crystal nucleation and growth in $62PbO-19B_2O_3-10SiO_2-9(Al_2O_3-K_2O-BaO-ZnO)$(in wt%) composition glass. This powder was heat-treated for 1 to 10 h at $445^{\circ}C$ for nucleation. DTA measurements were performed to obtain the crystallization peak with $5∼25^{\circ}C/min$ heating rates. DTA crystallization peak temperature increased with increasing the heating rate and decreased with increasing the heating time. Because the Avrami parameter (n) was approximately 1, the surface crystallization occurred. The maximum nucleation time was 2 h.

Optimum Potting Medium and Nitrogen and Phosphorus Levels in the Soil for Root Nodule Formation in Black Locust (Robinia pseudoacacia L.) Seedlings (아까시나무(Robinia pseudoacacia L.) 유묘의 뿌리혹 형성에 적절한 배양토, 질소, 인 수준 구명에 관한 연구)

  • The objectives of this study were to find out proper potting medium and nutrient levels in the soil to promote the root nodule formation in black locust (Robinia pseudoacacia L.) seedlings. Commercial potting medium, compost, organic fertilizer, molded forest fertilizer, and compound fertilizer were used at different mixing rates to bring in various levels of mineral nutrients in the soil. Seedlings were grown in pots in a greenhouse for three months. Commercial potting medium containing peatmoss, vermiculite, and geolite was not suited for early nodule formation due to lack of nutrients, even though it produced good total dry weight. Compost was the best medium to promote both high total dry weight production and nodule formation with providing the proper levels of nitrogen and phosphorus in the soil. Molded forest fertilizer was acceptable for nodule formation. Compound fertilizer and organic fertilizer was not suited for nodule formation. The potting medium should contain optimum levels of nitrogen (0.05-0.2%) and phosphorus (100-600ppm) to promote early nodule formation in black locust seedlings.

  • Ninety six Rambouillet ${\times}$ Kaghani intact male lambs of 18 months of age were divided into 6 groups of 16 lambs each, groups being G-1, G-2, G-3, G-4, G-5 and G-6. Average liveweight of animals on different treatments at day 0 of the experiment was G-1 (24.68 S.D 3.35), G-2 (24.56 S.D 2.55), G-3 (24.53 S.D 2.90), G-4 (24.51 S.D 3.38), G-5 (24.58 S.D 3.58) and G-6 (24.81 S.D 3.43). Animals on treatment G-1 were fed only maize silage ad libitum, G-2 had been offered maize silage ad libitum plus 4.8 kg of commercial concentrate (Sona Vanda) per group, G-3 maize silage ad libitum plus 4.8 kg of crushed maize grain per group. G-4 lambs were fed oat silage ad libitum plus 4.8 kg of crushed maize per group, G-5 oat silage plus 4.8 kg of commercial concentrate per group and G-6 only oat silage ad libitum. The results showed highly significant differences (p<0.01) among treatment groups in silage intake throughout the feeding trial. G-2 group showed the highest silage intake while treatment G-4 showed the lowest silage intake for the first two months and treatment G-6 for the last two months. The results of growth rates (g/d/animal) showed that treatments G-1 and G-6, where no supplementation was given, had a weight loss throughout the feeding trial. However, weight loss in treatment G-6 was more severe than treatment G-1. Treatments G-2, G-3, G-4 and G-5 all showed weight gain however, treatment G-2 had the most gain. G-2 group also showed the highest wool production while G-6 the lowest. These results indicated that silage when made from full bloom crops of oats and silage fed alone without any supplementation causes weight loss in sheep. Supplementation with concentrates having 19% CP is far better than the crushed maize grain and maize silage is better than oats silage. Maize silage is superior than oat silage however, better performance could be expected if silages were made at the early bloom (dough) stage of plant maturity. The results indicate that treatment G-2 shows highest response while treatment G-6 the lowest. So the response of different treatments on both the parameters of weight gain and wool production is almost similar.

Effects of Synchronization of Carbohydrate and Protein Supply in Total Mixed Ration with Korean Rice Wine Residue on Ruminal Fermentation, Nitrogen Metabolism and Microbial Protein Synthesis in Holstein Steers

  • Three Holstein steers in the growing phase, each with a ruminal cannula, were used to test the hypothesis that the synchronization of the hourly rate of carbohydrate and nitrogen (N) released in the rumen would increase the amount of retained nitrogen for growth and thus improve the efficiency of microbial protein synthesis (EMPS). In Experiment 1, in situ degradability coefficients of carbohydrate and N in feeds including Korean rice wine residue (RWR) were determined. In Experiment 2, three total mixed ration (TMR) diets having different rates of carbohydrate and N release in the rumen were formulated using the in situ degradability of the feeds. All diets were made to contain similar contents of crude protein (CP) and neutral detergent fiber (NDF) but varied in their hourly pattern of nutrient release. The synchrony index of the three TMRs was 0.51 (LS), 0.77 (MS) and 0.95 (HS), respectively. The diets were fed at a restricted level (2% of the animal's body weight) in a $3{\times}3$ Latin-square design. Synchronizing the hourly supply of energy and N in the rumen did not significantly alter the digestibility of dry matter, organic matter, crude protein, NDF or acid detergent fiber (ADF) (p>0.05). The ruminal $NH_3$-N content of the LS group at three hours after feeding was significantly higher (p<0.05) than that of the other groups; however, the mean values of ruminal $NH_3$-N, pH and VFA concentration among the three groups were not significantly different (p>0.05). In addition, the purine derivative (PD) excretion in urine and microbial-N production (MN) among the three groups were not significantly different (p>0.05). In conclusion, synchronizing dietary energy and N supply to the rumen did not have a major effect on nutrient digestion or microbial protein synthesis (MPS) in Holstein steers.

Effect of γ-Aminobutyric Acid (GABA) Producing Bacteria on In vitro Rumen Fermentation, Biogenic Amine Production and Anti-oxidation Using Corn Meal as Substrate

  • The effects and significance of ${\gamma}$-amino butyric acid (GABA) producing bacteria (GPB) on in vitro rumen fermentation and reduction of biogenic amines (histamine, methylamine, ethylamine, and tyramine) using corn meal as a substrate were determined. Ruminal samples collected from ruminally fistulated Holstein cows served as inoculum and corn was used as substrate at 2% dry matter (DM). Different inclusion rates of GPB and GABA were evaluated. After incubation, addition of GPB had no significant effect on in vitro fermentation pH and total gas production, but significantly increased the ammonia nitrogen ($NH_3$-N) concentration and reduced the total biogenic amines production (p<0.05). Furthermore, antioxidation activity was improved as indicated by the significantly higher concentration of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) among treated samples when compared to the control (p<0.05). Additionally, 0.2% GPB was established as the optimum inclusion level. Taken together, these results suggest the potential of utilizing GPB as feed additives to improve growth performance in ruminants by reducing biogenic amines and increasing anti-oxidation.

Clinical Characteristics and Survival Analysis of Breast Cancer Molecular Subtypes with Hepatic Metastases

  • Background: The liver is one of the most common metastatic sites of breast cancer, hepatic metastases developing in 6%-25% of patients with breast cancer and being associated with a poor prognosis. The aim of this study was to analyze the survival and clinical characteristics of patients with hepatic metastases from breast cancer of different molecular subtypes and to investigate the prognostic and predictive factors that effect clinical outcome. Methods: We retrospectively studied the charts of 104 patients with breast cancer hepatic metastases diagnosed at Sun Yat-sen University Cancer Center from December 1990 to June 2009. Subtypes were defined as luminal A, luminal B, human epidermal growth factor receptor 2 (HER2) enriched, triple-negative (TN). Prognostic factor correlations with clinical features and treatment approaches were assessed at the diagnosis of hepatic metastases. Results: The median survival time was 16.0 months, and the one-, two- three-, four-, five-year survival rates were 63.5%, 31.7%, 15.6%, 10.8%, and 5.4%, respectively. Median survival periods after hepatic metastases were 19.3 months (luminal A), 13.3 months (luminal B), 18.9 months (HER2-enriched), and 16.1 months (TN, P=0.11). In multivariate analysis, a 2 year-interval from initial diagnosis to hepatic metastasis, treatment with endocrine therapy, and surgery were independent prognostic factors. Endocrine therapy could improve the survival of luminal subtypes (P=0.004) and was a favorable prognostic factor (median survival 23.4 months vs. 13.8 months, respectively, P=0.011). Luminal A group of patients treated with endocrine therapy did significantly better than the Luminal A group of patients treated without endocrine therapy (median survival of 48.9 vs. 13.8 months, P=0.003). Conclusions: Breast cancer subtypes were not associated with survival after hepatic metastases. Endocrine therapy was a significantly favorable treatment for patients with luminal subtype.

Study of FAME components and total contents on Micro-algal Biodiesel derived from Dunaliella tertiolecta (Dunaliella tertiolecta를 이용한 미세조류 유래 바이오디젤의 FAME 성분 특성 연구)

  • Biodiesel has very similar physical properties (density, kinematic viscosity) and has even higher cetane number compare with conventional diesel. There are no necessity to change or modify the infra-structure & engine system. It is known that fatty acid methyl ester (FAME) is oxygen-contained components increasing the combustibility, biodegradability and reduced the exhaust harmful gas. These things made the biodiesel more popular as an alternative diesel fuel. But biodiesel's sources are controversial issues about $CO_2$ reduction effect at this time because those mainly come from edible plants such as soy, palm, rapeseed already spent lot of $CO_2$ to cultivate. Whereas micro-algae is focused because they are inedible and has rapid growth rates & high carbon-dioxide adsorption rate per area. In this study, we analyze the each FAME components using $GC{\times}GC$-TOFMS in stead of GC-FID and verify the previous total FAME contents method's applicability through the micro algal biodiesel derived from Dunaliella tertiolecta.

FREQUENCY OF COTTONSEED CAKE SUPPLEMENTATION FOR YEARLING HEIFERS ON BLUE GRAMA RANGE

  • Yearling replacement heifers were used in a two-year study at the Fort Stanton Experimental Ranch near Capitan, New Mexico, USA to compare frequency (once versus three times weekly) of feeding a cottonseed cake supplement. The study involved periods of 130 days (January 4 through May 14) in 1985, and 146 days (December 4 through April 29) in 1986. In each year, supplemental periods were just before the breeding season (natural breeding in multiple-sire groups) that lasted for 75 days during first year and 60 days during second year. During the first year, all (83) heifers (Angus $\times$ Hereford) were supplemented at a level equivalent to 45 kg/head daily and during second year, all (69) heifers (Angus $\times$ Hereford, Hereford) were fed at a level equivalent to .68 kg/head daily. In the first year, heifers were sorted by initial weight into two replications, i.e., those weighing under 223 kg were placed in replicate 1 and those weighing over 223 kg were placed in replicate 2. During the second year, heifers were sorted by breed into two replications, i.e., Angus $\times$ Hereford heifers were placed in replicate 1 and Hereford heifers in replicate 2. Each year, half of the heifers in each replication were supplemented once weekly and the other half three time weekly. Weight gains of heifers were similar for the two treatments over both years of study. During the first year, weight gains for heifers supplemented once or thrice weekly (.23 vs .21 kg daily, respectively) did not differ (p>.05) and the same trend for the second year (.15 vs .16 kg daily, respectively) was observed. Similarly, breeding performance was consistent with weight gains and no difference (p>.05) between treatments was observed either year. Lower conception rates during the second tear of study (92 vs 72% for years 1 and 2, respectively) were due mainly to involvement of Hereford heifers that had an average weight of 232 kg at the start of breeding season and conception rate of 54% (averaged across both supplemental treatments). Based on the combined results of both trials, frequency of supplementation did not affect growth rate of yearling heifers nor did it influence their subsequent conception rate.
