Effect of Growth Regulators on Organogenesis in Diploid and Tetraploid Codonopsis Ianceolata In Vitro Culture

  • This study aimed to investigate the effect of growth regulators on the formation of the organ in the in vitro propagation of diploid and tetraploid Codonopsis lanceolata, and gain the basic data for in vitro propagation of superior C. lanceolata. In the case of diploid C. lanceolata, the highestshoot formation (3.0) was observed at 0.5 mg·L-1 addition medium with low IBA concentration. The shoot formation of tetraploid C. lanceolata was suppressed by addition of IBA. In the addition of lAA, the shoot formation of diploid C. lanceolata was slightly higher at 1.0 mg·-L-1 addition medium than that of control group, whereas tetraploid C. lanceolata showed the highest number (5.4) from control group. In the case of NAA, the shoot formation of diploid and tetra C. lanceolata tended to decrease at higher concentration. In terms of BA addition, the shoot formation of diploid C. lanceolata was increased by the addition of BA, whereaswhile the growth of shoot was decreased by the addition of BA. In the case of tetraploid C. lanceolata, shoot was found to be formed by the addition of low concentration of BA, and the growth of shoot was inhibited with the higher addition concentration of BA. With the addition of kinetin, the shoot formation of diploid C. lanceolata was slightly higher than that of control group, and the formation of adventitious root was highest (5.3) in the control group. In the case of tetraploid C. lanceolata, the shoot formation was similar in all treatment groups, but the formation and growth of adventitious root were significantly lower than that of diploid C. lanceolata. In the case of TDZ addition, the shoot formation of diploid C. lanceolata showed the pronounced results at 5.0 mg·L-1 addition medium, and the growth of shoot was inhibited by the addition of TDZ. The formation of adventitious root was 5.3 and 4.9 in the control group and 0.1 mg·L-1 addition medium respectively. The formation of the shoot of tetraploid C. lanceolata showed better results with the higher concentration of TDZ, and the growth was better with the lower concentration of TDZ. The formation and growth of adventitious root were significantly slower than that of diploid C. lanceolata.

Growth Modeling of Perilla frutescens (L.) Britt. Using Expolinear Function in a Closed-type Plant Factory System (완전제어형 식물공장에서 선형지수함수를 이용한 들깨의 생육 모델링)

  • Growth modeling in plant factories can not only control stable production and yield, but also control environmental conditions by considering the relationship between environmental factors and plant growth rate. In this study, using the expolinear function, we modeled perilla [Perilla frutescens (L.) Britt.] cultivated in a plant factory. Perilla growth was investigated 12 times until flower bud differentiation occurred after planting under light intensity, photoperiod, and the ratio of mixed light conditions of 130 μmol·m-2·s-1, 12/12 h, red:green:blue (7:1:2), respectively. Additionally, modeling was performed to predict dry and fresh weights using the expolinear function. Fresh and dry weights were strongly positively correlated (r = 0.996). Except for dry weight, fresh weight showed a high positive correlation with leaf area, followed by plant height, number of leaves, number of nodes, leaf length, and leaf width. When the number of days after transplanting, leaf area, and plant height were used as independent variables for growth prediction, leaf area was found to be an appropriate independent variable for growth prediction. However, additional destructive or non-destructive methods for predicting growth should be considered. In this study, we created a growth model formula to predict perilla growth in plant factories.

A Study of the Effective Weed Control by Herbicides in a Nursery and Forests(I) -Weed Control in a Nursery for Silviculture and Landscape Architecture- (묘포장 및 산지에서 제초제를 이용한 효과적인 잡초방제에 대한 연구(I) -조림.조경용 수묘포장의의 제초관리-)

  • Three kinds of soil surface applied herbicides and three kinds of foliage applied herbicides were used to study weed control effect, appropriate concentration, phyto-toxicity against trees and economical efficiency of weed control on trees, Pinus thunbergii, Picea abies, Chamaecyparis obtusa, Quercus accutissima, and Fraxinus rhynchopylla in a nursery. The results were obtained as follows; 1.The control effect of the soil surface applied herbicides showed that the three herbicides; Alachlor, Pendimenthalin and Simazine were alike in their holding effect of weed development, and the sensibility of weed spp. on the herbicides was different. The mean control effect was 58~89%. 2. The weed control effect of the foliage applied herbicides demonstrated that Paraquat dichloride was the best of 84~95% and followed by Glufosnate ammonium and Glyphosate. Especially when the half of its standard amount was treated, the effect of Paraquat dichloride was higher than the other herbicides. 3. Generally, the weed control effect became better as the concentration of the herbcide getting higher. As the concentration of the herbicide got higher, the restraining effect of weed development was continued for a long time in the soil surface applied herbicides, while the weed control effect appeared earlier in the foliage applied herbicides. 4. As the result of the soil surface treatments, 20% of short seedings of Fraxinus rhychopylla treated with twice amount of standard Alachlor died and the rest seeding showed low growth. In the other treatments, there was neither harmful effect of herbicides on the seedings nor growth decrease of them compared to those weeded by men. 5. When treated with foliage applied herbcides, leaves were partially of and discolorated in most treatments. About 0.4~6.2% of Fraxinus thynchopylla, which had short seedings died, and there was no difference in growth with the other treatments. 6. The herbicides showed better economical efficiency over 68% than weed control by men. Especially, Alachlaor of the soil surface applied herbicides showed the highest efficiency(77.6%), while Paraquat dichloride of the foliage applied herbicides was the best(70.3%)..

  • Growth hormone (GH)-releasing peptides (GHRPs) and GH secretagogues (GHSs) are listed in the World Anti-Doping Agency (WADA) Prohibited List. In the present study, we developed and validated a method for the simultaneous analysis of seven GHRPs (alexamorelin, GHRP-1, -2, -4, -5, -6, and hexarelin) and three GHSs (anamorelin, ibutamoren, and ipamorelin) in human urine. Method validation was performed at minimum required performance levels specified by WADA technical documents (2 ng/mL) for all substances, and the method was validated with regard to selectivity (no interference), linearity (R2 > 0.9986), matrix effects (50.0%-141.2%), recovery (10.4%-100.8%), and intra- (2.8%-16.5%) and inter-day (7.0%-22.6%) precisions. The limits of detection for screening and confirmation were 0.05-0.5 ng/mL and 0.05-1 ng/mL, respectively.

The Effects of Defatted Punux ginseng on the Growth and Some Components in Rat. (탈지인삼이 흰쥐의 성장 및 체성분 함량에 미치는 영향)

  • In this paper it was attempted to observe the effect of defatted panax ginseng supplement of the growth rate, feed and protein efficiency ratios, and the contents of cholesterol, total lipid and protein in the serum, liver and aorta in Sprague-Dowley Albino male rat (weighing 83 ${\pm}$ 4 g). Seven kinds of experimental diets were prepared as follows : Stock (control) diet, ginseng control diets supplemented with 0.5, 1.0 and 3.0% of ginseng powder to the stock diet, and defatted ginseng Powder diets supplemented with 0.5, 1.0 and 3.0% of defatted ginseng powder to the stock diet. .All diets contained same level of lipid and protein, respectively. The results obtained are as follows : 1. The growth rate in the feeding group of 0.5% defatted ginseng powder diet for 16week were higher than other diet groups. 2. Feed and Protein efficiency in 0.5% defatted ginseng group showed similar tendency to that in body growth rate. 3. The total cholesterol contents in the serum of 0.5% defatted ginseng Powder diet$.$ group showed the tendency to decrease gradually for 4 to 12 weeks, maintaining higher level than other groups. The free cholesterol contents in the serum of defatted ginseng powder diet group in 8 the and 16th weeks were higher than all ginseng control diet group. The total and free cholesterol contents in the liver of all defatted ginseng diet groups in 16th week were higher than hose of all ginseng control groups. The total cholesterol content at 12h week and the free cholesterol content at 16th week i n the aorta of all defatted ginseng diet groups were lower than those of ginseng control groups, respectively. 4. The total lipid contents in the serum of 1.0 and 3.0% defatted ginseng diet groups at 2nd to I2th weeks were lower than other groups, and those in the liver and aorta of all defatted ginseng diet groups at 12th weeks were lower than those of ginseng control diet groups. 5. The protein contents of the serum and aorta were continuously increased throughout whole experimental period in all experimental groups. The protein content of the liver of all groups were decreased at 2nd week and after then no change was observed.

Effects of Feeding Bovine Colostrum on Growth and Feed Intake of Mouse (젖소초유 급여가 마우스의 성장과 사료섭취에 미치는 효과)

  • This studies were carried out to assess the effects of feeding bovine colostrum on growth and feed intake of mouse. A total of 30 mice were divided into three groups(10 mice for each groups), and treatments(colostrum, city milk, or no milk as the control) were randomly assigned. Average daily intake of solid feed during the experimental period were 4.73, 3.95 and 3.41g for control, normal milk, and colostrum, respectively. Average daily intake of milk were 0, 9.93, and 10.17g for control, normal milk, and colostrum, respectively. Average daily water intake were 6.28, 4.743, and 4.67g for control, normal milk, and colostrum, respectively. The growth rate of the colostrum group was 16.7% higher compared to the control group (P<0.05). There were differences in the concentrations of IL-4 and IL-13 associated with allergy in mouse blood at colostrum and city milk feeding as compared with control.

Effects of Supplemental Glutamine on Growth Performance, Plasma Parameters and LPS-induced Immune Response of Weaned Barrows after Castration

  • Two experiments were conducted to investigate the effects of supplemental glutamine on growth performance, plasma parameters and LPS-induced immune response of weaned barrows after castration. In experiment 1, forty-eight weaned male piglets were used and fed maize and soybean meal diets supplemented with 0 (Control) or 2% L-Gln (Gln+) for 25 days. The results indicated that the Gln+ group tended to increase average daily gain compared to control in stages of days 7 to 14 and 0 to 25. The Gln+ had significantly better feed efficiency than the control group did during days 14 to 25 and 0 to 25. The plasma blood urea nitrogen and alkaline phosphatase contents of Gln+ group were higher than those of the control group on day 14 post-weaning. In experiment 2, sixteen weaned male piglets were injected with E. coli K88+ lipopolysaccharide (LPS) on day 14 post-weaning. The results showed that the Gln+ group had lower concentrations of plasma adrenocorticotrophic hormone and cortisol than the control group on day 14 pre-LPS challenge. In addition, Gln+ group had higher plasma IgG concentration than the control group for pre- or post-LPS challenged on day 14 post-weaning. In summary, dietary supplementation of Gln was able to alleviate the stressful condition and inflammation associated with castration in weaned barrows, and to improve their immunity and growth performance in the early starter stage.

Effect of Crude Oil (WSF) on Early Life Stage of the Pacific Oyster, Crassostrea gigas (원유의 WSF (water soluble fraction)가 참굴의 초기생활사에 미치는 영향)

  • The observed the toxic effect of WSF (Water Soluble Fraction) on development time, development rate, attachment rate, survival rate and growth of the larva during the early stage of the Pacific oyster, Crassostrea gigas. The time required early juvenile from the fertilized eggs exposed to crude oil (WSF) was about 13 hours in control group. But that of the crude oil dose group takes 12 hours, with shorter compared to control group. Development and attachment rate of the oyster significantly higher in 0.4 and 0.8 mg/L WSF compared to control group (P<0.05) but significantly lower more than 1.6 mg/L WSF (P<0.05). Survival rate of the oyster larva significantly higher in 0.4 and 0.8 mg/L WSF compared to control group (P<0.05) but significantly lower more than 2.4 mg/L WSF (P<0.05). Survival rate of spot showed no significantly difference compared to control group (P>0.05). Shell growth of the oyster were significantly lower more than 1.6 mg/L WSF compared control group (P<0.05). The effect concentration of crude oil showed that more 0.4 mg/L in development and attachment rate and more 1.6 mg/L in growth. Also, chronic exposure of crude oil even effect also in low concentration.

Disease Control Efficacy of Chitosan Preparations against Tomato Leaf Mold (토마토 잎곰팡이병에 대한 키토산 제제의 방제 효과)

  • Chitosan has an antifungal activity and is widely used for control of various plant disease and plants growth in the field in Korea. Disease control efficacy of two preparations (SH-1, SH-2) of mixtures of high and low (chitooligosaccharide) molecular weight chitosan compounds against tomato leaf mold caused by Fulvia fulva was investigated under plastic greenhouse conditions. Both SH-1 and SH-2 formulations displayed potent disease control activity in two experiments. The protective activity of both preparations was comparable to synthetic thiophanate-M. The persistence activity of the formulations was sustained until 21 days after application. Effective concentration of the chtosan compounds for disease control was 1,200 mg a.i./L. In pot tests, chitosan preparations, at a concentration of 600 mg a.i./L, promoted plants growth. These results indicate that the chitosan preparations have a potential as an eco-friendly natural fungicide for the control of tomato leaf mold and plant growth regulator.

Changes in Shoot and Root Growth of Tomato Seedlings Stimulated by Brushing (브러싱 자극 토마토 공정묘의 지상부와 지하부 생육 변화)

  • Mechanical stimulation induce the morphological changes in plants. In this study, we investigated the growth changes of tomato seedlings applicated to mechanical stimulation. The brushing treatment was used for mechanical stimulation. The brushing treatment interval was 2 hr using transfer device attached acrylic film from 10 days after sowing. Growth parameter of tomato seedlings were measured 3-day intervals to investigate the growth changes during brushing treatment. The plant height and leaf area were decreased in brushing treatment than the control, and the fresh and dry weights of shoot didn't have significant difference in the control and brushing treatment. The total root length and root surface area were increased in brushing treatment compared than the control, and root volume has no significant difference in the control and brushing treatment. In conclusion, these results suggest that the application of brushing treatment on tomato seedling make shorten plant height and well-development root morphological characteristics.
