Prostatic acid phosphatase (PAP)는 전립선 암의 진단에 널리 사용되는 표지자로서 1935년 처음으로 동정되었고 인체 전립선에 가장 많이 존재하는 탈 인산화효소이다. PAP는 prostate epithelial cells에서 합성되는 전립선 특이적인 효소로서, 산성 환경에서 효소활성을 띠는 acid phosphatase 그룹에 속한다. PAP는 전립선액에 풍부히 존재하여 수정, 정자부족증, 만성통증의 감소에 관여한다. 그러나 가장 눈에 띄는 기능은 ERK1/2와 MAPK 경로에 관계된 HER-2와 PI3P의 탈 인산화를 유도하여 세포 성장 신호를 억제하고 전립선 암의 억제자로 작용하는 것이다. 최근 PAP DNA 백신을 이용하는 임상시험이 현재 진행 중이고, PAP를 이용한 immunotherapy를 통해 전립선 암을 치료하는 방법이 FDA의 승인을 받아 시행되고 있다. 이러한 PAP의 임상적 중요성에도 불구하고 현재까지 PAP의 분자적 조절기작에 대한 이해는 제한적이라 PAP에 대한 많은 연구가 필요한 실정이다. PAP는 NF-${\kappa}B$, TNF-${\alpha}$, IL-1 및 androgen과 androgen receptor에 의하여 promoter region이 조절된다고 알려졌다. 본 총설에서는 현재까지 밝혀진 PAP 유전자 및 단백질의 특징들과 더불어 전립선 암에서의 PAP의 기능, 발현 조절, 역할들을 종합하였다.
암의 성장과 신생혈관 억제인자로 알려진 thrombospondin-1의 생합성은 다양한 외부자극에 대해 전사단계에서 세포 특이적으로 조절된다. 이전의 연구에서 본 연구자들은 PMA가 정상 돼지 대동맥 내피세포(PAE)에서는 TSP-1의 발현을 감소시키는 반면 사람 간암 세포주인 Hep3B에서는 증가시키는 사실을 발견하였다. PMA 처치에 따른 정상 돼지 대동맥 내피세포에서의 TSP-1의 발현 감소현상은 tsp-1 유전자 조절부위의 염기서열 -767과 -723사이에 존재하는 염기서열이 억제 부위임을 밝혀 이러한 결과를 바탕으로 -767에서 -723 염기서열을 서로 부분 중복되도록 세 종류의 올리고 탐식자 (올리고 탐식자 a-1, -767∼-738; 올리고 탐식자 a-2, -759∼-730; 올리고 탐식자 a-3, -752∼723)를 제작하여 -767과 -723 부위의 특정 염기서열과 이에 결합하는 인자를 EMSA을 수행하여 분석하였다. 실험 결과, PMA 처치에 따른 정상 돼지 대동맥 내피세포에서의 TSP-1 감소는 -752에서 -730 사이의 염기서열이 저해 조절인자와 결합함과 더불어 -767에서 -760과 -752에서 -730 사이의 염기서열들에 촉진 조절인자들이 결합하지 못함으로서 기인된다는 실험적 사실을 관찰하였고. 특히, PMA 처치는 정상 돼지 대동맥 내피세포에서 저해 조절인자의 -752에서 -730 부위에 대한 친화력을 향상시켰으며 이러한 친화력은 c-Jun 항체에 의해 영향을 받지 않았다.
Fructan은 식물이 저온에 노출 되었을 때 다양한 조직에 축적됨으로써 여러 스트레스에 저항을 나타내는 것으로 알려져 있다. 본 연구에서는 fructan 생합성 경로에 관여하는 효소인 1-sst와 1-fft 유전자를 돼지감자 구근으로 부터 분리하였다. 분리한 1-sst와 1-fft 유전자는 Ti-plasmid vector인 KJG V-B2 vector에 35S promoter에 의해 발현할 수 있도록 형질전환용 벡터를 구축하였다. Agrobacterium tumefaciens법에 의해 1-sst와 1-fft 유전자의 형질전환 벼를 육성하였고, 1-sst, 1-fft 및 HPT 유전자 특이적인 primer를 사용하여 PCR 분석한 결과 유전자가 벼의 callus 게놈내에 안정적으로 삽입되었음을 확인하였다. 또한 Southern 및 RT-PCR 분석에서도 같은 결과를 얻었다. 형질전환 벼의 후대에서도 안정적으로 유전자가 발현되는 homo 계통을 선발하였고 이를 이용해 1-sst와 1-fft 유전자의 삽입이 확인된 형질전환 벼에서 유전자의 발현양상을 알아보기 위해 RT-PCR 및 Real-Time PCR를 수행한 결과 형질전환 벼에서 1-sst와 1-fft 유전자 모두 안정적으로 발현되고 있음을 확인하였다. 또한 1-sst와 1-fft 유전자가 삽입된 형질전환 벼를 이용한 기능 분석 연구를 통해 식물체가 저온에 노출되었을 때 1-sst와 1-fft의 작용에 의해 fructan 생합성량이 증가됨을 알 수 있었다. 따라서 본 연구를 통해 얻어진 fructan 생합성 관련 유전자가 삽입된 형질전환 벼는 탄수화물대사 및 저온, 건조 등의 환경 stress에 대한 내성에 대해 좋은 육종 소재로 이용 가능할 것으로 사료된다.
$\beta$-Amylase를 생산하여 전분 분해능을 갖는 산업용 효모를 개발하기 위해 산업용 Saccharomyces cerevisiae에서 Achlya bisexualis $\beta$-amylase (BAMY)유전자를 ADC1 promoter에 연결하여 구성적으로 발현시켰다. 효모의 형질전환은 $\delta$-서열을 재조합 부위로 하는integration 시스템을 이용하였다. Integration 시스템의 세균 유전자 부분은 제거되고 BAMY 유전자와 $\delta$-서열을 갖고 있는 짧은integrative cassette를 제조하였다. BAMY 유전자를 발현하는 재조합 S. cerevisiae 형질전환체는 세포외 배지로 45 kDa의 $\beta$-amylase를 분비하였고, $\beta$-amylase 활성은 A. bisexualis에 비해 약 18.5배 높았다. 형질전환체에 다중도입된 BAMY 유전자는 비선택배지에서 100세대 생장 후에도 안정되게 유지되었다. 각종전분을 기질로 했을 매 $\beta$-amylase의 활성은soluble starch를 기질로 했을 경우와 유사하게 높았고, 가수분해산물 분석 결과 maltose가 주 분해산물이었다.
Curcumin has diverse anticancer activities that lead to tumor growth inhibition of cancer cells and induction of apoptosis. Curcumin is involved in the regulation of multiple genes via transcription factors including NF-${\kappa}B$, STATs, AP1, and SP. Notch signaling plays critical roles in maintaining the balance between cell proliferation, differentiation and apoptosis, and thereby may contribute to the development of various cancers involving breast cancer. This study was to investigate the effects of curcumin on Notch1 gene expression and to explore the underlying mechanism. Here, we found that curcumin decreased the levels of Notch1 mRNA and protein in MDA-MB-231 human breast cancer cells, along with the downregulation of Sp family genes (Sp1, Sp2, Sp3, and Sp4). The repressive effect of curcumin on Notch1 gene transcription was confirmed by performing Notch1 promoter-driven reporter assay and three Sp-binding sites were identified on Notch1 promoter that may act as curcumin-respose elements. Moreover, treatment with mitramycin A, a specific Sp inhibitor, decreased the levels of Notch1 mRNA and protein in human breast cancer cells. Taken together, our results indicate that Notch1 gene expression is downregulated by curcumin, at least in part, through the suppression of Sp family, which may lead to apoptosis in human breast cancer cells.
Thrombospondin-1은 암성장과 신혈관생성 억제조절인자로 병리학적 조건과 세포외에서의 자극에 의해 세포 특이적으로 조절되어지고 이 TSP-1 유전자 발현조절은 암치료제 개발을 위한 새로운 접근으로 중요하다. Mistletoe는 기생식물로 면역조절과 항암제로 사용되어지고 있다. Helixor A는 mistletoe 추출물의 수용액부분이다. 여기서 우리는 Helixor A를 투여하면 간암세포주 (Hep3B)와 혈관내피세포주 (BAE)에서 TSP-1의 mRWA와 단백질 발현이 유도되는 것을 관찰하였다. TSP-1 promoter 활성분석으로 TSP-1유전자의 발현이 Helixor A에 의해 전사단계에서 조절되어 진다는 것을 확인하였다. 세포 침윤 분석에서 Helixor A를 처리한 배지를 두 세포주에 처리함으로 침윤된 세포의 수가 현저하게 줄어드는 것을 관찰하였고, Matrigel에서 혈관 내피 세포 (BAE)의 모세관 형성을 억제하는 것을 관찰하였다. 또한 세포 침윤과 모세관 형성에서 Helixor A를 처리했던 배지의 억제 효과가 TSP-1 중화 항체에 의해 그 효과가 상실되었다. 그러므로, 이 결과는 TSP-1이 Helixor A에 의해 유도되어진 혈관신생 억제 효과에 연관이 있음을 제시하였다. 이러한 결과를 종합 하였을 때 Helixor A가 TSP-1의 상승조절을 통해 혈관신생 억제 효과를 가질 것이라고 생각된다.
Octacosanol, which has prominent physiological activities and functions, has been recognized as a potential growth promoter in animals. A total of 392 1-d-old male Arbor Acres broiler chicks with similar body weight were randomly distributed into four dietary groups of seven replicates with 14 birds each supplemented with 0, 12, 24, or 36 mg octacosanol (extracted from rice bran, purity >92%)/kg feed. The feeding trial lasted for six weeks and was divided into the starter (day 1 to 21) and the grower (day 22 to 42) phases. The results showed that the feed conversion ratio (FCR) was significantly improved in broilers fed a diet containing 24 mg/kg octacosanol compared with those fed the control diet in the overall phase (day 1 to 42, p = 0.042). The average daily gain and FCR both showed linear effects in response to dietary supplementation of octacosanol during the overall phase (p = 0.031 and 0.018, respectively). Broilers fed with 24 or 36 mg/kg octacosanol diet showed a higher eviscerated yield, which increased by 5.88% and 4.26% respectively, than those fed the control diet (p = 0.030). The breast muscle yield of broilers fed with 24 mg/kg octacosanol diet increased significantly by 12.15% compared with those fed the control diet (p = 0.047). Eviscerated and breast muscle yield increased linearly with the increase in dietary octacosanol supplementation (p = 0.013 and 0.021, respectively). Broilers fed with 24 or 36 mg/kg octacosanol diet had a greater (p = 0.021) $pH_{45min}$ value in the breast muscle, which was maintained linearly in response to dietary octacosanol supplementation (p = 0.003). There was a significant decrease (p = 0.007) in drip loss value between the octacosanol-added and the control groups. The drip loss showed linear (p = 0.004) and quadratic (p = 0.041) responses with dietary supplementation of octacosanol. These studies indicate that octacosanol is a potentially effective and safe feed additive which may improve feed efficiency and meat quality, and increase eviscerated and breast muscle yield, in broiler chicks. Dietary supplementation of octacosanol at 24 mg/kg diet is regarded as the recommended dosage in the broilers' diet.
The present study was conducted to evaluate the effect of the dietary supplementation of Bacillus amyloliquefaciens-based direct-fed microbial (DFM) on growth performance, nutrient utilization, intestinal morphology and cecal microflora in broiler chickens. A total of two hundred and eighty eight 1-d-old Arbor Acres male broilers were randomly allocated to one of four experimental treatments in a completely randomized design. Each treatment was fed to eight replicate cages, with nine birds per cage. Dietary treatments were composed of an antibiotic-free basal diet (control), and the basal diet supplemented with either 15 mg/kg of virginiamycin as antibiotic growth promoter (AGP), 30 mg/kg of Bacillus amyloliquefaciens-based DFM (DFM 30) or 60 mg/kg of Bacillus amyloliquefaciens-based DFM (DFM 60). Experimental diets were fed in two phases: starter (d 1 to 21) and finisher (d 22 to 42). Growth performance, nutrient utilization, morphological parameters of the small intestine and cecal microbial populations were measured at the end of the starter (d 21) and finisher (d 42) phases. During the starter phase, DFM and virginiamycin supplementation improved the feed conversion ratio (FCR; p<0.01) compared with the control group. For the finisher phase and the overall experiment (d 1 to 42) broilers fed diets with the DFM had better body weight gain (BWG) and FCR than that of control (p<0.05). Supplementation of virginiamycin and DFM significantly increased the total tract apparent digestibility of crude protein (CP), dry matter (DM) and gross energy during both starter and finisher phases (p<0.05) compared with the control group. On d 21, villus height, crypt depth and villus height to crypt depth ratio of duodenum, jejunum, and ileum were significantly increased for the birds fed with the DFM diets as compared with the control group (p<0.05). The DFM 30, DFM 60, and AGP groups decreased the Escherichia coli population in cecum at d 21 and d 42 compared with control group (p<0.01). In addition, the population of Lactobacillus was increased in DFM 30 and DFM 60 groups as compared with control and AGP groups (p<0.01). It can be concluded that Bacillus amyloliquefaciens-based DFM could be an alternative to the use of AGPs in broilers diets based on plant protein.
An investigation was conducted to evaluate the effects of supplementing Alisma canaliculatum, Viscum album and Cornus officinalis probiotics on the growth performance and immune response in growing pigs. This experiment was conducted using 120 pigs (crossing of Landrace${\times}$Yorkshire and castrated) which were assigned to 5 treatments in 3 replications with 8 pigs per replications. The dietary treatments were NC group (without antibiotics), PC group (basal+Oxytetracycline 50ppm), AC group (basal+A. canaliculatum 0.5%), VA group (basal+V. album 0.5%) and COP group (basal+C. officinalis probiotics 0.5%). The initial body weights of pigs were 35kg on average and the experiment lasted for 9 weeks. The experimental animals were kept in the pens following a completely randomized design. They were provided the diets adequate for grower stage as recommended by NRC (ME:3,265 kcal/kg and CP:16%). COP fed pigs showed lower weight gain up to 6 weeks of age compared to NC group and other groups without significant differences (P>0.05). The carcass weights of pigs fed VA and COP were significantly higher compared to NC group (P<0.05), Back fat thicknesses groups fed three different additives were higher than NC group and lower then PC group (P<0.05). Crude fat contents in loin meat were significantly lower in groups fed three different additives while moisture contents of those three groups were higher than other groups (P<0.05). The thiobarbituric acid reaction substance (TBARS) value measured at fresh and $2^{nd}$ weeks was lower in additives fed groups but no statistical differences were observed among the treatments (P>0.05). Significantly highest PUFA (16.42g/100g) and ${\omega}$-3 fatty acids (ALA, EPA and DHA) content of meat were observed in COP fed pigs compared to NC group (P>0.05), which might mean that three additives function to enhance serum IgG in pigs. In consequence, it can be suggested that AC, VA and COP may have a potential to replace antibiotics as growth promoter and immune enhancer in the diets for growing pigs.
Objective: Hemicastration is a unilateral orchiectomy to remove an injured testis, which can induce hormonal changes and compensatory hypertrophy of the remaining testis, and may influence spermatogenesis. However, the underlying molecular mechanisms are poorly understood. Here, we investigated the impact of hemicastration on remaining testicular function. Methods: Prepubertal mice (age 24 days) were hemicastrated, and their growth was monitored until they reached physical maturity (age 72 days). Subsequently, we determined testis DNA methylation patterns using reduced representation bisulfite sequencing of normal and hemicastrated mice. Moreover, we profiled the testicular gene expression patterns by RNA sequencing (RNA-seq) to examine whether methylation changes affected gene expression in hemicastrated mice. Results: Hemicastration did not significantly affect growth or testosterone (p>0.05) compared with control. The genome-wide DNA methylation pattern of remaining testis suggested that substantial genes harbored differentially methylated regions (1,139) in gene bodies, which were enriched in process of protein binding and cell adhesion. Moreover, RNA-seq results indicated that 46 differentially expressed genes (DEGs) involved in meiotic cell cycle, synaptonemal complex assembly and spermatogenesis were upregulated in the hemicastration group, while 197 DEGs were downregulated, which were related to arachidonic acid metabolism. Integrative analysis revealed that proteasome 26S subunit ATPase 3 interacting protein gene, which encodes a protein crucial for homologous recombination in spermatocytes, exhibited promoter hypomethylation and higher expression level in hemicastrated mice. Conclusion: Global profiling of DNA methylation and gene expression demonstrated that hemicastration-induced compensatory response maintained normal growth and testicular morphological structure in mice.

회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
