Melatonin inhibits glycolysis in hepatocellular carcinoma cells by downregulating mitochondrial respiration and mTORC1 activity

  • Various mechanisms have been suggested to explain the chemopreventive and tumor-inhibitory effects of melatonin. Despite the growing evidence supporting melatonin-induced mitochondrial dysfunction, it remains largely unknown how this phenomenon modulates metabolic reprogramming in cancer cells. The aim of our study was to identify the mechanism underlying the anti-proliferative and apoptotic effects of melatonin, which is known to inhibit glycolysis. We analyzed the time-dependent effects of melatonin on mitochondrial respiration and glycolysis in liver cancer cells. The results showed that from a cell bioenergetic point of view, melatonin caused an acute reduction in mitochondrial respiration, however, increased reactive oxygen species production, thereby inhibiting mTORC1 activity from an early stage post-treatment without affecting glycolysis. Nevertheless, administration of melatonin for a longer time reduced expression of c-Myc protein, thereby suppressing glycolysis via downregulation of HK2 and LDHA. The data presented herein suggest that melatonin suppresses mitochondrial respiration and glycolysis simultaneously in HCC cells, leading to anti-cancer effects. Thus, melatonin can be used as an adjuvant agent for therapy of liver cancer.

Comparison of Storability and Quality of Sweet Pepper (Capsicum annum L.) Grown in Two Different Hydroponics Media

  • This study compared the effects of cocopeat and perlite growth media on the storability and quality of sweet pepper fruit stored using modified atmosphere packages (MAP) and carton boxes. The fruits were stored at 8℃ for 35 and 30 days, respectively. Perlite-grown fruits had a significantly lower size at harvest due to the medium's inability to hold plenty of water during the growing stage. Contrary to what is expected for small fruits, the result shows box-stored perlite-grown fruits to have lower weight loss and a longer shelf life than cocopeat-grown fruits, while MAP fruits have indifference. Perlite fruits also had a higher quality in terms of dry matter, soluble solids, and vitamin C, while box-stored fruits had a better visual quality. As expected, respiration and ethylene production rates were high, and fruits had similar after-storage firmness values. Based on the findings, perlite-grown sweet pepper fruits may have a better quality and give preference in a box storage condition.

Effect of Integrated Use of Organic and Fertilizer N on Soil Microbial Biomass Dynamics, Turnover and Activity of Enzymes under Legume-cereal System in a Swell-shrink (Typic Haplustert) Soil.

  • Quantifying the changes of soil microbial biomass and activity of enzymes are important to understand the dynamics of active soil C and N pools. The dynamics of soil microbial biomass C and N and the activity of enzymes over entire growth period of soybean-(Glycine max (L) Merr.)-wheat (Triticum aestivum L.) sequence on a Typic Haplustert as influenced by organic manure and inorganic fertilizer N were investigated in a field experiment. The application of farmyard manure at 4 to 16 $Mg{\cdot}ha^{-1}\;y^{-1}r^{-1}$ along with fertilizer nitrogen at 50 or 180 $kg{\cdot}ha^{-1}$ increased the mean soil microbial biomass from 1.12 to 2.05 fold over unmanured soils under soybean-wheat system. Irrespective of organic and chemical fertilizer N application, the soil microbial biomass was maximum during the first two months at active growing stage of the crops and subsequently declined with crop maturity. The mean annual microbial activity was significantly increased when manure and chemical fertilizer at 8 $Mg{\cdot}ha^{-1}$ and 50/180 N $kg{\cdot}ha^{-1}$, respectively were applied. The C turnover rate decreased by 47 to 72 % when the level of farmyard manure was increased from 4 to 8 and 16 $Mg{\cdot}ha^{-1}$. There were significant correlations between biomass C, available N, dehydrogenase, phosphatase and yield of the crops.

Integrating a Machine Learning-based Space Classification Model with an Automated Interior Finishing System in BIM Models

  • The need for adopting automation technologies to improve inefficiencies in interior finishing modeling work is increasing during the Building Information Modeling (BIM) design stage. As a result, the use of visual programming languages (VPL) for practical applications is growing. However, undefined or incorrect space designations in BIM models can hinder the development of automated finishing modeling processes, resulting in erroneous corrections and rework. To address this challenge, this study first developed a rule-based automated interior finishing detailing module for floors, walls, and ceilings. In addition, an automated space integrity checking module with 86.69% ACC using the Multi-Layer Perceptron (MLP) model was developed. These modules were integrated into a design automation module for interior finishing, which was then verified for practical utility. The results showed that the automation module reduced the time required for modeling and integrity checking by 97.6% compared to manual work, confirming its utility in assisting BIM model development for interior finishing works.

  • As amount of information in construction industry is growing, the role of information system in project management is becoming increasingly important. With the emerging IT application to the advancing construction industry, construction project management system with advanced technology has been progressed vigorously to improve construction productivity and management efficiency. Recently, a web-based Project Management Information System (PMIS) is developed to support decision-making process by efficiently managing project related information generated from various discipline. Many firms are in the process of developing the PMIS system or already have been applied the system to various projects. However, PMIS is still in its early stage of development to be applied at industrial plant construction projects that process management is significantly emphasized for the successful execution of the project. With the complexity of the industrial plant projects, the industry practitioners need to be able to visualize the construction schedule information to manage the project efficiently. This study suggests methodologies for improving PMIS specialized for industrial plant piping construction projects to estimate the baseline schedule and performance measurement more accurately by developing a framework for the piping construction projects. By using this developed system, the researchers expect that piping construction projects will be more efficiently managed on a real-time basis through measuring progress of piping at each and every state of progress milestone and provide management with opportunities to forecast the level of efforts required to execute the remaining work scope in a timely manner

Evaluation of Forage Production of Maize with Different Sowing Dates and Ridge Shape for Silage at Paddy Field in the Central Region of Korea

  • This study was conducted in 2021 using Kwangpyeongok and Gangdaok, that showed good yield performance both under upland and lowland conditions in the previous year. The experiments were carried out by sowing on April 22, May 14, May 30, June 19, and July 10, with aim to determine optimum sowing date in central region of Korea. The growing degree days (GDD) required to read the flowering stage were 1375.5-1725.3℃ for upland and 1582.7-1982.4℃ for lowland condition. The lowest GDD was observed in July 10 sowing regardless of ridge formation both under lowland and upland conditions for Kwangpyeongok. However, Gangdaok showed the lowest GDD under no-ridge in lowland and high-ridge in upland, both of which were sown on June 19. The difference in GDD between no-ridge and high-ridge treatment was little depending on the sowing date. In both lowland and upland, there was no significant difference between no-ridge and high-ridge treatments in stover dry matter, ear dry matter, and TDN between no-ridge and high-ridge treatments. Under upland condition, no significant difference in biomass and TDN was observed among sowing date treatments and between varieties. Under lowland condition, biomass production was severely reduced in May 30 sowing treatment, whereas no varietal difference was observed. Reduced biomass in May 30 sowing treatment may be due to excess waterlogging and lodging by rainfall.

머신러닝 스태킹 앙상블을 이용한 자율주행 자동차 RADAR 성능 향상 (Enhancing Autonomous Vehicle RADAR Performance Prediction Model Using Stacking Ensemble)

  • 레이다는 자율주행 차에 있어 필수적인 센서 부품으로, 레이다가 활용되는 시장은 점차 커지고 있으며 제품 종류도 다양해지고 있다. 본 연구에서는 평가 공정에서부터 레이다의 불량 여부를 예측해 자율주행의 안정성과 효율성을 높일 수 있도록 성능 예측 모델을 구축하고 평가하였다. 레이더 공정 과정의 39607개 입력 데이터로 모델을 학습하였으며, 결과적으로 17개 모델을 스태킹 앙상블했을 때 Meta Ridge 모델이 가장 높은 학습률을 나타내는 것을 확인하였다. 이러한 연구 결과가 제품의 불량을 공정 단계에서 우선 예측해 수율을 극대화하고 불량으로 인한 제품 폐기 비용을 감축하는 데 도움이 될 것으로 기대 한다.

은수원사시나무 벌목 후 맹아의 생장과 맹아가 주변 식생에 미치는 효과 (Sprout Growth and Its Effects on Vegetation after Lumbering of Populus albaglandulosa)

  • 도시의 식재림을 자연림으로 복원하기 위한 기초로서 서울시 강남구 청담동에 위치한 청담근린공원내에서 교목층을 구성하고 있는 은수원사시나무를 벌목한 후 초기 2년간 맹아의 생장 및 맹아가 식생에 미치는 효과를 조사하였다. 벌목된 후 맹아의 발생량에서 모체의 기저면적이 넓을수록 맹아의 수와 생체량이 증가하는 것으로 나타났으며 후자가 전자보다 더욱 관계가 깊었다. 그리고 맹아의 생산량은 벌목 첫 해에 552.7g?DW/㎡, 맹아의 제거 후 익년에 8.7 g?DW/㎡이었다. 따라서 맹아의 제거는 익년의 맹아발생을 급격히 감소하는 결과를 초래하는 것으로 나타났다. 맹아를 제거하지 않은 지역에서 익년의 지상부 건중량은 657.4 g?DW/㎡에 달하여 맹아를 제거하지 않은 지역의 2년간 생산량보다 많았다. 특히 2년생 맹아의 높이는 304 cm에 달하였다. 맹아의 생장은 4월말부터 8월말까지 이었으며 매우 늦은 시기까지 잎이 남아 있었다. 비엽적은 시간이 경과함에 따라, 상층으로 갈수록 다소 감소하였으나 큰 차이가 없었다. 따라서 은수원사시나무 맹아의 생장이 빠른 것은 생육기간이 길고 하층의 잎이 생육후기까지 남아 있기 때문으로 나타났다. 은수원사시나무 맹아를 제거한 경우, 관목층의 구성종은 변화가 없었으나 초본층의 경우 종 수가 현저히 증가하였으며 관목층의 피도는 증가하였으나 초본층에서는 큰 변화가 없었다. 맹아를 제거하지 않은 경우, 관목층과 초본층의 구성종과 피도 모두 변화가 적었다. 그리고 맹아를 제거하지 않은 지역의 초본의 주요 구성종은 초장이 큰 미국자리공, 서양등골나물 및 큰기름새이었다.

철원지역에서 재배한 양마 (Kenaf, Hibiscus cannabinus L.)의 재식거리와 품종, 수확시기에 따른 건물수량 및 사료성분의 변화 (Effects of Planting Density, Cultivar and Growing Days on the Dry Matter Yield and Forage Quality of Kenaf (Hibiscus cannabinus L.) in Cheorwon, Korea)

  • 본 연구는 양마 3품종을 재식거리, 품종 및 수확시기에 따른 건물수량과 사료성분의 변화를 검토하고자 강원도 철원지역에서 2005년 5월 20일부터 2005년 9월 12일까지 수행되었다. 시험배치는 3반복 분할구 배치로 주구는 재식거리와 수확시기를, 세구는 양마 3품종(Dowling, Everglade 및 Tainung)을 배치했으며, 결과를 요약하면 다음과 같다. 생육일수에 따른 건물 생산량은 93일까지는 재식거리가 $10{\times}10cm^2$ 구에서 높게 나타났다. 품종별 건물생산량에서는 모든 품종이 생육시기가 진행됨에 따라 건물수량이 높아지는 경향을 보였고, 특히 Bowling이 모든 수확시기에서 가장 높은 건물생산량을 보였다. 조단백질 함량은 생육일수가 길어짐에 따라 감소하는 경향을 보였다. 특히, 생육 중기(생육일수 84일)을 기준으로 생육초기에는 조단백질 함량은 큰 차이를 나타내지 않았지만, $10{\times}20cm^2$ 구에서 다소 높게 나타났다. 품종별로는 Bowling의 조단백질 함량이 다소 높은 것으로 조사되었으나, 큰 차이는 나타내지 않았다. 재식거리 및 품종에 따른 NDF와 ADF 함량의 차이는 조사된 3 품종 공히 생육시기가 지나감에 따라 다소 증가하는 경향을 보였지만, 큰 유의적 차이는 나타나지 않았다. 이상의 결과를 종합해 보면, 양마는 우리나라 중북부 지방에서 훌흉한 사료작물로서의 가능성이 검증되었고, 특히 Bowling 품종이 재식거리 $10{\times}10cm^2$로 파종하여 104일째 수확했을 때 가장 좋은 건물생산량과 품질을 보여주었다.

연초피복재배(煙草被覆栽培)에서 작토시(作土尸)의 이화학성(理化學性) 변화(變化)와 연초생장반응(煙草生長反應)에 관(關)한 연구(硏究) -3. 피복조건(被覆條件)이 연초생장(煙草生長)과 양분흡수(養分吸收)에 미치는 영향(影響) (A Study on Changes of Physico-Chemical Properties of Plow Layer Soil and its Response of Tobacco Growth under Polyethylene Film Mulching Condition -3.   • 재배방법(栽培方法)을 P.E film피복(被覆)과 무피복재배(無被覆栽培)로 하여 재배기간(栽培期間)동안 연초(煙草)의 생장반응(生長反應)과 양분흡수량(養分吸收量) 등에 대(對)하여 조사(調査)한 결과(結果)는 다음과 같다. 1. 지상부건물중(地上部乾物重) 150g/주(株)에 도달하는 기간(期間)은 피복조건(被覆條件)이 무피복조건(無被覆條件)보다 약 10일(日) 빨라서 재배기간(栽培其間)이 무피복재배(無被覆栽培)보다 약 25일(日)정도 앞당겨졌으며, 생육단계별(生育段階別) 상대생장율(相對生長率)도 피복조건(被覆條件)인 경우 이식후(移植後) 55일(日)째이후(以後)부터 생장(生長)이 둔화되었으나 무피복조건(無被覆條件)에서는 이식후(移植後) 70일(日)째 이후(以後)부터 생장(生長)이 둔화되었다. 2. 피복재배(被覆栽培)에서 엽중(葉中)의 질소흡수량(窒素吸收量)은 생육후기(生育後期)에 이르러 급격히 둔화되었으나 무피복(無被覆) 재배(栽培)에서는 생육후기(生育後期)까지 질소흡수(窒素吸收)가 계속되었고 엽중(葉中)의 질산태질소흡수량(窒酸態窒素吸收量)에서 그 경향이 더욱 뚜렷하였다. 건조후(乾燥後)의 잎담배중에서도 전질소(全窒素)와 질산태질소(窒酸態窒素) 함량(含量)이 무피복재배구(無被覆栽培區)에서 현저하게 높았다. 3. 무피복재배(無被覆栽培)에서 잎담배중의 환원당 함량은 피복재배(被覆栽培)의 경우보다 낮았으며 그 차이(差異)는 질산태질소(窒酸態窒素)가 가장 많이 함유(含有)된 하위엽(下位葉)에서 현저하였다. 4. 무피복재배(無被覆栽培)는 잎담배중의 함질소화합물(含窒素化合物)이 증가(增加)되고 환원당이 감소(減少)되어 잎담배 품질(品質)을 저하(低下)시켰다. 5. 엽위별(葉位別) 질산태질소(窒酸態窒素)와 칼륨함량(含量)의 분포(分布)는 서로 유사하여 두 성분(成分)은 흡수이동과정(吸收移動過程)에서 서로 Counter-ion으로 작용(作用)하는 것을 암시하였다.
