Annual yield characteristics in the log cultivation of Grifola frondosa (잎새버섯(Grifola frondosa) 원목매립재배시 연차별 수량특성)

  • We developed a log-burying cultivation technique for Grifola frondosa using oak logs and surveyed its annual yield characteristics. As a result of G. frondosa log cultivation, the harvesting period of the 'Yipsae1ho' cultivar was delayed by approximately 10-15 days compared to that of the Dabak cultivar, and the fruit color of the 'Yipsae1ho' cultivar was dark brown, while that of the 'Dabak' cultivar was grayish brown. Yield of the 'Yipsae1ho' cultivar was $16.0 kg/m^2$ in the first year, $15.4kg/m^2$ in the second year, $9.5 kg/m^2$ in the third year, $4.6 kg/m^2$ in the fourth year, and $4.6kg/m^2$ in the fifth year, while yield of the 'Dabak' cultivar was $12.3kg/m^2$ in the first year, $11.5kg/m^2$ in the second year, $12.7kg/m^2$ in the third year, $6.2 kg/m^2$ in the fourth year, and $8.2kg/m^2$ in the fifth year. Total yield of the 'Yipsae1ho' cultivar ($50.0 kg/m^2$) was slightly lower than that of the 'Dabak' cultivar ($50.8kg/m^2$). The optimum period for log-burying cultivation of Grifola frondosa is estimated to be 3 years.

Production of Exo-polysaccharide from Submerged Culture of Grifola frondosa and Its Antioxidant Activity

  • Exo-polysaccharide isolated from the culture of Grifola frondosa was modified by sodium periodate ($NaIO_4$) and sodium chlorite ($NaClO_2$) to delete polysaccharide part and phenolic compound, respectively, and was investigated what effect has each part of exo-polysaccharide against 2,2'-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress in porcine kidney epithelial cells (LLC-PK1). Oxidative stress on LLC-PK1 cell was measured by cell viability, lipid peroxidation, superoxide dismutase (SOD), and glutathione peroxidase (GSH-px) activity. Exposure of LLC-PK1 cells to 1 mM AAPH for 24 hr resulted in significant decrease in cell viability, SOD, and GSH-px action, and significant increase in lipid peroxidation. The treatment of exo-polysaccharide and $NaIO_4$ modified sample protected LLC-PK1 cells from AAPH-induced cell damage such as cell viability, lipid peroxidation, SOD, and GSH-px activity in a dose dependant manner (10, 100, and $500{\mu}g/mL$). However, the treatment of $NaClO_2$ modified sample did not affect for cell viability, lipid peroxidation, SOD, and GSH-px activity. The antioxidant activity of exo-polysaccharide was significantly decreased on AAPH-induced LLC-PK1 cell system when phenolic compound was deleted. The antioxidant activity was significantly correlated with the content of phenolic compound of exo-polysaccharide.

  • The personalized meal service is being developed to prevent and alleviate illnesses according to the individual's health condition. However, the current meal does not provide a fully customized service to individuals and a diet that meets the consumer's information needs. The cause is the lack of information on the ingredients and the difficulty of comparative analysis between the materials. Therefore, in this study, we propose basic analysis process for basic information acquisition and database construction for food composition before providing personalized food. In this study, we investigated the content of carbohydrate, reducing sugar and protein as basic components of Grifola frondosa and investigate the content of polyphenol as a biological active ingredients. Respectively. Studies on the hypoglycemic effect of the diabetic rat model have been carried out in relation to the prevention of diseases. Based on the results of this study, it is also possible to obtain information on the basic ingredients of the food and to analyze the information on the content and activity of the biological active ingredients. Using animal models, information on disease prevention and mitigation was also available. The process introduced in this study is applied to various food materials, accumulating data, and utilizing Database, this results will be an excellent tool for providing more efficient service by providing a proper dietary composition for consumers.

Effects of Organic Acid Addition on Fruiting Productivity and Primordium Formation in Grifola frondosa (유기산을 이용한 pH 조절 처리가 잎새버섯 자실체 발이 및 생산성에 미치는 영향)

  • This study was performed to develop a useful method for substrate pH adjustment in bottle cultivation of Grifola frondosa. Four types of organic acid solution-citric acid, malic acid, tartaric acid, and succinic acid-were added at 0.5% to 3.0% (w/v) into the substrate. The pH after the organic acid treatments ranged from 3.8 to 4.8 for the mycelial growth of G. frondosa after sterilization. Supplementation with succinic acid at 0.5% resulted in a shorter cultivation period and higher consistent yields compared to other organic acid treatments. These results indicate that the technique we developed is beneficial for higher production of G. frondosa using bottle cultivation.

Quality Characteristics and Antioxidant Activities of Grifola frondosa Tea with Different Pre-treatments (잎새버섯차의 전처리방법에 따른 품질특성 및 항산화활성)

  • Grifola frondosa is a promising new kind of cultivated mushroom owing to its excellent taste and functionality. However, more research is required to determine its value. In this study, the effects of pretreating Grifola frondosa tea by drying, steaming, and warming, on quality characteristics and antioxidant activities were analyzed. There was no difference in sugar, soluble solid, and nitrogen content between dried and steamed tea. The color of the warm pretreated tea was the darkest, and the amino acid content was 462.9 mg/L, which was 1.8 times higher than that of the other pretreatments. Moreover, the warm pretreated tea had the highest total polyphenol and flavonoid content, 14.6 mg/g and 2.2 mg/g, respectively. DPPH and ABTS radical scavenging activities significantly increased with the increase of phenolic compounds. Warming pre-treatment slightly increased the taste preference to 7.0. In conclusion, warming Grifola frondosa before drying was confirmed to improve the extraction of nutrients, antioxidants, and taste preference.

Determination of Proper Amounts of Media and Effects of Fermented Sawdust Addition on Yields of Grifola frondosa in the Bag Culture (잎새버섯 봉지재배시 적정 배지량 및 발효톱밥 첨가효과)

  • This study was conducted to determine proper amount of media and the effect of fermented sawdust on yield of Grifola frondosa in bag culture. The yield of Grifola frondosa was 227 g for 2 kg of media compared with 103 g for 1kg, 256 g for 2.5 kg, and 312 g for 3 kg. The greatest yield of Grifola frondosa per mass of media was 114 g/kg media for 2 kg of media. Days to harvest for 2 kg of media was 95 days compared with 93 days for 1kg, 99 days for 2.5kg, and 104 days for 3kg. Addition of fermented oak sawdust increased yield from 90 to 145 g/kg bag for Hambak cultivar and from 104 to 121 g/kg bag for Yipsae1ho cultivar. Culture duration was shorten 4-7 days by treating fermented oak sawdust.

  • This study was conducted to develop the log cultivation techniques of the maitake Grifola frondosa. We used two kinds of Grifola frondosa strains for this study. Fully incubated logs were buried into soil directly and were buried with soil inside of container box in 70% shaded vinyl house. In the vinyl house Yipsaeilho cultivar buried by the soil was delayed about 15days in harvest date compared with Dabak cultivar. Color of fruit body of Yipsaeilho cultivar was dark brown and that of Dabak cultivar was grayish brown. Yields of Yipsaeilho cultivar was 15-16 $kg/m^2$, more than that of Dabak cultivar, 11-13 $kg/m^2$. In the container box, Yipsaeilho cultivar buried by the soil was also delayed about 5days in harvest date compared with Dabak cultivar. Color of fruit body was similar to the result of vinyl house soil reclamation. But yields difference was not shown between two cultivars, 1.2-1.4 kg/box.

Anti-diabetic Effect of Crude Polysaccharides from Grifola frondosa in $KK-A^{y}$ Diabetic Mouse and 3T3-L1 Adipocyte (3T3-L1지방세포 및 제2형 당뇨모델($KK-A^{y}$)에서 잎새버섯(Grifola frondosa) 조다당체 추출물의 항당뇨 효과)

  • This study was performed to investigate the anti-diabetic mechanism of crude polysaccharides isolated from the fruiting bodies of Grifola frondosa. We treated 3T3-L1 adipocyte cells to observe whether the crude polysaccharides isolated from Grifola frondosa would stimulate insulin sensitivity. Significant insulin sensitizing activity was observed in the 3T3-L1 adipocytes, and giving the crude polysaccharide of Grifola frondosa with 1 nM of insulin caused glucose uptake to increase to a similar level as giving 50 nM of insulin alone. To confirm the mechanism for the anti-diabetic effect of the crude polysaccharides, we performed further examinations within $KK-A^{y}$ mice, an animal model of type 2 diabetes. The crude polysaccharides reduced blood glucose levels in the $KK-A^{y}$ mice for 2 weeks after feeding, and also significantly lowered plasma insulin levels. These results suggest that the anti-diabetic mechanism of the crude polysaccharide of Grifola frondosa is related to the enhancement of insulin sensitivity.

Hepatoprotective Effect of Grifola frondosa Water Extract on Carbon Tetrachloride-induced Liver Injury in Rats

  • The present study aimed at assessing the protective effect of water extract from fruit body of the Grifola frondosa (GFW) on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity. Rats orally administered with GFW 0.5, 1.0, 2.0 g/kg for 14 days were treated with $CCl_4$ to induce hepatotoxicity. Pretreatment with GFW remarkably prevented the elevation of serum AST, ALT, ALP, LDH, $\gamma$-GTP, and liver lipid peroxides in $CCl_4$-treated rat and GFW administration in liver injured rats by $CCl_4$ showed significant (p<0.05) protection of liver as evidenced from normal serum enzymes and malondialdehyde (MDA) levels. In the ultrastructural changes, administration of $CCl_4$-induced damage of hepatocytes with vacuolation, a highly damaged endoplasmic reticulum, and degenerating nuclei. However, pre-administration with GFW preserved normal ultrastructure of hepatocytes. These results suggest that GFW had an effect to inhibit $CCl_4$-induced liver injury in rat, and that it could be used as an effective hepatoprotective agent against chemical-induced liver damage.

Effect of brewers dried grain as a nutrient supplement in plastic vinyl bag cultivation of maitake (Grifola frondosa S. F. Gray) (잎새버섯 봉지재배시 영양원으로서 맥주박 효과)

  • This study was performed to determine most appropriate mixed ratio of brewers dried grain to wheat bran as a nutrient supplement in the plastic vinyl bag cultivation of Grifola frondosa. In the examination of an appropriate nutrient source, the IV substrate formulation (80 : 10 : 10, oak sawdust : wheat bran : brewers dried grain) resulted in 71.6 days to harvest and the highest yield of 142.6 g/bag. On the other hand, the investigation of the optimal mixing ratio of beer waste to wheat bran showed that the III substrate formulation (80 : 5 : 15, oak sawdust : wheat bran : brewers dried grain) resulted in the least days to harvest (61.8 days), with the highest yield of 140.8 g/bag.
