Isolation and Structural Determination of Aldose Reductase Inhibitor from Korean Fermented Soybean Paste

  • Aldose reductase catalyzes the conversion of glucose into sorbitol. Inhibiting this enzyme in diabetes mellitus can delay or prevent pathogenic process. Aldose reductase inhibitor was screened from Korean fermented soybean pastes (Doen-jang) and purified via sequential processes of ethanol extraction, HP-20 column chromatography, ethyl acetate extraction, silica gel column chromatography, and crystallization. Aldose reductase inhibitor was identified as genistein with molecular weight of 270 Da and molecular formula of $C_{15}H_{10}O_5$ based on UV spectrometry, $^1H$ and $^{13}C\;NMRs$, and mass spectrometry. Genistein inhibited aldose reductase of pig lens with $IC_{50}$ level of $20\;{\mu}M$. Because genistein was effective against aldose reductase of animal source, it may be a potential therapeutic agent for diabetic complications.

Introduction of Stay Green Mutant for the Development of Black Seed Coat and Green Cotyledon Soybean Variety (녹색자엽 검정콩 품종 육성을 위한 Stay green 변이체 활용)

  • The soybean stay green mutant genotype (SSG) derived from the nuclear gene, d1d2, and cytoplasmic gene, cytG, inhibit the breakdown of chloroplast in the leaves, pod walls, seed coats, and embryos during maturity. Soybean seed with black seed coat and green cotyledon (SBG) are preferred than black seed coat with yellow cotyledon (SBY) especially for cooking with rice and as source of traditional food in Korea. The researchers evaluated the seed's chlorophyll content of SSG and introduced SSG to the SBG variety breeding program. The seed chlorophyll content of SSG with d1d2 was $39.93{\sim}60.80\;{\mu}g/g$ and SSG with cytG $38.08{\sim}39.89\;{\mu}g/g$. The Korean SBG variety which was derived from SSG with cytG, contains $16.35{\sim}37.73\;{\mu}g/g$. The composition of seed chlorophyll differs according to the genetic background of SSG genotype. Inheritance study showed that cotyledon color was segregated 15:1 (yellow:green) at $F_2$ seed indicating two recessive genes control green cotyledon as revealed by previous study. Only less than 3% soybean lines showed black seed coat with green cotyledon among crosses SBY and SSG (d1d2). Results showed that SSG with d1d2 can be used as a good source for SBG with high chlorophyll content in the seed cotyledon, but due to the complex genetic behavior, breeding resource of SBG with d1d2 should be prepared to improve the breeding efficiency for development SBG variety.

Use of Unmanned Aerial Vehicle for Multi-temporal Monitoring of Soybean Vegetation Fraction

  • Purpose: The overall objective of this study was to evaluate the vegetation fraction of soybeans, grown under different cropping conditions using an unmanned aerial vehicle (UAV) equipped with a red, green, and blue (RGB) camera. Methods: Test plots were prepared based on different cropping treatments, i.e., soybean single-cropping, with and without herbicide application and soybean and barley-cover cropping, with and without herbicide application. The UAV flights were manually controlled using a remote flight controller on the ground, with 2.4 GHz radio frequency communication. For image pre-processing, the acquired images were pre-treated and georeferenced using a fisheye distortion removal function, and ground control points were collected using Google Maps. Tarpaulin panels of different colors were used to calibrate the multi-temporal images by converting the RGB digital number values into the RGB reflectance spectrum, utilizing a linear regression method. Excess Green (ExG) vegetation indices for each of the test plots were compared with the M-statistic method in order to quantitatively evaluate the greenness of soybean fields under different cropping systems. Results: The reflectance calibration methods used in the study showed high coefficients of determination, ranging from 0.8 to 0.9, indicating the feasibility of a linear regression fitting method for monitoring multi-temporal RGB images of soybean fields. As expected, the ExG vegetation indices changed according to different soybean growth stages, showing clear differences among the test plots with different cropping treatments in the early season of < 60 days after sowing (DAS). With the M-statistic method, the test plots under different treatments could be discriminated in the early seasons of <41 DAS, showing a value of M > 1. Conclusion: Therefore, multi-temporal images obtained with an UAV and a RGB camera could be applied for quantifying overall vegetation fractions and crop growth status, and this information could contribute to determine proper treatments for the vegetation fraction.

Protease Activities in Tenderizing Effect of Vegetables used as Cooking Material (조리용 채소의 단백분해효소 활성 및 연육효과)

  • Protease activities were measured in vegetables used as cooking material or a side dish. Proteases of green pepper (Kwari and Chungyang), perilla leaves, soybean sprout and mungbean sprout were showed high activities. Especially, protease in soybean sprout was the highest activity among them. After dialysis, remaining activities in mungbean sprout, green pepper (Kwari and Chungyang) and perilla leaves were 12, 23, 45% and 37%. In the results of thawing after freezing the proteases, remaining activities in sprout of mungbean and soybean were 100% and 65%. Protease in soybean sprout was showed higher activity and stability than others. Proteolytic effects of soybean sprout on myofibrillar and sarcoplasmic protein were showed higher than stroma protein. In SDS-PAGE, myosin heavy chain, actin and tropomyosin were hydrolyzed with increasing time. Protease activity was rapidly increased with increasing meat concentration in a early proteolysis reaction, but was slightly increased in later.

Effects of Solarization and Green Manual Crops for Promotion of Fall Patato Grawth (가을감자 生産性向上을 위한 綠肥作物栽培와 太陽熱 土壤消毒의 效果)

  • This study was conducted to effect promotion of fall potato growth in the place which green manual crops had been previously cultivated, crushed and tilled and polyethylene transparent vinyl had been mulched from Aug. 5 to Aug. 29 in 1997. The maximum temperature of below-ground at the depth of 10cm was $42^{\circ}C$ during the mulching period, and the pH and the organic compound content of soil was very increased by the cultivation of green manual crops and the mulching of polyethylene transparent vinyl. The total yields of green manual crops which had been previously cultivated were as the following orders; the hybrid of sudan grass, soybean, alfalfa, otherwise, fresh weight of stems and the tuber yield of fall potato were significantly affected in the plot of soybean + fall potato after the soil solarization with the mulching. As these results, it was possible to improve crops growth and to sterilize soil by the mulching treatment of polyethylene vinyl after cultivation of green manual crops during the high temperature of the summer season.

Synthesis of Poly(glycerol-succinic acid)-dithiocarbamate and Poly(glycerol-succinic acid)-1,3,4-thiadiazole Dendrimers and Their Use as Anti-Wear Oil Additives

  • A series of poly(glycerol-succinic acid) dithiocarbamate and 1,3,4-thiadiazole dendrimers, which have potential as anti-wear oil additives, were synthesized. Their anti-wear properties in three different oils (100N, DB-51, and soybean) were evaluated using a four-ball wear tester. The results indicated that thiocarbamate dendrimers have moderate anti-wear properties in DB-51 oil, and 1,3,4-thiadiazole dendrimers exhibited good anti-wear properties in 100N and DB-51 oils. However, dithiocarbamate and 1,3,4-thiadiazole dendrimers were not effective anti-wear additives in soybean oil.

  • cDNA of the glx1 gene encoding glyoxal oxidase (GLX) from Phanerochaete chrysosporium was isolated and expressed in Pichia pastoris. The recombinant GLX (rGLX) produces $H_2O_2$ over 7.0 nmol/min/mL using methyl glyoxal as a substrate. Use of rGLX as a generator of $H_2O_2$ improved the coupled reaction with recombinant manganese peroxidase resulting in decolorization of malachite green up to $150{\mu}M$ within 90 min.

  • Food materials usually contain much water, and thus are susceptible to decay be enzyme and microorganism. Cereals are usually consumed as staple food, but they have low protein content. Protein is a major component of body, and should be consumed as a nutrient for all ages. Animal protein like chicken and vegetable protein such as bean curd and soybean and good sources of high quality protein. Bean curd is a traditional food which have good nutrients, and so various cooking methods are needed to be developed for the supplement of high quality food. 1) Cereals such as rice, barley, and bean curd, soybean, green vegetables, tangle and brown seaweed are known as food for long life. 2) Soybean is able to suppress the formation of hydroperoxide by saponin and lecithin. 3) Curd in bean curd means soft in Chinese character, and bean curd may be the mother of cheese. 4) Bean curd have high nutrition and digestibility. As soybean is dipped longer in water for bean curd, the production rate is higher. 5) There are many kinds of bean curd products, and can be purchased inexpensively.

Variations in Seed Storage Protein among Different Colored Soybean Varieties

  • This study was carried out to know the variation of soybean seed proteins, 11S and 7S globulins, and their amino acid compositions among different colored soybean varieties, 'Danbaegkong' (yellow), 'Pureunkong' (green) 'Jinyulkong' (brown), and 'Geoumjeongkong l' (black). Soybean seed proteins showed a wide range in molecular size, but the electrophoresis patterns of total seed protein subunits showed a similarity among different colored soybean varieties. Amino acid compositions of total seed proteins were similar for all soybean varieties tested. However, soybean varieties showed low composition rates in sulfur containing amino acids. The composition rates of cysteine and methionine in the 11S globulins were higher than those of total seed proteins and 7S globulins. Glutamic acid and glycine were higher in the 11S and 7S globulins than those of total seed proteins. However, the levels of methionine and phenylalanine are high in the 11S globulins, but those of valine and lysin are slightly lower than the 7S globulins. By using HPLC, we tried to analyse the soybean seed proteins. The 11S globulin was composed of 10 major peaks whereas the 7S globulin was composed of 4 major peaks. The composition rates of 11S related proteins have a tendency to increasing during the maturing whereas those of 7S related proteins have a tendency to decreasing. Composition rates of each peaks among different colored soybean varieties suggested that soybean seed proteins are varied, although they showed similarity in the electrophoresis patterns, and understanding of this characteristics is important for the utilization of soybeans.

Green Tea Extract is an Effective Antioxidant for Retarding Rancidity of Yukwa (Rice Snacks) Fried in Soybean and Rice Bran Oils

  • Yukwa is a popular Korean traditional fried rice snack. The high fat content and porous structure of Yukwa cause it to rapidly become rancid, presenting difficult challenges for commercial distribution. In this study, an-tioxidant activities of green tea extracts (GTE) were evaluated in Yukwa fried in soybean oil (SBO), rice bran oil (RBO) and winterized rice bran oil (WRBO) during storage at 4$0^{\circ}C$ for 12 weeks. Lipid oxidation of Yukwa was determined by acid value (AV), peroxide value (POV), p-anisidine value(AnV), totox value and sensory evaluation. The addition of GTE to the oils reduced the increases in AV, POV, AnV, and totox. Totox increased most vapidly in Yukwa fried in SBO, fellowed by RBO>WRBO>SBO+200 ppm GTE>RBO+200 ppm GTE > WRBO + 200 ppm GTE (p<0.05). Sensory evaluation revealed that the addition of 200 ppm GTE delays rancidity in Yukwa by 7~8 weeks; providing compelling evidence that GTE is an effective antioxidant for Yukwa.
