A New Reporter Vector System Based on Flow-Cytometry to Detect Promoter Activity

  • In this study, we report the development of a new dual reporter vector system for the analysis of promoter activity. This system employs green fluorescence emitting protein, EGFP, as a reporter, and uses red fluorescence emitting protein, DsRed, as a transfection control in a single vector. The expression of those two proteins can be readily detected via flow cytometry in a single analysis, with no need for any further manipulation after transfection. As this system allows for the simultaneous detection of both the control and reporter proteins in the same cells, only transfected cells which express the control protein, DsRed, can be subjected to promoter activity analysis, via the gating out of all un-transfected cells. This results in a dramatic increase in the promoter activity detection sensitivity. This novel reporter vector system should prove to be a simple and efficient method for the analysis of promoter activity.

Generation of Protein Lineages with new Sequence Spaces by Functional Salvage Screen

  • A variety of different methods to generate diverse proteins, including random mutagenesis and recombination, are currently available, and most of them accumulate the mutations on the target gene of a protein, whose sequence space remains unchanged. On the other hand, a pool of diverse genes, which is generated by random insertions, deletions, and exchange of the homologous domains with different lengths in the target gene, would present the protein lineages resulting in new fitness landscapes. Here we report a method to generate a pool of protein variants with different sequence spaces by employing green fluorescent protein (GFP) as a model protein. This process, designated functional salvage screen (FSS), comprises the following procedures: a defective GFP template expressing no fluorescence is firstly constructed by genetically disrupting a predetermined region(s) of the protein, and a library of GFP variants is generated from the defective template by incorporating the randomly fragmented genomic DNA from E. coli into the defined region(s) of the target gene, followed by screening of the functionally salvaged, fluorescence-emitting GFPs. Two approaches, sequence-directed and PCR-coupled methods, were attempted to generate the library of GFP variants with new sequences derived from the genomic segments of E. coli. The functionally salvaged GFPs were selected and analyzed in terms of the sequence space and functional property. The results demonstrate that the functional salvage process not only can be a simple and effective method to create protein lineages with new sequence spaces, but also can be useful in elucidating the involvement of a specific region(s) or domain(s) in the structure and function of protein.

Combination of Epstein-Barr Virus-Based Plasmid and Nonviral Polymeric Vectors for Enhanced and Prolonged Gene Expression

  • An Epstein-Barr virus (EBV)-based plasmid contains the EBV nuclear antigen 1 (EBNA1) gene and EBV replication origin (oriP) sequence. Since EBNA1 (the only EBV-encoded protein) is combined with oriP, it is replicated simultaneously with chromosomal DNA in human, primate, and canine cells and is faithfully segregated at a stable copy number upon cell division. Consequently, it can be used to stably express gene inserts over a prolonged time in target cells. We have previously shown that the polyamidoamine (PAMAM) dendrimer can be surface-modified with L-arginine. Arginine is present at a high frequency in the transactivator of transcription (Tat) sequences of human immunodeficiency virus (HIV). It presents high membrane permeability and permits effective transfer of DNA inside the cells. In this study, we constructed two kinds of recombinant DNA by inserting the luciferase gene and enhanced green fluorescence protein (eGFP) gene as reporter genes into the pCEP4 plasmid vector. We measured dynamic light scattering (DLS) and zeta potential after preparing PAMAM-based cationic polymer/EBV-based plasmid complexes. We performed transfection of HEK 293 cell lines with the polyplexes, and monitored luciferase activity and green fluorescence protein (GFP) expression. Our results show that PAMAM-based cationic polymer/EBV plasmid complexes provide enhanced and sustained gene expression.

Expression of the Green Fluorescent Protein (GFP) in Tobacco Containing Low Nicotine for the Development of Edible Vaccine

  • This study was carried out to obtain basic information for gene manipulation in potent edible tobacco (Nicotiana tabacum cv. TI 516). N. tabacum cv. TI 516 is a plant for a possible candidate to use as an edible vaccine, since it contains a low level of nicotine. The effective plant regeneration system through leaf disc culture was achieved using a MS basal medium supplemented with 0.1 mg $1^{-1}$ NAA and 0.5 mg $1^{-1}$ BA. In order to transform the N. tabacum cv. TI 516 with the green fluorescent protein (GFP) gene, Agrobacterium tumefaciens LBA 4404 containing the GFP gene was used. Genomic PCR confirmed the integration of the GFP gene into nuclear genome of transgenic plants. Expression of the GFP gene was identified in callus, apical meristem and root tissue of transgenic N. tabacum cv. TI 516 plants using fluorescence microscopy. Western blot analysis revealed the expression of GFP protein in the transgenic edible tobacco plants. The amount of GFP protein detected in the transgenic tobacco plants was approximately 0.16% of the total soluble plant protein (TSP), which was determined by ELISA.

인간염색체 12q13에 내재한 마우스 Gamm1의 인간유전자 homolog, MYG1의 클로닝과 발현 (Cloning and Expression of a Human Homolog of Mouse Gamml, MVGI, Localized in 12q13)

  • 새로운 유전자를 클로닝하고 그 발현양상을 결정하는 것은 유전자의 기능을 이해하는데 필수적이다. 인간유전자 12q13의 고해상 물리지도를 작성하면서 이 지역의 D12S359와 D12S1618 사이에 내재하는 것으로 mapping된 stSG 3435 EST의 유전자를 클로닝하고 그 발현양상을 조사하였다. NIBI library를 조사하여 stSG 3435를 포함하는 클론 325E4를 분리하여 순차적 결실 방법으로 클로닝하여 자동염기서열분석으로 염기서열을 결정하였다. 1,331 bp의 염기서열을 가진 이 유전자는 Blast search에 의하면 376 개의 아미노산으로 이루어진 단백질로써 인간의 MYGI과 동일하며 마우스의Gamml, melanocyte proliferation gene 1과 86%의 동질성을 보였다. MYGI은 인간염색체의 12에 내재하며 마우스의 Gamml은 syntenic 부위인 마우스 염색체 15에 내재하므로 마우스의 Gamml의 homolog으로 간주된다. Northern blot analysis 결과 MYG1은 인간의 모든 조직에서 발현되며 정소에서 가장 강한 발현을 보였다. 이 유전자의 세포내 발현을 green fluorescence protein과 융합시켜 발현 귀착지를 confocal 현미경으로 동정한 결과 MYG1 단백질은 핵과 리소좀을 제외한 소기관에서 발현되는 것을 관찰하였다.

Direct Evaluation of the Effect of Gene Dosage on Secretion of Protein from Yeast Pichia pastoris by Expressing EGFP

  • Increasing the gene copy number has been commonly used to enhance the protein expression level in the yeast Pichia pastoris. However, this method has been shown to be effective up to a certain gene copy number, and a further increase of gene dosage can result in a decrease of expression level. Evidences indicate the gene dosage effect is product-dependent, which needs to be determined when expressing a new protein. Here, we describe a direct detection of the gene dosage effect on protein secretion through expressing the enhanced green fluorescent protein (EGFP) gene under the direction of the ${\alpha}$-factor preprosequence in a panel of yeast clones carrying increasing copies of the EGFP gene (from one to six copies). Directly examined under fluorescence microscopy, we found relatively lower levels of EGFP were secreted into the culture medium at one copy and two copies, substantial improvement of secretion appeared at three copies, plateau happened at four and five copies, and an apparent decrease of secretion happened at six copies. The secretion of EGFP being limiting at four and five copies was due to abundant intracellular accumulation of proteins, observed from the fluorescence image of yeast and confirmed by western blotting, which significantly activated the unfolded protein response indicated by the up-regulation of the BiP (the KAR2 gene product) and the protein disulfide isomerase. This study implies that tagging a reporter like GFP to a specific protein would facilitate a direct and rapid determination of the optimal gene copy number for high-yield expression.

피브로인 H-chain 재조합 단백질 발현시스템을 이용한 녹색형광실크 생산 (Production of fluorescent green silk using fibroin H-chain expression system)

  • 본 연구의 목적은 누에형질전환 기술을 이용하여 녹색형광실크를 개발하는 것으로서, 본 실험에서는 피브로인 H-chain의 N-말단과 C-말단을 이용하여 피브로인 재조합 단백질 발현 시스템을 제작하였고, 종결코돈이 없는 EGFP 유전자를 위의 발현 시스템에 클로닝하여 녹색형광실크를 제작하였다. 누에형질전환체 선발을 위해서는 3xP3 promoter와 DsRed2를 이용하여 선발하였고, 1200 개의 누에알에 microinjection 하여 F1 세대에서 8 bloods의 형질전환체를 선발하였다. 선발된 누에형질전환체는 초기배 단계의 눈과 신경조직, 유충과 번데기 그리고 성충의 눈에서 DsRed2 형광단백질이 발현되는 것을 확인 할 수 있었다. 또한 실크의 피브로인에서 EGFP 단백질이 발현되는 것을 확인하기 위해, F2세대의 누에형질전환체 중에서 5령 3일 유충을 해부하여 견사선을 형광현미경으로 관찰하였고, 중부와 후부 견사선에서 형광단백질이 발현되는 것을 확인할 수 있었다. 그리고 F2 세대의 고치와 저온에서 정련한 실크에서도 녹색형광단백질의 발현을 확인할 수 있었고, Western blot 분석에서도 EGFP 재조합 단백질이 피브로인 H-chain과 융합된 형태로 존재하는 것이 확인되었다. 이상의 결과에서 녹색형광실크를 생산하는 누에형질전환체가 성공적으로 제작 되었음을 확인할 수 있었고 이러한 결과를 토대로 새로운 산업소재로서 실크를 활용할 수 있을 것으로 기대한다.

  • 핵이식 방법은 형질전환 동물을 생산하기 위한 여러 가지 방법 중 최근에 많이 이용되고 있다. 본 실험은 형광단백질 유전자 (green fluorescence protein, GFP)가 도입된 태아섬유아세포를 이용 핵이식을 하여 형질전환 수정란의 생산효율을 검토하기 위하여 실시하였다. GFP 유전자는 임신 45-55 일령의 태아섬유아세포 (KbFF3)에 electroporation방법으로 transfection을 실시하였다. (중략)
