Continuous Stable production of won Willerand Factor Monoclonal Antibody in Spin Filter Bioreactor with Bleeding Technology

  • The characteristics of two different modes of perfusion culture, intermittent and continuous bleedings, were investigated by culturing the hybridoma cells producing von Willebrand Factor (vWF) monoclonal antibody (McAb) in a 15 L bioreactor without clogging the filter. Both culture methods exhibited similar profiles of cell density and metabolite concentrations during the culture period at the cell concentration of around 1${\times}$107 cells/mL. When the perfusion rate was increased, the intermittrnt bleeding culture showed problems of ammonia accumulation and decrease of cell viability. The continuous bleeding culture in terms of nutrient consumption and metabolite production kinetics. But the analysis of specific oxygen consumption rate showed that the specific oxygen consumption rate of intermittent bleeding culture was similar to that of exponential growth phase. The continuous bleeding culture showed higher specific oxygen consumption rate of intermittent bleeding culture. finally we proved the possibility of long-term operation of continuous bleeding culture and produced approximately 40 g of vWF McAb in a 15L bioreactor after one-month operation.

Sustenance and Enhancement of Soil Fertility for Organic Farming by Legumes and Green Manure (두과.녹비작물 재배를 통한 유기농법 토양비옥도의 유지와 증진)

  • An organic agriculture should be managed by mixed farming in farm unit as a closed recycling system. Due to restricted purchased of fertilizers from outside, organic farmer has to deal with limited amount of nutrient source in farm unit. Especially the supply of the essential nutrient, nitrogen, mostly depends on legumes fixing nitrogen optimizing the site-adapted crop rotation. Dynamics of humus and metabolic plant carbon and active soil carbon compartment in active and passive humuspool by rotation system was explained, and dynamics of potentially mineralizable nitrogen in organic nitrogen and biomass was discussed. It was also discussed comparison of ammonia emission, potential greenhouse effect, primary energy input, acidification potential, CO2 emission between organic and conventional farming, the nitrate-nitrogen dynamic in the soil profile by organic, integrated and conventional farming system. In conclusion, it was suggested for Korean Organic Agriculture that the importance of legumes and green manures in rotation system for increase/maintenance of soil ferfility, and was pointed out the need of investment for environment impact of Korean organic farming implement.

Green Purification System using Natural Hydrogen Generating Mineral Filter (천연 수소 발생 광물 필터를 이용한 녹조 정화 시스템)

  • In many regions of Korea, including the Four Major Rivers, the seriousness of the problem of algal blooms due to eutrophication of water quality is being raised.In this study, in order to solve these social problems, we manufactured a filter using natural mineral fusion (red illite, zeolite, germanium ceramic, selenium ceramic, carbon ceramic) and independently developed a tank system for green algae experiments to observe and determine the stages of change in water quality. In order to study ways to improve water quality through quantitative analysis, 1 ton of severely polluted green algae water from the Nak dong River region was stored in a water tank and exposed to ultraviolet rays in the same environment as the Nak dong River. Then, the same environment as the Nak dong River was created. The results were derived from a 5-week water quality test. The results of this experiment confirmed that green-colored cyano bacteria were significantly reduced just by the turbidity results. The results were obtained through a request to the Korea Testing & Research Institute located in Changwon-si, Gyeong sang nam-do. CI-(chlorine ion) and NH3-N(ammonia nitrogen) had the effect of saving every week. The device used in this study was made of natural minerals free of heavy metals that are harmless to the human body and nature through long-term consideration and exploration to kill and prevent various strains living in water. Green purification system using natural hydrogen generating mineral filter were effective a non-chemical and physical methods. The results of this study are one way to contribute to the serious problems caused by green algae in many countries, and will contribute to the water quality environment by preventing the waste of environmental resources, improving the health of the people, and increasing the ability to purify environmental water quality at home and abroad.

Effects of Different Additives on Fermentation Characteristics and Protein Degradation of Green Tea Grounds Silage

  • This study evaluated the fermentation characteristics and protein degradation dynamics of wet green tea grounds (WGTG) silage. The WGTG was ensiled with distilled water (control), or lactic acid bacteria (LAB), enzyme (E), formic acid (FA) and formaldehyde (FD) prior to ensiling. Three bag silos for each treatment were randomly opened at 0, 3, 7, 14, 28 and 60 days after anaerobic storage. For all the treatments, except for FA, there was a rapid decline in pH during the first 7 days of ensiling. LAB treatment had higher lactic acid content, lower ammonia-N ($NH_3$-N) and free-amino nitrogen (FAA-N) contents than other treatments (p<0.05). E treatment had higher lactic acid, water-soluble carbohydrates (WSC) and non-protein nitrogen (NPN) content than the control (p<0.05). FA treatment had higher $NH_3$-N and FAA-N content than the control (p<0.05). FD treatment had lower NPN and FAA-N content than the control, but it did not significantly inhibit the protein degradation when compared to LAB treatment (p>0.05). Results indicate that LAB treatment had the best effect on the fermentation characteristics and protein degradation of WGTG silage.

The adsorption efficiency of ceramic filter media prepared with the steel-making slag for the removal of VOCs (휘발성 유기화합물 제거를 위한 제강슬래그로 제조된 세라믹 여재의 흡착효율)

  • We studied the adsorption efficiency of steelmaking slag in removing volatile organic compounds (VOCs) for increasing the recycling rate of steel-making slag. Ceramic filter was prepared by mixing the steel-making slag and the diatomite which is used as adsorbents due to the advantage of the high specific surface area and regular mesopores. The adsorption efficiency for VOCs removal was about 80%, 96% and 85% in acetaldehyde, formaldehyde and ammonia, respectively. The adsorption efficiency over 80% for all The gases showed the practical possibility as the adsorption filter.

  • Deactivation of SCR catalysts applied in coal, orimulsion, and LNG power plants in Korea were studied for the regeneration of the deactivated catalyst. The catalysts were characterized by XRD, ICP-AES, BET and SEM, and were examined for ammonia SCR. Deactivation of SCR catalyst applied in coal power plant was mainly caused by the blockade of the pore due to the deposition of sulfate and particulate related to the ingredients of the fuel. The surface area of SCR catalyst applied in orimulsion power plant decreased considerably by the accumulation of the compounds of vanadium, sulfur, and magnesium on the surface of the catalyst. The compounds of vanadium and sulfur were related to the ingredients of the fuel, and the compound of magnesium was related to the additive of the fuel. The activity of the deactivated catalyst for ammonia SCR, however, decreased slightly. Despite the long use for more than two-year, deactivation of SCR catalyst applied in LNG power plants hardly occurred.

Solubilization of Inorganic Phosphates and Plant Growth Promotion by Pantoea Strains

  • Two phosphate solubilizing Pantoea strains (P. agglomerans and P. rodasii) were employed in elucidating their phosphate solubilizing potential under different carbon and nitrogen sources, pH, temperature and salt conditions. Plant growth promoting characteristics such as ACC deaminase activity, indole acetic acid (IAA), HCN, ammonia, and siderophore production of the two strains were assessed in vitro. Potential applicability of the strains as bio-inoculants was also evaluated in pot experiments conducted under green house conditions. Phosphate solubilization measured as the amount of phosphorous released into the medium was recorded as 810 and $788{\mu}g\;ml^{-1}$ respectively by P. agglomerans and P. rodasii. Glucose at the rate of 2% was found be the best carbon source, while $(NH_4)_2SO_4$ was the best nitrogen source for both strains. Despite a slight decrease in phosphate solubilization observed at higher temperature, pH and salt concentrations, both strains could withstand against a range of temperature ($30-35^{\circ}C$), pH (7-9) and the presence of NaCl (up to 5%) without much compromising the phosphate solubilization. Different plant growth promoting traits (ACC deaminase activity, IAA, HCN, ammonia, and siderophore production) of the strains and their ability to promote the growth of green gram seedlings indicate that both strains possess high potential to be used as bio-inoculants.

  • When rearing fish at recirculating filter systems, some pesticides are often used for controling various fish diseases. In this case, the pesticides are usually applied after stopping the filteration, from a view point of affecting the filtering ability of oxidation and decomposition by killing the decomposing bacteria in the filter system. Therefore, a great inconvenience is usually net. The authors carried out a series of experiments, and the influence of 3 kinds of treatments of pesticides, formalin 33 ppm+matachite green 0.15 ppm, dipterex 0.25 ppm, and furanace 0.1 ppm, to the filtering ability expressed as the consumption rate of dissolved oxygen and depletion of ammonia-nitrogen during the passing time through the filter layer was investigated, and it was found out that these pesticides with above mentioned concentrations which is normally used for controling several kinds of diseases and parasites during the time of eel culture, did not affect the filtering ability. Consequently, it can be said that these pesticides with above mentioned concentrations can he used without stopping filteration.

A Study on the Odor Removal Control System of Sewage Sludge

  • Purpose: The purpose of this study is to reduce odor complaints by identifying problems with odor management at the site of the water regeneration center and researching odor management methods. Due to the high population density of Korea, sewage treatment facilities are adjacent to residential and industrial areas. According to previous studies, the main malodor-emitting facilities of sewage treatment facilities were preliminary treatment facilities (2,220 times), sedimentation basins (4,628 times), and sludge treatment facilities (9,616 times). Research design, data and methodology: Compound malodors and designated malodor-producing substances were collected from five site boundaries of the water regeneration center and analyzed according to the official methods to test malodor, and a total of two times (August and September 2020) were conducted. Results: As a result of the measurement, in the green area in front of the center office, compound malodors were detected at a maximum of 8 times and at least 3 times during the dawn time. As for the designated malodor-producing substances, 0.1ppm of ammonia was detected in the green area in front of the center office and the park golf course. This is within 15 times the maximum allowable emission level of compound malodors and within 1ppm of the maximum allowable emission level of ammonia. Conclusions: Even if the dilution rate of the compound malodors did not exceed the maximum allowable emission level, the odor could be recognized, and more research is needed in the future to establish effective reduction measures according to the subjective and individual and seasonal odor characteristics.

Effects of sodium diacetate and microbial inoculants on fermentation of forage rye

  • Rye (Secale cereale L.) is a valuable annual forage crop in Korea but there is limited information about the impact of chemical and biological additives on fermentation characteristics of the crop. This experiment was conducted to investigate fermentation dynamics of wilted forage rye treated with the following six additives; control (no additive), sodium diacetate applied at 3 g/kg wilted forage weight (SDA3), 6 g/kg wilted forage weight (SDA6), inoculations (106 CFU/g wilted forage) of Lactobacillus plantarum (LP), L. buchneri (LB), or LP+LB. The ensiled rye sampled at 1, 2, 3, 5, 10, 20, 30, and 45 days indicated that the acidification occurred fast within five days of storage than the rest of the storage period. The microbial inoculants decline the pH of ensiled forage, more rapidly than the control or SDA treated, which accompanied by the decrease of water-soluble carbohydrates and increase of lactic acid. Compared with the control silage, all treatments suppressed ammonia-nitrogen formation below to 35 g/kg DM throughout the sampling period. Suppression of total microbial counting occurred in SDA6, LP, and LP + LB. The lactic acid production rates were generally higher in microbial inoculation treatments. Acetic acid concentration was lowest in the LP-treated silage and highest in the SDA- and LB-treated silages. The in vitro dry matter (DM) digestibility and total digestible nutrients were the highest in the silage treated with SDA (6 g/kg) at day 45 of ensiling. Based on lower ammonia-nitrogen concentrations and higher feed value, ensiling forage rye treated with SDA at 6 g/kg is promising through enhanced silage quality.
